When the leaves, roots and stem segments of seedling of Polygonatum odoratum were cultured on Murashige and Skoog medium with 2.0mg/l BAP, stem segments were the most efficient explants for adventitious shoot inductino. To observe the efficient combination of growth regulators on the adventitious shoot formation , stem segments were cultured on MS medium with various kinds of cytokinins (BAP, kinetin, zeatin). From this experiment, cytokinin treatement was prerequisite for theadventitious shoot formatino,especially BAP was the most effective. Auxin (NAA or IBA) in combination with cyotokinin highly enhanced the adventitious shoot formation. Twenty five percents of explants produced the adventitious shoots on medium with 2.0mg/l BAP solely, while 83% of explants produced the adventitious shoots on medium with 2.0mg/l BAP and 0.1mg/l IBA. Root formationform adventitious shoot was promoted after transfer to 1/2 MS medium supplemented with 0.1mg/l IBA and 0.5mg/l zeatin, thereafter the plantlets with shoots and roots were cultured on 1/2MS medium lacking growth regulators. When the stem segments were cultured to MS medium with 1.0mg/l 2,4 NAA and IBA , yellow and nodulous cali were formed from the stem segments which were developed into adventitious roots. These roots were actively grew after transferred to MS liquid medium lacking growth regulators.
Korean ginseng(Panax Ginseng C.A. Meyer) known as a oriental miracle drug is an important medicinal plant. Ginseng has been used for geriatric, tonic, stomachic, and aphrodisiac treatments for thousands years. Also, it is an antibiotic and has therapeutic properties against stress and cancer. Ginseng is widely distributed all over the world. Among them, Korean mountain ginseng has the most valuable effect on pharmaceuticals. The roots of mountain ginseng contained several kinds of ginsenosides that have many active functions for the human body. However, the study of mountain ginseng has a limit because the mountain ginseng is very expensive and rare. So, we artificially cultured mountain ginseng adventitious roots using the bioreactor culture system. We induced callus from original mountain ginseng, directly dug up in mountain and aged about one hundred ten years. Separated adventitious roots were precultured in 500ml conical flasks and then, transferred in 20L bioreactors. The adventitious roots of mountain ginseng were harvested after culturing for 40days, dried and then, extracted with several solvents. In this study, we investigated the whitening effect, anti-wrinkle effect and the safety of tissue cultured adventitious roots extract of mountain ginseng in order to identify the merit as a cosmetic ingredient. Particularly, extract of mountain ginseng adventitious roots showed whitening and anti-wrinkle effects. The inhibitory effect of this extract on the melanogenesis was examined using B-16 melanoma cell. When B-16 melanoma cells were cultured with adventitious root extract, there was a dramatically decrease in melanin contents of 8-16 melanoma cell. And we identified this extract inhibited Dopa auto-oxidation significantly. Also, when transformed mouse fibroblast L929 cells were treated with this extract, there was a significant increase in collagen synthesis. The results show significant inhibited melanization and wrinkle without inhibiting cell viability.
The effect of auxins and cytokinins on the formation of adventitious shoots, adventitious roots, trichomes, and calli in MS basal medium was investigated in leaf segments from ecotype Columbia of Arabidopsis thaliana. Adventitious shoots, adventitious roots, trichome, and calli were formed from leaf segments by a wide range of hormone concentrations and combinations. Adventitious shoots were formed respectively in treatment with 0.1mg/L IAA and 10 mg/L BA. Adventitious roots were formed in treatments with low concentration of IAA and NAA. Trichomes and calli were formed by increasing the concentration of IAA and NAA. The optimal combination was 0.5mg/L NAA and 0.1mg/L BA for trichome formation, 10mg/L NAA and 10mg/L BA for calli formation. When NAA was treated alone in culture media, adventitious roots were formed in 0.1mg/L, trichomes were formed in 2.0mg/L, and calli were formed in 10mg/L. Inductive time for formation of adventitious roots, trichomes and calli were determined at 6,7 and 18 days respectively by periodical transfer of leaf segments from NAA containing medium to NAA free medium.
In order to investigate the effects of elicitors on the growth and ginsenosides biosynthesis of ginseng adventitious roots, chitosan and jasmonic acid were treated with various concentrations. The growth rate of adventitious roots was increased with the addition of chitosan at higher concentrations (10 mg/L), but the best accumulation of ginsenosides was observed at the lower concentration (5 mg/L). Jasmonic acid was an effective elicitor for ginsenosides biosynthesis in ginseng adventitious roots. The maximum accumulation of ginsenosides was observed at the treatment of 10 uM jasmonic acid. But the jasmonic acid was found to decrease the growth rate of adventitious roots.

Korea mountain ginseng known as oriental miracle drug is an important medicinal plant. The effect of mountain ginseng adventitious roots extract has been described. The valuable root of mountain ginseng contained several kinds of ginsenosides that have been confirmed to have many active functions for the human body. However, the study of mountain ginseng has a limit because the price of wild ginseng is very expensive and rare. The mountain ginseng adventitious roots were derived from mountain ginseng callus that were induced from mountain ginseng roots. Adventitious roots were separated from callus and grown in solid media(Murachige and stoog media). It was cultured in a 20L bioreactor. After culturing for 40days, adventitious roots were harvested. Afterwards the harvested mountain ginseng adventitious roots were dryed and extracted. We examined the effect on melanogenesis of mountain ginseng adventitious roots extract. Here, we report the inhibitory effect of melanin biosynthesis on the adventitious roots extract of In vitro test. Also, we assessed the safety of adventitious roots extract. In vitro, cytotoxicity of adventitious roots extract was assessed in mouse fibroblast using two method: The neutral red uptake assay and the MTT assay. In vivo, the allergic and irritant were Patch teated in 30 patients. Consequently, extract of mountain ginseng adventitious roots have inhibitory effect on melanin biosynthesis in B-16 melanoma cell test, tyrosinase inhibitory test and DOPA auto-oxidation test. There were decreased 86%(0.5% concentration), 45%(1% concentration) and 61%(1% concentration), respectively.
The potency of adventitious root initiation was investigated in relation with germination days and ike end of culture days healed with root induction medium on the initiation of adventitious roots from explants of soybean (Glycine max L.) cotyledon. Also, the changes in the pattern of protein profiles related with adventitious mot initiation have been examined by two-dimensional polyauyl-amide gel elecoophoresis during the germination and the initiation of adventitious roots. The potency of adventitious root initiation from germinated cotyledons was high after 4 days, and adventitious roots were initiated from after 4 days and very high after 6 days healed with mot induction medium There was a reproducible quantitative change in 34 specific protein spots during germination and adventitious root initiation. A necessary basic protein for adventitious root initiation with rotative molecular weight of 27 kD was induced during the germination. And three basic protein groups with relative molecular weight 22 kD, 23 kD and 33 kD, and two acidic protein groups with relative molecular weight 27kD and 29 kD were induced during the initiation of adventitious roots.
Dendropanax morbifera (Araliaceae) is an endemic species in Korea and distributed in the southern part of Korea. The roots and stems of this plant have been used for folk medicine for the treatment of migraine headache, dysmenorrheal, and remove wind dampness and for Vanishes production. Production of adventitious roots in D. morbifera by in vitro cultures could be used as alternatives materials. Leaf, stem, and root segments from D. morbifera seedling were cultured on Murashige and Skoog (MS) medium supplemented with 3.0 mg/L IBA and 30 g/L sucrose. After 4 weeks of culture, the highest induction of adventitious roots was obtained from the leaf segment. Frequency of adventitious root formation on medium with various kinds of auxins (IAA, NAA, 2,4-D, and IBA) and various concentrations of IBA (0, 0.1, 0.5, 1.0, 3.0, and 5.0 mg/L) was tested. The maximum induction of adventitious root was obtained on medium with 1.0 mg/L IBA. In liquid culture, growth of root was best 1/2MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose. Adventitious roots were cultured in 5 L bioreactor containing 1/2 MS medium supplemented with 1.0 mg/L IBA and 30 g/L sucrose and mass-production of adventitious roots was successfully achieved. This study demonstrated for the first time to produce adventitious roots in D. morbifera.
In order to survey anthraquinone productivity from in vitro root culture, transformed hairy roots of Rumex crispus were induced from leaf segments by infection with Agrobacterium rhizogenes strain $A_4$ and compared with adventitious roots. The optimum condition of adventitious root formation from leaf segments was 5 $\mu$M NAA added to MS medium. Mannopine was detected in the extract of hairy roots by paper electrophoresis, but not in adventitious roots. Secondary root tips of both adventitious roots and hairy roots elongated without lateral root branching in hormone free MS medium, but primary root tips showed more rapid growth with extensive lateral root branching. MS basal medium was the best for growth of the adventious roots and hairy roots for anthraquinone content. Adventitious root tips and hairy root tips cultured in liquid MS medium supplemented with 0.05 $\mu$M NAA and 0.1 $\mu$M kinetin (contained 5% sucrose) showed the maximal growth and anthraquinone content. Anthraquinone content of hairy roots was increased by the culture periods, but was reduced after 25 days of culture.
An adventitious root formation protocol from Smilax china L. was established for in vitro production of dioscin, a steroidal saponin having various bioactivities such as anticancer, antifungal, antiviral, and antiobesity. Optimal medium for root initiation from leaf explant was MS medium containing $30\;g{\cdot}L^{-1}$ of sucrose supplemented with $1.0\;mg{\cdot}L^{-1}$ kinetin + $2.0\;mg{\cdot}L^{-1}$ NAA. The induction of adventitious roots from in vitro initiated root segments was most favorable to MS liquid medium with $0.1\;mg{\cdot}L^{-1}$ kinetin + $2.0\;mg{\cdot}L^{-1}$ NAA. Among the 20 different adventitious roots originated from different plants, strain No. 10 was selected based on production ability of dioscin, and its stability through the successive suspension culture. The maximum growth stage of adventitious roots was noticed at 5 weeks after subculture while that of dioscin production in the adventitious root was at 7 weeks after subculture in suspension culture system. These results provide that suspension culture of adventitious roots of Smilax china L. have a potential for in vitro mass production of dioscin.
Adventitious root culture was established in the Jawahar variety of Withania somnifera using MS basal medium supplemented with 0.5 (mg/l) IAA and 2.0 (mg/l) IBA. Root tips from germinated seedlings, MS0 maintained plants and adventitious roots were maintained in suspension medium (1/2 MS basal medium supplemented with 3% sucrose) for a period of 1 to 6 months. The weight gain in roots was noted and the withanolides were extracted from the dry roots using solvents petroleum ether, 50% ethanol and chloroform. The withanolides in the chloroform fractions of all root samples analyzed were compared using thin layer chromatographic analysis. Withanolide content in adventitious root sample was found to be superior compared to other roots at any given point of time during the 6month growth period.HPLC analysis of in vitro adventitious roots showed the presence of a new compound.
