Controls of the Hydrolysis of Ginseng Saponins by Neutralization of Organic Acids in Red Ginseng Extract Preparations (홍삼의 가열추출 과정중 유기산 중화에 의한 사포닌의 가수분해 억제)

  • Glucosidic bonds at the C20 position of the sapogenins were hydrolyzed easily in the lower pH, higher temperatures and longer times to give prosapogenins and sugars. The glucosidic bond of saponin at the C3 of ginsenoside-Rb1, which is secondary carbon, was relatively stable due to the low electron density of -0.2. But the bond of saponin at the C20 position, which is tertiary carbon with the relatively high electron density of -0.3, was liable to be hydrolyzed even in weakly acidic solution by the increase of heating time. On the other hand, red ginseng contained 13.34 mg/g of citric acid, 8.78 mg/g of malonic acid, 3.70 mg/g of oxalic acid, 2.13 mg/g of malic acid and 0.44 mg/g of succinct acid. Ginseng saponins were very stable in ginseng extract neutralized with sodium carbonate or sodium bicarbonate corresponding to the equivalent amount of the total organic acid in the red ginseng.

  • Great amounts of solid radioactive waste (second waste) and waste solution are generated from the remediation of uranium-contaminated soil. To reduce these, we investigated washing with a less acidic solution and recycling the waste solution after removal of the dominant elements and uranium. Increasing the pH of the washing solution from 0.5 to 1.5 would be beneficial in terms of economics. A high content of calcium in the waste solution was precipitated by adding sulfuric acid. The second waste can be significantly reduced by using sorption and desorption techniques on ampholyte resin S-950 prior to the precipitation of uranium at pH 3.0.

The Study on the Mordanting and Dyeing Properties of Sophora Japonica L. (괴화의 염색성에 관한 연구)

  • This paper surveys the mordanting and dyeing properties of Sophora japonica L. Appropriate extraction, dyeing and mordanting condition of Sophora japonica L. were determined, and the effect of mordanting method on dye uptake and color fastness of dyed fabric was investigated. The maximum absorbance of Sophora japonica L. solution was 367.6mn, rutin solution was 365.6mn. The color of Sophora japonica L. solution was affected at pH 2 and pH 8~9. The optimum temperature to extract Sophora japonica L. was during 1 hour in 8$0^{\circ}C$. The effective dyeing temperature and time of silk were 10$0^{\circ}C$, 60min. K/S value of dyeing fabrics was increased by pre-mordanting treatment, especially Fe, Sn, Cr. In the case of Sophora japonica L. light fastness was increased by Fe mordanting. Perspiration fastness was better in acidic solution than that in alkaline solution. Fastness to rubbing and dry-cleaning were good in general.

  • Streptozotocin, which is a naturally occurring nitrosoamide used extensively to produce diabetes mellitus in experimental animals, has been known to be chemically stable only under acidic condition (pH 4). The present study was conducted to determine whether its degradation products formed under various conditions can induce hyperglycemia in female rats. Streptozotocin in phosphate buffer saline (pH 7.4) resulted in spontaneous degradation rapidly. Furthermore, streptozotocin incubated with plasma isolated from rats was degraded even faster than those in neutral buffer. When streptozotocin was administered iv to rats, significant elevations in blood glucose level were observed within 24 hours. However, rats treated with equimolar concentration of streptozotocin degradation products under the phosphate buffer saline as well as the plasma did not lead to hyperglycemia. These results suggest that, when streptozotocin administered undergo spontaneous breakdown in vivo, its degradation Products do not induce the hyperglycemia in rats.

  • Dispersion of carbon nanotubes in biocompatible media are of particular interest for diverse biomedical and nanomedicine applications. Various biomolecules and biopolymers such as DNA, proteins, poly L-lysine, starch, gelatin, steroid biosurfactants, and chitosan have shown capability for the effective dispersion of carbon nanotubes in water. Chitosan has demonstrated capacity for effective dispersion of single-walled carbon nanotubes (SWCNTs) in acidic medium and it also showed tendency to preferentially disperse smaller diameter nanotubes. Chemical functionalizations of chitosan enable its solubility in neutral pH water by reducing the intra and inter molecular hydrogen bonding. Herein, we present a neutral pH water soluble chitosan derivative, chitosan-hydroxyphenyl acetamide (CHPA), obtained by functionalizing the amino groups of chitosan with 4-hydroxyphenyl acetic acid, as an efficient biocompatible dispersant for debundling and solubilization of SWNTs in neutral aqueous solutions. Various process conditions for individual dispersion of SWCNTs are analyzed based on optical absorption and Raman spectroscopy.

  • The effect of soil forming factors on the pedogenesis of basaltic volcanic ash soils and the influence of allophane material on soil properties have been investigated on 5 chronosequence soils situated from at the near sea coast up to the foot slope of Mt. Halla in Jeju Island. Time seems to be the important soil forming factor which today differentiates soil of the Island. Songag and Donghong soils developed in lower elevations are older and somewhat less influenced by ash shower. However, soils developed at higher elevations, Pyeongdae and Heugag, are rather younger and strongly influence by the ash. It is also proved that the parent materials are very heterogeneous. They mainly are basaltic with some contamination of acidic volcanic ashes and continental aeolian deposits where a considerable amount of quartz encountered in most soils studied. Many physico-chemical properties of soil, such NaF pH, phosphate sorption power, pH and extractable acidity are parameters to differentiate andepts and non-andeptic soils.

  • Dyeing and fastness characteristics of 100% meta-aramid fiber were investigated with cationic dyes and swelling agents under various dyeing conditions such as dyeing temperature and pH of dye bath. Dye exhaustion started at around $80^{\circ}C$ and settled down at $130^{\circ}C$. Among swelling agents used, N-methyl formanilide showed comparatively higher K/S values comparing to 1-phenoxypropan-2-ol. Under weak acidic conditions in the range pH 5 to 7, the exhaustion of cationic dyes could be enhanced leading to higher adsorption and stability of colorimetric property. Wash and rubbing fastness were generally good but low light fastness found can be attributable to the poor photo-stability of the cationic dyes.

  • A marine Streptomyces sp., which produce water-soluble blue pigment was isolated from shallow-sea muds. The effect of various nutritional conditions on growth of isolated strain were investigated to facilite the potential use of this organism in industry. The effect of carbon source was characterized as a 10g/1 of soluble starch. Growth was optimum at pH 7.0 and slightly affected with more alkaline range but was shown to decrease dramatically in acidic range. Under the optimum growth conditions, isolated strain produced substantial amounts of blue pigment and biomass.

  • The seeds of Zizyphus Jujuba have been used as an antianxiety agent for the treatment of insomnia from the earliest times. Sanjoinine-A, isolated from the seeds of Zizyphus Jujuba, have been found to have a minor tranquilizer activity. However this drug is poorly soluble in water. In order to increase the dissolution rate of sanjoinine-A, solid dispersions with PVP-MC and inclusion complex with ${\beta}-cyclodextrin$ were prepared and evaluated. All of these systems increased the dissolution rate of sanjoinine A comparing with sanjoinine-A free base. From pH-rate profile of sanjoinine-A at $60^{\circ}C$, it was found that sanjoinine A was relatively stable in acidic solution, but unstable in basic solution.

  • A bacteriocin-producing lactic acid bacteria was isolated from contaminated milk products, which was identified by using the API50 CH kit as Lactococcus lactis subsp. lactis with reliability of 98%. Fatty and analysia of the cell membrane showed that this strain contained same fatty acids profiles as type strain, Lactococcus lactis subsp. lactis ATCC 19435. The bacteriocin of Lactococcus sp. HY 449 showed relatively wide range of inhibition spectrum against gram positive and some gram negative bacteria such as Escherichia coli and maintained the inhibitory activity between pH2.0 and pH9.0 The thermostability of this bacteriocin was higher in acidic solution than in distilled water and was stable at 60$\circ $C for 1 hour.
