Anti-cancer Activity of Flavonoids from Aceriphyllum rossii (돌단풍(Aceriphyllum rossii)에서 분리한 플라보노이드의 항암활성)

  • The methanol extract from the aerial parts of Aceriphyllum rossii was fractionated into ethyl acetate, n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel column chromatography of EtOAc and n-BuOH layers afforded five flavonol glycosides. They were identified as astragalin (1), kaempferol 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (2), rutin (3), kaempferol 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$4)-${\alpha}$-L-rhamnopyranosyl 1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (4), and quercetin 3-O-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$4)-${\alpha}$-L-rhamnopyranosyl (1${\rightarrow}$6)-${\beta}$-D-glucopyranoside (5) on the basis of spectroscopic data. All of them showed an inhibition in farnesyl protein tranferase (FPTase) activity, and rutin (3) inhibited the growth of rat H-ras cell and the cell migration of human umbilical vein endothelial cells (HUVECs).

Antimicrobial Activities of Methanol Extracts Obtained from Several Ferns (양치식물류의 메탄올 추출물에 항균활성 분석)

  • Methanol extracts of the aerial and rhizome parts obtained from ten species of ferns has been screened for antimicrobial activities against Bacillus subtilis, Escherichia coli, Listeria monocytogenes, Propionibacterium acnes, Streptococcus mutans and Streptococcus sobrinus. Antimicrobial activities were carried out using broth microdilution method and paper disc diffusion assay and the extracts which showed clear zones more than 15mm in concentration of 2 mg/disc were tested for its antimicrobial activities at the $0.125{\sim}2\;mg{\cdot}mL^{-1}$ concentration of each extract for three days. The non-sterilized crude methanol extracts of Osmunda cinnamomea var. fokiensis rhizome showed the highest antimicrobial activities on B. subtilis(39%), E. coli (33%) and L. monocytogenes(58%) at the concentration of $2\;mg{\cdot}mL^{-1}$ after 72 hours. In P. acnes, frond extract of O. cinnamomea var. fokiensis showed most vigorous antimicrobial activities in the all extracts but it showed weak activity(clear zone diameters below 15 mm). All extracts has the antimicrobial activities on Streptococcus, but they exhibited weak activity. At the concentration of $2\;mg{\cdot}mL^{-1}$, only Osmunda japonica rhizome extracts showed 28 and 39% of antimicrobial activities on S. mutans and S. sobrinus after 72 hours and the other extracts showed below 10% of antimicrobial activities on S. mutans and S. sobrinus.

Gamma-ray Irradiation on Radio Sensitivity in Cnidium officinale Makino (천궁 돌연변이 유발을 위한 최적 감마선 조사량)

  • Background: Cnidium officinale Makino have been used in traditional medicine in Northeast Asia. Although gamma-ray mutagenesis has been used to develop breeding resources with novel characteristics, research on the radiation sensitivity of C. officinale Makino is limited. Hence, the optimal gamma-ray dosage for mutation breeding in C. officinale Makino was investigated. Methods and Results: Seedstocks were exposed to doses of gamma rays (5 Gy - 50 Gy), and subsequently planted in a greenhouse. After 30 days of sowing, the survival rates and growth decreased rapidly at doses above 20 Gy, while all individuals died at 50 Gy. The median lethal dose (LD50) was 25.65 Gy, and the median reduction doses (RD50) for plant height, number of stems, and fresh weight were 12.81, 9.32, and 23.26 Gy, respectively. Post-irradiation levels of malondialdehyde (MDA), peroxidase (POD), and chlorophyll in the aerial parts of the plant were quantified using spectrophotometry. Relative to the controls, the levels of MDA and POD increased, while the level of chlorophyll decreased at doses ≥ 10 Gy, indicating cellular damage. Conclusions: A dose of 20 Gy was found to be optimal for mutation breeding in C. officinale Makino.

Optimum Harvesting Time Based on Growth Characteristics of Four-year Ginseng (4년생 인삼에서 생육특성과 수확적기)

  • This study was carried out to find out the optimum harvesting time on the basis of physiological characteristics, yield and commercial value in four years old ginseng. For this purpose, the changes of agronomic growth characteristics of the aerial and underground parts with different growing stages were examined with fourth year ginseng. The leaf growth was dramatically increased from the mid-April (shooting stage) to the mid-May (flowering stage), and the growth was nearly completed by the mid-May, but the stem growth continued by the mid-August. The weight of fresh root decreased from the April (20.7g) to the June (18.2g), but increased from the June to the October(45. 1g). The yield of fresh root per "Kan" was 1.5 kg, 1.2kg and 1.3kg at the April, the May and the June, respectively, thereafter continually increased to 2.9 kg of the October, which showing the maximum yield. In conclusion, the ginseng root was generally harvested from the September to the October at farm-house, but it had better be harvested after the mid-October rather than from September to October considering the weight of dry root and the yield of fresh ginseng.h ginseng.

First Report of Stem Rot in Statice Caused by Rhizoctonia solani in Korea (Rhizoctonia solani에 의한 스타티스 줄기썩음병)

  • Stem rot of perennial statice (Limonium sinuatum) was observed in Un bong, Jeonbuk from 2006 to 2007. Affected plants were randomly distributed in the greenhouses and infection rate was more than 10%. Stem and leaf of statice at soil line were dried and turned brown, initially. As the disease became severe, other stem parts and crowns were turned dark brown and then sunken. The fungal isolates were showed initially white aerial mycelium and turned brown with age. They produced few sclerotia which small, irregularly shaped with pinpoint sized. Mycelia were branched at $90^{\circ}$ angles and multinucleate in one cell. The pathogenicity of causal organism was proved according to Koch's postulates. The causal fungus of stem rot was identified as Rhizoctonia solani based on the cultural and morphological characteristics. This is the first report on stem rot of statice by R. solani in Korea.

Effect of Sowing Density and Number of Seeds Sown on Panax ginseng C. A. Meyer Seedling Stands under Direct Sowing Cultivation in Blue Plastic Greenhouse (인삼 하우스 직파재배 시 파종입수 및 재식밀도가 입모율 향상에 미치는 영향)

  • This study was performed to investigate the effects of sowing density and number of seeds sown on the emergence rate and growth characters of Panax ginseng C. A. Meyer under direct sowing cultivation in a blue plastic greenhouse. Ginseng seedlings, derived from seeds sown directly at different densities (90, 108, 135, and 162 seeds per $162m^2$), were cultivated in sandy loam soil within a blue plastic greenhouse. In contrast to the emergence rate, which decreased with an increase of sowing density, number of survival plant showed an increasing trend. Interestingly, the emergence and number of survival plant were significantly enhanced when 2 or 3 seeds were sown per hole compared with when one seed was sown per hole. Growth of the aerial parts of ginseng were not markedly influenced by sowing density or the number of seeds sown. However, chlorophyll content (SPAD values) increased with an increase in sowing density. Root parameters, such as root length, diameter, and weight, and the number of lateral roots decreased with an increase in sowing density, but were not noticeably influenced by the number of seeds sown. Total saponin content was the highest in the treatment plot containing 135 seeds. Similarly, the content of each ginsenoside was also tended to be higher in this treatment than in other treatment plots. On the basis of the results obtained in this study, it was possible to determine the optimal sowing density and seed number for the direct sowing cultivation of ginseng in blue plastic greenhouse.

Composition of Essential Oil of Tagetes minuta L. (만수국아재비의 정유성분 조성)

  • The essential oils from the aerial parts of Tagetes minuta L. from two different locations in Korea were obtained by hydrodistillation in 0.52%(Goheung, Chonnam) and 0.48% yields(Puan, Chonbuk) as a dry weight base, respectively. A total of 69 components, accounting in 94.7% and 92.1% of the oil, respectively were identified by gas chromatography-mass spectrometric analysis and comparison of retention indices with those of authentic compounds. The main components in both samples(Goheung, Chonnam and Puan, Chonbuk) were limonene(5.83% and 6.41%, respectively), $cis-{\beta}-ocimene$ (4.87% and 47.73%), dihydrotagetone(14.78% and 52.83%), trans-tagetone(0.64% and 2.99%), cis-tagetone(1.13% and 2.50%), trans-tagetenone(1.15% and 11.45%) and cis-tagetenone(0.69% and 1.41 %). Although these components were present in both samples, the results showed large differences in the percentage composition of these components.

Comparison of Growth Characteristics and Active Ingredients in Atractylodes Inter-Specific Hybrid Cultivars (삽주 육성품종 간 생육특성 및 유효성분 분석)

  • Background: Atractylodes japonica Koidz. and Atractylodes macrocephala Koidz. belong to the family (Asteraceae). Their rhizomes, called white Atractylodes rhizomes, are used in traditional medicine. To address some issues with their cultivation, we recently developed eight hybrid cultivars by interspecific hybridization of A. japonica and A. macrocephala. This study was conducted to screen the hybrid cultivars that have high amounts of active ingredients and yield ability. Methods and Results: Experiments were conducted using the eight hybrid cultivars and A. macrocephala (control cultivar) in the experimental field of the department of Herbal Crop Research located in Eumseong, South Korea. We investigated the growth characteristics of the aerial and underground parts. Among the cultivars, 'Sanwon' had the highest rhizome dry weight (53.8 g/plant), followed by 'Dachul' (50.0 g/plant). In addition, the content of atractylenolide I, II, III and total active ingredients were investigated using high-performance liquid chromatography. Compared with A. macrocephala, most of the inter-specific hybrid cultivars had a higher content of active ingredients and yield ability. Conclusions: Through study, we established the superior quality of Atractylodes inter-specific hybrid cultivars. In particularly, it was found that 'Dachul' may be grown as a superior cultivar, with high amount of active ingredients as well as yield ability.

Changes in the Growth Characteristics and the Content of Decursin and Decursinol angelate in Angelica gigas Nakai by Mineral and Hormonal Agent Treatment (미네랄 및 호르몬 제제 처리에 따른 당귀의 생육특성 및 Decursin, Decursinol Angelate 함량 변화)

  • Background: Angelica gigas Nakai is a perennial herb belonging to the family Umbelliferae. Its roots are utilized in traditional medicine. The aim of this study was to increase the yield of and the content of two indicator components (decursin and decursinol angelate) in A. gigas Nakai. Methods and Results: The roots of A. gigas Nakai were harvested 4-times from late August to late October in 2017. Two agents (trace element-TE, and plant hormone-HM) were applied 4 times at intervals of 2 weeks. The content of the two indicator components were analyzed by high performance liquid chromatography. The HM treatment showed the greatest increase in underground part yield and root diameter. The content of the two indicator components in the control (non-treatment) group was the highest in the underground part, but was higher in the aerial parts in the agent treatment group. After treatment with the agents, the content of the indicator components tended to decrease in the underground part. However, the total content of the indicator components in the two agent treatment groups exceeded the level of 6% set by the Korean Pharmacopoeia. Conclusions: The highest underground part yield was found in the HM treatment group, while the highest content of decursin and decursinol angelate were found in the control group. This study provides basic information for yield improvement in A. gigas.

  • Sajabalssuk (Artemisia herba) was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc and n-BuOH fractions, four sterols were isolated through the repeated silica gel and ODS column chromatographies. From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the sterols were determined as ${\beta}-sitosterol$ (1), ergosterol peroxide (2), stigmasterol (3) and daucosterol (4). They were the first to be isolated from Sajabalssuk (Artemisia herba).
