Preventive effects of water extracts from Artemisiae Iwayomogi Herba on hyperlipiderma and liver damage induced by alcohol (한인진(韓茵蔯)이 알콜 투여로 유발된 흰쥐의 고지혈증과 간 손상의 예방에 미치는 영향)

  • Objectives : This study was performed to prove the preventive effect of water extracts from Artemisiae Iwayomogi Herba on hyperlipiderma and liver damage induced alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And Artemisiae Iwayomogi Herba extract was administrated for the same period. We measured the serum component in rat's blood. body weight and weight of liver. Result : At first, we observed preventive effects of Artemisiae Iwayomogi Herba on hyperlipiderma induced by alcohol. Artemisiae Iwayomogi Herba group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. Artemisiae Iwayomogi Herba group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. At second, we observed preventive effects of Artemisiae Iwayomogi Herba on liver damage induced by alcohol. Artemisiae Iwayomogi Herba group showed significant decrease of GOT, ALP and LDH in comparison with those of the control group. Artemisiae Iwayomogi Herba group showed significant increase of liver weight in comparison with those of the control group. Conclusion : Reviewing these experimental results, it appears that water extracts from Artemisiae Iwayomogi Herba have pharmaceutical preventive efficacy on hyperlipiderma and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of Artemisiae Iwayomogi Herba.

Oral Administration of Alcohol-Tolerant Lactic Acid Bacteria Alleviates Blood Alcohol Concentration and Ethanol-Induced Liver Damage in Rodents

  • Excessive alcohol consumption can have serious negative consequences on health, including addiction, liver damage, and other long-term effects. The causes of hangovers include dehydration, alcohol and alcohol metabolite toxicity, and nutrient deficiency due to absorption disorders. Additionally, alcohol consumption can slow reaction times, making it more difficult to rapidly respond to situations that require quick thinking. Exposure to a large amount of ethanol can also negatively affect a person's righting reflex and balance. In this study, we evaluated the potential of lactic acid bacteria (LAB) to alleviate alcohol-induced effects and behavioral responses. Two LAB strains isolated from kimchi, Levilactobacillus brevis WiKim0168 and Leuconostoc mesenteroides WiKim0172, were selected for their ethanol tolerance and potential to alleviate hangover symptoms. Enzyme activity assays for alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were then conducted to evaluate the role of these bacteria in alcohol metabolism. Through in vitro and in vivo studies, these strains were assessed for their ability to reduce blood alcohol concentrations and protect against alcohol-induced liver damage. The results indicated that these LAB strains possess significant ethanol tolerance and elevate ADH and ALDH activities. LAB administration remarkably reduced blood alcohol levels in rats after excessive alcohol consumption. Moreover, the LAB strains showed hepatoprotective effects and enhanced behavioral outcomes, highlighting their potential as probiotics for counteracting the adverse effects of alcohol consumption. These findings support the development of functional foods incorporating LAB strains that can mediate behavioral improvements following alcohol intake.

Conclusion : Reviewing these experimental results, it appears that water extracts from Injinho-tang and Haninjinho-tang have pharmaceutical preventive efficacy on hyperlipidemia and liver damage induced by alcohol. Therefore further additional study should be conducted to elucidate in depth the pharmaceutical efficacy of these.

  • Objectives : The Korean medical doctors use PMR(Polygoni Multiflori Radix) for nourishing the liver-kidney, loosing the bowel to relieve constipation, recovering from malaria, and clearing away heat and eliminating toxin, etc. But, this study was performed to investigate the effect of water extracts from PMR on prevention of hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And the extract was administrated for the same period. We measured the serum components in rat's blood, body weight and weight of liver. Results : At first, we observed effects of PMR on prevention of hyperlipidemia induced by alcohol. PMR group showed significant decrease of total cholesterol and triglyceride in comparison with those of the control group. PMR group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. At second, we observed effects of PMR on prevention of liver damage induced by alcohol. PMR group showed significant decrease of GOT, GPT, ALP, and LDH in comparison with those of the control group. PMR group showed significant increase of liver weight in comparison with those of the control group. Conclusions : Reviewing these experimental results, it appears that water extracts from PMR have pharmaceutical efficacy on prevention of hyperlipidemia and liver damage induced by alcohol. Methods : Except for the normal group, we fed rat on 25% alcohol for 55 days. And each extract was administrated for the same period. We measured the serum component in rat's blood, body weight and weight of liver. Results : At first, we observed effects of LW on hyperlipemia induced by alcohol. LW group didn't show significant increase of total cholesterol in comparison with those of the control group. LW group didn't show significant increase of HDL(High-Density lipoprotein) cholesterol in comparison with those of the control group. LW group showed significant decrease of triglyceride in comparison with those of the control group. LW group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. LW group showed significant increase of liver weight in comparison with those of the control group. At second, we observed effects of LW on liver damage induced by alcohol. Methods : Except for the normal group, we fed rats the control and Saengkankeonbi-tang groups with 25% alcohol for 55 days. For the same period we fed the Saengkankeonbi-tang group with Saengkankeonbi-tang extract as well and rats in normal and contrlo group on saline solution. We measured the serum components in rat's blood, body weight and weight of liver. Results : 1. At first, we observed effects of Saengkankeonbi-tang on prevention of hyperlipemia induced by alcohol. Saengkankeonbi-tang group showed significant decrease in total cholesterol levels in comparison with those of the control group. Saengkankeonbi-tang group showed significant increase in HDL-cholesterol levels in comparison with those of the control group. Saengkankeonbi-tang group showed significant increase of body weight in comparison with those of the control group in 4weeks and 8weeks. 2.   • Chronic alcohol and tobacco abuse plays a crucial role in the development of different liver associated disorders. Intake promotes the generation of reactive oxygen species within hepatic cells exposing their DNA to continuous oxidative stress which finally leads to DNA damage. However in response to such damage an entangled protective repair machinery comprising different repair proteins like ATM, ATR, H2AX, MRN complex becomes activated. Under abnormal conditions the excessive reactive oxygen species generation results in genetic predisposition of various genes (as ADH, ALDH, CYP2E1, GSTT1, GSTP1 and GSTM1) involved in xenobiotic metabolic pathways, associated with susceptibility to different liver related diseases such as fibrosis, cirrhosis and hepatocellular carcinoma. There is increasing evidence that the inflammatory process is inherently associated with many different cancer types, including hepatocellular carcinomas. The generated reactive oxygen species can also activate or repress epigenetic elements such as chromatin remodeling, non-coding RNAs (micro-RNAs), DNA (de) methylation and histone modification that affect gene expression, hence leading to various disorders. The present review provides comprehensive knowledge of different molecular mechanisms involved in gene polymorphism and their possible association with alcohol and tobacco consumption. The article also showcases the necessity of identifying novel diagnostic biomarkers for early cancer risk assessment among alcohol and tobacco users.

Effects of Dextrin and β-cyclodextrin on Protective Effect of Hovenia dulcis Fruit Extract Against Alcohol-induced Liver Damage in vivo (Dextrin과 β-cyclodextrin이 생체 내에서 헛개나무 추출물의 알코올성 손상으로부터 간보호에 미치는 영향)

  • ${\beta}$-cyclodextrin has an ability to protect compounds from oxidative reaction by collecting them within its ring-like structure. So, In harsh condition ($40^{\circ}C$), marker compound, quercetin, was dramatically reduced in Hovenia dulcis fruit extract containing dextrin at 4 and 8 week compared to 0 week, but not that containing ${\beta}$-cyclodextrin. To evaluate the effects of dextrin and ${\beta}$-cyclodextrin on protective effect of H.dulcis fruit extract against alcohol-induced liver damage, The mice were orally injected alcohol, H. dulcis fruit extract/dextrin (HD) and H. dulcis fruit extract/${\beta}$-cyclodextrin (HCD), respectively, for 7 days. The mice orally administrated with alcohol significantly enhanced the serum concentration of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) and the activity of lactate dehydrogenase (LDH) in serum compared to the control group. HD and HCD significantly decreased the levels of serum ALT and AST and serum LDH activities compared to alcohol group. And also alcohol group significantly increased the level of total cholesterol compared to the control group, but HD and HCD significantly reduced it compared to the alcohol group. However, the levels of TG in blood were not significantly changed in all groups. The activities of alcohol dehydrogenase (ADH) were significantly increased in HD and HCD group although those of aldehyde dehydrogenase showed an increasing tendency. This data suggested that HD and HCD were able to induce alcohol degradation in the liver tissues. All together, the results showed that HCD demonstrated their ability to protect liver from alcohol-induced damage on equal terms with HD.
