Effect of Cytokinin and Putrescine on Plant Regeneration from Leaf Explant of Rhodiola sachalinesis A. Bor (홍경천 (Rhodiola sachalinensis A. Bor) 잎절편으로부터 식물체 재분화에 미치는 Cytokinin과 Putresine의 영향)

  • The effects of cytokinin and putrescine on adventitious shoot induction from leaf explant of Rhodiola sachalinensis A. Bor were investigated. Among cytokinin used in this experiment, BA was more effective on adventitious shoot induction and shoot elongation than kinetin. Especially, 1 mg/L BA was the best to increase adventitious shoot induction (71%) and shoot elongation (3.0 mm). In addition, 100 mM putrescine in MS medium with 1 mg/L BA was higher in adventitious shoot induction (93%) and shoot elongation (3.8 mm) than single treatment of 1 mg/L BA. Adventitious shoots induced in this experiment rooted on 1/2 MS medium and acclimated over 95% on composed soil (peatmoss:sand=1:1).

  • Xanthoceras sorbifolium Bunge (yellowhorn) is a woody tree in the soapberry family, Sapindaceae, native to northern China. This species has been identified as a major woody bioenergy plant for bio-diesel production because of high oil content in seed. But the flowers do not bear fruit well while the many flowers blooming. This study was performed to regenerate in vitro plantlet using adventitious shoot formation. To establish the protocol of plant regeneration, adventitious shoots formation rate in the culture of cotyledon of immature zygotic embryos was 68.6% in 1/2 MS medium with 0.5 mg l-1 BA and 3% sucrose (w/v). In the culture of cotyledons of mature zygotic embryos, induction of adventitious shoots was needed to contain high sucrose in pre-culture medium and the frequency of shoot induction was 64.4%. Multiple shoots were induced in 0.5 mg l-1 TDZ, and rooting of shoot was induced 4.0 mg l-1 IBA. Flow cytometry analysis revealed that all the regenerated plantlets were diploid.

  • Plant regeneration of Pulsatilla koreana was achieved via adventitious shoot formation indirectly from callus and directly from adventitious root segments. For the callus induction from leaf or petiole explants, combination of 2,4- dichlorophenoxyaceticacid (2,4-D) with $2.22\;{\mu}M$ 6-benzyladenine (BA) was effective. Adventitious shoot induction from callus was enhanced by the combined treatment with $0.1\;{\mu}M$ polyvinylpyrrolidone (PVP) compared to cytokinin treatment alone. Adventitious roots were induced from the petiole segments on 1/2 MS medium with $4.93\;{\mu}M$ IBA. High frequency direct adventitious shoot formation from the segments of adventitious roots was achieved on medium with $4.92\;{\mu}M$ 2-isopentenyladenine (2-ip). Elongated shoots were rooted on half-strength MS medium containing $5.71\;{\mu}M$ indole acetic acid (IAA). Regenerated plantlets with well-developed shoots and roots were successfully transferred to soil. This in vitro propagation protocol might be useful for mass propagation as well as conservation of this plant.

  • A simple, high-frequency and reproducible protocol for induction of adventitious shoot buds and plant regeneration from leaf-disc cultures of Jatropha curcas L. has been developed. Adventitious shoot buds were induced from very young leaf explants of in vitro germinated seedlings as well as mature field-grown plants cultured on Murashige and Skoog's (MS) medium supplemented with thidiazuron (TDZ) ($2.27{\mu}M$), 6-benzylaminopurine (BA) ($2.22{\mu}M$) and indole-3-butyric acid (IBA) ($0.49{\mu}M$). The presence of TDZ in the induction medium has greater influence on the induction of adventitious shoot buds, whereas BA in the absence of TDZ promoted callus induction rather than shoot buds. Induced shoot buds were multiplied and elongated into shoots following transfer to the MS medium supplemented with BA ($4.44{\mu}M$), kinetin (Kn) ($2.33{\mu}M$), indole-3-acetic acid (IAA) ($1.43{\mu}M$), and gibberellic acid ($GA_3$) ($0.72{\mu}M$). Well-developed shoots were rooted on MS medium supplemented with IBA ($0.5{\mu}M$) after 30 days. Regenerated plants after 2 months of acclimatization were successfully transferred to the field without visible morphological variation. This protocol might find use in mass production of true-to-type plants and in production of transgenic plants through Agrobacterium/biolistic-mediated transformation.

Factors influencing Adventitious Shoot induction from Leaves of Populus deltoides Bartr (미류나무 잎절편체 조직배양을 통한 식물체 재분화에 미치는 요인)

  • We have investigated several factors influencing adventitious shoot induction from the Populus deltodies bartr. leaf segments. To obtain in vitro materials, at first, stem segments from ex vivro were tested for axillary bud breaking on the five macronutrients levels of two differenr media. WPM was better both in bud breaking and leaf expansion than MS medium. The amount of NH$_{4}$NO$_{3}$ added in the medium did not seem to affect axillary bud breaking, significantly and subsequent shoot elongation from the stem segments of P. deltodies. However, other components in WPM might have played important roles in axillary bud breaking and shoot elongation. Regenerability from three sections (the distal, the middle and the base) of leaves cultured on WPM supplemented with TDZ and NAA combinations appeared to be different; middle and basal sections of leaves produced more organogenic sites than those of top section on WPM and those sesctions produced the highest ogranogenic sites on the same medium supplemented with 0.01 mg/l TDZ and 0.02mg.l NAA. Among 3 carbon sources tested for adventitious shoot elongation, fructose seemed to be stimulating the elongation of adventitious shoots. Sucrose and glucose added in the medium resulted in the necrosis which caused dying of adventitious shoots, eventually.

Influence of plant growth regulators on adventitious shoot formation of Glehnia littoralis Fr. Schmid (갯방풍의 기내부정아 형성에 미치는 식물생정조절물질의 영향)

  • Petiole explant of Glehnia littoralis Fr. Schmidt was in vitro cultured MS plant medium(DUCHEFA co.) supplemented with various plant growth regulators and examined to find out their optimum combination and concentration for plantlet regeneration. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on MS plant medium with various kinds of plant growth regulators. Embryogenic calli and adventitious shoot formation were greatly influenced by plant growth regulators. Embryogenic calli induction showed a good response on MS medium supplemented with NAA and BA than others. Especially, combination of 1.0 mg/L NAA and 0.5 mg/L BA on MS medium led to the greatest frequency in adventitious shoot. The results suggest that plant regulator selection be important factor to achieve an efficient regeneration.

  • To acquire the normal regeneration of plantlets, we investigated combinations and concentrations of plant growth regulations for optimal conditions of adventitious root formation. Based on the previous study, we performed callus and shoot induction. When induced shoot was transferred into a rooting medium containing plant hormones, it wilted and died. Thus, the shoot proliferated on 1/2 MS medium for 10 days and was then treated with MS medium supplemented with 3.0 mg/L NAA for 3 days. Adventitious root formations were observed after shoot planlets were transferred to 1/3 MS medium. The concentrations of salt and sucrose were gradually reduced in MS medium and the rooted plantlets were transferred for acclimatization into a mixture of peatmoos : perlite (3 : 2).

  • Micropropagation of Ulmus davidiana Planch was established via adventitious shoot formation from the segments of adventitious roots. Adventitious roots were produced directly from root segments of seedlings on a 1/2 SH medium plus various concentrations of IBA. The maximum growth of adventitious roots was observed in the presence of 2.0 mg/L IBA. After the segments of adventitious roots were cultured on various cytokinins (zeatin, 2-iP, BA, kinetin) and cytokinins plus auxin (IBA), formation of adventitious shoot was investigated. Among cytokinin treated, kinetin was the most effective on both adventitious shoot induction and number of shoots. Especially, 2.0 mg/L kinetin was the best to increase adventitious shoot induction (95.8%) and a number of shoots (8.4). Adventitious shoots were rooted on 1/2 WPM medium and the plantlets were acclimated 100% on composed soil (peatmoss : vermiculite 1 : 1).

Cytohistological Study of Development of Callus and Adventitious Shoots from Cultured Stem of Vigna radiata (녹두 줄기 조직배양에서 캘러스와 부정아 형성에 관한 세포조직학적 연구)

  • This study was carried out to establish a reproducible culture system for callus formation and adventitious shoot development from young stem segments of Vigna radinta, and histological work for orgin of callus tissue and adventitious shoot. Induction of callus from young stem explants of Vigna radiata was very effective on MS inorganic salts supplemented with 0.5 mg/L 2,4-D and 1.0 mg/L kinetin. For the adventitious shoot regeneration from the callus tissues, the hormone combination of 0.75 mg/L NAA, 1.5 mg/L kinetin and MS salts resulted in about 21% efficiency. Histological examination showed that callus tissues originated from out-growths by callus cambium rings with do novo meristematic activities, which were localized at the outside of the vascular cambium. Adventitious shoots were developed from shoot apical meristem originated from the surface of callus masses. The shoot apical meristem produced leaf primordium, which then became leaf.

Effects of BA and NAA on Adventitious Shoot Formation from Mature Zygotic Embryos of Stewartia koreana Nakai (노각나무 성숙 접합자 배로부터 부정아 유도에 미치는 BA 및 NAA 효과)

  • Zygotic embryos of Stewartia korean Nakai were cultured to determine the effects both of BA and NAA on adventitious shoot induction. Multiple shoots (about 11 per explant) were formed when the embryos were treated with 1.0 mg/L BA alone. On the other hand, NAA appeared to inhibit shoot induction when treated with BA. Adventitious shoots looked differ in form and color by the combinations of BA and NAA treated. When both BA and NAA were present, the shoots became short and developed dark color. The highest rooting was observed at 0.5mg/L NAA. The results could be useful for the establishment of in vitro regeneration system for Stewartia koreana Nakai.
