기혈관절방(氣血關節方)A(CWS-A)가 CFA로 유발(誘發)된 관절염(關節炎)에 미치는 영향(影響) (The effect of Gihyeolgwanjeolbang-A(CWS-A) on adjuvant-induced arthritis in rats)

  • Objective : To investigate effects of herbal preparations formulated(Gihyeolgwanjeolbang-A, CWS-A) on complete Freund's adjuvant (CFA)-induced arthritis in rats. Method : Arthritis was induced by injecting CFA subcutaneously into the left knee joint and paw, and the herbal preparations formulated(CWS-A I, 82.3 mg/kg ; CWS-A II, 164.6 mg/kg) was administered orally (i.g.) for 10 consecutive days beginning on day 10 after the injection. External shape, paw edema, inflammatory cytokines tumor necrosis factor alpha (TNF-alpha) and interleukin-1 beta (IL-1beta), and histological observation were assessed. Result : In swelling of the paw, CWS-A I and CWS-A II group in 15 days and 20 days were significantly reduced compared to controls. In serum ALT, CWS-A I and CWS-A II group were significantly reduced compared to controls. In TNF-${\alpha}$, CWS-A I and CWS-A II group were a tendency reduced compared to controls. In HGB, HCT, MCHC of erythrocytes, CWS-A II group was increased compared to controls. In histological observations, CWS-A II group was observed synoviocytes more than control group and was observed proteoglycans in the deep layers. Conclusion : The data suggest that CWS-A significant anti-arthritic effects that may be mediated by suppressing inflammatory parameters.

Gene Expression Profile of T-cell Receptors in the Synovium, Peripheral Blood, and Thymus during the Initial Phase of Collagen-induced Arthritis

  • Background: Current management strategies attempt to diagnose rheumatoid arthritis (RA) at an early stage. Transcription profiling is applied in the search for biomarkers for detecting early-stage disease. Even though gene profiling has been reported using several animal models of RA, most studies were performed after the development of active arthritis, and conducted only on the peripheral blood and joint. Therefore, we investigated gene expression during the initial phase of collagen-induced arthritis (CIA) before the arthritic features developed in the thymus in addition to the peripheral blood and synovium. Methods: For gene expression analysis using cDNA microarray technology, samples of thymus, blood, and synovium were collected from CIA, rats immunized only with type II collagen (Cll), rats immunized only with adjuvant, and unimmunized rats on days 4 and 9 after the first immunization. Arrays were scanned with an Illumina bead array. Results: Of the 21,910 genes in the array, 1,243 genes were differentially expressed at least 2-fold change in various organs of CIA compared to controls. Among the 1,243 genes, 8 encode T-cell receptors (TCRs), including CD3${\zeta}$, CD3${\delta}$, CD3${\varepsilon}$, CD8${\alpha}$, and CD8${\beta}$ genes, which were down-regulated in CIA. The synovium was the organ in which the genes were differentially expressed between CIA and control group, and no difference were found in the thymus and blood. Further, we determined that the differential expression was affected by adjuvant more than Cll. The differential expression of genes as revealed by real-time RT-PCR, was in agreement with the microarray data. Conclusion: This study provides evidence that the genes encoding TCRs including CD3${\zeta}$, CD3${\delta}$, CD3${\varepsilon}$, CD8${\alpha}$, and CD8${\beta}$ genes were down-regulated during the initial phase of CIA in the synovium of CIA. In addition, adjuvant played a greater role in the down-regulation of the CD3 complex compared to CII. Therefore, the down-regulation of TCR gene expression occurred dominantly by adjuvant could be involved in the pathogenesis of the early stage at CIA.

강황(薑黃)약침이 CFA로 유발된 류마티스성 관절염에 미치는 영향 (Effects of Curcuma longa $L_{INNE}$ Pharmacopuncture at $ST_{36}$ on Adjuvant Induced Arthritis in Rats)

  • Objectives : The objective of this study was to investigate the effects of Curcuma longa $L_{INNE}$ pharmacopuncture at $ST_{36}$ on Complete Freund's Adjuvant(CFA)-induced arthritis in rats. Materials and methods : Arthritis was induced by injecting CFA subcutaneously into the left knee joint and paw, and Curcuma longa $L_{INNE}$ pharmacopuncture(CLL-A. $0.0343{\mu}g/kg$; CLL-B. $0.171{\mu}g/kg$; CLL-C. $0.343{\mu}g/kg$) was injected at $ST_{36}$ each other day for 5 times beginning on day 10 after the CFA injection. Paw edema, withdrawal response, hematological, serological and histological observation were assessed. Results : In paw edema volume all 3 groups(CLL-A, CLL-B, CLL-C) showed significant decrease compared to the CFA control group. In withdrawal response to reaction time and withdrawal response to force all 3 groups(CLL-A, CLL-B, CLL-C) showed significant increase compared to the CFA control group. In serum AST, group CLL-C showed significant decrease compared to the CFA control group. In histological observations, in all 3 groups, more normal chondrocytes were observed compared to the CFA control group and safranin O stain showed high positive reaction in the cartilage tissue close to the bone tissue. Conclusions : The results suggest that Curcuma longa $L_{INNE}$ Pharmacopuncture at $ST_{36}$ has a suppressing inflammation effect on Freund's adjuvant arthritis in rats.

금주사액약침(金注射液藥鍼)의 Adjuvant 관절염 유발 흰쥐의 진통효과(鎭痛效果) 및 그 기전(機轉)에 관한 연구 (The Experimental Study on the Analgesic Effect and Mechanism of Gold Injection Aqua-acupuncture to Adjuvant Induced Arthritis in Rats)

  • Objective: In this study, the analgesic effect of gold injection aqua-acupuncture on CFA induced arthritis in rats was inverstigated. Furthermore, to determine the antinociceptive mechanism of gold injection aqua-acupuncture, naloxone and yohimbine were administrated intraperitoneally 15min prior to the gold injection aqua-acupuncture. Methods: The experimental groups were divided into 6 groups. Control group, NSI, AMI, AMI+SL : AMI+Nx, AMI+Yb. Thereafter we measured the chages of TFL between control and experimental groups. Results: 1. The tail flick latency induced by CFA was significantly increased on AMI as compared with the NSI group. 2. Effect of sodium aurothiomalate aqua-acupuncture in TFL on CFA was decreased after application of naloxone. The anti-nociceptive effect of AMI was completely antagonized by naloxone(an opioid receptor antagonist). 3. Effect of sodium aurothiomalate aqua-acupuncture in TFL on CFA was decreased after application of yohimbine. The anti-nociceptive effect of AMI was completely antagonized by yohimbine(${\alpha}2$-adrenoceptor antagonist). Conclusions: According to the results, gold injection aqua-acupuncture has significant analgesic effects on adjuvant induced arthritis in rats and the effect of anti-nociception was mediated by opiodergic system and ${\alpha}2$-adrenergic system.

$^{99m}Technetium-methylene$ Diphosphonate의 전신 골격주사술을 이용한 관절염의 핵의학적 진단 (Scintigraphic Detection of Adjuvant-induced Arthritis in Rats Using $^{99m}Technetium-methylene$ Diphosphonate)

  • 본 연구는 랫드에서 Freund's complete adjuvant를 랫드의 후지에 투여하여 인의적으로 유발된 관절염을 진단하기 위한 방법으로 $^{99m}Technetium-MDP (^{99m}Tc_MDP$)을 이용한 골 스캔의 임상적 의의를 찾고자 하였다. 성숙 Sprague-Dawley 암컷 랫드를 대조군과 실험군으로 분리하고, 골의 스캔은 0.05mCi의 $^{99m}Tc_MDP$를 정맥 주사하여 3시간 후에 전신 영상을 얻었다. 골의 스캔과 방사선학적 평가를 비교한 겨과 관절염 유발 7일 후부터 관절염의 병변이 관찰되었지만, 방사선학적 소견에서는 14일 후부터 병변이 관찰되었다. 골의 스캔에서 관절염 유발 7일 후에 유의적으로 골의 이상적 섭취 증각가 관찰되었고(p<0.05), 21일 후에는 전지와 후지의 모든 부위에서 골의 이상적 섭취 증가가 관찰되었지만, 방사선학적 소견에서는 후지에서만 병변 부위가 관찰되었다. 이상의 결과에서 $^{99m}Tc_MDP$을 이용한 골의 스캔은 류마티스 관절염을 임상적으로 진단하는데 방사선학적 진단보다 효과적인 방법임을 알 수가 있었고, 골의 스캔은 류마티스 관절염을 초기에 진단하는데 유효한 진단방법임을 확인하였다.

Adjuvant에 의해 유발된 관절염 백서모델에 있어서 대방풍탕의 진통효과 (Analgesic Effect of Daebangpung-tang on a Rat Model of Adjuvant-induced Arthritis)

  • Daebangpung-tang(DBPT) is one of the prescriptions used for the treatment of rheumatoid arthritis(RA) in oriental medicine. The present study aimed to examine the analgesic effect of DBPT on a rat model of CFA-induced arthritis, which is not identical to human auto-immune arthritis although it does have many features in common with RA, and the relation between DBPT-induced analgesia and steroid hormones. CFA-induced arthritis rat model used to test the effect of DBPT was chronic pain model. After the induction of arthritis, rats subsequently showed a reduced stepping force of the affected limb for at least the next 18 days. The reduced stepping force of the limb was presumably due to a painful knee, since oral injection of indomethacin produced temporary improvement of weight bearing. DBPT dissolved in water was orally administrated. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 4 hours. DBPT produce significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 3 hours. The magnitude of this improvement was equivalent to that obtained after an oral injection of 3mg/kg of indomethacin and this improvement of stepping force was interpreted as an analgesic effect. The reduced stepping force was divided into three stages(10-30g, 30-50g, and 50-70g). All experiments was performed at 50-70g of stepping force, since both DBPT and indomethacin showed the most excellent analgesic effect at 50-70g of stepping force. DBPT produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner and showed analgesic effect on neuropathic pain as well. DBPT-induced analgesic effect could not be blocked by systemic injection of steroid antagonist mifepristone. The present study suggest 1) that DBPT produces a potent analgesic effect on the chronic knee arthritis pain model in the rat and 2) that steroids system does not mediate DBPT-induced analgesia.

Prevention of Arthritic Inflammation Using an Oriental Herbal Combination BDX-1 Isolated from Achyranthes bidentata and Atractylodes japonica

  • An oriental herbal combination (BDX-1) was isolated from Achyranthes bidentata and Atractylodes japonica. We previously tested the clinical effectiveness of BDX-1 in rheumatoid arthritis (RA) patients and found that it has a beneficial therapeutic effect. Here, we provide experimental evidence for the effectiveness of BDX-1 on RA in murine models. The oral administration of BDX-1 was found to markedly inhibit collagen-induced arthritis, adjuvant-induced arthritis, and zymosan-induced inflammation. It also inhibited carrageenan-induced acute edema and acetic acid-induced writhing response. In addition, the biological activity of BDX-1 was found to be strongly increased by fermentation. Our results suggest that BDX-1 could be useful for the treatment of rheumatoid arthritis.

목과(木瓜)의 Adjuvant 관절염(關節炎)에 대한 실험적(實驗的) 연구(硏究) (The Experimental Study on Chaenomelis Fructus(木瓜) in Case of the Arthritis Induced by Injecting Freund's Complete Adjuvant)

  • 9Objective : We carried out this experimental study in order to investigate the effect of Chaenomelis Fructus(木瓜) extract and herbal-acupuncture on the arthritis induced by injecting Freund's complete adjuvant Method : 1. We categorized SD-rats into five groups as followings. (1) Control: Without any Treat (2) Treat I: treated by acupuncture (3) Treat II: treated by drinking 5% saline (4) Treat III: treated by drinking Chaenomelis Fructus(木瓜) extract (5) Treat IV: treated by Chaenomelis Fructus(木瓜) herbal-acupuncture 2. We established ten items for decision of experimental results as followings Edema rate, Pain endurance, WBC, Hemoglobin, Platelet, Total protein, Albumin, Globulin, RA factor, CRP Results : The groups that showed significant effects compared with Contol group were explained as followings. 1. Edema rate : Treat I , Treat III 2. Pain endurance : Treat I, Treat IV 3. WBC : Treat IV 4. Hemoglobin : Treat III 5. Plat99elet : Treat IV 6. Total protein : Treat I, Treat IV 7. Albumin : None 8. Globulin : Treat I, Treat IV 9. RA factor : Treat I, Treat IV 10. CRP : Treat IV Conclusion : Observing above experimental results, we can guess that the effects of acupuncture and Chaenomelis Fructus(木瓜) herbal-acupuncture on Freund's adjuvant arthritis are more superior than other ways. In addition, we can find that the latter is more better than the former in effect by two items(platelet, CRP).

고려홍삼의 콜라겐 유도 관절염의 예방과 억제효과 (Preventive and Inhibitory Effect of Korean Red Ginseng on Collagen-Induced Arthritis in Mice)

  • Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by joint inflammation and progressive cartilage and bone erosion. Korean red ginseng (KRG) has been shown to have an anti-inflammatory effect by inhibiting the secretion of inflammatory cytokines like $TNF-{\alpha}$, IL-1, -6, and -8, and $IPN-{\gamma}$. In this study, whether KRG extract has an inhibitory effect on the collagen-inducible development of arthritis in DBA/1J mice was investigated. To induce arthritis, type II collagen emulsified in Complete Freund's Adjuvant was intradermally injected into the base of the tails of mice. Three weeks after the initial injection, a booster injection of type II collagen emulsified in Incomplete Freund's Adjuvant was administered. The oral administration of KRG extract for 8${\sim}$10 weeks at the dose of 300 mg/kg (three days a week) inhibited the development of arthritis in the experimental group, compared to the control group which was given saline. While the administration of KRG extract three times a week demonstrated both preventive and inhibitory effects, the administration of KRG extract once a week had little inhibitory effect. In other studies, the regimen of KRG administration has been shown to decrease the plasma level of inflammatory cytokines like IL-8 and TNF-${\alpha}$, but the plasma levels of these cytokines were not decreased in the present study. The results of the present study suggest that KRG has preventive and inhibitory effects on collagen-induced arthritis.

백출(白朮)의 콜라겐 유도 관절염 마우스에서의 관절염 개선 효과 연구 (Effects of Atractylodis Rhizoma Alba extract on collagen-induced arthritis in mice)

  • Objectives : The present study was undertaken to evaluate whether Atractylodis Rhizoma Alba ethanol (ARA-E) extract, which is the pericarp of $Atractylodes$ $japonica$ Koidz. has an effect on collagen-induced arthritis (CIA) in mice. Methods : Male DBA/1J mice were induced by intradermal injection of bovine collagen-II in Freund's incomplete adjuvant (IFA). The CIA mice in the onset of arthritis were treated daily with oral administration of ARA-E extract at dose of 50 mg/kg/bw for 28 days. Arthritis index, histopathological changes and the levels of anti-type II collagen (CII) IgG and inflammatory cytokine, TNF-${\alpha}$ in sera of mice were measured to evaluate the antiarthritic effect of ARA-E. Results : ARA-E extract significantly decreased the arthritic scores and inhibited pathological changes of knee joint tissues in CIA mice. ARA-E extract also significantly decreased the serum levels of anti-CII IgG and TNF-${\alpha}$ in CIA mice. These results indicate that ARA-E extract may effectively prevent arthritic damages in CIA mice, at least partially, by inhibiting the production of autoantibodies and inflammatory cytokine. Conclusions : This studies suggest that ARA-E has a therapeutic potential in inflammatory joint diseases such as rheumatoid arthritis.
