Phenolic Compounds from the Flower Buds of Camellia japonica

  • Hot water extracts of Camellia japonica flower buds were found to have the higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity than the other solvent extracts. Five phenolic compounds were isolated and purified from the ethyl acetate soluble-neutral fraction of hot water extracts by Sephadex LH-20 column chromatography and octadecyl silane-high performance liquid chromatography using the guided assay of DPPH radical scavenging. Based on mass spectrometer and nuclear magnetic resonance, the isolated compounds were identified as p-hydroxybenzaldehyde (1), vanillin (2), dehydroxysynapyl alcohol (3), 7S,7'S,8R,8'R-icariol $A_2$ (4), and (-)-epicatechin (5). Four compounds (1-4) except for 5 were newly identified in this plant. Their DPPH radical scavenging activities as 50% scavenging concentration decreased in order to 5 $(20\;{\mu}M)>{\alpha}-tocopherol$ $(29\;{\mu}M)>4$ $(67\;{\mu}M)>3$ $(72\;{\mu}M)>1=2$ ($>250\;{\mu}M$). These results indicate that the antioxidant effect of the hot water extract of C. japonica flower buds may partially due to 5 isolated phenolic compounds.

  • The fruits of Prunus davidiana were extracted with 80 % aqueous methanol at room temperature. The concentrated extract was partitioned as ethyl acetate (EtOAc), n-butyl alcohol, and water fractions. From the EtOAc fraction, three triterpenoids were isolated through the repeated silica gel ($SiO_2$) and octadecyl $SiO_2$ column chromatographies. Based on physico-chemical and spectroscopic data including nuclear magnetic resonance, mass spectrometry, and infrared, the compounds were identified to be ursolic acid (1), corosolic acid (2), and ${\alpha}-amyrin$ (3).

Effects of Horse Bone Extracts on the Induced Postmenopausal Osteoporosis in Rats (백서의 폐경기 골다공증 모델에서 말뼈 추출물의 효과)

  • Osteoporosis is a metabolic bone disease associated with an imbalance of bone remodeling. Osteoporosis is characterized by decreased bone mass and increased bone fractures. In this study, we investigated the effects of horse bone extracts (HBEs) in vivo. Horse bone was extracted with 80% alcohol (HBE-A) at $100^{\circ}C$ or water (HBE-W) at $120^{\circ}C$. Animal model of postmenopausal osteoporosis was used, in which osteoporosis was induced by ovariectomy of female S.D. rats (female rats were divided into 5 groups), and HBEs were administered to ovariectomized rats every day for 8 weeks. After 8 weeks, the rats were sacrificed and the following osteoporotic factors were measured: body weight, bone mineral density (BMD), uterine/body weight ratio, serum estradiol (E2), and serum alkaline phosphatase (ALP). The results showed that the administration of HBE-W decreased the changes of body weight in ovariectomized rats. HBE-W increased the uterine/body weight ratio and BMD. In addition, HBEs decreased the ALP. Therefore, HBEs may be used for the prevention or treatment of bone disease.

Isolation and Charactrization of Lactic Acid Bacteria and Yeast from Traditional Andong sikhe (전통 안동식혜로부터 젖산균 및 효모의 분리 및 그 특성)

  • We isolated seventy two lactic acid bacteria and two hundred yeast from traditional Andong sikhe. Microorganisms were isolate from 4 kinds of traditional Andong sikhe. The optimum temperature and pH for growth of sikhe yeast, No. SCS 5, was $30^{\circ}C$ and 4.5, respectively. SCS 5 produced $CO_2$ gas and 1.5% of alcohol in malt extract broth. SCS 5 was identified as S. cersvisiae from the observation of shape of vegetative reproduction, morphological and cultural chararteristics, fermentation and assimilation of carbon sources, and physiological characteristics.

Analysis of Total Sugar by Extraction Condition and Material to Develope the Extraction Process of Ginseng Polysaccharide (인삼 다당체 추출 공정 개발을 위한 인삼의 추출 조건 및 원료에 따른 총당 변화)

  • This study analyzed the extraction condition for large-scale extraction process to be used industrially. The total sugar content of 5-year Red ginseng in viewpoint of the ginseng materials was highest by $32\%$ and it of 5-year Keumsan ginseng was $31\%$. Therefore 5-year Keumsan ginseng was used by test sample. The next extraction condition, that is the total sugar content of the internal white among the parts of ginseng, the extraction efficiency under the condition of optimal temperature $80^{\circ}C$ and optimal extract time 6hrs, was highest. And the amount of total sugar extracted from ginseng treated with protease, ${\alpha}-amylase$ and cellulase was increased about $20\%$. Total sugar recovery in methods of alcohol concentration $70\%$ and freezer-dry method was highest.

Anti-Obesity and Lipid Metabolism Effects of Ulmus davidiana var. japonica in Mice Fed a High-Fat Diet

  • The root bark of Ulmus davidiana var. japonica (Japanese elm) is used in Korea and other East Asian countries as a traditional herbal remedy to treat a variety of inflammatory diseases and ailments such as edema, gastric cancer and mastitis. For this study, we investigated the lipid metabolism and anti-obesity efficacy of ethyl alcohol extract of Ulmus davidiana var. japonica root bark (UDE). First, HPLC was performed to quantify the level of (+)-catechin, the active ingredient of UDE. In the following experiments, cultured 3T3-L1 pre-adipocytes and high-fat diet (HFD)-fed murine model were studied for anti-obesity efficacy by testing the lipid metabolism effects of UDE and (+)-catechin. In the test using 3T3-L1 pre-adipocytes, treatment with UDE inhibited adipocyte differentiation and significantly reduced the production of adipogenic genes and transcription factors PPARγ, C/EBPα and SREBP-1c. HFD-fed, obese mice were administered with UDE (200 mg/kg per day) and (+)-catechin (30 mg/kg per day) by oral gavage for 4 weeks. Weight gain, epididymal and abdominal adipose tissue mass were significantly reduced, and a change in adipocyte size was observed in the UDE and (+)-catechin treatment groups compared to the untreated control group (***p < 0.001). Significantly lower total cholesterol and triglyceride levels were detected in UDE-treated HFD mice compared to the control, revealing the efficacy of UDE. In addition, it was found that lipid accumulation in hepatocytes was also significantly reduced after administration of UDE. These results suggest that UDE has significant anti-obesity and lipid metabolism effects through inhibition of adipocyte differentiation and adipogenesis.

Anti-Inflammatory Effect of Hexane Fraction from Eisenia bicyclis on Lipopolysaccharides-Treated RAW 264.7 Cells (LPS로 유도된 RAW 264.7 대식세포에 대한 대황(Eisenia bicyclis) 헥산 분획물의 항염증 효과)

  • Eisenia bicyclis is known to have secondary metabolites exhibiting various biological activities. In a preliminary study, the n-hexane fraction obtained from the ethanolic extract of E. bicyclis showed higher anti-inflammatory activity than the ethyl acetate and butyl alcohol fractions based on the inhibition of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production in RAW 264.7 cells. Using this fraction (E. bicyclis hexane fraction, EHF), we investigated the molecular mechanisms underlying its anti-inflammatory effect in LPS-stimulated RAW 264.7 cells. Pretreatment of the cells with up to 50 ㎍/mL EHF significantly inhibited NO and prostaglandin E2 production as well as their responsible enzyme proteins and mRNAs, in a dose-dependent manner (P<0.05). Similarly, EHF markedly reduced the production of pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6 and tumor necrosis factor (TNF)-α as well as their mRNA levels. Nuclear translocation of nuclear factor-kappa B (NF-κB) was strongly suppressed by EHF treatment. EHF significantly reduced the phosphorylation of mitogen-activated protein kinases and phosphatidylinositol 3-kinase/Akt in LPS-stimulated cells. Moreover, EHF reduced ear edema in phorbol myristate acetate (PMA)-induced mice. These results indicate that EHF contains potent anti-inflammatory compounds, which may be used as a dietary supplement for the prevention of inflammatory diseases.

Effects of Nature Herb Mixture Containing Laminaria japonica Areschoung on Fine Dust-induced Bronchitis in Mice (미세먼지로 유도된 기관지염증에 다시마가 함유된 복합물의 효과)

  • The purpose of this study was to investigate the effects of natural herb mixture containing Laminaria japonica Areschoung (LAM) on fine dust-induced bronchitis in mice. Laminaria japonica Areschoung is main content of LAM, which is including fucoidan. Fucoidan extracted from phaophyta is known to prevent bronchitis and to defend against bacteria and virus infection. In this study, we experienced the effect of LAM on bronchitis and investigated gene expressions (e.g ; IL-8, COX-2, MCP-1) and bio-markers (e.g ; IL-8, PGE2, MUC5AC) associated with bronchitis by using A549 cells. Also, we investigated whether LAM can suppress the bronchitis in fine dust-induced animal models. We injected fine dust (50 ㎕) twice as INT (Intra-Nasal-Trachea) method. Then LAM (200 mg/kg and 400 mg/kg) were oral administered for 14 days. We analyzed the number of immune cells, immunoglobulin E, bio-markers level associated with bronchitis. LAM significantly decreased bio-marker (IL-1β, IL-6, IL-8, TNF-α, Histamine, PGE2), immune cells (white blood cell, neutrophil, lymphocyte, monocyte), and immunoglobulin E, that are increased by fine dust. Taken together, this study suggest that LAM can be used as effective herbal extract for bronchitis.

Natural Product Extracts for Depression: Analysis of Patent Status in South Korea (우울증에 대한 천연물 추출물 국내 특허 동향 분석)

  • Objectives: To analyze the trend of Korean patents of natural product extracts for depression. This study also aimed to enhance the development and application of natural product extracts for depression in Korean herbal medicine. Methods: We searched the Korea Intellectual Property Rights Information Service and Science ON to collect Korean patent data of natural product extracts for depression. Two authors independently screened and assessed full texts of screened patents for eligibility. Included patents were analyzed both quantitatively and qualitatively. Results: A total of 62 patents from 2002 to December 2021 were included for analysis. The number of patents has constantly increased since 2002. Glycyrrhiza uralensis Fischer, Perilla frutescens Britton var. acuta Kudo, Zizyphus jujuba Miller var. inermis Rehder were frequently used among 62 patents. Alcohol extraction was mostly used, followed by water extraction and ethyl extraction regarding extract methods. Eight patents were herbal medicine used for treating depression in Korean Medicine. Conclusions: Various Korean herbal medicine and minerals were used as natural-products for treating depression. These results provide fundamental data that can be used for inventing new patents using Korean herbal medicine, developing new natural product extracts for depression, and extending the range of application of these products in clinical setting.

Analysis of ethyl carbamate in alcoholic beverages (주류 중 에틸카바메이트 분석)

  • In order to survey the contents of ethyl carbamate in alcoholic beverages, GC/MS-SIM method was used after extraction with dichloromethane in solid phase extract cartridge contained alcoholic beverages. In the applied GC/MS-SIM method, the values of recoveries and relative standard deviation were ranged from 85.2 % to 87.9 % and from 0.7 % to 1.9 %, the limit of detection and quantification were $2{\mu}g/kg$ and $10{\mu}g/kg$. Depending on alcoholic beverage kinds, the levels were variable and the average level was $194{\mu}g/kg$ for liquor, $105{\mu}g/kg$ for fruit wine, $62{\mu}g/kg$ distilled spirit, $28{\mu}g/kg$ for sake, $15{\mu}g/kg$ for yakju, $12{\mu}g/kg$ for other alcohol beverages, ND for soju, respectively.
