아토피피부염 환아에서 CTLA-4 exon 1과 promoter 유전자 다형성 (Polymorphisms of the CTLA-4 promoter(-318) and exon 1(+49) genes with atopic dermatitis in Korean children)

  • 목 적 : Cytotoxic T lymphocyte-associated antigen-4(CTLA-4)를 표현하는 유전자는 IgE 조절과 T 세포의 활성에 중요한 역할을 하는 후보유전자로 알려져 있다. 본 연구에서는 CTLA-4 exon 1(+49 position)과 promoter 유전자(-318 position)의 다형성과 한국 소아의 아토피피부염의 유전적 감수성 및 아토피피부염의 임상양상의 관계에 대해 알아보고자 하였다. 방 법 : 아토피성 습진 145명, 비아토피성 습진 69명과 대조군 96명을 대상으로 restriction fragment length polymorphism방법으로 CTLA-4 promoter(-318 T/C)와 exon 1(+49 A/G)의 다형성을 조사하였다. 결 과 : CTLA-4 exon 1과 promoter의 유전자 다형성이 아토피성 습진 환아군이나 비아토피성 습진 환아군에서 대조군에 비해 의의있는 분포의 차이를 보이지 않았으며, 아토피피부염의 중증도, IgE 농도, 호산구의 수와도 유의한 관계가 없없다. 결 론 : 본 연구에서는 CTLA-4 유전자 다형성이 한국 소아의 아토피피부염의 유전적 감수성 및 아토피피부염의 임상양상에 관여하지 않는 것으로 생각된다.

화피 추출물이 염증 및 알레르기 반응에 미치는 영향 (Effect of Phellinus igniarius Quel Extract on the Anti-inflammatory, Anti allergy, Anti-oxidant, Anti-wrinkle reaction)

  • Objectives : Betula platyphylla var. japonica extract (BPE) was used to determine the modulation of cytokine secretion, the activation of inflammatory and allergic factor and the inhibition of gene expression. Inflammatory and allergic cytokines as IL-$1{\beta}$, IL-2, IL-4, IL-5, IL-6, IL-8, TNF-${\alpha}$, NO and COX-2 were measured to use effectively on improvement or treatment of atopic dermatitis. Methods : We used NC/Nga mouse induced by atopic dermatitis to observe the effects of BPE on the weight, water and feed, blood test, weight of organs, histological change, total IgE and histological change of main organs. Results : BPE is effective on anti-inflammatory and allergic reaction. However, further study is needed to prove which component of BPE indicates effective pharmacological action. Conclusions : The above results suggest that Phellinus igniarius Quel extract could be applicable for improvement of several skin functions.

진도견(珍島犬)의 혈액상(血液像) (Hematologic values of normal Jindo dogs)

  • 임상적으로 건강한 진도개 203두의 혈액치(血液値)를 조사(調査)하였다. 203두에는 암수가 다 포함되어 있었으며 개의 연령은 3개월에서 10세까지 이었다. 적혈구수(赤血球數), 혈색소량(血色素量), 적혈구용적(赤血球容積) 그리고 호산구수(好酸球數) 등은 개의 연령과 함께 증가하였다. 진도개에서 나타난 높은 호산구수(好酸球數)는 심장사상충증(心臟絲狀蟲症)과 벼룩의 감염에 의한 피부염(皮膚炎)때문인 것으로 생각된다.

Probiotics as a Potential Immunomodulating Pharmabiotics in Allergic Diseases: Current Status and Future Prospects

  • The prevalence of allergic disorders has dramatically increased over the past decade, particularly in developed countries. Apart from gastrointestinal disorders, neoplasia, genital and dermatological diseases etc., dysregulation of gut microbiota (dysbiosis) has also been found to be associated with increased risk of allergies. Probiotics are increasingly being employed to correct dysbiosis and, in turn, to modulate allergic diseases. However, several factors like strain variations and effector metabolites or component of them in a bacterial species can affect the efficacy of those as probiotics. On the other hand, host variations like geographical locations, food habits etc. could also affect the expected results from probiotic usage. Thus, there is a glaring deficiency in our approach to establish probiotics as an irrefutable treatment avenue for suitable disorders. In this review, we explicate on the reported probiotics and their effects on certain allergic diseases like atopic dermatitis, food allergy and asthma to establish their utility. We propose possible measures like elucidation of effector molecules and functional mechanisms of probiotics towards establishing probiotics for therapeutic use. Certain probiotics studies have led to very alarming outcomes which could have been precluded, had effective guidelines been in place. Thus, we also propose ways to secure the safety of probiotics. Overall, our efforts tend to propose necessary discovery and quality assurance guidelines for developing probiotics as potential immunomodulatory 'Pharmabiotics.'

Potential Anti-Allergy and Immunomodulatory Properties of Lactococcus lactis LB 1022 Observed In Vitro and in an Atopic Dermatitis Mouse Model

  • Lactococcus lactis is a lactic acid bacterium and used in the dairy food industry. The ameliorating effects of Lactobacillus species on atopic dermatitis (AD) have been extensively studied, but the specific effect of L. lactis strains has not yet been investigated. In this study, the efficacy of L. lactis LB 1022, isolated from natural cheese, was evaluated using RAW 264.7, HMC-1 and HaCaT cell lines and an ovalbumin-sensitized AD mouse model. L. lactis LB 1022 exhibited nitric oxide suppression and anti-allergy and anti-inflammatory activity in vitro. Oral administration of L. lactis LB 1022 to AD mice significantly reduced the levels of IgE, mast cells, and eosinophils, and a range of T cell-mediated T helper Th1, Th2, and Th17-type cytokines under interleukin (IL)-10, transforming growth factor-β (TGF-β), thymus and activation-regulated chemokine (TARC), and thymic stromal lymphopoietin (TSLP). In addition, L. lactis LB 1022 treatment increased the concentration of short-chain fatty acids. Overall, L. lactis LB 1022 significantly modulated AD-like symptoms by altering metabolites and the immune response, illustrating its potential as candidate for use in functional food supplements to alleviate AD.

지방자치단체 중심의 아토피 예방관리 프로그램 효과 평가 (Evaluation of Effectiveness for an Atopy Prevention Program Operated by a Local Government)

  • Objectives: We evaluated the effects of an allergy control program on children with atopic dermatitis (AD) in five schools located within Gyeonggi-do. Methods: Based on the results of the 12-month prevalence of AD symptoms from an International Study of Asthma and Allergies in Childhood (ISAAC) questionnaire of all students (n=2109; 1040 boys, 1069 girls) at five randomly selected schools, 227 students with AD symptoms were screened. Finally, 188 students with greater than 1 on SCORAD (SCORing Atopic Dermatitis) determined by a physician were selected. The allergy control program includes education related to allergic diseases, skin prick tests, and SCORAD evaluation, as well as keeping a daily journal to check the usage of moisturizer at least once a day. To determine the effectiveness of the allergy control program, SCORAD evaluation was performed again six weeks later. Results: The most common symptom for 12-month prevalence was rhinorrhea (33.9%), followed by atopic skin lesions (14.8%) and wheezing (4.0%). The 12-month prevalences of the three allergic diseases were higher for boys than for girls, and a significant difference was found for allergic rhinitis (P<0.001). The mean SCORAD index decreased significantly from 26.1 to 17.5 after the completion of the six-week control program (P<0.01). In particular, these differences were more pronounced for the group which used the daily journal (P<0.001). Conclusions: Our findings indicate that the allergy control program including usage of a daily journal as well as regular monitoring could be a promising tool for preventing and alleviating the symptoms of allergic diseases.

약용식물추출물의 아토피성피부염에 대한 항염증 및 항알레르기효과(제 1보) (Anti-inflammatory and Anti-allergic Effects of Herbal Extracts on Atopic Dermatitis(Part I))

  • Atopic dermatitis is a chronic inflammatory skin disease associated with cutaneous hyper-reactivity to environmental triggers. In order to develop effective therapeutic herbal extracts for atopic dermatitis, cytotoxicity, antioxidant, anti-inflammatory and anti-allergic activities were investigated for various herbal extracts. Among candidate extracts, we selected Aloe vera L. (AV), Viola mandshurica W. Becker (VM), Punica granatum L. (PG), Dendrobium nobile L. (DN) and mixture of the above extracts (MX) for further investigations. All of them did not show cytotoxic activities to macrophage RAW264.7 cells below the concentration of 100 ppm. All showed antioxidant, anti-inflammatory and anti-allergic effects, although to various extents. In antioxidant effects, AV showed the highest effect, followed by PG and VM, while DN did the lowest. In evaluation for anti-inflammatory activities in macrophage RAW264.7 cells, AV and DN inhibited almost completely the LPS-induced production of nitric oxide, while AV, DN and VM showed strong inhibitory activities on the LPS-induced production of TNF-${\alpha}$. In anti-allergy effect in mast cell HMC-1, DN showed the highest effect, followed by AV and PG, while VM did the lowest. In the topical allergy reaction induced by compound 48/80 in Sprague-Dawley rat, DN exhibited significant anti-allergic effect, while PG, VM and AV did slight effect. These results suggest that AV, VM, PG and DN have antioxidant, anti-inflammatory and anti-allergic activities, and thus have the potential to reduce and alleviate the symptoms of atopic dermatitis.

청소년 알레르기성 질환의 복합성과 중증도가 자살 생각에 미치는 영향 (Effect of Allergy Related Disease on Suicide Ideation among Adolescents in Korea)

  • Background & Objectives: There were increasing evidence about the relationship between allergy related disease such as asthma, atopic dermatitis and allergic rhinitis and suicide ideation. However little was known about the concrete relatedness between severity and comorbidity of allergy related disease with suicide ideation. The objective of this study was to investigate the cases of the prevalence of suicide ideation among adolescents with allergy related disease such as asthma, atopic dermatitis and allergic rhinitis, and examine the association between allergy related disease and suicidal ideation among adolescents in South Korea. Methods: Data was based on Korean Youth Risk Behavior Web-based Survey(2014) which was a cross-sectional study containing 34,874 Korean middle and high school students who diagnosed with allergy related disease. We used the weights, strata and primary sampling unit information provided by the public use dataset to compute descriptive statistics and logistic regressions. Computations were done with SPSS version 20.0. Results: 19.9%, 15.6%, 13.8% of adolescents who suffered from one, two and three of allergy related diseases respectively reported having been thought of suicide ideation. Socio-demographic factors were adjusted as control variables. Students with greater severity of disease were more likely to have suicide ideation. Odds ratio for students who were absent one to three days from school because of allergies was 1.96(95% CI 1.51-2.46), and odds ratio for those who were absent more than four days from school was 3.60(95% CI 2.46-5.28). Conclusions: Given that adolescents' severity and comorbidity of allergy related disease were clearly associated with suicide ideation, suicide prevention programs for adolescents with allergy related disease should be improved by strategic approaches towards the severity and comorbidity of disease.
