$UPLC^{TM}$를 이용한 화장품 중 보존제 8종(파라벤 6종, 페녹시에탄올, 클로페네신)의 동시분석 (Simultaneous Determination of 8 Preservatives (6 Parabens, 2-Phenoxyethanol, and Chlorphenesin) in Cosmetics by $UPLC^{TM}$)

  • 미생물의 오염을 통해 화장품이 변질되거나 분해되는 것을 방지하여 소비자를 보호하기 위해 화장품에 보존제가 사용된다. 파라벤류는 제형화하기 쉽고, 활성 범위가 넓으며, pH에 화학적으로 안정하면서 저렴하여 거의 모든 종류의 화장품에 널리 사용된다. 페녹시에탄올과 클로페네신 역시 화장품에 일반적으로 사용되는 보존제로 보통 파라벤과 함께 사용된다. 파라벤의 독성은 일반적으로 낮지만, 손상된 피부에는 자극을 유발할 수 있으며, estrogenic 잠재성, 마취 효과 및 생식 독성의 가능성에 대한 논란이 있어 왔다. 따라서 파라벤은 배합한도 원료로 지정 관리되고 있으며, 페녹시에탄올과 클로페네신도 마찬가지로 최대 허용량이 지정되어 있다. 그러므로 제품 중 보존제의 함량을 관리하는 것은 중요하다. 그러나 일반적으로 사용되는 역상 액체크로마토그래프법으로는 이성질체를 포함한 6종의 파라벤과 페녹시에탄올 및 클로페네신을 동시에 분리 분석하기 어려웠다. 용출 시간이 길어져, 피크 모양이 나쁘고 분리능이 좋지 않아 정확한 정량이 불가능하였다. 본 연구에서는 ultra performance liquid $chromatography^{TM}\;(UPLC^{TM}$)를 이용하여 8종의 보존제를 10 min 이내에 동시분석을 시도하였다. 또한, International conference on harmonisation (ICH) 가이드라인의 밸리데이션 방법에 근거하여 본 시험법의 적합성을 검증하고, 로션, 팩트, 파운데이션 및 립글로스 등 다양한 제형에 적용이 가능함을 보였다. 본 시험법은 파라벤류를 포함한 다양한 보존제를 함유한 화장품 중 보존제의 함량을 단시간에 간편하고 정확하게 정량하는데 활용될 수 있을 것이다.360 nm)에서 3개의 피이크로 분리되었다. 분리된 3가지 성분은 luteolin, quercetin 및 kaempferol이었으며, 그들의 성분비는 각각 18.24 %, 58.79 %, 22.97 %로 quercetin의 함량이 가장 큰 것으로 나타났다. 루이보스 추출물의 ethylacetate 분획의 TLC 크로마토그램은 7개의 띠로 분리되었고, HPLE 크로마토그램은 9개의 피이크를 보여주었다. TLC와 HPLC의 띠와 피이크를 확인한 결과, HPLC의 9개의 피이크는 용리순서로 peak 1 (조성비 14.71 %)은 isoorientin, peak 2 (28.84 %)는 orientin peak 3 (5.63 %)은 vitexin, peak 4 (12.73 %)는 rutin과 isovitexin, peak 5 (9.24 %)는 hyperoside, peak 6 (5.40%)은 isoquercitrin, peak 7 (1.48 %)은 luteolin, peak 8 (17.61 %)은 quercetin 및 peak 9 (4.59 %)는 kaempferol로 확인되었다. Aglycone 분획은 elastase 저해활성($IC_{50}$)이 $9.08\;{\mu}g/mL$로 매우 큰 활성을 나타내었다. 이상의 결과들은 루이보스 추출물이 $^1O_2$ 혹은 다른 ROS를 소광시키거나 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리키며, 루이보스 성분에 대한 분석과 ethylacetate 분획의 당 제거 실험 후 얻어진 aglycone 분획의 큰 elastase 저해활성으로부터

금전초 추출물 및 분획물의 항산화 활성 및 세포 보호 효과 (Antioxidative and Cellular Protective Effects of Lysimachia christinae Hance Extract and Fractions)

  • 본 논문에서는 금전초로부터 50% 에탄올 추출물, 에틸아세테이트 분획 및 아글리콘 분획을 제조하였고 이들의 항산화 활성, 세포 보호 효과 및 성분 분석에 대한 연구를 수행하였다. 항산화능 평가에서 50% 에탄올 추출물, 에틸아세테이트 분획 및 아글리콘 분획의 라디칼 소거 활성($FSC_{50}$)은 각각 146.8, 22.2 및 $27.2{\mu}g/mL$이었고, 총 항산화능($OSC_{50}$)은 29.3, 2.9 및 $4.5{\mu}g/mL$이었다. 에틸아세테이트 분획의 자유라디칼 소거 활성 및 총 항산화능이 가장 크게 나타났다. 또한 $^1O_2$로 유도된 적혈구 광용혈에 대한 세포 보호 효과(${\tau}_{50}$)는 금전초 50% 에탄올 추출물, 에틸아세테이트 분획 및 아글리콘 분획 $5{\mu}g/mL$에서 각각 26.9, 57.5 및 103.9 min을 나타냈다. 특히 아글리콘 분획물의 ${\tau}_{50}$은 (+)-${\alpha}$-tocopherol (37.7 min)보다 훨씬 큰 세포 보호 효과를 나타내었다. UVB로 유도된 세포 손상에서 에틸아세테이트 분획물은 최대 90.1%까지 세포 생존율을 증가시켰다. 과산화수소로 유도된 세포 손상에서도 에틸아세테이트 분획물은 $5-25{\mu}g/mL$에서 농도 의존적으로 세포 보호 효과를 나타내었다. 금전초 에틸아세테이트 분획물의 성분 분석을 위해 TLC, HPLC, UV-vis spectrum, LC-MS로 분석한 결과, quercetin, kaempferol 및 그 배당체가 주성분들로 확인되었다. 결론적으로 금전초가 자외선에 노출된 피부를 보호하는 항산화제로서 작용할 수 있고, 기능성 화장품 원료로 응용 가능성이 있음을 시사한다.

$^{11}$C 표지 자동합성장치에서 루프법을 이용한 ($^{11}$C)(R)-PK11195의 간편한 합성법 (A Convenient Radiolabeling of [$^{11}$C](R)-PK11195 Using Loop Method in Automatic Synthesis Module)

  • 목적: (R)-1-(2-Chlorophenyl)-N-methyl-N-1-(1-methyl-propyl)-3-isoquinoline carboxamide ((R)-PK11195)는 말초형 벤조디아제핀 수용체 (PBR)의 친화성이 높으며, 활성화된 소교세포(microglia)의 PBR에 선택적으로 결합하는 것으로 알려져 있다. 본 연구에서는 말초형 벤조디아제핀 수용체 (PBR)의 FET용 리간드인[$^{11}$C](R)-PK11195를, 절차가 빠르고 간단한 [$^{11}$C]CH$_3$I 자동합성장치 및 루프법을 도입해서 합성하였다. 대상 및 방법: 사이클로트론에서 $^{14}$N(p,a)$^{11}$C핵반응에 의하여 생산된 [$^{11}$C]Co$_2$를 0.2 M LiAlH$_4$/THF (0.2 mL)로 환원한 다음 HI (1 mL)과 반응하여 [$^{11}$C]CH$_3$I를 생산하였다. 반응용매인 NaH DMF complex (0.1 mL)와 녹인 전구물질 (R)-N-desmethyl-PK11195 (1 mg)을 녹인 DMSO (0.1mL)를 섞은 혼합액을 HPLC의 루프에 미리 주입하고 [$^{11}$C]CH$_3$I를 상온에서 5분 동안 질소가스로 불어준 뒤에 semi-preparative HPLC로 [$^{11}$C](R)-PK11195를 분리하였다. 결과: 전구물질과 [$^{11}$C]CH$_3$I의 [$^{11}$C]메틸화 반응에서의 표지효율은 71.8$\pm$8.5% 이었다. 분리 후 얻은 [$^{11}$C](R)-PK11195의 비방사능은 11.8$\pm$6.4 GBq/$\mu$mol이었으며, 방사화학적 순도는 99.2% 이상이었다. C-11 표지 후 얻어진 (R)-PK11195의 물질의 질량 분석은 m/z 353.1 (M+1)으로 물질구조를 확인 할 수 이었다. 결론: 뇌염증에 의한 활성화 소교세포의 영상화를 위한 PET용 방사성의약품인 [$^{11}$C](R)-PK11195를 반복적으로 생산해야 하는 임상 적용을 위해, 합성절차가 빠르고 간단한 [$^{11}$C]CH$_3$I 자동화 합성장치를 1차 루프 방법을 이용하여 $^{11}$C을 표지 할 수 있었다. 이 연구를 통해서 [$^{11}$C](R)PK11195 표지 할 때 반응 단계를 줄이고 반응을 실온에서 할 수 있어서 표지과정을 보다 단순화할 수 있었으며, NaH의 DMF현탁액을 사용함으로써 보다 안전하게 생산할 수 있게 되었다.

BV-2 미세아교세포에서 왕귀뚜라미 유래 Teleogryllusine의 신경염증 억제 효과 (Anti-neuroinflammatory Effect of Teleogryllus emma Derived Teleogryllusine in LPS-stimulated BV-2 Microglia)

  • 최근 중추신경계에서 면역기능을 담당하는 미세아교세포(microglia)의 염증반응을 효율적으로 조절하는 것은 알츠하이머 병, 파킨슨 병 및 헌팅턴 병과 같은 퇴행성 뇌질환의 치료를 위한 중요한 타겟으로 인식되고 있다. 왕귀뚜라미(Teleogryllus emma)는 다양한 치료효능으로 인해 세계적으로 널리 이용되고 있으며, 본 연구팀에서는 최근 왕귀뚜라미의 전사체 분석을 통하여 항균활성을 가지는 다양한 종류의 새로운 항균 펩타이드(antimicrobial peptide; AMP) 후보들을 선별한 바 있다. 항균 펩타이드는 미생물에서부터 포유류까지 매우 다양한 종으로부터 발견되었으며, 현재는 항균활성뿐만 아니라 염증반응과 같은 다양한 질병의 치료제 개발을 위한 후보 물질로 관심을 받고 있다. 본 연구에서는 선행연구를 통하여 선별된 왕귀뚜라미 유래 항균 펩타이드들 중에서 Teleogryllusine(VKWKR-LNNNKVLQKIYFVKI-NH2)으로 명명된 항균 펩타이드의 신경염증 억제 효능을 관찰하였다. Teleogryllusine의 신경염증 억제 효능을 관찰 하기 위하여 immortalized mouse microglia 세포주인 BV-2 세포에 Teleogryllusine을 1시간 전처리 한 후 LPS를 이용하여 BV-2 세포의 염증 반응을 유도하였다. 그 결과 Teleogryllusin은 최대 처리 농도인 80 ㎍/ml까지 세포독성 없이 nitric oxide (NO) 생성을 현저히 감소시킴을 확인할 수 있었다. 또한 염증반응 매개인자인 iNOS와 COX-2 및 cytokine (Il-6, TNF-α)의 발현을 유전자 수준과 단백질 수준에서 확인한 결과 Teleogryllusine 처리농도에 의존적으로 감소됨을 확인할 수 있었다. 또한 Teleogryllusine의 신경염증 억제작용 기작을 확인한 결과 mitogen activated protein kinases (MAPKs)와 IκB의 인산화 및 proteosome에 의한 IκB의 분해를 억제함으로서 BV-2 세포의 신경염증반응이 조절됨을 확인할 수 있었다. 이러한 결과로 보아 왕귀뚜라미 유래 Teleogryllusine 펩타이드는 신경염증반응에 의해 유도되는 퇴행성 뇌질환 치료 및 예방을 위한 의료용 소재로 사용될 수 있을 것으로 기대된다.

Violacein을 생산하는 Massilia sp. EP15224 균주 (Production of Violacein by a Novel Bacterium, Massilia sp. EP15224 Strain)

  • 비올라세인은 항균, 항암, 항산화, 항말라리아, 항설사 활성과 같은 다양한 생리활성을 가지는 보라색소 물질로 알려져 있다. 본 연구에서는 한국의 산림토양에서 분리되었으며 보라색 색소를 생산하는 EP15224 균주를 선발하여 16S rRNA 유전자의 염기서열을 분석하였다. 그 결과 Massilia sp. BS-1과 97%의 가장 높은상동성을 보였다. 또한 16S rRNA 유전자에 근거한 계통분류학적 분석에서 EP15224는 기존 보고된 비올라세인 생산 세균들과는 별개로 구분되는 Massilia속의 새로운 종임을 확인하였다. 이 균주가 생산하는 보라색 소물질을 분리하여 LC/MS/MS로 분석한 결과, 분자량이 343.34인 비올라세인과 일치하였다. 또한 비올라세인의 생산효율을 높여주는 최적의 배지성분[glucose 2 g/l, $(NH_4)_2SO_4$ 1 g/l, $Na_2HPO_4{\cdot}7H_2O$ 2 g/l, $KH_2PO_4$ 1 g/l, $MgSO_4{\cdot}7H_2O$ 0.1 g/l, $\small{L}$-tryptophan 0.24 g/l]과 배양조건[$25^{\circ}C$에서 72시간 배양, 250 rpm, 10% 접종량]을 조사하였으며 이 조건에서 액체배양 하였을 때 리터 당 280 mg의 비올라세인이 생산되었다.

인공위성 원격탐사를 이용한 백두산 화산 감시 연구 리뷰 (A Review on Monitoring Mt. Baekdu Volcano Using Space-based Remote Sensing Observations)

  • 백두산은 중국과 북한의 국경 경계에 위치하고 있는 성층화산으로 신생대 올리고세 이후 주요 분화 단계를 거쳐 형성된 것으로 알려져 있다. 2010년 이후 마그마 재활동으로 인한 백두산 화산활동 여부에 대한 관심이 증대되고 있다. 백두산 화산 활동을 감시하기 위한 연구는 기상청, 한국지질자원연구원 등 국가기관 중심으로 활발히 수행되고 있다. 2018년에는 한-중 백두산 공동 관측 장기연구 과제가 선정되었으며 이로부터 화산특화연구센터가 설립되기도 하였다. 그러나 백두산은 우리나라로부터 지리적으로 멀리 떨어져 있어 접근에 대한 제약이 있을 뿐만 아니라 백두산 화산 주변에 설치되어 있는 현장 관측 장비로부터 수집된 현장 자료의 공유 혹은 접근이 쉽지 않은 상황이다. 원격탐사는 직접적인 물리적 접촉 없이 대상 물체에 대한 특성을 원격으로 측정하는 수단으로서, 대상물의 관측을 위해 자동차, 무인기, 항공기, 인공위성 등 여러 형태의 플랫폼이 사용된다. 지난 수십 년 간, 다양한 파장 대역에서의 전자기파를 이용한 원격탐사 자료를 활용하여 화산 감시 연구가 수행되어 왔다. 특히 레이더 원격탐사는 주야조건, 기상조건에 관계없이 자료를 획득할 수 있을 뿐만아니라 위상정보를 이용한 레이더 위상간섭기법을 통한 미세 지표 변위 관측이 가능하여 매우 널리 이용되고 있는 화산 감시 기술이다. 본 논문의 목적은 백두산 화산 관측을 위해 수행된 기존 원격탐사 연구 문헌을 수집하고 동향을 파악하는 것이다. 또한 지속적인 화산 감시를 위한 가용 영상레이더 위성정보를 조사하여 향후 이를 바탕으로 백두산 화산 지표 변위의 주기적 탐지 연구를 수행하는데 활용할 예정이다.

Cefoperazone(T-1551)의 약리학적 연구 (Pharmacological Studies of Cefoperazone(T-1551))

  • The pharmacological and microbiological studies of Cefoperazone (T-1551, Toyama Chemical Co., Japan) were conducted in vitro and in vivo. The studies included stability and physicochemical characteristics, antimicrobial activity, animal and human pharmacokinetics, animal pharmacodynamics and safety evaluation of Cefoperazone sodium for injection. 1) Stability and physicochemical characteristics. Sodium salt of cefoperazone for injection had a general appearance of white crystalline powder which contained 0.5% water, and of which melting point was $187.2^{\circ}C$. The pH's of 10% and 25% aqueous solutions were 5.03 ana 5.16 at $25^{\circ}C$. The preparations of cefoperazone did not contain any pyrogenic substances and did not liberate histamine in cats. The drug was highly compatible with common infusion solutions including 5% Dextrose solution and no significant potency decrease was observed in 5 hours after mixing. Powdered cefoperazone sodium contained in hermetically sealed and ligt-shielded container was highly stable at $4^circ}C{\sim}37^{\circ}C$ for 12 weeks. When stored at $4^{\circ}C$ the potency was retained almost completely for up to one year. 2) Antimicrobial activity against clinical isolates. Among the 230 clinical isolates included, Salmonella typhi was the most susceptible to cefoperazone, with 100% inhibition at MIC of ${\leq}0.5{\mu}g/ml$. Cefoperazone was also highly active against Streptococcus pyogenes(group A), Kletsiella pneumoniae, Staphylococcus aureus and Shigella flexneri, with 100% inhibition at $16{\mu}g/ml$ or less. More than 80% of Escherichia coli, Enterobacter aerogenes and Salmonella paratyphi was inhibited at ${\leq}16{\mu}/ml$, while Enterobacter cloaceae, Serratia marcescens and Pseudomonas aerogenosa were somewhat less sensitive to cefoperagone, with inhibitions of 60%, 55% and 35% respectively at the same MIC. 3) Animal pharmacokinetics Serum concentration, organ distritution and excretion of cefoperazone in rats were observed after single intramuscular injections at doses of 20 mg/kg and 50 mg/kg. The extent of protein binding to human plasma protein was also measured in vitro br equilibrium dialysis method. The mean Peak serum concentrations of $7.4{\mu}g/ml$ and $16.4{\mu}/ml$ were obtained at 30 min. after administration of cefoperazone at doses of 20 mg/kg and 50 mg/kg respectively. The tissue concentrations of cefoperazone measured at 30 and 60 min. were highest in kidney. And the concentrations of the drug in kidney, liver and small intestine were much higher than in blood. Urinary and fecal excretion over 24 hours after injetcion ranged form 12.5% to 15.0% in urine and from 19.6% to 25.0% in feces, indicating that the gastrointestinal system is more important than renal system for the excretion of cefoperazone. The extent of binding to human plasma protein measured by equilibrium dialysis was $76.3%{\sim}76.9%$, which was somewhat lower than the others utilizing centrifugal ultrafiltration method. 4) Animal pharmacodynamics Central nervous system : Effects of cefoperazone on the spontaneous movement and general behavioral patterns of rats, the pentobarbital sleeping time in mice and the body temperature in rabbits were observed. Single intraperitoneal injections at doses of $500{\sim}2,000mg/kg$ in rats did not affect the spontaneous movement ana the general behavioral patterns of the animal. Doses of $125{\sim}500mg/kg$ of cefoperazone injected intraperitonealy in mice neither increased nor decreased the pentobarbital-induced sleeping time. In rabbits the normal body temperature was maintained following the single intravenous injections of $125{\sim}2,000mg/kg$ dose. Respiratory and circulatory system: Respiration rate, blood pressure, heart rate and ECG of anesthetized rabbits were monitored for 3 hours following single intravenous injections of cefoperazone at doses of $125{\sim}2,000mg/kg$. The respiration rate decreased by $3{\sim}l7%$ at all the doses of cefoperazone administered. Blood pressure did not show any changes but slight decrease from 130/113 to 125/107 by the highest dose(2,000 mg/kg) injected in this experiment. The dosages of 1,000 and 2,000 mg/kg seemed to slightly decrease the heart rate, but it was not significantly different from the normal control. All the doses of cefoperazone injected were not associated with any abnormal changes in ECG findings throughout the monitering period. Autonomic nervous system and smooth muscle: Effects of cefoperazone on the automatic movement of rabbit isolated small intestine, large intestine, stomach and uterus were observed in vitro. The autonomic movement and tonus of intestinal smooth muscle increased at dose of $40{\mu}g/ml$ in small intestine and at 0.4 mg/ml in large intestine. However, in stomach and uterine smooth muscle the autonomic movement was slightly increased by the much higher doses of 5-10 mg/ml. Blood: In vitro osmotic fragility of rabbit RBC suspension was not affected by cefoperazone of $1{\sim}10mg/ml$. Doses of 7.5 and 10 mg/ml were associated with 11.8% and 15.3% prolongation of whole blood coagulation time. Liver and kidney function: When measured at 3 hours after single intravenous injections of cefoperaonze in rabbits, the values of serum GOT, GPT, Bilirubin, TTT, BUN and creatine were not significantly different from the normal control. 5) Safety evaluation Acute toxicity: The acute toxicity of cefoperazone was studied following intraperitoneal and intravenous injections to mice(A strain, 4 week old) and rats(Sprague-Dawler, 6 week old). The LD_(50)'s of intraperitonealy injected cefoperazone were 9.7g/kg in male mice, 9.6g/kg in female mice and over 15g/kg in both male and female rats. And when administered intravenously in rats, LD_(50)'s were 5.1g/kg in male and 5.0g/kg in female. Administrations of the high doses of the drug were associated with slight inhibition of spontaneous movement and convulsion. Atdominal transudate and intestinal hyperemia were observed in animals administered intraperitonealy. In rats receiving high doses of the drug intravenously rhinorrhea and pulmonary congestion and edema were also observed. Renal proximal tubular epithelial degeneration was found in animals dosing in high concentrations of cefoperazone. Subacute toxicity: Rats(Sprague-Dawley, 6 week old) dosing 0.5, 1.0 and 2.0 g/kg/day of cefoperazone intraperitonealy were observed for one month and sacrificed at 24 hours after the last dose. In animals with a high dose, slight inhibition of spontaneous movement was observed during the experimental period. Soft stool or diarrhea appeared at first or second week of the administration in rats receiving 2.0g/kg. Daily food consumption and weekly weight gain were similar to control during the administration. Urinalysis, blood chemistry and hematology after one month administration were not different from control either. Cecal enlargement, which is an expected effect of broad spectrum antibiotic altering the normal intestinal microbial flora, was observed. Intestinal or peritoneal congestion and peritonitis were found. These findings seemed to be attributed to the local irritation following prolonged intraperitoneal injections of hypertonic and acidic cefoperazone solution. Among the histopathologic findings renal proximal tubular epithelial degeneration was characteristic in rats receiving 1 and 2g/kg/day, which were 10 and 20 times higher than the maximal clinical dose (100 mg/kg) of the drug. 6) Human pharmacokinetics Serum concentrations and urinary excretion were determined following a single intravenous injection of 1g cefoperazone in eight healthy, male volunteers. Mean serum concentrations of 89.3, 61.3, 26.6, 12.3, 2.3, and $1.8{\mu}g/ml$ occured at 1,2,4,6,8 and 12 hours after injection respectively, and the biological half-life was 108 minutes. Urinary excretion over 24 hours after injection was up to 43.5% of administered dose.

  • The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure. The present study suggests a correlation between regions of the low wall shear stress and the development of anastomotic neointimal fibrous hyperplasia(ANPH) in end-to-end anastomoses. 30523 T00401030523 ^x Air pressure decay(APD) rate and ultrafiltration rate(UFR) tests were performed on new and saline rinsed dialyzers as well as those roused in patients several times. C-DAK 4000 (Cordis Dow) and CF IS-11 (Baxter Travenol) reused dialyzers obtained from the dialysis clinic were used in the present study. The new dialyzers exhibited a relatively flat APD, whereas saline rinsed and reused dialyzers showed considerable amount of decay. C-DAH dialyzers had a larger APD(11.70$\pm$1.32mmHg/min)compared to CF dialyzers(4.32$\pm$0.55mmHg/min)(p<0.05). However, there was no observable difference in the UFR between the two dialyzers. Neither APD nor UFR showed any significant increase with an increasing number of reuses for up to more than 20reuses. A substantial number of failures observed in APD(larger than 20mmHe/min)on the reused dialyzers(2 out of 40 CP and S out 26 C-DAK) were attributed to the Possible damage on the fibers. The CF 15-11 HFDs which failed APD test did not show changes in the UFR compared to normal dialyzers indicating that APD is a more sensitive test than UFR test to evaluate the integrity of the fibers. 30527 T00401030527 ^x For quantitative measurement of reflected light from a clinical diagnostic strip, a prototype old reflectance photometer was designed. The strip loader and cassette were made to obtain more accurate reflectance parameters. The kubelka-munk coefficient and reflection optical density were determined ar four different wavelengths(500, 550, 570 and 610nm) for blood glucose strip. The mice were divided into two groups. One group was injected with Photofrin II, intravenously (2.5mg/kg body weight). The other group received no Photofrin II. Mice from both groups were irradialed for approximately 15 minutes at 100, 300, or 500 mW/cm2 with the argon (488nm/514.5 nm), dye(628nm) and gold vapor (pulsed 628 nm) laser light. A photosensitizer behaved as an added absorber. Under our experimental conditions, the presence of Photolfrin II increased surface temperature by at least 40% and the temperature rise due to 300 mW/cm2 irradiation exceeded values for hyperthermia. Light and temperature distributions with depth were estimated by a computer model. The model demonstrated the influence of wavelength on the thermal process and proved to be a valuable tool to investigate internal temperature rise. 30536 T00401030536 ^x We investigated the structural geometry of thirty-eight Korean femurs. The purpose of this study is to identify major geometrical differences between Korean femurs 3nd others that we believe belong to Caucasians so that we would be able to get insights into the femoral component design that fits Asians including Koreans. We utilized computerized tomography (CT) images of femurs extracted from cadavers. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data. The major results show that the geometry of the Korean femurs is significantly different from that of Caucasians: (1) the anteversion angle and the canal flare index are greater by the amount of approximately 8˚ and 0.5, respectively, (2) the shape of the isthmus cross section is more round, and (3) the distance between the teaser trochanter and the proximal border of the isthmus is shelter by about 15 mm. The results suggested that the femoral component suitable for Asians should be different from the currently-used components designed and manufactured mostly by European or American companies. 30537 T00401030537 ^x It is well known that nonlinear propagation characteristics of the wave in the tissue may give very useful information for the medical diagnoisis. In this paper, a new method to detect nonlinear propagation characteristics of the internal vibration in the tissue for the low frequency mechanical vibration by using bispectral analysis is proposed. In the method, low frequency vibration of f0( = 100Hz) is applied on the surface of the object, and the waveform of the internal vibration x (t) is measured from Doppler frequency modulation of silmultaneously transmitted probing ultrasonic waves. Then, the bispectra of the signal x (t) at the frequencies (f0, f0) and (f0, 2f0) are calculated to estimate the nonlinear propagation characteristics as their magnitude ratio, w here since bispectrum is free from the gaussian additive noise we can get the value with high S/N. Basic experimental system is constructed by using 3.0 MHz probing ultrasonic waves and the several experiments are carried out for some phantoms. The operating system and database management system were SCO UNIX and SCO foxbase, respectively. For program development, we used Xbase language provided by SCO foxbase. The C language was used for interface purpose. To make the system use friendly, pull-down menu was used. The system connected to our hospital information system via application program interface (API), so the information related to patient and request details is automatically transmitted to our computer. Our system interfaced with fwd complete blood count analyzers(Sysmex NE-8000 and Coulter STKS) for unidirectional data tansmission from analyzer to computer. Because the point of maximun temperature of laser-irradiated gelatin exists below the surface due to surface cooling, evaporation at the boiling temperature is made explosively from below the surface. The important parameters of this process are the conduction loss to laser power absorption (defined as the conduction-to-laser power parameter, Nk), the convection heat transfer at the surface to conduction loss (defined as Bi), dimensionless extinction coefficient (defined as Br.), and dimensionless irradiation time (defined as Fo). Dependence of Fo on Nk and Bi has been observed by experiment, and the results have been compared with the numerical results obtained by solving a 2-dimensional conduction equation. Fo and explosion depth (from the surface to the point of maximun temperature) are increased when Nk and Bi are increased.To find out the minimum laser power for explosive evaporative removal process, steady state analysis has been also made. We obtained a two-dimensional current density distribution within a phantom by processing the real and imaginary MR images from a 4.77 NMR machine. Optimal stimulus parameters, which can minimize muscle fatigue and can achieve effective muscle contraction were determined in slow and fast muscles in Sprague-Dawley rats. Stimulus patterns of our stimulator were designed to simulate electromyographic activity monitored during locomotion of normal subjects. Muscle types of the lower extremity were classified according to their mechanical property of contraction, which are slow muscle (msoleus m.) and fast muscle (medial gastrocneminus m., rectus femoris m., vastus lateralis m.). Optimal parameters of electrical stimulation for slow muscles were 20 Hz, 0.2 ms square pulse. For fast muscle, 40 Hz, 0.3 ms square pulse was optimal to produce repeated contraction. Higher stimulus intensity was required when synergistic muscles were stimulated simultaneously than when they were stimulated individually. Electrical stimulation for each muscle was designed to generate bipedal locomotion, so that individual muscles alternate contraction and relaxation to simulate stance and swing phases. Portable electrical stimulator with 16 channels built in microprocessor was constructed and applied to paraplegic patients due to lumbar cord injury. The CT images were transformed into bitmap data by using a film scanner, and then analyzed by using a commercially available software called Image v.1.0 and a Macintosh IIci computer.The resulting data were compared with already published data.
