  • Nitrogenous compounds in hydrolysate of skipjack processing by-product(SPB) was increased 6.4 times in amount comparing that of raw SPB. The major compounds in hydrolysate were anserine, histidine, leucine, hydroxyproline, arginine, phenylalanine and taurine, and composed 56.25% of total nitrogenous compounds. In fatty acid composition, the highest amount was saturated fatty acids in both samples. Polyunsaturated fatty acids such as $C_{20:5}$ and $C_{22:6}$ were increased after hydrolysis. A total of 99 volatile compounds was detected in raw and hydrolysate of SPB by vacuum simultaneous steam distillation-solvent extraction/gas chromatography/mass chromatorgraphy. Of these, 75 compounds were identified in raw SPB, while 72 compounds were identified in hydorlysate of SPB. There compounds were composed mainly of 28 aldehydes, 20 ketones, 19 alcohols, 5 nitrogen containing compounds, 5 aromatic hydrocarbons, 4 furans and 12 miscellaneous compounds. Levels of aldehydes and aromatic hydrocarbons decreased after hydrolysis, whereas heterocyclic compounds such as pyrazies, furans increased.

Purification and Characterization of Antioxidative Peptides from Enzymatic Hydrolysate of Cod Teiset Protein (대구고니 단백질의 효소적 가수분해물로부터 항산화성 펩타이드의 분리${\cdot}$정제 및 특성)

  • In order to utilize by-products which would normally be discarded in marine processing plants, cod teiset protein was hydrolyzed and antioxidative actiTity of the hydrolysate was investigated. AntioxidatiTe peptide was isolated using ultrafiltration membrane, ion-exchange chromatography on a SP-Sephadex C-25 column, gel filtration on a Sephadex G-15 column, high performance liquid chromatography on an ODS column, and capillary electrophoresis chromatography. Antioxidative activities of the cod teiset hydrolysate were compared with ${\alpha}-tocopherol$, one of the commercial antioxidant. The hydrolysate passed through a membrane with molecular weight cut-off (MWCO) 1 kDa was shown the strongest antioxidative activity, and the activity was higher $10{\%}$ as compared with ${\alpha}-tocopherol$. In addition, the peptide isolated by ion-exchange chromatography, gel filtration, and HPLC, respectively, was higher $53{\%}$ as compared with ${\alpha}-tocopherol$, and the amino acid sequence was Ser-Asn-Pro-Glu-Trp-Ser-Trp-Asn.

  • A compound stimulatory to the growth of S. thermophilus CH-1 was isolated from the cell-free filtrate of L. helveticus YM-1 in milk medium. The stimulant was identified as a peptide with a molecular weight of approximately 5000 and exhibited positive ninhydrin reaction. Some kinds of amino acids confirmed as aspartic acid, alanine, valine, glutamic acid, lysine, proline, leucine were rich in the stimulatory peptide hydrolysate. Among them, glutamic acid was most abundant. Judging from bioautographic results, glutamic acid and phenylalanine were expected to exert an important role for the stimulation.

  • The hydrolysis reaction of soybean oil was conducted experimentally by various source enzymes. The analytical result of hydrolysate of soybean oil showed that the compositions were linoleic acid, olic acid, palmitic acid, and stearic acid in order. The enzymes CR-E and CC-E from Candida rufosa and Candida cylindracea had two hold or more hydrolysis conversions than those of Lipase 16, Novozyme 871, and Lipolase-100L under the same conditions. Therefore CR-E and CC-E were selected for further experiments. These two enzymes had similar ranges of optimun conditions as follows: pH 3-6, $35-45^{\circ}C$, and water to soybean oil ratio of 3.3 or above. They finally got conversions 95% above.

  • Lignin hydrolysates contain many different chemical species, including ferulic acid, coumaric acid, vanillic acid, vanillin, syringaldehyde and furfural. From the perspective of biofuels, these compounds are problematic and can cause downstream loss of product if not removed prior to beginning the fermentative process. In contrast, a search for these compounds within the literature turns up many papers where the same compounds have beneficial properties pertaining to human health, including as antioxidants and in cancer prevention, or are involved in bacterial cell-to-cell signaling. Consequently, this article reviews the dual nature of these and other compounds found in lignin hydrolysates, highlighting both their detrimental and beneficial activities.

  • Production of calcium lactate very useful for medical supplies of Ca-therapy was obtained by lactic acid fermentation of lactobacillus sporogenes, a spore forming lactic acid bacterium. Corn steep liquor 1%, soybean enzyme hydrolysate 3%, yeast extract powder 2% can substitute for yeast extract and peptone as nutrient sort traces in fermentation medium using 10% glucose concentration. In the calcium lactate production medium containing yeast extract powder 2%, glucose 18%, CaCO3 12%, the lactic acid fermentation was carried out at 45$^{\circ}C$ for 4days with continuous agitation of 100 rpm. As results, fermentation yield was 97.5%. The five steps such as protein coagulation, decolorizing evaporating, crystallizing, and drying were carried out to harvest calcium lactate from 10l of supernatant of fermented medium to be removed cell and CaCO3. As results, 2065.0g of white crystal calcium lactate dihyrate was recovered and a yield of 84.9% was obtained.

Preparation and Chemical Characteristics of Food Protein Hydrolysates (식품단백질 효소분해물의 제조 및 이화학적 특성)

  • In this study, food protein hydrolysates were prepared from six types of food protein: purified meat protein, whole egg protein, casein, isolated soy protein, concentrated rice protein, and gluten. Food proteins were hydrolyzed with pepsin and ethanol (80%)-soluble fractions of pepsin hydrolysates were employed for analysis. The products were colorless and odorless powders with low fat content and good solubility. The MW (molecular weight) of the protein hydrolysates was confirmed to be $200{\sim}1,800$ via gel filtration. Free amino acid contents accounted for less than 5% of the samples. The results of our amino acid analysis revealed that all food protein hydrolysates preserved their original amino acid compositions and nutritional values of their source proteins with highly pure oligopeptide mixtures. These results show that the food protein hydrolysates prepared in these investigations should prove excellent dietary nitrogen sources for a variety of applications.

  • The carpophores of a mushroom, Naematoloma fasciculare (Fr.) Karst, were extracted with 0.2 N NaOH and the extract was dialyzed through visking tube. It was found to contain an antitumor activity against sarcoma 180 implanted in mice. The components of this aqueous extract were found to be a polysaccharide and a protein by color reactions including Anthrone and Lowery-Folin tests. The hydrolysis of the polysaccharide with 3% HCL-Me-OH and trimethylsily lation of the hydrolysate yielded five monosaccharides, i. e. glucose, frutose, mannose, galactose and xylose, which were detected and analyzed by GLC. After hydrolysis of the protein with 6N HCl, 15 amino acids, including aspartic acid and glutamic acid, were detected and analyzed by an auto amino acid analyzer.

  • The optimal conditions for producing levulinic acid from office waste paper were investigated. Glucose was produced by enzymatic hydrolysis and its yield maximized by varying the soaking time of the substrate and amounts of enzyme and substrate. The optimal conditions to produce levulinic acid using the hydrolysate were determined by response surface methodology, with reaction temperature and catalyst (sulfuric acid) concentration as independent variables. The production model was assessed with an ANOVA regression analysis, and the results indicate its suitability for levulinic acid production (p, F, and lack-of-fit values were 0.003, 20.1, and 0.058, respectively). The optimal conditions were a reaction time of 56.27 min and catalyst concentration of 5.9% with a predicted yield of 2.588 g/L. We verified the findings under the same conditions and obtained 2.323 g/L of levulinic acid.

  • Infant formula is classified into standard cow's milk-based and special formulas. This review aimed at summarizing the types of special milk formulas currently sold in Korea, and the appropriate indications for the use of these formulas; lactose free formula, soy-based formula, protein hydrolysate formula, amino acid-based formula, preterm formula, medium chain triglyceride formula, low-phosphorus formula, protein-energy-enriched formula, and formulas for inborn errors of metabolism.
