The Study on Production and Performance of Crossbred Korean Native Chickens (KNC) (토종 순종계를 이용한 토종닭 생산 및 생산성 연구)

  • The current work was carried out to investigate the effect of crossbred Korean native chickens (KNC) on performance and carcass ratio. Seven hundred twenty 1-d-old chicks were divided into groups by strain (A, B, C and D) and sex (male and female). Strains were A) (KNC egg-meat type C strains $\times$ KNC meat type S strains) $\times$ KNC meat type H strains, B) (KNC egg-meat type C strains $\times$ KNC meat type H strains) $\times$ KNC meat type S strains, C) (KNC native R strains $\times$ KNC meat type S strains) $\times$ KNC meat type H strains and D) (KNC native L strains $\times$ KNC meat type H strains $\times$ Ross broiler. Experimental diets consisted of 3 phases such as starter(0~5 weeks; CP 20.0%, ME 3,050 kcal/kg), earlier (5~8 weeks; CP 18.0%, ME 3,100 kcal/kg) and finisher (8~12 weeks; CP 16.0%, ME 3,150 kcal/kg). Body weight (BW) and feed intake (FI) was measured every week and carcass ratio(CR) was calculated at 5 and 10 week after starting experiment. There was no difference in BW among strains until 5 weeks (P>0.05), however D strain resulted in a higher BW after 5 weeks (P<0.05). Body weight gain (BWG) and FI in D strain were also significant higher compared to the other strains for all periods. However, D strains showed the lowest (P<0.05) fee conversion ratio (FCR). The other strains except D showed a similar BW, BWG, FI and FCR among strains. In addition, there were no differences in carcass weight (CW) and carcass ratio (CR) among strains at 5 weeks, however D strain showed higher CW and CR at 10 weeks. These results suggested the basic data that needed to develope the new strains.

Chemical Composition and Meat Quality of Crossbred Korean Native Chickens (KNC) (교배 조합 토종닭의 이화학적 성상 및 육질 특성)

  • This work was carried out to investigate chemical composition and meat quality of crossbred Korean native chickens (KNC). Ninety 1-d male chicks were used in this work and were divided into 4 groups as A: (KNC egg-meat type C strains $\times$ KNC meat type S strains) (female) $\times$ KNC meat type H strains (male), B: (KNC egg-meat type C strains $\times$ KNC meat type H strains) (female) $\times$ KNC meat type S strains (male), C: (KNC native R strains $\times$ KNC meat type S strains) (female) $\times$ KNC meat type H strains (male), D: (KNC native L strains $\times$ KNC meat type H strains) (female) $\times$ Ross broiler (male) strains. They were fed the broiler diets for 12 weeks at the flat house and twenty seven chickens were slaughtered at week 5 and 10. Chicken thigh and breast were weighed and physicochemical compositions and sensory characteristics were investigated. Physical compositions of meats have no difference among strains at 5 week, and B strains differed from other strains at 10 week (P<0.05). The pH, moisture, and collagen content of meat from B strains were lower than other strains at 5 week. Ash and collagen of A strains were the lowest at 10 week (P<0.05), but others have no difference among strains. In sensory test, juiciness of D strains was the highest at 5 week, and tenderness of B strains was the lowest at 10 week (P<0.05). In conclusion, the crossbreeding of KNC did not affect physical traits but affected chemical composition of the chicken meat slaughtered at 5 week.

Microbial and Pathogenic Contamination of Ready-to-eat Fresh Vegetables in Korea (한국에 유통중인 신선편이 채소류의 미생물 품질 및 병원성 세균의 오염도 조사)

  • The purpose of this study was to evaluate microbiological contamination of fresh vegetables in Korea. Twenty types of vegetables were tested for total aerobic bacteria, coliforms, Escherichia coli, yeast and mold, and pathogenic bacteria such as Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus, Salmonella, E. coli O157:H7, Cronobacter sakazakii, Shigella, and Campylobacter. Levels of total aerobic bacteria and coliform on 20 vegetables were between 3.74 and 8.04 log CFU/g, and 0.16 and 5.02 log CFU/g, respectively. The highest contamination levels of total aerobic bacteria were observed on water dropwort, sprouts, mungbean sprout, and ballflower root. There was no significant difference (p>0.05) in microbial contamination levels of total aerobic count, coliform, E. coli, yeast and mold between organic and nonorganic vegetables. When isolation methods using selective agars were applied, L. monocytogenes, B. cereus, Salmonella and Campylobacter were isolated from some fresh vegetable samples. Results of API kit tests showed that L. monocytogenes was identified on Chinese cabbage, cucumber, soybean sprouts, and iceberg lettuce while Salmonella was identified on Korean leek. Furthermore, Campylobacter jejuni was also identified in more than 50 of the 100 samples. However, when positive samples from API kit were tested for real-time PCR or 16S rRNA sequencing method, only B. cereus from perilla leaf, carrot, water dropwort, and sprouts showed positive results. These results indicate that selective agar and API kit detection methods might result in false positive results for some pathogens. Therefore, studies need to improve isolation or confirmation methods for such pathogens.

Varietal Difference of Oil Content and Omega Fatty Acid omposi tion in Korea Local Perilla (들깨 지방수집종의 기름함량 및 오메가 지방산 조성 차이)

  • This study was conducted to obtain basic information for breeding materials on the oil quality improvement of perilla seeds. Oil contents and omega(${\omega}$)fatty acid compositions of seeds were investigated to 317 varieties of domestic perillas. Oil contents of 317 perilla varieties ranged from 35.6 to 55.3 with 44.5% of varietal mean. The major omega fatty acids contained in the oil were oleic acid(${\omega}$-9) 15.2%, linoleic acid(${\omega}$-36) 13.9% and linolenic acid(${\omega}$-3) 63.1% in the mean value. Varietal variation of $\omega$-9, 6 and 3 fatty acids ranged of 9.5~21.4%, 9.1~20.4% and 50.6~70.5% respectively. Unsaturated fatty acid were averaged 92.2% of seed oil in fatty acid composition. The ratios of ${\omega}$-6 to ${\omega}$-3 ranged of 0.13~0.34%(0.22% in mean value). The highest linolenic acid variety was Yecheonjong being 70.5%. The lowest variety in rations of ${\omega}$-6 to ${\omega}$-3 was Goseongjong being 0.13%. Oil content showed positive correlation with stearic acid and linolenic acid, while the negative correlation with oil content and linoleic acid. On the other hand, A significant negative correlation were showed between lionolenic acid and the ratios ${\omega}$-6 / ${\omega}$-3 fatty acid, saturated fatty acid. Saturated fatty acid was highly correlated with unsaturated fatty acid negatively being r=-0.723$^{**}$

Genetic Diversity and Relationship by SSR Markers of Korean Soybean Cultivars (한국 콩 육성품종의 SSR마커에 의한 유전적 다양성과 유연관계)

  • Genetic diversity of 91 Korean soybean cultivars was assessed with 20 simple sequence repeat (SSR). Twenty SSR loci generated a total of 149 alleles. The number of alleles for each SSR locus ranged from 3 to 15 with a mean of 7.5 alleles. Genetic diversity estimated by PIC value of 91 cultivars was ranged from 0.424 to 0.905 with an average of 0.711. Cluster analysis based on Nei's genetic distances classified 91 soybean cultivars except Geomjeongkong 4 into 7 groups. The majority groups were I, IV, and VI which included 26, 24, and 18 cultivars, respectively. Obvious differences in genetic diversity appeared to be related with the released periods of cultivars and utilization type of cultivars, but not with breeding sites. Cultivars released in 1970's and in 1990's showed the lowest and the highest genetic diversities with 0.576 and 0.706, respectively. Soybean cultivars for vegetable and early maturity showed the lowest genetic diversity with 0.514, while those for soy sauce and tofu showed the highest genetic diversity with 0.691. Genetic distance between soybean cultivar groups developed before 1969 and during 1970's was the nearest, while genetic distance between those developed in 1970's and 1990's was the furthest. Cultivar group for vegetable and early maturity showed the furthest genetic distance with cultivar group for soy sauce and tofu, while it showed the nearest genetic distance with cultivar group for cooking with rice. Genetic distance was greater between soybean cultivar groups developed in Suwon and Iksan than between those developed in Milyang and Iksan.

Review of the study on the surfactant-induced foliar uptake of pesticide (계면활성제에 의해 유도되는 농약의 엽면 침투성 연구 현황)

  • Research trends in the measurement of foliar uptake of pesticides and the recently proposed action mechanism of the surfactant-induced uptake of pesticides were reviewed with the related reports and studies. Major techniques used in those fields are bioassay, radiotracer techniques with leaves or cuticular membrane. Recently, a new method using Congo Red as a tracer was proposed. The limiting factor in the pesticides uptake into leaves is the waxy layer which consists of the epicuticular and cuticular wax. Physico-chemical parameters such as molar volume, water solubility and partition coefficient of pesticides have limited influences on the pesticide uptake into leaves. Polydisperse ethoxylated fatty alcohol surfactants are well known as the good activator for many pesticides. It is now generally agreed that uptake activation is not related to the intrinsic surface active properties of surfactants such as surface activity, solvent property, humectancy and critical micelle concentration. Recent studies using ESR-spectroscopy revealed that the surfactants have an unspecific plasticising effect on the molecular structure of the wax and cuticular matrix, leading to increased mobilities of pesticides. Penetration of surfactants into waxy layer altered the pesticide mobility in wax and the partition coefficient of pesticide, and then the pesticides penetration into leaves was enhanced temporally. The enhancing effect of surfactant could be significantly different depending on the carbon number of aliphatic moiety and the number of ethoxy group in polyoxyethylene chain of surfactants. It is suggested that the rate of penetration of surfactants should have a significant relationship with the rate of penetration of pesticides.

Evaluation of Ventilation Systems in an Enclosed Nursery Pig House (무창자돈사의 환기시스템 정립 및 환기효율 평가)

  • An experiment was conducted to establish the most suitable ventilation system for the enclosed nursery pig house in Korea, comparing four different ventilation systems ; i) air enters through perforated ceiling and exhausts through chimney (NA), ii) air enters through perforated ceiling and exhausts through side walls (NB), iii) air enters through perforated ducts and exhausts through side walls (NC) and iv) air enters through perforated ducts and exhausts through chimney(ND). The experiment was carried out during winter and summer separately. The experimental pigs were weaned at fourteen days old in winter (December-February) and at twenty one days old in summer (June-August). The main results of the experiment are as follows : A preliminary experiment showed that in the NC system during summer, air can reach all the pig rooms in the house and the air flow rates of the upper, middle (1.2 m height of the room) and low (at the height of pig stature) parts of the room were measured at 7.0-8.08, over 0.5 and over 0.2 m/s, respectively, which flow rates were much higher(p$<$0.05) than those in other system. At the minimum ventilation efficiency during winter, air flow rates of upper, middle and low parts of the room equipped with the NC system were detected at over 1, less than 0.5 and around 0.07 m/s, respectively. It is concluded that the separated ventilation system air-entering through ducts is the most suitable for the ventilation system of the enclosed nursery pig house and the exhausting system through side walls is more efficient for ventilation than the system through roof. Furthermore, to sustain proper temperature and reduce energy waste as well as heat consumption, a future research should be carried out to develop the environmental control system in relation to developing a heat regulator.

Residual Characteristics of Fluquinconazole and Flusilazole in Tomatoes during Greenhouse Cultivation and Processing (토마토 중 fluquinconazole과 flusilazole의 포장 및 가공에 따른 잔류특성)

  • This study was carried out to investigate residue dissipation of fluquinconazole and flusilazole in field-sprayed tomatoes during greenhouse cultivation and processing. The test pesticide, fluquinconazole+flusilazole 8.5 (7+1.5)% SC, was sprayed onto the tomatoes growing in a greenhouse according to Korea preharvest intervals and then samples were collected on 0 (3 hours after spraying), 1, 3, 5 and 7 days after last application for decline test. For processing test, tomatoes collected at harvest on 5 day after last application were processed to puree and juice. Limits of quantitation of fluquinconazole and flusilazole were 0.005 mg/kg in both tomatoes and their processed products. Recoveries for validation of the analytical methods for fluquinconazole and flusilazole in tomatoes and their processed products ranged from 74.8 to 97.5%. Biological half-lives of fluquinconazole and flusilazole in tomatoes under greenhouse conditions found to be 5.2 and 6.4 days, respectively. Average persistent residue levels of fluquinconazole and flusilazole were 37.34 and 79.53% after washing, 8.95 and 28.75% in filtrates after boiling, 3.58 and 14.66% in puree, and 3.34 and 13.52% in juice, respectively. These results indicated that the test pesticide residues on tomatoes could be largely removed through washing and boiling.

Functional Activities of Cacao Nibs and Couvertures according to Process Conditions (카카오닙과 커버춰의 가공 조건에 따른 기능성 분석)

  • This study was conducted to provide information regarding changes in antioxidant activity in response to conching temperatures, conching times, and cacao mass content (CMC) in dark chocolate. The radical scavenging activities and functional components of cacao nibs were highest for raw cacao nib (R0) under all conditions. Moreover, antioxidant activities and functional compounds increased during roasting for 25 min. As the conching temperature increased, the radical scavenging activities and functional components increased. Quantitative analysis of major catechin-derived compounds by HPLC revealed that R0 had the highest value for other roasted cacao nibs in all aspects (P<0.05). The content of procyanidin B2, catechin, and epicatechin increased during roasting for 25 min. Finally, evaluation of couvertures revealed that procyanidin B1 content increased as conching time increased to 48 h, except for 70% CMC and conched at $60^{\circ}C$ (HH) and 70% CMC and conched at $50^{\circ}C$. Overall, HH48 contained the richest catechin-derived components.

Bionomics and a Lana Sampling Method of Coppery Wireworm, Selatosomus puncticollis (Motschulsky), (Coleoptera: Elateridae) in Potato Field (청동방아벌레(Selatosomus puncticollis Motschulsky)의 생태적 특성 및 감자포장내 유충밀도 조사법)

  • The occurrence pattern and bionomics of Selatosomus puncticollis (Motschulsky) were surveyed in three net house (6$\times$70m) in the field and in a laboratory ($20^{\circ}C$, RH$75\%$, L16/DB). Emergence of hibernated adults of S. puncricollis started from early May, reached peak at early June and diminished in late June. Occurrence rate to total number was $42.1\%$ in early June and $31.7\%$ in late May, when most of hibernated adults emerged at these periods. Adults started to oviposit from mid June, and eggs hatched from early July Larvae turned into pupae from mid July and emerged to adults from mid August. Egg-period was 23 days and pupal period was 21 days. It is estimated that preovipositional period and larval period were approximately 10 months and 30 months, respectively. Wireworms were distributed at the soil depth of 10-5 cm, $56.8\%$ in 1997 and $45.8\%$ in 1998. To establish bait techniques to attract wireworms in the soil, six baits: pieces of potato, carrot, and sweet potato, wheat grains, corns, and flour dough wrapped with gauze, were buried at 15 cm of soil depth, and collected after 5 and 10 days. The numbers of wireworms attracted by potato pieces, wheat grains and sweet potato pieces after 10 days were 1.8, 1.6, and 1.4/bait, respectively. Therefore, burying potato pieces at 15 cm of soil depth and collected after 10 days could be recommended as a wireworm baiting technique.
