Subtilisin YaB, produced by alkalophilic Bacillus strain YaB, is an extracellular alkaline serine protease having 55% homology to subtilisin BPN'. It is synthesized as a 378-amino acid preproenzyme and secreted into the culture medium as a 265-amino acid mature protease. To examine the role of pro-sequence for the secretion of subtilisin YaB, we have studied the expression, in Bacillus subtilis, of a mutant preprosubtilisin YaB in which active site Ser214 is substituted with Cys. The use of a six protease-deficient strain, WB600, was required for its efficient production. The prosubtilisin YaB, thus produced, was indeed secreted into the culture medium and was processed to its mature form upon treatment with exogenously added active subtilisin YaB. From these results, we have concluded that the processing of pro-sequence is not essential for the secretion of the enzyme.

Fundamental studies at the CSIRO division of Wool technology and ICI on the diffusion of dyes into wool〔1,2〕have let to development of a new approach to wool dyeing. In this method, the cell membrane complex of wool is modified before dyeing by treatment under mildly alkaline conditions with a special chemicals. Wool pretreated with ethoxylated quaternary ammonium salt has an increased rate of dyebath exhaustion and dye penetration early in the dyeing cycle. This enables the treated material to be dyed below the boil for a similar time to the conventional cycle. This technique can be used on untreated and shrinkresist-treated wool and wool/nylon blends. In addition to good macro-levelness and excellent coverage of tippiness, the low temperature dyeing process give higher exhaustion levels of dyestuffs and insect-resist agent and hence cleaner effluent liquors, compared with conventional dyeing process. Low Temperature Dyeing process cause significantly less fiber damage than conventional way. The reduction in damage is reflected in improved processing performance of the dyed wool.
Bromelain has been used as a meat-tenderizing agent in food processing. To increase the availability of bromelain, microencapsulation into liposome was carried out by the dehydration and rehydration method. Small unilamellar vesicles prepared by sonication treatment showed higher encapsulation efficiency (EE) than by the French press method. In the preparation of liposome, the effect of pH and centrifugal force on EE was also investigated and it showed a higher EE at acidic pH than at alkaline pH with centrifugation at 80, 000$\times$g. The actual EEs except unencapsulated bromelain which bound on the outside surface of liposome by electrostatic interaction also were investigated, and the optimal EE was at pH 4.6, at 0.6 of a ratio of bromelain to phosholipid(18.2%, w/w). Release of bromelain from liposomes was stimulated as the temperature increased at neutral pH.
The recovery of tellurium from Te-cement obtained during the processing of copper anode slimes was carried out by a series of hydrometallurgical processes, i.E. leaching Te-cement in NaOH solution, precipitation of impurities with the addition of ${Na}_{2}S$ followed by direct electrowinning of tellurium in NaOH solution. The optimum conditions for each process were determined and discussed in terms of various parameters associated with each step to maximize the recovery and purity of tellurium. The final purity of tellurium from electrownning in alkaline solution was found to be more than 99.9%.
In an attempt to evaluate the fertilizer of sediments obtained from coastal farming areas, chemical composition, bacteriological quality and heavy metals in the sediments alkalized by quicklime and magnesium hydroxide were analyzed. The optimum conditions of alkalization were also measured. A perfect reaction was possible by the addition of quicklime of $30{\%}$ at the rate of $25{\%}$ of sediment and $100{\%}$ of livestock wastes. According to the classification standard for compost constituent by Higgins, all composts had the intermediate or high grade in $T-N, K_2O,\;CaO\;and\;MgO$, but below the low grade in $P_2O_5$, Stabilization by quicklime and magnesium hydroxide has been known to inhibit bacterial decomposition of organic matter and activity of pathogenic organisms. In this study, raising pH of stabilized sediments to 12 for 2 hours (PSRP rriteria of EPA) allowed $99.9{\%}$ of coliform group, fecal group and viable cell count to be reduced. As a result, sediments of coastal farming areas are likely to be used to produce the organic fertilizer by alkaline stabilization.
Active lipase-producing bacterium Burkholderia gladioli Bps-1 was rapidly isolated using a modified trypan blue and tetracycline, ampicillin plate. The electro-phoretically pure enzyme was obtained by purification using ethanol precipitation, ion-exchange chromatography, and gel filtration chromatography. The molecular weight was 34.6 kDa and the specific activity was determined to be 443.9 U/mg. The purified lipase showed the highest activity after hydrolysis with $p-NPC_{16}$ at a pH of 8.5 and $50^{\circ}C$, and the $K_m$, $k_{cat}$, and $k_{cat}/K_m$ values were 1.05 mM, $292.95s^{-1}$ and $279s^{-1}mM^{-1}$, respectively. The lipase was highly stable at $7.5{\leq}pH{\leq}10.0$. $K^+$ and $Na^+$ exerted activation effects on the lipase which had favorable tolerance to short-chain alcohols with its residual enzyme activity being 110% after being maintained in 30% ethanol for 1 h. The results demonstrated that the lipase produced by the strain B. gladioli Bps-1 has high enzyme activity and is an alkaline lipase. The lipase has promising chemical properties for a range of applications in the food-processing and detergent industries, and has particularly high potential for use in the manufacture of biodiesel.

The effects of glass composition on the wettability and reactivity with $ZrO_2$substrate was evaluated and fabrication variables and glass compositions was investigated. Various glass compositions was investigated. Alkaline earth silicate glass show good wettability and lower viscosity and crystallization of glass could be prevented by $B_2O_3$.The sealant glass begin to wet on $ZrO_2$substrate below $900^{\circ}C$ and porosity occurred in various glass compositions, the crystallization and porosity in the glass could be prevented by the addition of flux into glass composition. But flowability and reactivity of glass with $ZrO_2$substrate was enhanced. Processing variables should be optimized to reduce the porosity by enhancing the sintering of glass powder. Many silicate glasses were investigated for the applications of high temperature sealants. Wetting and bonding of glass was good enough to seal together between $ZrO_2$and other ceramic components of SOFC. But porosity and reaction layer were occurred in the sealant glass. It will be possible to produce glass sealant without porosity and reaction layer at the interface by optimization of processing variable and modify the glass compositions. In present study, wettability of glass-filler composite was investigated. The porosity, shape of filler and interfacial reactions of sealant glass with fillers were examined.

To utilize effectively fish skin wasted from fish processing, a yellowfin sole skin gelatin was prepared by alkaline extraction method and the physico-chemical properties were examined. Conditions for the suitable extraction and decolorization for gelatin preparation from yellowfin sole skin are as follows: the skin is limed with 1.5% calcium hydroxide solution at $5^{\circ}C$ for 5 days, washed throughly with tap water, extracted with 6 volumes of water ($pH5.0{\sim}7.0$) to dehydrated skin for 3 hours at $50^{\circ}C$, and then bleached with 3% activated carbon. Though yellowfin sole skin gelatin was prepared under above condition, the physico-chemical property values such the melting point and gelling point of that were lower than those of pork skin gelatin. Therefore, the purified yellowfin sole skin gelatin requires a suitable modification operation for more a good quality gelatin manufacture.
This paper examines the hydration and physical properties of alkali-blast furnace slag cement activated by Na$_2$SiO$_3$, Na$_2$CO$_3$, NaOH, Na$_2$SO$_4$. Four levels of Na$_2$O content in mixtures, 1, 3, 5, and 7 wt%, were investigated, and a W/S ratio 0.5 was used to prepare paste and mortar specimens. Compressive strength measurement of mortars was carried out adding alkali activated slag 30 wt% to OPC. The main hydration products with alkali activator kinds were C-S-H,C$_4$AH$\_$13/, AFt and Al(OH)$_3$ etc. For using Na$_2$CO$_3$ activated slag, hydration ratio of slag was higher than that of different activators, and Na$_2$SO$_4$ activated slag mortar appeared the highest compressive strength values at 28 days with activator content of 5 and 7 wt%.
