Effect of Arsenic on Immunity, Oxidative Enzyme and Various Hematological Parameters in Cross Bred Calves

  • An experiment was conducted on crossbred male calves to study the effect of arsenic (As) on immunity status and certain hematological parameters. Ten crossbred male calves of 3-4 months of age were distributed into two equal groups. Group I was kept as control, whereas, group II was supplemented daily with 50 ppm As (as $As_sO_3$) up to 90 days, in the diet. Calves of both groups were fed as per ICAR standards and their requirements were fulfilled by feeding concentrate mixture and green oats. All calves were kept under similar managemental conditions. Blood samples were collected at fortnightly intervals to estimate various haematological parameters and superoxide dismutase (SOD) enzyme activity. Serum Ig and serum glutamic pyruvate transaminase (SGPT) were also measured. Cell-mediated immune responses of the calves were monitored at 0, 45 and 90 of experimental feeding, through lymphocyte proliferation. No change in blood total leukocyte counts (TLC), differential leukocyte counts (DLC), packed cell volume (PCV), haemoglobin (Hb) and SGPT was observed with As supplementation. A decrease in SOD activity was noticed in group II calves. Stimulation index (SI) for lymphocyte proliferation decreased from 1.14 to 0.79 in group II calves during 90 days experimental feeding, whereas, there was no change in SI values in group I indicating significant decrease in immune response of As supplemented calves. Blood As concentration increased in group II calves with the decrease in immune response. Short term supplementation of As to growing calves suggested suppressive effects on cell-mediated immunity. However, long term experiments are required to demonstrate clearly the efects of this toxic metal in calves.

  • Extracellular nuclease(s) in buffalo rumen fluid were purified from strained rumen fluid by a procedure involving Seitz filtration, acetone fractionation and gel filtration on Sephadex G-100. The enzyme resolved into two peaks exhibiting both DNase and RNase activities. The molecular weight of enzyme corresponding to peaks I and II were approximately 30,000 and 12,000 respectively. The properties of enzymes from the two peaks, however, were same. Optimum temperature for both DNase and RNase activities was at $50^{\circ}C$. Whereas DNase activity was stable upto $60^{\circ}C$, RNase activity was stable only up to $50^{\circ}C$. DNase activity recorded two pH optima, one at pH 5.5 and the other at pH 7.0. RNase activity recorded a broad pH optimum between pH 6.0-8.0. pH stability of the enzyme coincided with pH optima for both the activities. DNase activity was stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. RNase activity was also stimulated by $Mg^{2+}$ and $Mn^{2+}$ and inhibited by $Cu^{2+}$, $Fe^{2+}$, $Zn^{2+}$, $Hg^{2+}$ and $Ag^+$. Reducing agents stimulated both the activities.

GC-MS Analysis of Diterpene Quinone Constituents of Salviae Miltiorrhizae Radix and Biological Activity

  • The ether extract of Salviae miltiorrhizae Radix(SMR) was fractioned to give five subfractions, so that two subfractions of them were recrystallized to yield each pure diterpene quinone pigment. On the basis of spectral evidence, these two compounds were identified as tanshinone II and crytotanshinone. Cryptotanshinone exhibited both of a potent platelet anti-aggregating activity in vitro and a potent antimicrobial activity. GC-MS analysis of the other extract showed that tanshinone II was contained in the largest proportion of all the diterpene quinones. In addition, GC-MS analysis gave other valuable analytical informations.

국내산 이탈리안 라이그라스 사일리지와 열풍 건초 위주의 TMR 급여가 홀스타인 착유우의 반추 활동량, 우유 생산성 및 경제성 분석에 미치는 영향 (Effects of TMR Feeding Using Italian Ryegrass Silage and Hay of Those Dried by Hot-Air Dry System on Rumination Activity, Milk Productivity and Economic Feasibility in Holstein Dairy Cows)

  • 본 연구는 수입산 티모시 건초, 국내산 IRG 사일리지 및 열풍 건초 IRG를 이용한 TMR의 혼합 급여가 착유우의 유 생산량, 유질, 반추 활동 및 조사료 별 경제성 비교 분석 등을 통해 국내산 IRG 사일리지와 열풍 건초 국내산 IRG의 사료가치 평가를 수행하였다. 체중은 차이를 보이지 않았으며, 반추위 활동 시간과 온도는 정상범위로 확인되었다. 우유 내 지방, 단백질 및 유당은 차이를 보이지 않았으며, 체세포수는 1등급의 범위를 보였다. 경제성 분석 결과, 수입산 티모시에 비해 국내산 IRG 사일리지와 열풍 건초 IRG를 이용한 TMR 급여에서 두당 402.8원/일과 331.4원/일의 수익이 증가할 것으로 생각된다. 그러므로 국내산 IRG 사일리지와 열풍 건초 국내산 IRG는 수입산 티모시 건초를 대체하여 착유우에 이용될 수 있을 것으로 사료된다.

Antibacterial Activity of Crude Aronia melanocarpa (Black Chokeberry) Extracts against Bacillus cereus, Staphylococcus aureus, Cronobacter sakazakii, and Salmonella Enteritidis in Various Dairy Foods: Preliminary Study

  • In this study, the antibacterial activity of Aronia melanocarpa (black chokeberry) ethanol extract against Bacillus cereus, Staphylococcus aureus, Cronobacter sakazakii, and Salmonella Enteritidis was investigated using the spot-on-lawn assay. The results showed that this extract exhibited antibacterial activities against Bacillus cereus (complete inhibition) and Staphylococcus aureus (partial inhibition), but did not inhibit the growth of Cronobacter sakazakii and Salmonella Enteritidis. This study shows that the Aronia melanocarpa (black chokeberry) ethanol extract was more effective against Gram-positive bacteria than Gram-negative bacteria. Hence, it is suggested that Aronia melanocarpa could be a useful food supplement, and could be utilized as a naturally derived additive for maintaining the safety of various dairy products. Furthermore, future research should be conducted to examine the possibility of using such products as functional ingredients for improving the anti-oxidative and anti-inflammatory activities of food products.

  • The roots of Cichorium intybus L. (chicory) have many nutritional qualities and also have been used for health promotion as traditional medicine. The antimicrobial activity of Cichorium intybus L.'s (chicory) ethanol extract on Bacillus cereus ATCC 10876 were investigated using spot on lawn method in this study. Ethanol extract exhibited antimicrobial activities against Bacillus cereus ATCC 10876 with significance. Also, the results produced the larger zones of inhibition against Bacillus cereus ATCC 10876 tested, with increasing concentration of Cichorium intybus L.'s (chicory) ethanol extracts,. Hence, Cichorium intybus L. (chicory) could be served for keeping the safety of various dairy products as natural antimicrobial agents.

Lipase-producing Filamentous Fungi from Non-dairy Creamer Industrial Waste

  • Lipase-producing fungi have been isolated from environments containing lipids. The non-dairy creamer industrial waste has a high amount of lipids so it is a potential source for the isolation of lipase-producing fungi. However, the study of fungi that secrete lipase from this industrial waste has not been reported. The purpose of this study was to obtain lipase-producing filamentous fungi from non-dairy creamer industrial waste. Mineral salt and potato dextrose agar were used as media for the isolation process. The qualitative screening was conducted using phenol red agar medium and the quantitative screening using broth medium containing glucose and olive oil. Isolates producing the highest amounts of lipase were identified with molecular methods. We found that 5 out of 19 isolated filamentous fungi are lipase producers. Further analysis showed that isolate Ms.11 produced the highest amount of lipase compared to others. Based on ITS sequence Ms.11 was identified as Aspergillus aculeatus. The lipase activity in medium containing 1% glucose + 1% olive oil at pH 7.0 and 30℃ after 96 and 120 h of incubation was 5.13 ± 0.30 U/ml and 5.22 ± 0.59 U/ml, respectively. The optimum lipase activity was found at pH 7.0, 30℃ and using methanol or ethanol in the reaction tube. Lipase was more stable at 20-30℃ and maintained 85% of its activity. It was concluded that isolate Ms.11 is a potential source of lipase that catalyzes transesterification reactions. Further studies are required to optimize lipase production to make the strain suitable for industry purposes.

발효유제품에서 박테리오파지의 특성, 검출과 제어 (The Characteristics, Detection and Control of Bacteriophage in Fermented Dairy Products)

  • This study was to review the classification, detection and control of bacteriophage in fermented dairy products. Bacteriophage has lytic and/or lysogenic life cycles. Epidemiologically speaking, detected major phages are c2, 936 and p335. Among them p335 has been the largest concern in dairy industry. Traditionally, various analytical technologies, such as spot, starter activity, indicator test, ATP measurement and conductimetric analysis, have been used for the phage detection. In recent years, advanced methods such as flow cytometric method, petrifilm, enzyme linked immunosorbent assay (ELISA) and multiflex PCR diagnostic kit have been deveoloped. The phage contamination has been controlled by using heat, high-pressure treatment, and the combinations of heat and pressure, and/or chemical. Also some starter cultures with phage-resistant character have been developed to minimize the concentration of phages in dairy product. Bacteriophage inhibition media such as calcium medium was also mentioned. To prevent the contamination of bacteriophage in dairy industry, further researches on the detection and control of phage, and phage resistant starters are necessary in the future.

Lactobacillus gasseri 505를 사용한 꾸지뽕잎 추출물 첨가 신바이오틱스 발효유의 면역활성 조절 연구 (Immunomodulatory Effects of Fermented Milk based on Synbiotic Interaction between Cudrania tricuspidata Leaf Extract and Lactobacillus gasseri 505)

  • This study examined the immunomodulatory effects of milk fermented with based on the synbiotic interaction between Cudrania tricuspidata leaf extract (CT) and Lactobacillus gasseri 505. Raw 264.7 macrophages were treated with milk (Control), milk supplemented with 0.2% CT (CT-milk), or CT-milk fermented with L. gasseri 505 (FCT). FCT-treated cells showed higher NO production than the other groups. ELISA revealed that FCT produced the highest enhancement of the production of pro-inflammatory cytokines such as $TNF-{\alpha}$, $IFN-{\gamma}$, $IL-1{\beta}$, IL-6, IL-2, IL-12, and IL-10. Furthermore, FCT also increased the gene expression of pro-inflammatory mediators such as iNOS and COX-2 in a concentration-dependent manner. Collectively, these results indicate that the synbiotic interaction between CT and L. gasseri 505 might enhance their immunomodulatory effects of fermented milk.

Reproductive Performance of Dairy Buffaloes Supplemented with Varying Levels of Vitamin E

  • The effect of vitamin E supplementation on plasma ${\alpha}$-tocopherol level, total antioxidant level and reproductive performance in Murrah buffaloes was studied during periparturient period. Twenty-four advance pregnant buffaloes were randomly divided into four equal groups as $T_1$, $T_2$, $T_3$ and $T_4$ and were supplemented with 0, 1,000, 1,500 and 2,000 IU of ${\alpha}$-tocopheryl acetate (Merck) from 60 days prepartum to 30 days postpartum and 0, 500, 750 and 1,000 IU from 30 to 60 days postpartum, respectively. Blood samples were collected at -60, -45, -30, -15, -7, 0, 7, 15, 30 and 60 days of parturition and were analyzed for plasma ${\alpha}$-tocopherol and total antioxidant activity (TAA). The intake of DM, CP and TDN did not vary among different groups. Plasma ${\alpha}$-tocopherol and TAA around parturition (-7 to 15 day) in $T_3$ and $T_4$ were significantly higher than the control group. There was 17% reduction in retention of fetal membranes (RFM) and metritis in $T_4$ than control. The post partum estrus interval averaged 58.00, 55.33, 51.83 and 43.00 days in $T_1$, $T_2$, $T_3$ and $T_4$ respectively. There was significant reduction in days open in both $T_3$ and $T_4$ in comparison to $T_1$ group (127,130 Vs.146). All the vitamin E supplemented groups showed reduction in days open than their previous lactation performance. Supplementation of vitamin E at $1,500IU\;d^{-1}$ from 60 day prepartum to 30 day post partum to buffaloes exhibited beneficial effect on plasma ${\alpha}$-tocopherol level and TAA around parturition and continuation of its supplementation at $1,000IU\;d^{-1}$ from 30 to 60 days of lactation improved post partum reproductive performance of buffaloes.
