Glycine max Merr enhances the viability and adhesion ability of Lactobacillus buchneri in gastrointestinal condition in vitro.

  • Probiotics are microorganisms that have beneficial effects on the health of the host. The health promoting effect by probiotics influences suppressing harmful bacteria, prevention of constipation, blood cholesterol reduction and regulation of blood pressure. Prebiotics are used to promote the growth or activity of microorganisms. Synbiotics, which are a mixture of probiotics and prebiotics, synergize in the intestines by complementing each other. Synbiotics not only improves the viability of the probiotics while passing through the gastrointestinal tract, maintain intestinal homeostasis, but also regulate balance of harmful and useful bacterial growth. Glycine max Merr (GMM) has been widely used in Asian countries to treat cancer, obesity, oxidative stress and imbalanced immune diseases. In addition, it has been reported that dietary fiber-rich grains promote bowel movements and prevent constipation. In this study, we investigated the viability of LactobacillIus buchneri (L.buchneri) strains, known as lactic acid bacteria under conditions of gastric fluid and intestinal fluid to determine the suitability of L.buchneri as probiotics. The adhesion ability of L.buchneri to caco-2 cells was also confirmed. The present studies showed that GMM extract promoted the growth and activity of L.buchneri strains as prebiotics. Also, this results suggested that the mixture of L.buchneri and GMM extract can helps maintain intestinal health and healthy body as synbiotics and health functional food material.

The Effects of Peucedani Radix Herbal Acupuncture at St36 on Ovalbumin-induced Asthma in C57BL Mouse (전호약침(前胡藥鍼)이 Ovalbumin-induced Asthma Mouse Model에 미치는 영향)

  • Objectives : The aim of this study was to investigate the effect of Peucedani Radix herbal acupuncture(PR-HA) at St36(joksamni) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week) C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (3times a week). One of the two experimental groups was just treated with needle-prick on St36 and the other group was treated with 1% concentrations of PR-HAS at St36, for the later 8 weeks (3times /week). Results : 1. The weight and total cells of lung of the mice group treated with PR-HA decreased significantly compared with those of control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated with PR-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated with PR-HA in Photomicrographs decreased significantly compared with those of control group. 4. According to Histological analysis of lung sections, it decreased significantly adhension of collagen in PR-HA than those of control group 5. The concentration of IgE, IL-4, IL-5, in BALF and IL-4, IL-5, Il-13 in serum of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The number of Gr-1+/CD11b+, CD11b+, CD3-/CCR3+, CD4+, CD3e+/CD69+ , CD23+B220+ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The cytokine's manifestation of mRNA of the mice group treated with PR-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : We conclude that PR-HA is effective on OVA-induced asthma of C57BL/6 mouse.

  • The resolving capacities of xanthophyll pigments on thin-layers of Silica Gel, Hyflo super-Cel, and Micro-Cel C with varying concentrations of acetone in petroleum ether as the developing solvent were compared. The results showed that the resolving capacity of Micro-Cel C thin-layer was superior to others and satisfactory for the separation of leaf carotenoids in clearly separated six bands; carotenes, lutein-zeaxanthin, antheraxanthin, violaxanthin, an unidentified band, and neoxanthin, when it was developed with $13\%$ acetone-petroleum ether solution for 15 to 20minutes in an unsaturated chamber. Adhension of Micro-Cel C to glass was adequate without binder. Calcium sulfate used as a binder appeared to inactivate the resolving capacity of Micro-Cel C. Removing an about 0.2cm-wide layer on bo side of thin-layer slide helped to prevent 'edge effect' which gave tailing and faster solvent ascending along the side than the center. An adequate thickness of thin-layer was obtained when a 3 ml aliquot of the suspension in which l0g powdered Micro-Cel C was suspended in 75 ml distilled water was coated on a $2\times20cm$ glass slide.

  • The plasma source ion implantation(PSII) technique which is a method using high negative voltage pulse in plasma system has the potential to change the surface properties of polymer. PSII technique increase the surface free energy by introducing polar functional groups on the surface so that it improves reactivity, hydrophilicity, adhension, biocompatability, etc. However, the mobility of polymer chains enables the modified surface layers to adapt their composition to interfacial force. This hydrophobic recovery interrupts the stability of modified surfaces to keep for the long time. In this study, poly(methyl methacrylate)(PMMA), poly(2-hydroxyethyl methacrylate)(PHEMA), and polu(2-hydroxypropyl methacylate)(PHPMA) for contact lens application, were modified to improve the wettability with PSII technique and were investigated the surface stabilities. Polymer film was prepared with solution casting(3 wt.% solution) and was annealed at 11$0^{\circ}C$ under vacuum oven to remove solvent completely and to eliminate physical ageing. The thickness of the film measured by scanning electron microscopy (SEM) and surface profilometer was about 10${\mu}{\textrm}{m}$. Polymers were treated with different kinds of gases, pulse frequency, pulse with, pulse voltage, and treatment time. Even though PMMA, PHEMA, and PHPMA have similar repeat unit structure, the optimal treatment conditions and the tendency to hydrophobic recovery were different. PHPMA, more hydrophilic polymer than PMMA and PHEMA showd better wettability and stability after mild treatment. Surface tensions were obtained by water and diiodomethane contact angle measurements to monitor the relation between hydrophobic recovery and polymer structure. Different ion species in plasma change the polar component and dispersion component of polymer surface. For better wettability surface, the increase of polar component was a dominant factor. We also characterized modified polymer surfaces using x-ray photoelectron spectroscopy(XPS), secondary ion mass spectrometry(SIMS), Fourier Transform infrared spectroscopy(FT-IR), and SEM.

NMR structural studies on Human CD99 Type I

  • Human CD99 is a ubiquitous 32-kDa transmembrane protein encoded by the mic2 gene. The major cellular functions of CD99 protein are related to homotypic cell adhension, apoptosis, vesicular protein transport, and differentiation of thymocytes or T cells. Recently it has been reported that expression of a splice variant of CD99 transmembrane protein (Type I and Type II) increases invasive ability of human breast cancer cells. To understand structural basis for cellular functions of CD99 (Type I), we have initiated studies on hCD99$^{TMcytoI}$ and hCD99$^{cytoI}$ using circular dichroism (CD) and multi-dimensional NMR spectroscopy. CD spectrum of hCD99$^{TMcytoI}$ in the presence of 200mM DPC and CHAPS displayed an existence $\alpha$-helical conformation. The solution structure of hCD99$^{cytoI}$ determined by NMR is composed of one N-terminal $\alpha$-helix, $\alpha$A, two C-terminal short $\alpha$-helix segments, $\alpha$B and $\alpha$C. While $\alpha$A and $\alpha$B are connected by the long flexible loop, $\alpha$B and $\alpha$C connected by type III$\beta$-turn. Although it has been rarely figured out the correlation between structure and functional mechanism of hCD99$^{TMcytoI}$ and hCD99$^{cytoI}$, there is possibility of dimerization or oligomerization. In addition, the feasible mechanism of hCD99$^{cytoI}$ is that it could have intramolecular interaction between the N- and C- terminal domain through large flexible AB loop.

  • This study is based on the evaluation of a single component injection type adhesive repair material and a new 2 component type used in leakage of underground concrete structures. The studies showed that based on different mix ratio of the 2 component type adhesive sealant comprised of an asphalt main and latex mixture agent the viscosity of the material compound differed significantly. Based on a permeability testing, injection and economic efficiency, and performance, the appropriate ratio was determined to be 6:1 and was proceeded to the KS F 4935 evaluation method. The 2 component type adhesive sealant uses a static mixer and a cartridge type container for the injection procedure and was able to satisfy the evaluation criteria outlined in the KS F 4935 standard.

Methods : C57BL/6 mice were sensitized and challenged with ovalbumin(OVA) once a week for twelve weeks. The experimental group was treated with 1% concentrations of TS-HA at Joksamni(ST36) three times a week for the last eight weeks. Results : 1. The weight and total lung cells of the mice in the group treated with TS-HA decreased significantly compared with those of control group. 2. Total leukocytes and eosinophils in Bronchoalveoler-lavage fluid(BALF) of the mice in the group treated with TS-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice in the group treated with TS-HA in photomicrographs decreased significantly compared with those of control group. 4. According to histological analysis of lung sections, adhension of collagen in TS-HA decreased significantly compared with that of control group. 5. Improved Treatment Technique for the Reuse of Waste Solution Generated from a Electrokinetic Decontamination System (동전기제염장치에서 발생한 폐액의 재사용을 위한 개선된 처리기술)

  • A large amount of acidic waste solution is generated from the practical electrokinetic decontamination equipments for the remediation of soil contaminated with uranium. After filtration of uranium hydroxides formed by adding CaO into the waste solution, the filtrate was recycled in order to reduce the volume of waste solution. However, when the filtrate was used in an electrokinetic equipment, the low permeability of the filtrate from anode cell to cathode cell due to a high concentration of calcium made several problems such as the weakening of a fabric tamis, the corrosion of electric wire and the adhension of metallic oxides to the surface of cathode electrode. To solve these problems, sulfuric acid was added into the filtrate and calcium in the solution was removed as $CaSO_4$ precipitate. A decontamination test using a small electrokinetic equipment for 20 days indicated that Ca-removed waste solution decreased uranium concentration of the waste soil to 0.35 Bq/g, which is a similar to a decontamination result obtained by distilled water.

Clostridium difficile Toxin A Inhibits Erythropoietin Receptor-Mediated Colonocyte Focal Adhesion Through Inactivation of Janus Kinase-2

  • Previously, we demonstrated that the erythropoietin receptor (EpoR) is present on fibroblasts, where it regulates focal contact. Here, we assessed whether this action of EpoR is involved in the reduced cell adhesion observed in colonocytes exposed to Clostridium difficile toxin A. EpoR was present and functionally active in cells of the human colonic epithelial cell line HT29 and epithelial cells of human colon tissues. Toxin A significantly decreased activating phosphorylations of EpoR and its downstream signaling molecules JAK-2 (Janus kinase 2) and STAT5 (signal transducer and activator of transcription 5). In vitro kinase assays confirmed that toxin A inhibited JAK 2 kinase activity. Pharmacological inhibition of JAK2 (with AG490) abrogated activating phosphorylations of EpoR and also decreased focal contacts in association with inactivation of paxillin, an essential focal adhesion molecule. In addition, AG490 treatment significantly decreased expression of occludin (a tight junction molecule) and tight junction levels. Taken together, these data suggest that inhibition of JAK2 by toxin A in colonocytes causes inactivation of EpoR, thereby enhancing the inhibition of focal contact formation and loss of tight junctions known to be associated with the enzymatic activity of toxin A.
