Studies on the Selectivity of the Herbicide Alachlor;II. A Metabolic Approach to Selectivity (제초제 Alachlor 의 선택성에 관한 연구;II. 대사론적 접근)

  • Absorption, translocation, and metabolism of the herbicide alachlor in soybean, Chinese cabbage, and barnyard grass seedlings were examined and compared with each other using [phenyl-U-$^{14}C$] alachlor in search of a primary factor contributing to the selectivity of alachlor. When root of each seedling was immersed into the solution containing [$^{14}C$]alachlor, the amount of absorbed radioactivity/mg dry matter of seedling which was suggested to be correlated with the susceptibility of plants to alachlor decreased in the order of soybean ${\gg}$ Chinese cabbage ${\geq}$ barnyard grass and the rate of translocation to shoot was Chinese cabbage ${\geq}$ barnyard grass ${\gg}$ soybean. These orders did not consistently explain the selective phytotoxicity of alachlor. Analyses of extracts by reverse phase chromatography showed that alachlor was detoxified by conjugation with glutathione in all three plants and the rate of glutathione conjugation of soybean, the resistant species to alachlor, was the greatest, while that of barnyard grass, the susceptible, was the lowest among three plants. This result explained well the selective phytotoxicity of alachlor. Both absorption and translocation contribute undoubtedly to the selectivity by influencing the active internal concentration of alachlor. However, neither of them appeared to be a primary factor. It was concluded that the most important primary factor was the rate of glutathione conjugation, which detoxifies alachlor and plays an important role in selectivity.

  • Present work has been initiated to see if inherent biochemical difference among plants is, in any way, related to the observed selectivity characteristics of preemergence herbicide, alachlor. Application of aqueous solution of alachlor onto three intact plants, soybean, chinese cabbage and barnyard grass resulted in phytotoxicity responses in the testt plants in varying degree. Examination of glutathione (and homoglutathione) contents of the test plants indicated that the phytotoxicity is inversely proportional to the peptide contents of the test plants. It was also noted that four to five water soluble metabolites are readily formed in intact seedling treated with labelled alachlor and glutathionealachlor and homoglutathionealachlor conjugates were tentatatively identified as major metabolites. It is concluded that conjugation reaction involving glutathiones and xenobiotic alachlor, a typical phase II reaction, acts as detoxification reaction in the three test plants and this would, in turn, contribute to observed selectivity of alachlor.

  • Two of the degradation products of alachlor in soil. product 1, 2,6-diethyl-N-(methoxymethyl) acetanilide and product 2,2-hydroxy-2',6'-diethyl-(methoxymethyl) acetanilide were synthesized from alachlor reacting with 3N-hydrochloric acid in the presence of zinc powder at room temperature and a saturated sodium bicarbonate solution at $90^{\circ}C$ for 78 hr, respectively. At the concentrations of both $5{\times}10^{-4}M\;and\;1{\times}10^{-3}M$, product 2 exhibited almost the same phytotoxicity to rice seedlings, in particular, as alachlor, whereas product 1 lost its phytotoxic effectiveness. It seems that substitution of chlorine atom by hydroxyl group did not affect the phytotoxicity of alachlor, whereas substitution by hydrogen atom did.

Effect of Seeding Method and Pre-emergence Herbicides on Plant Growth and the Production of Sorghum × Sudangrass Hybrid (파종 방법과 토양처리용 제초제 처리가 수수 × 수단그라스 교잡종의 생육 및 생산성에 미치는 영향)

  • The following experiments were used to evaluate the effect of seeding method and pre-emergence herbicides on weed control and the production of the $sorghum{\times}sudangrass$ [sorghum bicolor (L.) Moench] hybrid under field and in vitro conditions. Under the in vitro condition, we evaluated how different kinds of herbicides affect the rate of seed germination and the growth of early seedlings in the $sorghum{\times}sudangrass$ hybrid treated with herbicides. Simazine, alachlor, and methabenzthiazuron were used, and their concentration levels were 0%, 40%, 70%, and 100%. The germination ratio of the seeds was reduced by the application of simazine, methabenzthiazuron, and alachlor compared with the control (distilled water). The alachlor application severely decreased the sorghum germination ratio. However, seed germination was not greatly influenced by the concentration level of the herbicides. Root and shoot length were increased in the following order: control > simazine > methabenzthiazuron > alachlor. Overall, alachlor significantly reduced root length more than shoot length. The differential tolerance of the $sorghum{\times}sudangrass$ hybrid to pre-emergence herbicides by seeding method was then tested under the field condition. The plants were severely injured by the alachlor application, leading to a great reduction in the number of stems. However, for the simazine and methabenzthiazuron applications the number of stems lost was slightly lower than it was for the alachlor and simazine + alachlor applications. In the seeding method, drilling was more effective in reducing herbicide injury than broadcasting. All of the herbicides effectively controlled weeds. The simazine and methabenzthiazuron applications showed a significantly higher dry matter (DM) yield. However, the alachlor and simazine + alachlor applications severely reduced the DM yield. This result suggests that the alachlor application has a significant effect on plant production. Similarly, the total digestible nutrient (TDN) yield was greatly reduced by the alachlor and simazine + alachlor applications. From the results presented above, we concluded that the yield of DM was the highest for broadcasting with no herbicide. and drilling was the most-effective way to reduce herbicide injury. The simazine application was the most-effective herbicide, showing the highest forage yield and TDN yield.

A Study of Mode of Action of Alachlor - II. Effect of Alachlor on Peroxidase Synthesis in Oat(Avena sativa L.) (Alachlor의 제초기구(除草機構)에 관한 연구(硏究) - II. Alachlor가 귀리의 Peroxidase합성(合成)에 미치는 영향(影響))

  • The effect of alachlor treatment on peroxidase synthesis in oat root tips was studied. Alachlor caused increase in the amount of soluble peroxidase in oat root tips, peroxidase activity increase as the rate of alachlor application increased, Alachlor treatment of oats with $1{\times}10^{-6}M$, peroxidase activity increased 0.20 unit higher than that of nontreatment. After 12hr, 65mM of peroxide treatment of oats inhibited 16% root growth, and 130 mM peroxide treatment caused 59% inhibition. With PAGE of peroxidase extracted from normal root tips, PAGE give 4 species($P_2$, $P_3$, $P_4$, and $P_5$ band) of peroxidase. Alachlor significantly activated isoperoxidase. Three isoperoxidase($P_1$, $P_6$, and $P_7$) are synthesized at a increased concentration of alachlor, SDS-PAGE analysis of proteins extracted from oat root tips showed that they were made up of subunits blow 100 kD polypeptide.

  • In order to illustrate adsorption phenomena of herbicides(alachlor and paraquat) on soils, absorption equation of herbicides and the relationships between soil properties and adsorption constants were investigated with 22 soils. The results were as follows : 1. The shaking time for approaching equillibrium reaction of herbicides(alachlor and paraquat) with woils were about 30 minutes for paraquat and 4 hours for alachlor, respectively. 2. The distribution coefficients of alachlor were inbetween 0.81-33.83 in 5 ppm and 0.09-15.52 in 50 ppm, respectively. 3. The adsorption of alachlor was positively correlated with organic matter and paraquat was with clay content of soils. 4. Both paraquat and alachlor were highly adsorbed in Chunpo series soil containing low contents of organic matter and clay on account of different mechanism from other soils, 5. Freundlich's adsorption constant(K) was greater than distribution coefficient(Kd), and the differences between K and Kd's were to be increased with increasing equillibrium concentrations.

Effects of Alachlor and Endosulfan on the Survival and Malformation of Bombina orientalis Embryos (제초제 Alachlor과 살충제 Endosulfan이 무당개구리 (Bombina orientalis) 배아의 생존 및 기형유발에 미치는 영향)

  • The chloroacetanilide herbicide alachlor (2-chloro-2', 6'-diethyl-N-(methoxymethyl)-acetanilide) and organochlorine insecticide endosulfan (6, 7, 8, 9, 10, 10-hexachloro-l, 5, 5a, 6, 9, 9a-hexahydro-6, 9-methano-2, 3, 4-benzodioxathiepin-3-oxide)are the highly toxic agricultural chemicals. Bombina orientalis is one of the most common amphibians in the world and comprises a large proportion of their total number. B. orientalis spawns in the farming regions at Spring when the massive application of agricultural chemicals occurs. These chemicals in farmland may threaten the reproduction of this frog. Therefore, we examined the embryotoxic and survival effects of alachlor and endosulfan at various concentrations in B. orientalis embryos. The survival rates of embryos at 312h post fertilization treated with alachlor and endosulfan were decreased with concentration dependent manner. Also, developmental malformations appeared by alachlor and endosulfan in B. orientalis embryos. The malformations showed in order of frequency with bent trunk, tail dysplasia, bent tail, thick-set body and ventral blister in alachlor treated embryos. The exposure of endosulfan produced 7 types of severe external malformations with tail dysplasia, pectoral blister, bent trunk, bent tail, cephalic dysplasia, ventral blister, and thick-set body. Following exposure to alachlor and endosulfan the types of malformations were diverse, suggesting these chemicals target multiple events in embryonic and larval development in this species. These results suggested that alachlor and endosulfan were detrimental for survival and development of B. orientalis embryos.

  • The effects of alachlor [2-chloro-2', 6' diethyl-N-(methoxymethyl) acetanilide] treatment on nucleic acid, amino acid and protein synthesis were studied. The amide herbicide alachlor blocks the biosynthesis of the amino acids isoleucine, valine and aromatic amino acid in oat root tips. Nucleic acid was inhibited, but was not proportional to reduction in protein synthesis. $1{\times}10^{-4}M$ of alachlor treatment of oat roots inhibited 36% DNA synthesis, but DNA synthesis was not inhibited at $1{\times}10^{-5}M$. RNA synthesis was inhibited by $1{\times}10^{-5}M$ and $1{\times}10^{-4}M$ of alachlor 16 and 27%, respectively, while inhibition of protein synthesis did occur at same concentrations. Inhibition of protein synthesis also did not occur at concentration below $1{\times}10^{-4}M$ alachlor. It suggest that inhibition of protein sythesis caused significantly by alachlor($1{\times}l0^{-3}M$) result from secondary action.

Tolerance of Corn, Sorghum, Sorghum-Sudangrass Hybrid, and Pearl Millet to Simazine and Alachlor (옥수수, 수수, 수수-수단그라스 교잡종 및 진주조의 Simazine과 Alachlor에 대한 저항성)

  • In 1988 the tolerance of corn, sorghum, sorghum-sudangrass. and pearl millet hybrids to simazine and alachlor was tested in field during the growing season and pots during the summer and fall. In field and summer pot experiments(Exp.) the above mentioned four crops were tested at the ratios of simazine WP (50% ai, g/10a) ; alachlor EC (43.7% ai. ml/10a) of 130: 0, 100: 0, 70: 200, 0: 300 and 0: 400 and a sorghum hybrid was tested at 0, 50, 100, 200, 300, and 400ml/10a of alachlor and 70g/10a of simazine+ 200ml/10a of alachlor in fall pot Exp. In corn emergence rate, percent stand, plant height of seedlings, and dry matter(DM) yield were not affected by simazine and alachlor in all Exps. In sorghum and sorghum-sudangrass early growth and DM yield were not affected by simazine and alachlor in field Exp. In contrast, simazine reduced height and dry weight of seedlings slightly without any deterimental effects on emergence and survival rates. but alachlor reduced survival rate, plant height, and dry weight of seedlings significantly in summer pot Exp. In fall Exp. alachlor did not affect emergence rate of a sorghum hybrid, but survival rate, plant height, and dry weight of seedlings reduced with increased levels of alachlor when applied higher than 100ml/10a. In pearl millet simazine did not affect emergence rate, plant height, and DM yield in field, but reduced survival rate, plant height, and dry weight of seedlings in summer pot Exp. However, alachlor reduced DM yield significantly due to a lower percent stand even in the field. In summer pot Exp. although emeregence rate was slightly reduced, all seedlings were dead after emergence. Simazine did not control grasses such as Digitaria sanguinalis, Setaria viridis, Echinochloa crusgalli effectively, but controlled broadleaf weeds. Alachlor controlled all grasses, Porluraca oleracea, and Amaranthus mangostanus, but did not control Acalypha australis and Chenopodium album. A combination of simazine and alachlor controlled weeds more effectively than either simazine or alachlor alone.

Thyroid Hormone-like Activity of Alachlor as R Endocrine Disruptor in Rats and HeLaTRE Cell Culture (랫드와 HeLaTRE Cell에서의 Alachlor에 의한 갑상선 호르몬성 영향 연구)

  • This study was designed to investigate the suitability of the pubertal assay and the enhanced TG 407 as methods for detection of endocrine-mediated effects, especially thyroid function. Male and female Sprague-Dawley rats were gavaged daily with 0, 12.5, 25, 50 mg/kg alachlor in corn oil during 30 days. The effects of alachlor on thyroid gland, the genital organs and thyroid hormone were measured in male and female rats. Dose of alachlor 25, 50 mg/kg/day increased relative weight of testis and thyroid gland in exposed male rats and decreased relative weight of vagina in exposed female rats. Relative weight of thyroid gland was decreased in alachlor 25 mg/kg/day exposed female rats. Dose of alachlor 25, 50 mg/kg/day decreased plasma T4 and testosterone in female rats. Another purpose of this study was to investigate the effects of endocrine disruptors as like thyroid hormone in vitro. Luciferase activity was measured to dectect reaction of test chemicals and thyroid hormone response elements in HeLaTRE cell. Dose of alachlor 1 nM-1000 nM increased 100-134% luciferase activity compared with control.
