Antibacterial Activities of Essential Oil from Zanthoxylum schinifolium Against Food-Borne Pathogens (산초 정유성분의 식중독균에 대한 항균 활성)

  • In this study, the antibacterial activities of essential oil from Zanthoxylum schinifolium against four Gram-positive bacteria and six Gram-negative bacteria were investigated. The antibacterial activity of the oils was determined using the agar-well diffusion assay, MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration). In particular, essential oil from Z. schinifolium showed higher antibacterial activity against Gram-positive bacteria than against Gram-negative bacteria. Essential oil from Z. schinifolium displayed large inhibition zones especially against Bacillus cereus (31 mm). At concentrations between 0 and $20\;{\mu}g/mL$ the oils showed an antibacterial effect against both Gram-negative and Gram-positive bacteria. The minimum inhibitory concentration (MIC) values against nine bacteria ranged from 1.25 to $5\;{\mu}g/mL$. The minimum bactericidal concentration (MBC) values against eight bacterial ranged from 2.5 to $20\;{\mu}g/mL$, except Shigella sonnei. Furthermore, our finding on the antibacterial activities of essential oils from Zanthoxylum schinifolium validated the use of this plant for medical purposes.

  • The amylase was0 incubated in a 9mm hole on the starch agar gel plate bored with a cork borer. When 0.1N-iodine solution sprayed on the plate, the formed uncolored zone were observed. An activity of amylase has been determined by measurement of diameter of the uncolored zone. We named this method 'cork borer method'. When amylase was incubated on the starch agar gel plate, the following points were obtained. 1) The optimum pH for the formation of the zone in case of amylases(Biotex, Spitase) which produced by Bacillus is neutrality and alkali, while that for Glucoamylase, Biodiase, and Mucorrennin which produced by Rhizopus and Mucor is from 5 to 7. 2) The diameter of the zone was increased by the incubation time and amylase activity. 3) The zone was easily formed at low level of starch concentration and was formed much bigger than at $50^{\circ}C\;than\;at\;10^{\circ}C$. From the above results, after malting the starch agar gel plate, keeping constant concentration of the starch, the measurement of amylase activity is in efficiency upon the constant reaction time and temperature.

  • An agar degrading bacterium, This strain was isolated from sea water in Jeju. The strain is Gram-negative, rod and strictly aerobe for growth. The identification bases on the 16S rRNA gene sequencing showed that the strain was closely related to the genus Agariovarans sp. and named Agariovorans sp. JA1. The strain grew on agar as a sole carbon and energy source and produced an extra cellular agarase. For the increase of agarase productivity, 0.5% agar, yeast extract and $NaNO_3$ were used as carbon, organic and inorganic nitrogen source, respectively. For effective production of agarase and growth, the pH, temperature and NaCl concentration were was pH 7, $25^{\circ}C$ $^{\sim}$ $30^{\circ}C$ and 2%, respectively.

Antibacterial Action against Food-Borne Pathogens by the Volatile Flavor of Essential Oil from Chrysanthemum morifolium Flower (국화 꽃 휘발성 향기성분의 식중독균에 대한 항균 작용)

  • The aim of this study was to investigate antibacterial activities of essential oil from C. morifolium against four Grampositive bacteria and six Gram-negative bacteria. The antibacterial activity of the oils was determined by agar-well diffusion assay, minimum inhibitory concentration(MIC), and minimum bactericidal concentration(MBC). Essential oil of C. morifolium had a large inhibition zones especially against Salmonella enterica(21 mm) and Bacillus cereus(19 mm). Essential oil of C. morifolium generally showed higher antibacterial activity against Gram-positive bacteria than Gram-negative bacteria. MIC of essential oil from C. morifolium was 5 ${\mu}g/m{\ell}$ against ten food-borne pathogens. MBC values were determined to be from 5 to 20 ${\mu}g/m{\ell}$ against eight bacteria except Salmonella choleraesuis and Listeria monocytogenes. Therefore, the essential oil of C. morifolium and its components have a potent antibacterial activity against food-borne pathogens, and is expected to be used as a novel food preservative.

Medium Composition and Growth Regulator on Organogenesis Platycodon grandiflorum (Jacq.) A. DC. with Yellow Green Petals ('녹색 꽃잎 도라지'의 기관분화에 미치는 배지조성 및 생장조절제의 영향)

  • This study was carried out to determine the optimal medium composition and growth regulators for the micropropagation of Platycodon grandiflorum (Jacq.) A. DC. Nodes containing yellow green petals were used as plant materials to execute the study. The best performance of adventitious root development was found in 1/4 strength of MS basal salt and the growth was satisfactory in the concentration of 1/2 MS medium. The best condition for adventitious root development and growth was observed in the higher concentration (5%) of sucrose and activated charcoal free 1/4MS medium respectively. Adventitious roots were developed at the controlled culture medium at pH 4.8 with a tendency of suppression with higher levels of pH. However, it was prevailed that the development and growth depended on the concentration of agar. The lower concentration of agar (0.4%) was performed better than that of higher concentration (1.2%), whereas the agar concentration (0.4%) showed the best performance for the development and growth of adventitious roots. For the development of shoots containing node, BA combined with IAA was more effective than kinetin with IAA or NAA. The highest shoot development (3.9 shoots per explant) was performed on MS medium supplemented with 0.1 mg/L BA and 0.5 mg/L IAA.

Effect of Some Factors Affecting Callus Formation of Red Clover (레드 클로버의 캘러스 유도에 미치는 몇가지 요인의 영향)

  • This experiment was conducted to determine factors affecting callus formation of some varieties of red clover. Growth regulators. basal medium, medium pH, agar concentration, and plant age for explant were investigated. The result obtained were as follows. As auxin source, 0.1 mg/ l picloram was found to be effective on callus formation. Callus formation was the highest on PC medium among several growth media. The callus produced on PC medium was more friable than those produced on the other media. The medium pH 5.8 gave the best response for callus formation. The effective seedling age for callus formatiom was around 5-7 days old. In agar concentration, 0.5-0.8%(WN) was suitable for callus formation.

Sutdies on Quality Characteristics of Jeju Mandarin Orange Jelly for the Aged (고령자용 감귤젤리의 품질 특성 연구)

  • This study was carried out to determine the quality characteristics of Jeju mandarin orange jelly with various gelling agent such as agar, ${\kappa}-carrageenan$ and gellan gum for the aged. The concentration of agar was 0.4-0.6% and that of ${\kappa}-carrageenan$ and gellan gum was 0.2-0.4%. The color value, gelling temperature, melting temperature, break down rate, textural properties and sensory acceptance test of Jeju mandarin orange jelly with various gelling agent were measured. Average age of the subjects for acceptance test was 78.23. Redness and yellowness of Jeju mandarin orange jelly with agar was lower than that with ${\kappa}-carrageenan$. The gelling and melting temperature of Jeju mandarin orange jelly with agar was lowest among the jellies. Break down rate of Jeju mandarin orange jelly with agar was highest among the jellies. Above results showed that the stability of Jeju mandarin orange jelly with agar was inferior than that with ${\kappa}-carrageenan$ and gellan gum. Hardness, adhesiveness and springiness of Jeju mandarin orange jelly with agar was lowest among the jellies and sensory acceptance of Jeju mandarin orange jelly with gellan gum was highest among them. Thus, gellan gum was appropriate gelling agent for the Jeju mandarin orange jelly with regard to the acceptability and the depression of sour taste in Jeju mandarin orange jelly could improve the acceptability for the aged.

Evaluation of Antimicrobial Activity of the Methanol Extracts from 8 Traditional Medicinal Plants

  • The methanol extract of 12 medicinal plants were evaluated for its antibacterial activity against Gram-positive (5 strains) and Gram-negative bacteria (10 strains) by assay for minimum inhibitory concentration (MIC) and minimum bacterial concentration (MBC). The antibacterial activity was determined by an agar dilution method (according to the guidelines of Clinical and Laboratory Standard Institute). All the compounds (12 extracts) of the 8 medicinal plants (leaf or root) were active against both Gram-negative and Gram-positive bacteria. Gram-negative showed a more potent action than Gram positive bacteria. The MIC concentrations were various ranged from $0.6\;{\mu}g/ml$ to $5000\;{\mu}g/ml$. The lowest MIC ($0.6\;{\mu}g/ml$) and MBC ($1.22\;{\mu}g/ml$) values were obtained with extract on 4 and 3 of the 15 microorganisms tested, respectively.

Stydies on the Selective Screening Method of Bifidobacteria Used in Yoghurt (호상 요구르트 제품에 이용되는 Bifidobacteria 의 선택적 검출 방법)

  • In fermentation of dairy products, bifidobacteria is used in conjunction with other lactic acid bacteria, such as L. acidophilus, L. bulgaricus and S. thermophilus, rendering the enumeration of bifidobacteria difficult. In order to develop optimum conditions for selective enumeration of bifidobacteria, we examined MIC of several antibiotics against various bifidobacteria and other lactic acid bacteria. The growth of L. acidophilus, L. bulgaricus and S. thermophilus were inhibited by lithium chloride at the concentration of less than 4 mg/ml, whereas growth inhibition of bifidobacteria occurred at concentrations over 6-10 mg/ml. Tetracycline and chloramphenicol were also found to selectively inhibit growth of other lactic acid bacteria at the concentration of 1-3 $\mu$g/ml. Addition of 6 mg/ml lithium chloride, 1 $\mu$g/ml, tetracycline or 3 $\mu$g/ml chloramphenicol to medium was found to be optimal for selective enumeration of bifidobacteria. By using these three inhibitory chemicals in the TPY medium, higher number of bifidobacteria were selectively isolated than with NPNL agar and LP agar.

Evaluation of Antifungal Activities of Nanoparticles against Cladosporium cladosporioides Spore Bioaerosols (Cladosporium cladosporioides 포자에 대한 나노입자의 항진균 특성 평가)

  • The antifungal activity of silver, copper, and titania nanoparticles against fungal spores was investigated. Cladosporium cladosporioides spores were aerosolized and sampled on a solid agar plate using an Anderson impactor. The solid agar plate contained different concentration of nanoparticles ranging from 0 to $500{\mu}g/mL$. Silver and copper nanoparticles were shown to be an effective antifungal agent, while titania nanoparticles were not. Antifungal activity of these effective nanoparticles appeared at $300{\mu}g/mL$ concentration.
