Combining the Power of Advanced Proteome-wide Sample Preparation Methods and Mass Spectrometry for defining the RNA-Protein Interactions

  • Emerging evidence has shown that RNA-binding proteins (RBPs) dynamically regulate all aspects of RNA in cells and involve in major biological processes of RNA, including splicing, modification, transport, transcription and degradation. RBPs, as powerful and versatile regulatory molecule, are essential to maintain cellular homeostasis. Perturbation of RNA-protein interactions and aberration of RBPs function is associated with diverse diseases, such as cancer, autoimmune disease, and neurological disorders. Therefore, it is crucial to systematically investigate the RNA-binding proteome for understanding interactions of RNA with proteins. Thanks to the development of the mass spectrometry, a variety of proteome-wide methods have been explored to define comprehensively RNA-protein interactions in recent years and thereby contributed to speeding up the study of RNA biology. In this review, we systematically described these methods and summarized the advantages and disadvantages of each method.

  • Tumors of the brain are the deadliest, with a life expectancy of only a few years for those with the most advanced forms. Diagnosing a brain tumor is critical to developing a treatment plan to help patients with the disease live longer. A misdiagnosis of brain tumors will lead to incorrect medical treatment, decreasing a patient's chance of survival. Radiologists classify brain tumors via biopsy, which takes a long time. As a result, the doctor will need an automatic classification system to identify brain tumors. Image classification is one application of the deep learning method in computer vision. One of the deep learning's most powerful algorithms is the convolutional neural network (CNN). This paper will introduce a novel deep learning structure and image gradient to classify brain tumors. Meningioma, glioma, and pituitary tumors are the three most popular forms of brain cancer represented in the Figshare dataset, which contains 3,064 T1-weighted brain images from 233 patients. According to the numerical results, our method is more accurate than other approaches.

  • This paper aimed to summarize the current situation of prognostication for patients with an expected survival of weeks or months, and to clarify future research priorities. Prognostic information is essential for patients, their families, and medical professionals to make end-of-life decisions. The clinician's prediction of survival is often used, but this may be inaccurate and optimistic. Many prognostic tools, such as the Palliative Performance Scale, Palliative Prognostic Index, Palliative Prognostic Score, and Prognosis in Palliative Care Study, have been developed and validated to reduce the inaccuracy of the clinician's prediction of survival. To date, there is no consensus on the most appropriate method of comparing tools that use different formats to predict survival. Therefore, the feasibility of using prognostic scales in clinical practice and the information wanted by the end users can determine the appropriate prognostic tool to use. We propose four major themes for further prognostication research: (1) functional prognosis, (2) outcomes of prognostic communication, (3) artificial intelligence, and (4) education for clinicians.

  • Exposure to ultraviolet (UV) light is often associated with skin damage, sometimes very serious, and in recent times has received particular attention as a health risk. As a result, the proper use of sunscreen has long been recommended to protect against skin damage. The continued increase in the use of sunscreen may be linked to increased information about the risk of melanoma and non-melanoma skin cancer caused by prolonged exposure to ultraviolet rays. Natural and harmless materials that block and prevent UV light have emerged as essential household items in the field of skin beauty. New materials need to be considered and evaluated in relation to ultraviolet rays and their harmful effects. This study aims to explain the effect of UV exposure on human skin, the classification of sunscreens, the application of zeolite, nano clay, and LDH in sunscreen formulations, as well as the regulation of this service in various countries around the world.

  • Although radiation and chemotherapy methods for cancer therapy have advanced significantly, surgical resection is still recommended for most cancers. Therefore, intraoperative imaging studies have emerged as a surgical tool for identifying tumor margins. Intraoperative imaging has been examined using conventional imaging devices, such as optical near-infrared probes, gamma probes, and ultrasound devices. However, each modality has its limitations, such as depth penetration and spatial resolution. To overcome these limitations, hybrid imaging modalities and tracer studies are being developed. In a previous study, a multi-modal laparoscope with silicon photo-multiplier (SiPM)-based gamma detection acquired a 1 s interval gamma image. However, improvements in the near-infrared fluorophore (NIRF) signal intensity and gamma image central defects are needed to further evaluate the usefulness of multi-modal systems. In this study, an attempt was made to change the NIRF image acquisition method and the SiPM-based gamma detector to improve the source detection ability and reduce the image acquisition time. The performance of the multi-modal system using a complementary metal oxide semiconductor and modified SiPM gamma detector was evaluated in a phantom test. In future studies, a multi-modal system will be further optimized for pilot preclinical studies.

췌장 선암의 절제 가능성 평가 (Radiologic Evaluation for Resectability of Pancreatic Adenocarcinoma)

  • 췌장 선암의 발견, 진단, 절제 가능성 평가, 병기 설정 및 치료 방침 결정에 영상 검사는 중요한 역할을 담당하고 있다. 최근에는 진단 당시 경계절제성 혹은 국소 진행성 췌장암을 진단 받은 경우 선행 보조 치료 또는 완화 치료 후 가능하면 근치적 절제술을 고려할 것이 권고된다. 이 종설에서는 췌장 선암의 절제 가능성 평가, 원격 전이의 진단 및 선행 보조 치료 또는 완화 치료 후 절제 가능성을 재평가할 때 영상 검사의 해석, 판독 기준 및 고려할 사항에 대하여 설명하고자 한다.

Confocal Laser Endomicroscopy in the Diagnosis of Biliary and Pancreatic Disorders: A Systematic Analysis

  • Background/Aims: Endoscopic visualization of the microscopic anatomy can facilitate the real-time diagnosis of pancreatobiliary disorders and provide guidance for treatment. This study aimed to review the technique, image classification, and diagnostic performance of confocal laser endomicroscopy (CLE). Methods: We conducted a systematic review of CLE in pancreatic and biliary ducts of humans, and have provided a narrative of the technique, image classification, diagnostic performance, ongoing research, and limitations. Results: Probe-based CLE differentiates malignant from benign biliary strictures (sensitivity, ≥89%; specificity, ≥61%). Needle-based CLE differentiates mucinous from non-mucinous pancreatic cysts (sensitivity, 59%; specificity, ≥94%) and identifies dysplasia. Pancreatitis may develop in 2-7% of pancreatic cyst cases. Needle-based CLE has potential applications in adenocarcinoma, neuroendocrine tumors, and pancreatitis (chronic or autoimmune). Costs, catheter lifespan, endoscopist training, and interobserver variability are challenges for routine utilization. Conclusions: CLE reveals microscopic pancreatobiliary system anatomy with adequate specificity and sensitivity. Reducing costs and simplifying image interpretation will promote utilization by advanced endoscopists.

Stilbenes contribute to the anticancer effects of Rheum undulatum L. through activation of apoptosis

  • Rheum undulatum L. (R. undulatum) is a medicinal plant used for the treatment of inflammatory diseases in East Asian countries. Numerous stilbenes isolated from R. undulatum have been revealed to possess anticancer effects. The aim of the present study was to evaluate the effect of extracts and compounds isolated from R. undulatum on human gastric cancer cell viability and to elucidate their molecular mechanism of action on the apoptosis pathway. The results demonstrated that aloe-emodin and chrysophanol 1-O-β-D-glucopyranoside, isolated from the methanolic extract of dried rhizomes of R. undulatum, exhibited anti-proliferative effects on the human gastric carcinoma cell line AGS, with IC50 values of 84.66±0.44 and 68.28±0.29 µM, respectively. The percentage of apoptotic cells increased significantly following treatment with each compound at a concentration of 100 µM, compared with that in the non-treated group in the image-based cytometry assay. Western blot analysis revealed that these compounds activated the caspase cascade and inhibited B-cell lymphoma-2, an anti-apoptotic protein.

End-of-Life Care for End-stage Heart Failure Patients

  • Efforts to improve end-of-life (EOL) care have generally been focused on cancer patients, but high-quality EOL care is also important for patients with other serious medical illnesses including heart failure (HF). Recent HF guidelines offer more clinical considerations for palliative care including EOL care than ever before. Because HF patients can experience rapid, unexpected clinical deterioration or sudden death throughout the disease trajectory, choosing an appropriate time to discuss issues such as advance directives or hospice can be challenging in real clinical situations. Therefore, EOL issues should be discussed early. Conversations are important for understanding patient and family expectations and developing mutually agreed goals of care. In particular, high-quality communication with patient and family through a multidisciplinary team is necessary to define patient-centered goals of care and establish treatment based on goals. Control of symptoms such as dyspnea, pain, anxiety/depression, fatigue, nausea, anorexia, and altered mental status throughout the dying process is an important issue that is often overlooked. When quality-of-life outweighs expanding quantity-of-life, the transition to EOL care should be considered. Advanced care planning including resuscitation (i.e., do-not resuscitate order), device deactivation, site for last days and bereavement support for the family should focus on ensuring a good death and be reviewed regularly. It is essential to ensure that treatment for all HF patients incorporates discussions about the overall goals of care and individual patient preferences at both the EOL and sudden changes in health status. In this review, we focus on EOL care for end-stage HF patients.

엑스선 조사처리된 신선편의 채소류의 미생물학적, 이화학적, 관능적 품질 평가 (Microbiological, physicochemical, and organoleptic evaluation of fresh-cut vegetables irradiated using X-rays)

  • 본 연구는 면역저하 환자용 신선편의 채소류의 위생화를 위해 당근, 오이고추, 방울토마토, 파프리카를 모델식품으로 선정하여 다양한 흡수선량으로 엑스선 조사처리한 후 미생물학적, 이화학적, 관능적 품질 평가를 수행하였다. 당근과 오이고추의 경우 0.4 kGy, 방울토마토와 파프리카의 경우 0.2 kGy 이상의 흡수선량으로 엑스선 조사처리 시 멸균이 확인되었으며, 총 호기성세균 및 대장균군을 제외한 병원균들은 1 log 수준의 검출한계 이하로 나타났다. 모델식품에 접종된 E. coli. L. monocytogenes, S. Typhimyrium에 대한 엑스선의 D10값은 0.11-0.32 kGy로 나타나, 0.4 kGy의 흡수선량을 신선편의 채소류의 멸균을 위한 잠정적인 최소 흡수선량으로 설정하였다. 최대 0.6 kGy의 흡수선량으로 엑스선 조사처리된 신선편의 채소류에 대한 전체 선호도의 경우 7점 척도에서 모두 5.0 점 이상으로 나타나 이들의 관능적 품질이 수용 가능한 수준인 것으로 판단하였다. 또한, 암환자를 대상으로 엑스선 멸균처리(0.4 kGy)된 신선편의 채소류에 대한 선호도 설문조사에서 오이고추와 방울토마토, 파프리카의 경우 모든 항목에 대한 평균 선호도 점수는 5점 척도에서 모두 4.0점 이상으로 나타났다. 따라서 엑스선으로 멸균처리된 오이고추, 방울토마토, 파프리카는 면역력이 저하된 환자들을 위한 위생학적으로 안전한 신선편의 채소로 제공이 가능할 것으로 판단된다.
