Understanding and Importance-Performance Analysis of Food Allergen Labeling System (알레르기 유발식품 표시의 이해도와 중요도-수행도 분석 -서울·경기지역의 식품업체 종사자를 대상으로-)

  • The purpose of this research is to investigate and analyze food allergy labelling for the arrangement of improvement plans. Survey was done as a quantitative research targeting food industry employees with 399 random workers in Seoul and Gyonggi area. The data was analyzed using SPSS windows (ver. 12.0) for frequency analysis, t-test and factor analysis. The importance and performance of the food allergen labelling were divided by 15 items. Only 43.1% of the workers understood the operation allergic food labels. The first improvement on allergic food labels was "using boldface for food allergen labes". The importance of all of these factors was significantly higher than performance. The selection attributes with relatively low the performance but high importance(2 quadrant) were "consumer education for allergic food labels" and "training of professional counselors on food allergy". Therefore, the factors to be improved through the IPA were consumer education and training of professional counselors. With this research and extended efforts for revision of laws, reliability of food industry and accuracy of food labelling would improve, thereby boosting the productive commercial activities in labelling code.

Effects of Seokgokamibokhapbang(SGBHB) on anti-CD40-and recombinant IL-4-induced cytokine production and immunoglobulin E and histanine release in highly purified mouse B cells (석고가미복합방이 Anti-CD40과 rIL-4로 유도된 생쥐의 B 세포에서 싸이토카인 생성 및 면역글로블린 E에 미치는 효과)

  • In the present study, we exarnined anti-allergic effect of SGBHB in cultured B cells. B cells were prepared from isolated murine splenocytes and activated by co-treatment of anti-CD40 monoclonal antibody and recombinant IL-4 allergens. Anti-allergic effects of SGBHB in activated B cells were determined by measuring B cell surface activated molecules (CD23+ and CD11a+), and expression levels of IL-$1{\beta}$, IL-6, IL-10, TNF-$\alpha$, IgE, and HRF. The major findings are summarized as follows. 1. SGBHB treatment did not produce significant cytotoxic effects on mouse lung fibroblast cells. 2. SGBHB produced significant inhibitory effect on the expression of B cell surface activated molecules (CD23+ and CD11a) in activated B cells. 3. SGBHB treatment significantly inhibited expression levels of IL-$1{\beta}$, IL-6, and TNF-$\alpha$ mRNAs in activated B cells.IL-6 protein levels were significantly decreased by $100{\mu}g/m{\ell}$ of SGBHB treatrrient, and TNF-$\alpha$ protein levels were decreased compared to the control group, but statistically insignificant. 4. SGBHB treatment significantly increased IL-10 at both mRNA and protein levels in activated B cells. 5. SGBHB treatment significantly inhibited levels of IgE production. Thus, the present data suggest that SGBHB has an anti-allergic effect on activated B cells by controlling irnmune responses, and further implicates the possibility on clinical application as a therapeutic agent.

Comparison of IgE induction in mice by pollens from three pine tree species

  • Background: Over the years, pine pollens have been excluded as an allergen due to its relatively large size, low protein content, and waxy hydrophobic layer, despite their abundance. However, recent studies suggest the possibilities of pine pollens being allergens, and it has been reported that allergy symptoms were highly prevalent in areas with considerably large pine forests and high possibility of exposure to the pollen. Therefore, we conducted a comparative analysis of the allergenicities of the pollens from the dominant species of Korean pines, red pine (Pinus densiflora), black pine (Pinus thunbergii), and pitch pine (Pinus rigida), in mice. Methods: The protein composition of the pollens from the three pine species was compared via sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The pine pollens and proteins extracted from the pollens were introduced to BALB/c mice by nasal inhalation and application to exposed skin and the IgE produced by the mice were extracted from blood and analyzed via ELISA. Results: SDS-PAGE showed differing protein compositions of the pollens of the three pine species. Analysis of blood IgE compositions showed a similar amount of IgE produced when pollens were applied to skin. In contrast, when mice inhaled the pollens, P. densiflora was shown to induce significantly more IgE production than those of the other two species. Conclusions: The experimental results demonstrate that the pollens of all three South Korean pine species induce IgE production, and this production was more pronounced when the pollens were inhaled than when they were applied to the skin. Of the three species, the pollen of P. densiflora was found to induce the highest level of IgE production.

A Clinical study of Paranasal Sinusitis (副鼻洞炎에 關한 臨床的 硏究)

  • Paranasal sinusitis, especially chronic is one of the most common diseases in the field of otolaryngology. It is similar to Bee Yeun(鼻淵) in oriental medicine. Most cases of sinusitis are due to anatomical abnormalities within ostiomeatal unit or disturbed mucocilliary flow. The ostiomeatal unit is the first place of contact with bacteria and allergens during aspiration, and it can be obstructed easily by minute mucosal swelling due to anatomical narrowness. Therefore the treatment of paranasal sinusitis is not easy and often leads to recurrences in spite of long term treatment or surgical therapy. We studied 83 patients who had visited our hospital with complaints of nasal symptoms; they had been diagnosed as having paranasal sinusitis through an endoscopy or CT scan in another hospital and were diagnosed as the paranasal sinusitis through a PNS series. The results were as follows: 1. Age and sex distribution: The most common occurence was found between 6-10 and 1-5 years old. The Males Were 52($62.7\%$) and Females were 31($37.3\%$). 2. By residence 58 cases lived in apts: 25 lived in houses. 3. The longest duration of disease varied from a lower of 13 to a higher of 36 months in 22 cases and from 7 to 12 months in 21 cases. 4. The most common complication & past history with otolaryngologic or allergic disease were adenoid or tonsil hypertrophy & tonsillectomy and adenoidectomy (21 cases). In decreasing order the others were atopic dematitis, otitis media with effusion and allergic rhinitis. 5. Distribution of paranasal sinus disease was most common in both maxillary sinuses in 52 cases. 12 cases showed a normal PNS X-ray series but these had been diagnosed as paranasal sinusitis with an endoscopy or CT scan in another hospital. 6. Common sinusitis - related symptoms were from highest incidence to lowest nasal obstruction, postnasal drip rhinorrhea, frontal headache, cough with sputum. 7. The most administered of prescription was Gamibangpoongtongsungsan and Sunbangpaedoksan extract. 8. In 26 cases the subjects showed significant improvement symptoms in PNS X-ray series : In 21 case showed partial improvement symptoms.

  • Egg allergy has been reported as the most prevalent food hypersensitivity among children. One of the major egg allergens is ovalbumin (OVA). OVA is the major protein in the egg white, comprising 54% of its total protein content. Curcumin isolated from Curcuma longa has been used as folk remedies in order to treat many chronic diseases for many years. In the present report, we present biochemical evidence that curcumin inhibits the NF-${\kappa}B$ activation induced by OVA. Curcumin also inhibits OVA-induced iNOS expression and nitrite production. These data suggest new approaches for the development of efficient anti-allergic strategies.

Evaluation of the Agreement between Immunodot Assays and Intradermal Skin Testing or Favrot Diagnostic Criteria in Canine Atopic Dermatitis

  • This study was undertaken to identify differences between atopic and non-atopic dogs in three rapid screening immunodot assays as well as the ability of the assays to predict the results of intradermal skin testing (IDST) or Favrot diagnostic criteria (FDC). Twenty-nine dogs diagnosed with canine atopic dermatitis (CAD) were selected as the atopic group. Twenty-five dogs without CAD were included as the non-atopic group. Three types of immunodot assays were conducted on all serum samples from both groups: Allercept E-screen 2nd generation (ES2G), Canine Allergic Tendency Reference Test (ALERT), and Asan Easy Test Canine IgE (AETC). IDST, which included 39 allergens, and immunodot assays were performed concurrently in 13 dogs from the atopic group and compared. While there were no significant differences in positivity between the two groups in the evaluation of ALERT (P = 0.435) and AETC (P = 0.313), positivity in ES2G testing was significantly higher in the non-atopic group than the atopic group (P = 0.038). The ES2G, ALERT, and AETC results showed fair (${\kappa}=0.235$), slight (${\kappa}=0.133$), and slight (${\kappa}=0.014$) accordance with IDST, respectively. The outcomes of ES2G, ALERT, and AETC indicated poor (${\kappa}=-0.211$), slight (${\kappa}=0.106$), and slight (${\kappa}=0.087$) agreement with FDC. In conclusion, rapid screening immunodot assays were not useful for the diagnosis of CAD. These assays may provide a supplementary method for predicting the results of IDST in atopic dogs.

Screening of Herbal Extracts to Reduce PAR-2 and Cytokine Expression Related to Atopic Dermatitis in Keratocytes (피부세포에서 아토피 피부염의 유발과 관련된 PAR-2 및 사이토카인의 발현을 감소시키는 한약재 탐색)

  • The prevalence of atopic dermatitis has markedly increased in recent years but the mechanism has not been clearly revealed. Recent study exhibited that atopic dermatitis was exacerbated by the increase of proteinase-activated receptor (PAR)-2 expression, which activated $I{\kappa}B$ kinase --> nuclear factor kappa B. Therefore, we determined whether the allergens of dust mites induced the expression of PAR-2, intercellular adhesion molecule-1 (ICAM-1, adehision molecule), interleukins (IL)-6 in HaCaT keratocytes and which herbal 1,3-butylene glycol extracts (Mori Cortex Radicis, Sanguisorba officinalis L., Arctium lappa Linne, Torilis japonica DC, Melia azedarach Linne var. japoinca Makino) suppressed their expression. Dust mite allergen increased PAR-2, ICAM-1 and IL-6 expression in HaCaT cells in a dose-dependent manner up to $3{\mu}g/mL$ but their expression reached the plateau over the dosages. The allergen ($3{\mu}g/mL$) also secreted more cytokines such as tumor necrosis factor (TNF)-${\alpha}$ and IL-6 into the media. Among five different herbal extracts ($50{\mu}g/mL$), Mori Cortex Radicis and Sanguisorba officinalis L. suppressed the PAR-2, ICAM-1 and IL-6 expression in HaCaT cells, which was activated by dust mite allergen ($3{\mu}g/mL$) and they also reduced the secretion of TNF-${\alpha}$ and IL-6 into the media. In conclusion, Mori Cortex Radicis and Sanguisorba officinalis L. can effectively reduce the prevalence and progression of atopic dermatitis by dute mite allergen.

Clinical Study of MAST CLA and Sasang Constitution in Allergic Rhinitis Patients (알레르기비염 환자의 체질분포 및 특이적 IgE 측정법(MAST CLA)의 임상적 의의)

  • Background and Objectives The diagnosis of allergic rhinitis includes detailed clinical history, physical examination and the use of either in vivo or vitro tests for relevant allergens. Skin test has been used the most commonly. Recently MAST CLA is used for determination of serum spcecific IgE, This study attempted to find out the distribution of Sasang Constitution and to compare the MAST CLA with skin tests in allergic rhinitis patients. Methods Skin tests, MAST CLA and Sasang Constitution study were performed for 35 allergic rhinitis patients who visited Kyunghee Oriental Medical Center from Sept. 2001 to Nov. 2001. Results 1. The ratio between male and female was 1:1.5. the peak age was the thirties(42.9$\%$) 2. 45.7$\%$ of patients had family history of allergic diseases and allergic rhinitis was the most common. 3. 51.4$\%$ of patients lived in A.P.T. and in preference of cool and warm, 54.3$\%$ of patients prefered both of cool and warm. 4. Among 24 cases who were consulted to dept. of Sasang, 45.8$\%$ was Taeumin. 5. 65.7$\%$ of patients reacted positive to skin test and the common offending allergen was D. pteronyssinus(82.6$\%$). 6. 25.7$\%$ of patients reacted positive to MAST CLA and the common offending allergen was D. farinae(88.9$\%$). 7. Among 22 cases who was performed skin test and MAST CLA the sensivity and specificity of MAST CLA was 27.4$\%$ and 94.9$\%$. There was significant correlations between MAST CLA and skin test(p=0.005, r=0.574, 1, spearman correlation coefficienct).

Genomics Approach to Identify the Cause of the Missing Omega-5 Gliadin Protein in O-Free Wheat

  • A previous work developed and identified a new omega-5 gliadin deficient wheat line named O-free by crossing Keumkang and Olgeuru, which is nutritionally quite meaningful in that omega-5 gliadin is one of the known wheat allergens. To verify the characteristics of the O-free, we performed RNA sequencing (RNAseq) analysis of the O-free and the two parent lines (Keumkang and Olgeuru). The results of the similarity analysis with the ESTs for gliadins and glutenins showed that the O-free ESTs had no similarity with the omega-5 gliadin sequences but had similarity to other gliadins and glutenins. Furthermore, mapping results between the raw RNAseq data from the O-free and the omega-5 gliadin sequence showed a clear deletion of the N-terminal sequences which are an important signature of omega-5 gliadin. We also designed specific PCR primers that could identify omega-5 gliadin in the genomic DNA. The results showed that no omega-5 gliadin fragments were detected in the O-free. According to these results, we confirmed that the deficiency of omega-5 gliadin in the O-free is not caused by post-transcriptional or post-translational regulations such as epigenetic phenomena but by a simple deletion in the chromosome. Furthermore, we showed that the low-molecular weight glutenin subunit (LMW-GS) gene in the O-free had a single nucleotide polymorphism (SNP) causing a premature stop codon, resulting in a truncated polypeptide. We expect that the O-free line may serve as an excellent source of wheat that could prevail in the hypo-allergen wheat market, which has recently gained interest world-wide.

Monte Carlo Simulation of Irradiation Treatment of Peaches (Prunus persica L. Batsch) (몬테카를로 시뮬레이션을 이용한 복숭아의 방사선 조사)

  • Food irradiation is important not only in ensuring safety but also improving antioxidant activity of peaches. Our objective was to establish the best irradiation treatment for peaches by calculating dose distribution using Monte Carlo simulation. 3-D geometry and component densities of peaches, extracted from CT scan, were entered into MCNP to obtain simulated dose distribution. Radiation energies for electron beam were 1.35 MeV (low energy) and 10 MeV (high energy). Co (1.25 MeV) and the Husman irradiator, containing three sealed Cs source rods in an annular array, were used for gamma irradiation. At 1.35 MeV electron beam simulation, electrons penetrated well beyond the peach skin, enough for surface treatment for microorganisms and allergens. At 10 MeV electron beam simulation, for top-beam only treatment, doses at the core were the highest and for double beam treatment, the electron energy was absorbed by the entire sample. At Co source, the radiation doses were presented on the whole area. At Cs source, the dose uniformity ratios were 2.78 for one source and 1.48 for three ones at 120 degrees interval. Proper control of irradiation treatment is critical to establish confidence in the irradiation process.
