고지방 식이로 유도된 비만 쥐에서 실리빈 (Silybin)이 체중 및 내당 능에 미치는 영향 (Effect of Silybin on Body Weight and Glucose Tolerance in High-fat-diet Induced Obese Mice)

  • In this study, we investigated the effects of silybin on body weight and glucose tolerance in mice fed high fat diet mice. We found that body weight, plasma TG contents, fat size, glycerol, 3-hydroxybutyrate and total cholesterol were significantly decreased in silybin (500 mg/kg) supplemented groups compared to high fat diet group. Whereas, total food intake was not changed between high fat diet group and high fat diet plus silybin group. Futhermore, supplement of high fat elevated the glucose intolerance and was improved in silybin supplement group. Finally, we examined the effect of silybin on circulating adipocytokine level to explore the possible mechanism by which silybin improves high fat diet-induced obesity and diabetes. The silybin supplement significantly reduced the level of adipocytokine, such as leptin, resistin, IL-6, and MCP-1 induced by high fat diet. These results suggest that silybin can be used to improve obesity and diabetes.

운동 트레이닝을 통한 심폐체력 반응의 차이가 복부비만 여성의 심혈관계 위험요인과 아디포싸이토카인에 미치는 영향 (Effects of different cardiorespiratory fitness response to exercise training on cardiovascular disease and adipocytokine in abdominal obesity women)

  • 본 연구는 복부비만 여성을 대상으로 운동 트레이닝을 통해 심폐체력의 반응의 차이가 심혈관계 위험요인 및 아디포싸이토카인 분비 및 유전자 발현에 미치는 영향을 알아보는 데 그 목적이 있다. 본 연구는 운동 트레이닝 실시 후 심폐체력 향상의 유무를 이용하여 복부비만 여성 총 48명을 운동-반응 집단(n=34)과 운동-비반응 집단(n=14)으로 나누었다. 운동 트레이닝은 주당 1200kcal를 소모하도록 설계되었으며, 중등도-활발한 운동강도, 걷기나 간단한 조깅의 형태로 12주간 실시하였다. 측정 변인으로 실험 참가 전, 후에 신체조성, 혈압, 심폐체력, 혈중지질, 글루코스, 인슐린 그리고 유리지방산을 측정하였으며, 혈중 아디포싸이토카인과 복부지방 아디포싸이토카인 유전자 발현을 분석하였다. 본 연구의 결과 운동 트레이닝 후 두 집단 모두 비만지표에서 통계적으로 유의하게 감소하였지만, 허리둘레(p=0.040), 체지방률(p=0.031) 그리고 중성지방(p=0.023)과 수축기 혈압(0.046)은 운동-반응 집단과 운동-비반응 집단에서 상호작용 효과를 보였다. 혈중 렙틴은 두 집단에서 상호작용 효과(p=0.022)가 나타났으나 복부지방 렙틴 유전자는 두 집단 모두에서 트레이닝 후(p<0.001)에 유의하게 감소하였다. 이상의 결과를 종합해보면 12주간 운동 트레이닝 실시 후 심폐체력의 향상을 보인 운동-반응 집단은 운동-비반응 집단에 비해 심혈관계 질환 위험요소와 아디포싸이토카인 분비 및 유전자 발현에 더 효과적인 것으로 나타났다. 또한 운동 트레이닝 실시 후 심폐체력의 개선이 없었던 운동-비반응 집단에서도 비만지표와 혈중 렙틴 및 렙틴 유전자 발현이 감소되는 것을 살펴봤을 때 규칙적인 운동 자체가 비록 심폐체력의 개선이 없더라도 인간의 건강상의 이점을 주는 것으로 나타났다. 따라서 심폐체력이 향상되지 않더라도 규칙적인 운동 은 비만 관련 위험요인의 감소 및 예방에 좋은 영향을 주는 것으로 사료된다.

생강 성분인 (6)-Gingerol이 3T3-L1에서 지방세포 증식과 분화 과정에 미치는 영향 (Effects of (6)-gingerol, ginger component on adipocyte development and differentiation in 3T3-L1)

  • 본 연구에서는 한국 음식의 양념류, 향신료로 많이 사용해온 생강의 여러 성분 중 (6)-gingerol을 3T3-L1 preadipocyte에 처리하였을 때 지방세포의 증식과 분화되는 과정에 미치는 영향에 대해 관찰해보고자 하였다. 실험 결과 (6)-gingerol의 첨가량이 증가할수록 세포의 증식이 유의적으로 억제되었다. 지방 분화과정 중에서의 (6)-gingerol은 분화초기에는 효과가 나타나지 않았지만, 지방세포로의 변화가 가속화되는 중기 과정에 관여하는 $PPAR{\gamma}$, $C/EBP{\alpha}$는 (6)-gingerol의 처리로 두 유전자의 발현이 억제되는 것을 확인할 수 있었고, 후기 관련 유전자인 FABP4, AP2의 발현도 (6)-gingerol의 처리군에서 발현이 유의적으로 감소되었다. 또한, 지방세포에서만 분비되는 adipocytokine 중 leptin 발현에는 (6)-gingerol의 처리가 유의적으로 억제되었으나, adiponectin의 경우에는 유의적인 효과는 나타나지 않았다.

The Difference in Bone metabolism markers and Adipocytokine according to the applying Modern Dance with Osteopenia elderly women

  • 본 연구는 골감소증 노인 여성들을 대상으로 모던댄스 적용에 따른 골대사 지표 및 아디포사이토카인의 변화를 분석하여 골다공증 예방을 위한 효과적인 운동 방법을 제안하는데 있다. 연구의 대상자는 T-score -1.0 ~ -2.5사이 여성노인을 선정하였고, 모던댄스 그룹(n=10), 컨트롤 그룹(n=10)으로 무선 배정했다. 모던댄스는 주3회, 60분씩 12주간 실시하였고 자료 분석은 SPSS를 이용해 이원반복분산분석(two-way repeated measures ANOVA)으로 분석했다. 본 연구 결과 모던댄스 그룹에서 osteocalcin(p<.01), T-score(p<.05)가 유의하게 증가했다. Adiponectin(p<.05)은 모던댄스 그룹에서 유의하게 증가했고, TNF-α(p<.05), IL-6(p<.05)는 모두 모던댄스 그룹에서 유의하게 감소하였다. 이상의 결과를 종합해 보면 모던댄스는 골감소증 노인여성의 골다공증 예방을 위해 효과적인 전략이 될 것으로 사료되며 더불어 신진대사 및 기능 향상에도 긍정적인 영향을 미칠 것으로 생각된다.

Obesity and Metabolic Syndrome in Adults with Prader-Willi Syndrome

  • Body fat distribution in patients with Prader-Willi syndrome (PWS) is characterized by reduce lean body mass (LBM), increased total body fat mass (FM), and lower percentage of visceral adipose tissue (VAT). Individuals with PWS seem to have a lower risk for insulin resistance with high levels of adiponectin, an anti-atherogenic adipocytokine that is decreased in visceral fat hypertrophy subjects compared to simple obese subjects, both in children and in adults. The mechanism of the reduction in visceral adiposity in PWS is still unclear. It might be related to qualitative intrinsic characteristics of adipocyte or novel genetic influences on the control of fat distribution. However, obesity remains a critical problem, and obesity status plays a crucial role in individual metabolic risk clustering and development of metabolic syndrome (Mets) in PWS children and adults. Long-term growth hormone (GH) treatment after cessation of skeletal growth improved body composition, with an increase in lean body mass and a reduction in total body fat and subcutaneous and visceral fat in PWS adults. Thus, the role of GH is important after childhood because it might attenuate obesity and Mets in PWS adult by adipocyte modification.

Corn silk extract improves cholesterol metabolism in C57BL/6J mouse fed high-fat diets

  • BACKGROUNG/OBJECTIVES: Corn silk (CS) extract contains large amounts of maysin, which is a major flavonoid in CS. However, studies regarding the effect of CS extract on cholesterol metabolism is limited. Therefore, the purpose of this study was to determine the effect of CS extract on cholesterol metabolism in C57BL/6J mouse fed high-fat diets. MATERIALS/METHODS: Normal-fat group fed 7% fat diet, high-fat (HF) group fed 25% fat diet, and high-fat with corn silk (HFCS) group were orally administered CS extract (100 mg/kg body weight) daily. Serum and hepatic levels of total lipids, triglycerides, and total cholesterol as well as serum free fatty acid, glucose, and insulin levels were determined. The mRNA expression levels of acyl-CoA: cholesterol acyltransferase (ACAT), cholesterol 7-alpha hydroxylase (CYP7A1), farnesoid X receptor (FXR), lecithin cholesterol acyltransferase (LCAT), low-density lipoprotein receptor, 3-hyroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA reductase), adiponectin, leptin, and tumor necrosis factor ${\alpha}$ were determined. RESULTS: Oral administration of CS extract with HF improved serum glucose and insulin levels as well as attenuated HF-induced fatty liver. CS extracts significantly elevated mRNA expression levels of adipocytokines and reduced mRNA expression levels of HMG-CoA reductase, ACAT, and FXR. The mRNA expression levels of CYP7A1 and LCAT between the HF group and HFCS group were not statistically different. CONCLUSIONS: CS extract supplementation with a high-fat diet improves levels of adipocytokine secretion and glucose homeostasis. CS extract is also effective in decreasing the regulatory pool of hepatic cholesterol, in line with decreased blood and hepatic levels of cholesterol though modulation of mRNA expression levels of HMG-CoA reductase, ACAT, and FXR.

Relations of Serum Visfatin and Resistin Levels with Endometrial Cancer and Factors Associated with its Prognosis

  • Background: The aims of this study were compare the serum visfatin and resistin levels between endometrial cancer (EC) patients and controls and evaluate their power to predict prognosis. Materials and Methods: This prospective study was conducted between March 2013 to June 2014 on the Gynecologic Oncology Department of the University of Selcuk, Konya, Turkey. A total of 42 EC patients and 42 controls were included and assessed for differences in serum visfatin and resistin levels, along with prognostic factors. Results: Endometrial cancer patients had significantly higher visfatin levels than control s (p: 0.011), associated with deep myometrial invasion (p: 0.019). In contrast the serum level of resistin did not significantly differ between EC patients and controls (p: 0.362). However, high resistin level in EC patients was associated with increase lymph node metastasis (p: 0.009). On logistic regression analysis, we found that serum visfatin elevation was associated with risk of myometrial invasion (OR: 1,091; 95%CI: 1.021-1.166; p: 0.010) and serum resistin with risk of lymph node metastasis (OR: 1.018; 95%CI: 1.000-1.035; p: 0.046). For myometrial invasion prediction, a serum visfatin level greater than 26.8 ng/mL demonstrated a sensitivity and specificity of 66.6 % and 96.4%, respectively. For lymph node metastasis prediction, the best cut-off for serum resistin level was 599ng/mL. A serum resistin level greater than this demonstrated a sensitivity and specificity of 87.5% and 77.1%, respectively. Conclusions: Our data suggest that serum visfatin is elevated in patients with EC and serum visfatin and resistin levels could be used to predict the risk of advance stage lesions.

Influence of Adipocytokines and Periprostatic Adiposity Measurement Parameters on Prostate Cancer Aggressiveness

  • Background: The relationship between obesity and prostate cancer aggressiveness is controversial in recent studies, partly because BMI is the only generally applied marker of obesity. Our study aimed at evaluating the correlation of periprostatic fat (PF) on magnatic resonance imaging (MRI) and adipocytokines with prostate cancer aggressiveness. Patients and method: A total of 184 patients who underwent radical retropubic prostatectomy (RRP) were analyzed retrospectively; different fat measurements on MRI slices and levels of adipocytokines were compared with the clinical and pathologic factors using SSPS ver.13.0. Result: The PF rates showed a statistically significant variation (p=0.019, 0.025) among groups, that is to say, more adipose tissue was distributed in periprostatic areas of high risk patients. Logistic regression analysis adjusted for age revealed a statistically association between the PF, the ratio and the risk of having high-risk disease (p=0.031, 0.024). The levels of IL-6, leptin and c-reactive protein (CRP) significantly increased with the aggressiveness of prostate cancer, and also with PF and its ratio. The strongest correlation was seen between IL-6 and PF (Pearson r coefficient=0.67, P<0.001). No association was observed between adipocytokines and BMI. Conclusion: Periprostatic adiposity not only affects prostate cancer aggressiveness, but also influences the secretion of adipocytokines. IL-6, PF and CRP have promoting effects on progression of prostate cancer.

조각자(皂角刺)가 estradiol valerate로 유발된 백서(白鼠)의 다낭성 난소에 미치는 영향 (Effects of Gleditsiae Spina(GS) on the Polycystic Ovary Induced by Estradiol Valerate in Rats)

  • Purpose: In the theory of traditional medicine, Glenditsia spina(GS) can resolve carbuncle, relive swelling, dispel wind and destroy parasites. This study was designed to investigate the effects of GS on gene expression of ovarian tissue in polycystic ovary syndrome(PCOS) rats. Methods: In this experiment, female rats injected with a single dose of 2 mg estradiol valerate(EV) and GS was given for 5 weeks. The genetic profile for the effects on ovarian tissue in PCOS rats was measured using microarray technique, and the functional analysis on these genes was conducted. Results: 985 genes were increased in control and restored to normal level in GS group. (B), 733 genes were decreased in control group and restored to normal level in GS group. (F). Metabolic pathways related in B group genes were Graft-versus-host disease, Allograft rejection, Autoimmune thyroid disease, Cytokine-cytokine receptor interaction, Small cell lung cancer, Type I diabetes mellitus. Metabolic pathways related in F group genes were Antigen processing and present, Adipocytokine signalling pathway, Focal adhesion, ECM-receptor interaction, Pancreatic cancer, Notch signalling pathway, Tight junction. The network of total protein interactions was measured using cytoscape program, and some key molecules, such as c-Fos, c-Myc, ABL1 related in B group, MAPK8, RASA1, CALR related in F group that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conclusion: These results suggest possibility of GS as anti-cancer and anti-hyperplasia drug in PCOS. In addition, the present author also suggests that related mechanisms are involved in suppression of proto-oncogene such as c-Fos, c-Myc and ABL1, and in regulation of cell cycle such as RASA1.

Confirmation of genotypic effects for the bovine APM1 gene on marbling in Hanwoo cattle

  • Background: Our previous study had identified the SNP (g.81966377T > C) and indel (g.81966364D > I) located in the promoter of APM1 to have a significant effect on marbling in Hanwoo. APM1 encodes an adipocytokine called adiponectin, which plays a significant role in lipogenesis. The aim of this study was to verify and validate the effect of the SNP and indel on marbling and other carcass traits in a large, representative, countrywide population of Hanwoo cattle. The carcass traits measured were marbling (MAR), backfat thickness (BFT), loin eye area (LEA), and carcass weight (CAW). Results: Primers were designed to amplify 346 bp of the genomic segment that contained the targeted SNP (g.81966377) and the indel (g.81966364). After data curation, the genotypes of 8,378 individuals identified using direct sequencing analysis estimated frequencies for C (0.686) and T (0.314) respectively showing genotype frequencies for CC (0.470), CT (0.430) and TT (0.098). The genotypes were significantly associated with MAR, BFT and LEA. The indel had significant effect on marbling (P < .0001) with strong additive genetic effects. The allele frequencies was estimated at (DEL, 0.864) and insertion (INS, 0.136) presenting genotypes of D/D (75.63 %), D/I (21.44 %), and I/I (2.92 %). Significant departure from Hardy-Weinberg equilibrium was not detected for both the SNP and the indel. Conclusion: The SNP genotypes showed significant association with MAR, BFT and LEA with strong additive genetic effects, while the indel was significantly associated with MAR. The results confirmed that the variants can be used as a genetic marker for improving marbling in Hanwoo.
