Extraction of Total Flavonoids from Lemongrass Using Microwave Energy: Optimization Using CCD-RSM (마이크로웨이브 에너지를 이용한 레몬그라스로부터 플라보노이드 성분의 추출: CCD-RSM을 이용한 최적화)

  • In this study, we measured total flavonoids after extracting the total flavonoids from lemongrass which is known to have a high content of antioxidant ingredients when using microwave energy. Also, optimal extraction conditions of active ingredients using central composite design-response surface methodology (CCD-RSM) were presented. Both ultrapure water and alcohol were used as extraction solvents and the volume ratio of ethanol/ultrapure water, microwave irradiation time, and microwave irradiation power were set as independence variables. And the extraction yield and total flavonoids were measured. The optimal extraction conditions using CCD-RSM were the volume ratio of ethanol/ultrapure water = 56.3 vol.%, the microwave irradiation time = 6.1 min, and the microwave irradiation power = 574.6 W. We could also obtain expected results of yield = 17.2 wt.% and total flavonoids = 44.7 ㎍ QE/mL dw under the optimum conditions. The comprehensive satisfaction degree of this formula was 0.8562. The P-value was calculated for the yield of 0.037 and the total flavonoids content of 0.002. The average error from actual experiments established for the verification of conclusions was lower than 2.5%. Therefore, a high favorable level could be obtained when the CCD-RSM was applied to the optimization of extraction process.

Study on safety performance evaluation of stationary SOFC stack (건물용 고체산화물연료전지 스택 안전성능평가 연구)

  • The code and standards related to fuel cells were analyzed to derive the SOFC(Solid Oxide Fuel Cell) stack safety performance evaluation items and evaluation methode. Safety performance evluation of the SOFC stack was tested by quoting derived test items. The stack used in the test is an anode-supported type 2 Cell stack (Active surface area : 220cm) manufactured by MICO Inc, and SOFC stack safety performance evaluation system used for the test is self-manufactured. We conducted a leakage test, current voltage characteristic test, rated output test, and power response characteristics test. In the safety performance evaluation test, the stack showed no gas leakage, the maximum output and rated output was recorded to 65.6 W(1.41 V, 46.5 A, $422mA/cm^2$), 62.3 W(1.57 V, 40 A, $363mA/cm^2$). In the power response characteristics test verified that the output is kept stable within two seconds. At the maximum load (40 A) and the minimum load (8 A), the output was recorded 62 W and 16W in $750^{\circ}C$. This study will contribute to the universalization and to provide much safe environment of operating the solid oxide fuel cell system.

Electrochemical Analysis of CuxCo3-xO4 Catalyst for Oxygen Evolution Reaction Prepared by Sol-Gel Method (Sol-Gel법을 이용한 CuxCo3-xO4 산소 발생 촉매의 합성 및 전기화학 특성 분석)

  • Transition metal oxide is widely used as a water electrolysis catalyst to substitute for a noble metal catalyst such as $IrO_2$ and $RuO_2$. In this study, the sol-gel method is used to synthesize the $Cu_xCo_{3-x}O_4$ catalyst for the oxygen evolution reaction (OER),. The CuxCo3-xO4 is synthesized at various calcination temperatures from $250^{\circ}C$ to $400^{\circ}C$ for 4 h. The $Cu_xCo_{3-x}O_4$ synthesized at $300^{\circ}C$ has a perfect spinel structure without residues of the precursor and secondary phases, such as CuO. The particle size of $Cu_xCo_{3-x}O_4$ increases with an increase in calcination temperature. Amongst all the samples studied, $Cu_xCo_{3-x}O_4$, which is synthesized at 300?, has the highest activity for the OER. Its onset potential for the OER is 370 mV and the overpotential at $10mA/cm^2$ is 438 mV. The tafel slope of $Cu_xCo_{3-x}O_4$ synthesized at $300^{\circ}C$ has a low value of 58 mV/dec. These results are mainly explained by the increase in the available active surface area of the $Cu_xCo_{3-x}O_4$ catalyst.

A Study on the Radiometric Correction of Sentinel-1 HV Data for Arctic Sea Ice Detection (북극해 해빙 탐지를 위한 Sentinel-1 HV자료의 방사보정 연구)

  • Recently, active research on the Arctic Ocean has been conducted due to the influence of global warming and new Arctic ship route. Although previous studies already calculated quantitative extent of sea ice using passive microwave radiometers, melting at the edge of sea ice and surface roughness were hardly considered due to low spatial resolution. Since Sentienl-1A/B data in Extended Wide (EW) mode are being distributed as free of charge and bulk data for Arctic sea can be generated during a short period, the entire Arctic sea ice data can be covered in high spatial resolution by mosaicking bulk data. However, Sentinel-1A/B data in EW mode, especially in HV polarization, needs significant radiometric correction for further classification. Thus, in this study, we developed algorithms that can correct thermal noise and scalloping effects, and confirmed that Arctic sea ice and open-water were well classified using the corrected dual-polarization SAR data.

The Need and the Direction to Improve the System of Measures for the Preservation of Buried Cultural Heritage (매장문화재 보존조치 제도의 개선 필요성과 방향)

  • After inspecting ground surface or excavating cultural heritage site, the government can take measures for the preservation of buried cultural heritage based on the related law. It means that the provisions complement the limitation of current cultural heritage management system by extending the scope of cultural heritage to be preserved. But we neither have set any matters about implementing the measures for preservation of the buried cultural heritage, nor manage the heritage and its surroundings after implementing the measures. Due to these insufficiency of the law, there arise several problems in the field. For example, the measures for preservation are not complete, or preserved cultural heritage is damaged due to inappropriate management. We have to resolve the problems in order to accomplish the original purpose of taking measures for preservation of the heritage. First, it is necessary to make sure of the legal status of preserved buried cultural heritage, to establish the standards to decide whether to preserve it or not. We need to have regulations by law or internal rule on the daily management, who should manage it, what and how the manager should manage. It is also important for local residents to take an active part in preserving and utilizing the cultural heritage as the owner. Through building up the foundation mentioned above, the meaning and value of preserved buried cultural heritage can be expanded widely.

A Preliminary Study on Micro-earthquakes Occurred from 2010 to 2017 in Busan, Korea (2010-2017년 부산지역의 미소 지진 예비 탐색)

  • Although the knowledge of current seismicity is a critical information for making and implementing effective earthquake-related policy, the detailed seismicity information of the metropolitan areas with high-population density has been largely underestimated due to the high-level of cultural noise and small earthquake magnitude. This study presents 12 earthquakes including 2 earthquakes previously known and 10 additional earthquakes occurred from 2010 to 2017 in Busan, but they were unreported by the Korea Meteorological Administration. Matched filter technique is used to detect micro-earthquakes. Although the epicenters of micro-earthquakes though present a distinguished linearity, a correlation with faults in the area is unknown. A repeated micro-seismicity suggests that there are subsurface structures responsible for observed events. If large earthquakes occur along the fault in Busan, they may cause catastrophic natural disasters. Given the fact that the recent earthquakes did not accompany any surface signatures, it is highly recommended that the current micro-seismicity be investigated, and updated seismicity information be incorporated into establishing active fault maps in Korea.

Concepts of System Function and Modulation-Demodulation based Reconstruction of a 3D Object Coordinates using Active Method (시스템 함수 및 변복조 개념 적용 능동 방식 3차원 물체 좌표 복원)

  • In this paper we propose a novel approach to representation of the 3D reconstruction problem by employing a concept of system function that is defined as the ratio of the output to the input signal. Akin to determination of system function (or system response), this paper determines system function by choosing (or defining) appropriate input and output signals. In other words, the 3D reconstruction using structured circular light patterns is reformulated as determination of system function from input and output signals. This paper introduces two algorithms for the reconstruction. The one defines the input and output signals as projected circular light patterns and the images overlaid with the patterns and captured by camera, respectively. The other one defines input and output signals as 3D coordinates of the object surface and the image captured by camera. The first one leads to the problem as identifying the system function and the second one leads to the problem as estimation of an input signal employing concept of modulation-demodulation theory. This paper substantiate the proposed approach by providing experimental results.

Comparative study of thermal gelation properties and molecular forces of actomyosin extracted from normal and pale, soft and exudative-like chicken breast meat

  • Objective: The objectives of this study were to investigate the thermal gelation properties and molecular forces of actomyosin extracted from two classes of chicken breast meat qualities (normal and pale, soft and exudative [PSE]-like) during heating process to further improve the understanding of the variations of functional properties between normal and PSE-like chicken breast meat. Methods: Actomyosin was extracted from normal and PSE-like chicken breast meat and the gel strength, water-holding capacity (WHC), protein loss, particle size and distribution, dynamic rheology and protein thermal stability were determined, then turbidity, active sulfhydryl group contents, hydrophobicity and molecular forces during thermal-induced gelling formation were comparatively studied. Results: Sodium dodecyl sulphate-polyacrylamide gel electrophoresis showed that protein profiles of actomyosin extracted from normal and PSE-like meat were not significantly different (p>0.05). Compared with normal actomyosin, PSE-like actomyosin had lower gel strength, WHC, particle size, less protein content involved in thermal gelation forming (p<0.05), and reduced onset temperature ($T_o$), thermal transition temperature ($T_d$), storage modulus (G') and loss modulus (G"). The turbidity, reactive sulfhydryl group of PSE-like actomyosin were higher when heated from $40^{\circ}C$ to $60^{\circ}C$. Further heating to $80^{\circ}C$ had lower transition from reactive sulfhydryl group into a disulfide bond and surface hydrophobicity. Molecular forces showed that hydrophobic interaction was the main force for heat-induced gel formation while both ionic and hydrogen bonds were different significantly between normal and PSE-like actomyosin (p<0.05). Conclusion: These changes in chemical groups and inter-molecular bonds affected protein-protein interaction and protein-water interaction and contributed to the inferior thermal gelation properties of PSE-like meat.

Application of Experimental Design Methods for Minimum Weight Design and Sensitivity Evaluation of Passive-Type Deck Support Frame for Offshore Plant Float-Over Installation (해양플랜트 플로트오버 설치 공법용 수동형 갑판 지지 프레임의 최소중량설계와 민감도 평가를 위한 실험계획법 응용)

  • This paper presents the findings of a comparative study on minimum weight design and sensitivity evaluation using different experimental design methods for the structural design of an active-type deck support frame (DSF) developed for the float-over installation of an of shore plant topside. The thickness sizing variables of the structural members of a passive-type DSF were considered the design factors, and the output responses were defined using the weight and strength performances. The design of the experimental methods applied in the comparative study of the minimum weight design and the sensitivity evaluation were the orthogonal array design, Box- Behnken design, and Latin hypercube design. A response surface method was generated for each design of the experiment to evaluate the approximation performance of the design space exploration according to the experimental design, and the accuracy characteristics of the approximation were reviewed. Regarding the minimum weight design, the design results, such as numerical costs and weight minimization, of the experimental design for the best design case, were evaluated. The Box- Behnken design method showed the optimum design results for the structural design of the passive-type DSF.

Synthesis and Characterization of Zeolite Using Water Treatment Sludge (정수슬러지를 이용한 제올라이트의 합성 및 특성연구)

  • Zeolite was synthesized hydrothermally using the water-treatment sludge, and the effects of various synthesis parameters like reaction temperature, reaction time, and Na2O/SiO2 molar ratio on the crystallization of zeolite were investigated. Crystal structure, physical property, and thermal stability of zeolite crystals were characterized by X-ray powder diffraction, FTIR spectroscopy, BET nitrogen adsorption, and TGA measurements. The removal efficiencies of nitrogen in ammonia, heavy metal ions, and TOC were calculated to evaluate zeolite's adsorption capacity. The primary chemical composition of water-treatment sludge was 28.79% Al2O3 and 27.06% SiO2. The zeolites were synthesized by merely employing the water-treatment sludge as silica and alumina sources without additional chemicals. Zeolite crystals synthesized through the water-treatment sludge were confirmed as an A-type zeolite structure. Zeolite A had the highest crystallinity obtained from a gel with the molar composition 2.1Na2O-Al2O3-1.6SiO2-65H2O after 5 h at a temperature of 90 ℃. The specific surface area of zeolite obtained was 55 ㎡ g-1, which was higher than commercial zeolite A. The removal efficiency of nitrogen in ammonia was 68% after 3 h of reaction time, while the removal efficiencies of Pb2+ and Cd2+ ions were 99.1% and 99.3%, respectively. These results indicate active ion exchange between Pb2+ or Cd2+ ion and Na+ ion in the zeolite framework. The adsorption experiments on the different zeolite addition conditions were performed for 3 h with 300 ppm humic acid. Based on the results, TOC's highest efficiency was 83% when 5 g of zeolite was added.
