The Hepatoprotective Effects of Polysaccharides Isolated from Submerged Fermentation of Ganoderma lucidum

  • A neutral polysaccharide, GP, was isolated from a fermentation broth of Ganoderma lucidum Acid hydrolysis and a paper chromatography analysis indicated that the polysacchride was composed of glucose, xylose, and mannose. The molecular weight was estimated to be $2.9{\times}10^4$. The oral administration of GP to mice showed that it can inhibit liver damage induced by GalN and $CCl_4$.

Memory-Enhancing Effects of Silk Fibroin-Derived Peptides in Scopolamine-Treated Mice

  • Although enzyme-hydrolyzed silk fibroin has been reported to enhance cognitive function before, it has been still unknown which peptides can improve memory. Here we report that amino acid sequences of three novel peptides were identified from fibroin hydrolysate. Fibroin hydrolysate was obtained by hydrolysis with protease after partial hydrolysis with 5M $CaCl_2$. Synthesized peptides derived from these sequences improved scopolamine-induced memory impairments in mice. We confirmed this hydrolysate had effects that improved learning and memory abilities by performing the Rey-Kim test. From this hydrolysate of silk fibroin, amino acid sequences of eight peptides were identified by LC-MS/MS. Three peptides (GAGAGTGSSGFGPY, GAGAGSGAGSGAGAGSGAGAGY, and SGAGSGAGAGSGAGAGSGA) were synthesized to investigate whether they could improve memory. Passive avoidance test and Morris water maze test were performed, and all peptides showed memory-enhancing abilities on scopolamine-induced memory impairments in mice. In this study, we identified three novel peptides that could improve memory, and that silk fibroin hydrolysate was a mixture of various active peptides that could enhance memory.

  • A new method for felbinac, which is a potent anti-inflammatory agent, is described. Friedel-Crafts reaction of biphenyl with ethyl ${\alpha}-chloro-{\alpha}-(methylthio)acetate(1)$ afforded ethyl 2-methylthio-2-(4-biphenylyl)acetate(4). Felbinac (7) was synthesized by desulfurization of compound (4) with zinc dust in acetic acid, followed by hydrolysis of the resultant ethyl 2-(4-biphenylyl)acetate (6).

  • Hydrolysis of inulin was investigated employing various commercially available resin catalysts for the production of high grade fructose syrup. The particle size and porosity of the resin significantly affected the distribution of the products, indicating that the intraparticle diffusion of reactants controls thc selectivity as well as the reaction rate. To confirm the effect of the intraparticle diffusion, two different types of resin catalysts were prepared: the one having sulfonic acid group distributed uniformly throughout genular microparticles (A-type) and the other having sulfonic acid group located mainly at the exterior surface of genular microparticles (E-type). The results were found that the reaction rate and the selectivity of the E-type catalyst were higher than those of the A-type catalyst.

Inulin의 산 가수분해 반응에서 초음파가 미치는 영향 (Effect of Ultrasound on the Acid Hydrolysis of Inulin)

  • 부산물이 거의 생기지 않는 반응조건에서 inulin의 산 가수분해 반응에 미치는 초음파의 영향을 조사했다. 반응조건은 50~$60^{\circ}C$와 염산 0.1~0.3%(w/w)의 상당히 완화된 범위였으며, 항온조 내 반응기 위치와 기계적인 교반에 대한 영향을 조사했다. 초음파를 사용한 경우와 하지 않은 경우 fructose생성속도를 온도와 산의 농도변화에 따라 비교했으며 두 경우 활성화에너지는 25kcal/mol로 같았다. 초음파에 의한 반응속도 증가는 평균 22%였다.

  • A convenient method for the synthesis of indobufen, which is a potent antiinflammatory agent, was described. Ethyl 2-phenylbutyrate(4) was prepared by Friedel-Crafts reaction of benzene with ethyl $\alpha$-chloro-$\alpha$-(methylthio)acetate(l) followed by ethylation and desulfurization of the resultant ethyl 2-(methylthio)phenylacetate(2). Ethyl 2-(p-aminophenyl)butyrate(6) was prepared by nitration of (4) and successive reduction of ethyl 2-(p-nitrophenyl) butyrate(5). Indobufen was obtained by condensation reaction of (6) with phthalic anhydride followed by reduction and hydrolysis of the resultant ethyl 2-[p-(1, 3-dioxo-2-isoindolinyl)phenyl]butyrate(7).

Enzymatic Synthesis of Cephaloglycin

  • Cephaloglycin was synthesized directly from D-${\alpha}$ -phenylglycine methyl ester and 7-aminocephalosporanic acid using whole cell enzyme of Xanthomonas citri (IFO 3835). Some optimal conditions for cephaloglycin synthesis were investigated, and yield improvements for its production by several methods were attempted. Using the whole cell enzyme system, the reaction kinetic model for cephaloglycin synthesis is proposed, and the kinetic constants for D-${\alpha}$ -phenylglycine methyl ester hydrolysis, cephaloglycin synthesis, and cephaloglycin hydrolysis were determined. The $K_m$ values of D-${\alpha}$-phenylglycine methyl ester, 7-aminocephalosporanic acid, and cephaloglycin were 11 mM, 24 mM, and 167 mM, and $K_i$ value of D-${\alpha}$-phenylglycine was 15 mM, respectively. The pattern of product inhibition was found to be competitive one.

NMR Study of Poly(γ-Glutamic Acid)Hydrogels Prepared by γ-Irradiation : Characterization of Bond Formation and Scission

  • Hydrogels were prepared from poly( Υ-glutamic acid) (PGA) solution by g-irradiation of 90 kGy and 170 kGy. The hydrogels were more cross-linked with a higher dosage g-irradiation and completely hydrolyzed at 85℃ within 4 hours resulting in homogeneous solution. NMR techniques were employed to clarify chemical bond formation and scission involved during γ-irradiation and hydrolysis. Characterization of these samples was carried out by taking both liquid state and solid state NMR spectra of PGA and hydrolyzed hydrogels and comparison of these spectra with the solid state NMR spectra of hydrogels. Our results indicate that complicated chemical bond formation and scission have occurred during hydrolysis and γ-irradiation. The samples prepared with higher dosage of γ-irradiation showed more diverse chemical bond formation and scission.

식물체 플라보노이드 성분 분석을 위한 적정 가수분해 조건 (Determination of Optimum Hydrolysis Conditions for Flavonoid Analysis in Plant Leaves)

  • 식물체의 플라보노이드 성분 함량 분석에 필요한 적정 가수분해 조건을 구명하기 위하여 은행잎을 이용하여 중심합성법으로 설계한 산 가수분해 시간과 HCl 농도에 따라 myricetin, quercetin, kaempferol 함량을 HPLC로 분석하고, SAS의 반응표면분석법으로 산 가수분해 조건의 최적화를 시도하였다. HCl 0.5~2.5 M, 0.5~2.5시간 범위 내에서 10개의 구간을 잡아서 최적화한 결과 1.5~1.9 M의 HCl 농도와 1.4~2.0시간의 가수분해 조건에서 myricetin, quercetin, kaempferol 성분이 가장 높았다. Superimposing하여 구한 식물체의 플라보노이드의 적정 가수분해 조건은 HCl 1.5 M, 1.5시간이었으며, 이 조건에서 구한 플라보노이드 함량의 예측치와 측정치는 95% 이상 일치하였다. 본 연구에서 구한 적정 가수분해 조건을 이용하여 녹차잎, 무화과잎, 상동나무잎의 플라보노이드 함량을 분석하여 보았다.

전두유의 가수분해조건에 따른 이화학적 특성 변화 (Physicochemical Property Changes of Whole Soymilk Dependent on Hydrolysis Conditions)

  • 본 연구에서는 전두유의 가수분해 조건에 따른 품질 특성을 조사하였다. 그 결과 효소제(KMF-G) 농도가 증가함에 따라 전두유 가수분해물의 당도, 칼슘 내인성, 및 총 유리아미노산 함량은 증가하였으며, SDS-PAGE 분석 결과 단백질의 분자량 변화는 0.20%(w/w)와 0.35%(w/w) 농도에서 유사한 패턴을 나타냈다. 전두유 가수분해물은 효소제(KMF-G) 농도 0.20%(w/w), 가수분해 시간 60분에서 가장 우수하게 나타났다. 전두유 가수분해물을 115, 130 및 $145^{\circ}C$에서 15초간 살균하였을 때 품질변화는 크지 않았다. 이상의 결과 전두유의 가수분해 최적조건은 효소제(KMF-G) 농도 0.20%(w/w), 가수분해 시간 60분으로 설정할 수 있었으며 식품소재로의 다양한 활용이 기대된다.
