  • In a comparison the Logistics system for Agro-food of Korea and the Logistic system of Europe and Japan, the more developed countries of Europe have better systems. This well-formed rationalization of Logistics on Agro-food, and perfectly-equipped packaging system measurements for Agro-food make 100% pallet freight possible. Also, this means that a unit load system has been achieved. In Japan, the Logistic system is mainly accomplished by auction. However, it has been rapidly changing into the same type as the developed countries these days. Logistic system in Korea is also mainly accomplished by auction under the influence of Japan. However it also has been rapidly changing in to the same type as the developed countries these days. In the future, Korea has to build up international competitiveness in the face of FTA(Free Trade Agreement) era. This can happen by having better-organized Agro-food Logistics system and unit load system which results in accurate packaging system measurements.

  • It is necessary to construct the supply system which is stable and quality assured for professional performance of agro-food export. The purpose of this study is to present the basic information about the improvement directions on the Leading Group of Agro-food Export. The result of analysis is presented by the achievement of business objective, farm-organization and large scale-up, marketing achievement, and the fidelity of group operations etc.

  • Japanese agriculture has faced to several threats: aging and decrease of farmer population, global competition, and the risk of climate change as well as harsh and variable weather. On the other hands, the number of large scale farms is increasing, because farm lands have been being aggregated to fewer numbers of farms. Cost cutting, development of efficient ways to manage complicatedly scattered farm lands, maintaining yield and quality under variable weather conditions, are required to adapt to changing environments. Information and communications technology (ICT) would contribute to solve such problems and to create innovative technologies. Thus we have been developing an early warning and decision support system to reduce weather and climate risks for rice, wheat and soybean production in Japan. The concept and prototype of the system will be shown. The system consists of a weather data system (Agro-Meteorological Grid Square Data System, AMGSDS), decision support contents where information is automatically created by crop models and delivers information to users via internet. AMGSDS combines JMA's Automated Meteorological Data Acquisition System (AMeDAS) data, numerical weather forecast data and normal values, for all of Japan with about 1km Grid Square throughout years. Our climate-smart system provides information on the prediction of crop phenology, created with weather forecast data and crop phenology models, as an important function. The system also makes recommendations for crop management, such as nitrogen-topdressing, suitable harvest time, water control, pesticide spray. We are also developing methods to perform risk analysis on weather-related damage to crop production. For example, we have developed an algorism to determine the best transplanting date in rice under a given environment, using the results of multi-year simulation, in order to answer the question "when is the best transplanting date to minimize yield loss, to avoid low temperature damage and to avoid high temperature damage?".

  • The mcr-1 gene is a new "superbug" gene discoverd in China in 2016 that makes bacteria highly resistant to the last-resort class of antibiotics. The mcr-1 gene raised serious concern about its possible global dissemination and spread. Here, we report a potential anti-resistant strategy using the CRISPR/Cas9-mediated approach that can efficiently induce mcr-1 gene knockout in Escherichia coli. Our findings suggested that using the CRISPR/Cas9 system to knock out the resistance gene mcr-1 might be a potential anti-resistant strategy. Bovine myeloid antimicrobial peptide-27 could help deliver plasmid pCas::mcr targeting specific DNA sequences of the mcr-1 gene into microbial populations.

Antioxidant Activity and Protective Effects of Anthocyanins-Rich Fraction from Korean Purple Sweet Potato Variety, "Shinjami" against Oxidative Stress in HepG2 Cell (자색고구마 신자미로부터 분리한 안토시아닌 분획물의 항산화 활성과 산화스트레스에 대한 간세포 보호 효과)

  • Anthocyanins, a class of flavonoids, are natural water-soluble pigments, which are mainly found in vegetables and fruits. Anthocyanins have attractive pharmacological activities, such as anti-oxidant, anti-inflammatory, anti-cancer, and anti-diabetic. The purpose of this study was to investigate the protective effects of anthocyanins-rich fraction (ANF) from Korean purple sweet potato variety, "Shinjami", against hydrogen peroxide ($H_2O_2$)-induced oxidative stress. In our results, pre-treatment of HepG2 cells with ANF ($10{\mu}g/mL$) significantly prevented cell death and maintained cell integrity, following exposure to 0.9 mM hydrogen peroxide. The $H_2O_2$-dependent production of intracellular ROS was also significantly decreased by pre-treatment with ANF (6 h, $10{\mu}g/mL$). In addition, ANF increased the mRNA levels of antioxidant enzymes, catalase and glutathione level in $H_2O_2$-treated HepG2 cells. These results indicated that ANF protected HepG2 cells against $H_2O_2$-induced oxidative stress by inducing protective system.

  • The purpose of this study was to identify the candidate genes involved in selenium content in brown rice. Rice (Oryza sativa L.) was important crop including diverse functional substance such as carbohydrate, protein, lysine and tocopherol, mineral. Especially, selenium as nutritionally important minerals, it was known to activate the immune system, antioxidant effect and inhibition of carcinogenesis. Also recommended daily requirements of the United States and the United Kingdom were 55 to 90 ug for selenium. Therefore, selenium content in brown rice of core-set were analyzed by using ICP-MS (Inductively Coupled Plasma Mass Spectrometer) and GWAS (Genome Wide Association Study) was conducted to search for candidate genes in this study. The new natural variants identified through haplotyping analysis would be useful to develop new rice varieties with improved storage ability of the valuable mineral through the future molecular breeding.

  • This study examined the regulation status on wastewater treatment in agro-food processing and a case study on effluent treatment m ethods was carried out to evaluate any change after the mitigation of regulation. First, in order to clarify the area of investigation, the definition of small-scale agro-processing facilities was reviewed through literature survey and local government ordinance. The current law were separately analyzed into four areas; effluent treatment facilities, development of agro-processing industries, land use and food processes equipment. The exclusion clauses on wastewater discharging facility in the enforcement regulation were defined in detail, which can be served in practice. Site survey, after the questionnaire survey of the person in charge of the local unit, was carried out. As the result, this survey confirmed the positive effects of the deregulation on promoting sewer system service in rural areas, introducing the new processing construction and so on. In addition, it was found that some matters to be considered to determine whether to introduce wastewater treatment plan for public food processing facilities.

Evaluation of Microbiological Safety of Lettuce and Cultivation Area (상추와 생산환경의 미생물 안전성 평가)

  • Produce, including leafy vegetables, has been implicated in several outbreaks of food illness. To evaluate microbiological safety of lettuce and it's cultivation area, a total of 147 samples were collected from lettuce farms and post harvest facility at Icheon, Gyeonggi province. The collected samples were assessed for presence of sanitary indicator microorganisms (Aerobic plate count, coliform count, Escherichia coli) and foodborne pathogens (Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus). The population of APC was over 4.0 log CFU from most of the samples. While the numbers of APC, and coliform of lettuce at 62 days after transplanting were 4.18 log CFU/g, and 1.00 log CFU/g, respectively, those of 10 days after transplanting were 5.37 log CFU/g, and 2.87 log CFU/g, respectively. B. cereus was highly detected from soil and balance which were contaminated with 3.5 log CFU/g, and 2.6 log CFU/100 $cm^2$, respectively. The number of E. coli recovered from gloves was 3.5 log CFU/hand. However, E. coli O157:H7, Salmonella spp., and L. monocytogenes were not detected. These data suggested that risk management system should be introduced to lettuce farms to enhance safety of lettuce.

Expression of Codon Optimized β2-Adrenergic Receptor in Sf9 Insect Cells for Multianalyte Detection of β-Agonist Residues in Pork

  • ${\beta}_2$-adrenergic receptor (${\beta}_2-AR$) was expressed efficiently using Bac-to-Bac Baculovirus Expression System in Sf9 cells as a bio-recognition element for multianalyte screening of ${\beta}$-agonist residues in pork. Sf9 cells were selected as the expression system, and codon optimization of wild-type nucleic acid sequence and time-dependent screening of expression conditions were then carried out for enhancing expression level and biological activity. Under optimum conditions of multiplicity of infection (MOI) = 5 and 48 h post transfection, the protein yield was up to 1.23 mg/ml. After purification by chromatographic techniques, the purified recombinant protein was applied to develop a direct competitive enzyme-linked receptor assay (ELRA) and the efficiency and reliability of the assay was determined. The IC50 values of clenbuterol, salbutamol, and ractopamine were 28.36, 50.70, and $59.57{\mu}g/l$, and clenbuterol showed 47.61% and 55.94% cross-reactivities with ractopamine and salbutamol, respectively. The limit of detection (LOD) was $3.2{\mu}g/l$ and the relevant recoveries in pork samples were in the range of 73.0-91.2%, 69.4-84.6%, and 63.7-80.2%, respectively. The results showed that it had better performance compared with other present nonradioactive receptorbased assays, indicating that the genetically modified ${\beta}_2-AR$ would have great application potential in detection of ${\beta}$-agonist residues.

The Effect of Age on the Myosin Thermal Stability and Gel Quality of Beijing Duck Breast

  • The effect of age (22, 30, 38, and 46 days) on Beijing duck breast myosin gels was investigated. The results showed that the water holding capacity (WHC) and gel strength were markedly improved at the age of 30 days. Differential scanning calorimetry suggested that the myosin thermal ability increased at the age of 30 and 38 days (p<0.05). A compact myosin gel network with thin cross-linked strands and small regular cavities formed at the age of 30 days, which was resulted from the higher content of hydrophobic interactions and disulfide bonds. Moreover, the surface hydrophobicity of myosin extracted from a 30-day-old duck breast decreased significantly under temperature higher than 80℃ (p<0.05). This study illustrated that myosin extracted from a 30-day-old duck's breast enhanced and stabilized the WHC, thermal stability and molecular forces within the gel system. It concluded that age is an essential influencing factor on the myosin thermal stability and gel quality of Beijing duck due to the transformation of fibrils with different myosin character.
