The effects of black garlic (Allium satvium) extracts on lipid metabolism in rats fed a high fat diet

  • BACKGROUD/OBEJECTIVES: The mechanism of how black garlic effects lipid metabolism remains unsolved. Therefore, the objectives of this study were to determine the effects of black garlic on lipid profiles and the expression of related genes in rats fed a high fat diet. MATERIALS/METHODS: Thirty-two male Sqrague-Dawley rats aged 4 weeks were randomly divided into four groups (n=8) and fed the following diets for 5 weeks: normal food diet, (NF); a high-fat diet (HF); and a high-fat diet + 0.5% or 1.5% black garlic extract (HFBG0.5 or HFBG1.5). Body weights and blood biochemical parameters, including lipid profiles, and expressions of genes related to lipid metabolism were determined. RESULTS: Significant differences were observed in the final weights between the HFBG1.5 and HF groups. All blood biochemical parameters measured in the HFBG1.5 group showed significantly lower values than those in the HF group. Significant improvements of the plasama lipid profiles as well as fecal excretions of total lipids and triglyceride (TG) were also observed in the HFBG1.5 group, when compared to the HF diet group. There were significant differences in the levels of mRNA of sterol regulatory element binding protein-1c (SREBP-1c), acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS), and glucose-6-phosphate dehydrogenase (G6PDH) in the HFBG1.5 group compared to the HF group. In addition, the hepatic expression of (HMG-CoA) reductase and Acyl-CoA cholesterol acyltransferase (ACAT) mRNA was also significantly lower than the HF group. CONCLUSIONS: Consumption of black garlic extract lowers SREBP-1C mRNA expression, which causes downregulation of lipid and cholestrol metahbolism. As a result, the blood levels of total lipids, TG, and cholesterol were decreased.

숙성 흑마늘 추출액 첨가가 빵용 밀가루 반죽의 레올로지 특성에 미치는 영향 (Effects of Aged Black Garlic Extracts on the Rheology of Flour Dough)

  • 제빵용 밀가루인 강력분에 흑마늘 추출액을 0, 5, 10, 15% 첨가하여 반죽의 레올로지 특성으로 Falling number, RVA에 의한 호화 특성, Farinograph, Alveograph 등을 분석하였다. Falling number는 흑마늘 추출액의 첨가량이 증가함에 따라 대조구에 비하여 낮아졌으나 제빵적성의 범위에 있었다. 호화도 특성에서 흑마늘 추출액의 첨가량이 증가함에 따라 호화개시 온도, 최고점도, 유지강도, 최종점도, break down, set back 등 모두가 대조구에 비해 증가하였다. Farinograph 측정에서 흑마늘 추출액 첨가량이 증가할수록 반죽의 강도와 흡수율은 증가하였고, 반죽형성시간은 감소하였으며 안정도와 약화도는 급격히 감소하는 경향을 나타냈다. 반죽의 내성(MTI)은 대조구에 비해 흑마늘 추출액 첨가량이 증가할수록 증가하였으며, FQN은 대조구가 가장 높았다. Alveograph 측정에서 $P_{max}$는 대조구가 가장 높았고 L, G 값은 흑마늘 추출액 15% 첨가구가 가장 높았으며 W값은 15% 첨가구에서 가장 낮았다. 이상의 실험 결과 흑마늘 추출액 5%까지는 첨가가 가능하나 그 이상은 반죽의 레올로지 특성을 저하시켜 제빵 적성이 나빠질 것으로 생각된다.

Effects of garlic intake on cancer: a systematic review of randomized clinical trials and cohort studies

  • BACKGROUND/OBJECTIVES: Due to the rapid increase of global cancer incidence and mortality and a high level of interest in cancer prevention, a systematic review of garlic intake and cancer risk is needed. SUBJECTS/METHODS: We implemented a systematic review to examine the effects of varying levels of garlic intake on cancer. We conducted comprehensive literature searches in three electronic databases (MEDLINE, Embase, and Web of Science) for studies published between database inception and July or September of 2018. Two investigators independently screened abstracts and full-texts, extracted data, and assessed risk of bias (RoB). A total of one medium-quality randomized controlled trial (RCT) and 13 cohort studies graded as high RoB were included. RESULTS: The 1-year follow-up results from a RCT showed that a significant decrease in the number and size of colorectal adenomas among participants with colorectal adenomas who received high-dose aged garlic extract (AGE) compared with those who received low-dose AGE (P < 0.05). The results of prospective observational studies provided inconsistent associations of colorectal cancer risk with garlic supplements and garlic intake as food. CONCLUSIONS: In summary, the AGE was effective in reducing the number and magnitude of colorectal adenomas in one RCT, but there were inconsistent associations between garlic intake and colorectal cancer in cohort studies. Therefore, we could not draw a firm conclusion regarding the effects of garlic on cancer, because the current strength of evidence is inadequate due to a lack of number of high-quality RCTs.

  • In this study, we investigated the hepatoprotective effects of aged black garlic (ABG) in rodent models of liver injury. ABG inhibited carbon tetrachloride-induced elevation of aspartate transaminase (AST) and alanine transaminase (ALT), which are markers of hepatocellular damage, in SD rats. D-galactosamine-induced hepatocellular damage was also suppressed by ABG treatment. However, ABG does not affect the elevation of alkaline phosphatase (ALP), a marker of hepatobilliary damage, in rats treated with carbon tetrachloride or D-galactosamine. We also examined the effect of ABG on high-fat diet (HFD)-induced fatty liver and subsequent liver damage. ABG had no significant effect on body weight increase and plasma lipid profile in HFD-fed mice. However, HFD-induced increase in AST and ALT, but not ALP, was significantly suppressed by ABG treatment. These results demonstrate that ABG has hepatoprotective effects and suggest that ABG supplementation might be a good adjuvant therapy for the management of liver injury.

Independent beneficial effects of aged garlic extract intake with regular exercise on cardiovascular risk in postmenopausal women

  • The purpose of the study was to assess the effects of a 12 weeks aged garlic extract (AGE) regimen with regular exercise on cardiovascular disease (CVD) risk in postmenopausal women. A total of 30 postmenopausal women ($54.4{\pm}5.4$ years) were randomly divided into the following four groups: Placebo (Placebo; n = 6), AGE intake (AGEI; n = 8), exercise and placebo (Ex + Placebo; n = 8), exercise and AGE (Ex + AGE; n = 8) groups. The AGE group consume 80 mg per day, and exercise groups performed moderate exercise (aerobic and resistance) three times per week. After 12 weeks of treatment, body composition, lipid profile, and CVD risk factors were analyzed. Body weight was significantly decreased in AGEI, Ex + Placebo, and Ex + AGE groups compared to baseline. Body fat % was significantly decreased in the AGEI and Ex + Placebo groups. Body mass index (BMI) was significantly decreased in the AGEI, Ex + Placebo, and Ex + AGE groups. Fat-free mass was significantly decreased in the AGEI group. Total cholesterol (TC) was significantly lower in the Ex + Placebo compared to the Placebo group. AGE supplementation or exercise effectively reduced low-density lipoprotein (LDL-C). Triglyceride (TG) was significantly increased in the AGEI group. Malondialdehyde (MDA) levels were significantly decreased in the AGEI, Ex + Placebo, and Ex + AGE compared to the placebo group. AGE supplementation reduced homocysteine levels regardless of whether the women also exercised. The present results suggest that AGE supplementation reduces cardiovascular risk factors independently of exercise in postmenopausal women.

  • 본 연구에서는 흑마늘 열수 추출물(ABG)의 암세포 전이억제 효능을 검정하였다. AGS AGS 인체위암세포의 이동성과 침윤성을 억제하였으며, 이는 TER의 증가와 연관성이 있었다. 또한 ABG에 의한 AGS 위암세포의 침윤성 억제는 TIMPs 발현 증가에 의한 MMPs의 발현 및 활성 저하에 의한 것임을 알 수 있었다. 아울러 AGS 위암세포에서 과발현을 나타내는 TJ 단백질인 claudins의 발현 저하 등이 ABG의 항전이 효과에 연관되어 있음을 알 수 있었다. 이상의 결과는 흑마늘 열수 추출물이 단순한 암세포의 증식억제를 통한 암예방 및 항암활성 뿐만 아니라 암세포의 전이 또한 효과적으로 억제할 수 있음을 보여주며, 이와 연관된 보다 구체적인 분자세포생물학적 접근 및 in vivo 연구의 필요성이 요구됨을 의미한다.

S-Allyl Cysteine(SAC)이 제대혈 유래 중간엽 줄기세포 증식에 미치는 영향 (Effect of S-Allyl Cysteine(SAC) on the Proliferation of Umbilical Cord Blood(UCB)-derived Mesenchymal Stem Cells(MSCs))

  • To improve the growth of human mesenchymal stem cells(hMSCs) under general cell culture conditions(20% $O_2$ and 5% $CO_2$), we examined the effect of s-allylcysteine(SAC), which is known as an antioxidant and the main component of aged-garlic extract, on hydrogen peroxide-induced cellular stress in hMSCs. We found that SAC blocked hydrogen peroxideinduced cell death and cellular apoptosis, but that SAC did not improve the growth of hMSCs during short-term culture. To evaluate the protective effect of SAC, we examined the endogenous expression of the antioxidant enzymes catalase (CAT), superoxide dismutase(SOD), and glutathione peroxidase(Gpx) in hMSCs. Hydrogen peroxide was found to downregulate the expression of CAT, SOD, and Gpx at the protein level. However, in the pre-treatment group of SAC, SAC inhibited the hydrogen peroxide-induced down-regulation of CAT, SOD, and Gpx. Unfortunately, treatment with SAC alone did not induce the up-regulation of antioxidant enzymes and the cell proliferation of hMSCs. Surprisingly, SAC improved cell growth in a single cell level culture of hMSCs. These results indicate that SAC may be involved in the preservation of the self-renewal capacity of hMSCs. Taken together, SAC improves the proliferation of hMSCs via inhibition of oxidative-stress-induced cell apoptosis through regulation of antioxidant enzymes. In conclusion, SAC may be an indispensable component in an in vitro culture system of human MSCs for maintaining self-renewal and multipotent characterization of human MSCs.

S-benzyl-cysteine-mediated Cell Cycle Arrest and Apoptosis Involving Activation of Mitochondrial-dependent Caspase Cascade through the p53 Pathway in Human Gastric Cancer SGC-7901 Cells

  • S-benzyl-cysteine (SBC) is a structural analog of S-allylcysteine (SAC), which is one of the major water-soluble compounds in aged garlic extract. In this study, anticancer activities and the underlying mechanisms of SBC action were investigated and compared these with those of SAC using human gastric cancer SGC-7901 cells. SBC significantly suppressed the survival rate of SGC-7901 cells in a concentration- and time-dependent manner, and the inhibitory activities of SBC were stronger than those of SAC. Flow cytometry revealed that SBC induced G2-phase arrest and apoptosis in SGC-7901 cells. Typical apoptotic morphological changes were observed by Hoechst 33258 dye assay. SBC-treatment dramatically induced the dissipation of mitochondrial membrane potential (${\Delta}{\Psi}m$), and enhanced the enzymatic activities of caspase-9 and caspase-3 whilst hardly affecting caspase-8 activity. Furthermore, Western blotting indicated that SBC-induced apoptosis was accompanied by up-regulation of the expression of p53, Bax and the down-regulation of Bcl-2. Taken together, this study suggested that SBC exerts cytotoxic activity involving activation of mitochondrial-dependent apoptosis through p53 and Bax/Bcl-2 pathways in human gastric cancer SGC-7901 cells.

In vitro에서 항산화 효능이 있는 흑마늘 추출물의 MMP-2 및 MMP-9에 대한 활성 억제효과 (In vitro Inhibitory Effect of Aged Black Garlic Extract with Antioxidant Activity on MMP-2 and MMP-9 Related to Metastasis)

  • 지질, 단백질 및 DNA의 산화적 손상이 관절염, 간염, 위염, 대장염 및 치주 질환과 같은 만성 염증뿐만 아니라 암 전이에 관련되어 있다고 알려져 있다. 이러한 질환의 발생을 예방하기 위하여 독성이 없는 천연 화합물을 개발하는 것이 최근의 주요 연구 관심 대상이다. 산화적 스트레스와 관련 있는 DPPH radical, hydrogen peroxide, hydroxyl radical 및 과산화수소와 같은 활성산소에 대한 흑마늘(ABGE)의 소거능력이 연구되었다. 뿐만 아니라 TBARS assay를 이용하여 본 연구에서 사용된 산화방법으로 Fenton반응에 의하여 hydroxyl radical에 노출된 세포에서 ABGE의 항산화 효과도 조사되었다. ABGE는 활성산소종 중에서 특히 과산화수소에 대한 항산화 효능이 우수하였고 hydroxyl radical에 노출된 genomic DNA의 산화에 대한 보호 효과도 관찰되었다. 살아있는 세포에 대한 산화적 스트레스도 ABGE의 존재 하에서 억제되었다. 뿐만 아니라, 암전이와 관련 있는 MMP-2 및 MMP-9의 활성과 발현에 대한 ABGE의 효과를 gelatin zymography 및 western blot을 이용하여 조사하였다. ABGE는 PMA로 자극한 사람 섬유아육종세포로부터 분비된 MMP-2 및 MMP-9의 활성과 발현을 동시에 억제 하였으므로 암을 억제 할 수 있는 하나의 생리활성물질로 개발 될 수 있으리라 판단된다.

채취시기 및 가공방법에 따른 섬애약쑥의 이화학적 특성과 항산화활성 (Physicochemical characteristics and antioxidant activity of Sumaeyaksuk depending on harvest times and processing methods)

  • 경남 남해군에 자생하는 고유품종으로 품종보호 등록된 섬애약쑥의 채취시기와 건조방법에 따른 이화학적 특성과 항산화활성을 평가하였다. 섬애약쑥은 5, 6, 7월에 동일한 재배지에서 수확한 후 각각을 12일 동안 음건(SD) 및 $60^{\circ}C$에서 7일간 숙성한 다음 $90^{\circ}C$에서 220분간 저장하여(AD) 건조하였다. 전처리방법을 달리하여 건조한 섬애약쑥의 유리당을 분석한 결과 glucose만이 검출되었으며 함량은 $0.42{\pm}0.02{\sim}0.43{\pm}0.01g/100g$$0.41{\pm}0.02{\sim}0.47{\pm}0.04g/100g$이었다. 총 페놀화합물의 함량은 AD($1.29{\pm}0.08{\sim}2.90{\pm}0.08g/100g$)와 비교하여 SD($1.85{\pm}0.09{\sim}3.45{\pm}0.14g/100g$)에서 더 높았다. 물 추출물을 31.5, 62.5, 125, 250, $500{\mu}g/mL$의 농도로 제조하여 항산화활성을 평가한 결과 DPPH와 ABTS 라디칼 소거활성은 SD 5월 시료와 AD 7월 섬애약쑥 추출물의 활성이 가장 높았으며, FRAP도 이들 시료에서 여타 시료에 비해 유의적으로 활성이 높았다. ${\beta}$-Carotene에 대한 탈색 저해활성도 SD의 5월과 AD의 7월 시료에서 각각 $25.53{\pm}2.85{\sim}81.43{\pm}2.56%$, $35.98{\pm}2.22{\sim}79.00{\pm}1.42%$로 가장 활성이 높았다. 이상의 결과 섬애약쑥은 5월 채취하여 음건하였을 때와 7월에 채취하여 숙성가공하였을 때 유용성분의 섭취가 더 용이하며, 높은 항산화활성을 나타내어 기능성 식품 소재로 활용도가 높을 것으로 생각된다.
