Effect of the addition of protein and lipid on the quality characteristics of Yakju (단백질과 지방첨가가 약주의 품질특성에 미치는 영향)

  • In this study, the fermentation characteristics of Yakju were investigated by addition of protein and lipid. These are classified according to raw material (rice, glucose) and inducing substance (rice protein, rice lipid). Alcoholic fermentation occurred at $25^{\circ}C$, after 14 days. The results of this study were as follows: Alcohol content of Yakju with rice protein was higher than those of other samples. The pH and glucose of rice Yakju were detemied to be 4.86~5.13 and 4.17~4.86, respectively. Titratable acid and the total amino acid content of the Yakju with rice protein were the highest among other samples. The optical density contents of the rice Yakju and glucose Yakju were 0.52~0.653 and 0.27~0.61, respectively. The concentration of organic acids in rice Yakju (433.98~519.31 mg%) was higher than that of glucose Yakju (303.76~387.50 mg%). The major organic acid components of the Yakju were succinic, citric, acetic and lactic acids. The nitrogen compound concentrations of rice Yakju (4377.38~10208.06 ppm) was higher than that of glucose Yakju (671.20~9368.93 ppm). The protein odor correlation coefficient was 0.98 (p<0.001) showing a very high correlation coefficient, while lipid odor coefficient showed a negative correlation with -0.038 (p<0.458).

Effect of Alcohols and Carboxylic Acids on Extraction Characteristics for 1,3-Propanediol by Aqueous Two Phases Systems (수상이성분계에 의한 1,3-프로판디올 추출특성에 대한 알콜과 카르복실산의 영향)

  • 1,3-Propandiol is a promising chemical which can be produced from fermentation of glycerol because the application of 1,3-propanediol is mainly in the production of bio-polytrimethylene terephthalate (bio-PTT). However, the cost of downstream processes in the biological production of 1,3-propanediol can make a high portion in the total production cost due to the large amount of water and the by-produced carboxylic acids such as succinic, lactic and acetic acids in 1,3-propanediol fermentation broth. In this study, aqueous two-phases systems composed of hydrophilic alcohols and phosphate salts were applied to the recovery of 1,3-propanediol from its artificial aqueous solution. Formation of aqueous biphases in hydrophilic alcohols and phosphate salts was due to the salting-out effect of salts in bottom phase, thereby 1,3-propanediol in bottom phase was moved into top phase. Extraction efficiency for 1,3-propanediol was proportional to the polarity of hydrophilic alcohols and the basicity of salts and the maximum value of extraction efficiency was more than 98%. In the aqueous two-phases systems after extraction, there was no carboxylic acid in top phase. Therefore, it was concluded that the aqueous two-phases systems composed of hydrophilic alcohols and phosphate salts were effective for the selective recovery of 1,3-propanediol without any coextraction of carboxylic acids.

Effect of Wheat Flour Brew with Bifidobacterium bifidum on Rheological Properties of Wheat Flour Dough (Bifidobacterium bifidum을 이용한 밀가루 brew가 반죽의 이화학적 성질에 미치는 영향)

  • In order to economically utilize flour brew with Bifidobacterium bifidum as a bread improver, the effect of flour brew on the rheological properties of dough, growth curve and acid production, and symbiosis with yeast were investigated. Growth of bifidobacteria was not increased more than initial seed volume but was consistent during 24 hours of incubation. pH was decreased and T.T.A was increased up to 12 hours of incubation. Symbiosis between bifidobacteria and yeast was little. Bifidobacteria produced more lactic acid than acetic acid in flour brew and the opposite in skim milk broth. This result was inferred from Lactobacillus sp. inherent in flour. On rheological properties of dough, farinograms of flour showed progressively decreasing baking absorption, mixing time and stability as the amount of flour brew increased. The validation of extensograms showed that R/E ratio linearly increased with increment of flour brew, and nearly doubled in all treatments comparing to that of control, which suggest the reduction of actual fermentation time. On visco/amylograms, malt index increased with addition of flour brew, accordingly showing the decrease in viscosity. Break down and set back value decreased with increment of flour brew, suggesting that staling rate of bread can be delayed.

Rapid Processing of the Fish Sauce and Its Quality Evaluation (속성어간장 제조 및 품질 평가)

  • Changes in chemical characteristic, microflora, and sensory evaluation of fish sauce extracted at an interval of one week from fermented solution were investigated. pH was reduced from 6.0 to 4.5, and trimethylamine oxide from 132.5 to 87.2 mg/100g during fermenting periods. Trimethylamine increased from 5.6 to 50.2 mg/100g, and volatile basic nitrogen from 48.3 to 232.5 mg/100g. Bacterial flora isolated from the fish sauce were 70% Lactobacillus sp. and 13% Bacillus sp. Among the free amino acids, alanine, glutamic acid, valine, and methionine contents constitute 40% of the total free amino acids. Major non-volatile organic acid of the fish sauce was lactic acid (76%). Sensory evaluation results of the fish sauce were higher than the traditional soybean sauce after 28 days of fermentation.

Solid-Liquid Equilibria and Excess Molar Volumes, Refractive Indices and Deviation in Viscosity for Binary Systems of C3-C6 Carboxylic Acids (Carboxylic acid 이성분계의 고-액 상평형과 과잉물성, 굴절률 및 점도 편차)

  • Recently, bio-butanol is being promoted as environmentally friendly sustainable energy. However, some problems are still obstacle for commercialization of bio-butanol: the development of cheap biomass and enhancement of fermentation ratio and preparation of economical separation process for fermented products. In the conventional ABE biobutanol fermentation process, organic acids with acetone, butanol, and ethanol are produced. Therefore, it is necessary to study phase equilibrium data and mixture properties for the design and operation of separation process. However, there is lack of design data for organic acids except acetic acid contained system. In this study, therefore, binary solid-liquid equilibria (SLE) and mixture properties: the excess molar volumes ($V^E$), molar refraction deviation (${\Delta}R$) and deviation of viscosity (${\Delta}v$) at 298.15 for $C_3-C_6$ organic acid were reported. The experimental SLE data were correlated with the NRTL and UNIQUAC activity coefficient model with less than 0.5 K of root mean square deviation (RMSD). In addition, $V^E$, ${\Delta}R$ and ${\Delta}v$ for the same binary systems were satisfactorily fitted using the Redlich-Kister polynomial with less than ca. 0.004 standard deviation.

Hepatoprotective and Curative Properties of Kombucha Tea Against Carbon Tetrachloride-Induced Toxicity

  • Kombucha tea(KT) is sugared black tea fermented with a symbiotic culture of acetic acid bacteria and yeasts, which is said to be tea fungus. KT is claimed to have various beneficial effects on human health, but there is very little scientific evidence available in the literature. In the present study, KT along with black tea(BT) and black tea manufactured with tea fungus enzymes(enzyme-processed tea, ET) were evaluated for hepatoprotective and curative properties against $CCl_4$-induced toxicity, using male albino rats as an experimental model by analyzing aspartate transaminase, alanine transaminase, and alkaline phosphatase in plasma and malondialdehyde content in plasma and liver tissues. Histopathological analysis of liver tissue was also included. Results showed that BT, ET, and KT have the potential to revert the $CCl_4$-induced hepatotoxicity. Among the three types of teas tried, KT was found to be more efficient than BT and ET. Antioxidant molecules produced during the fermentation period could be the reason for the efficient hepatoprotective and curative properties of KT against $CCl_4$-induced hepatotoxicity.

Molecular Weight, Protein Binding Affinity and Methane Mitigation of Condensed Tannins from Mangosteen-peel (Garcinia mangostana L)

  • The objectives of this study were to determine the molecular weight of condensed tannins (CT) extracted from mangosteen (Garcinia mangostana L) peel, its protein binding affinity and effects on fermentation parameters including total gas, methane ($CH_4$) and volatile fatty acids (VFA) production. The average molecular weight ($M_w$) of the purified CT was 2,081 Da with a protein binding affinity of 0.69 (the amount needed to bind half the maximum bovine serum albumin). In vitro gas production declined by 0.409, 0.121, and 0.311, respectively, while CH4 production decreased by 0.211, 0.353, and 0.549, respectively, with addition of 10, 20, and 30 mg CT/500 mg dry matter (DM) compared to the control (p<0.05). The effects of CT from mangosteen-peel on in vitro DM degradability (IVDMD) and in vitro N degradability was negative and linear (p<0.01). Total VFA, concentrations of acetic, propionic, butyric and isovaleric acids decreased linearly with increasing amount of CT. The aforementioned results show that protein binding affinity of CT from mangosteen-peel is lower than those reported for Leucaena forages, however, the former has stronger negative effect on IVDMD. Therefore, the use of mangosteen-peel as protein source and $CH_4$ mitigating agent in ruminant feed requires further investigations.

Effects of Nitrogen Application Rate on the Yields, Nutritive Value and Silage Fermentation Quality of Whole-crop Wheat

  • Whole-crop wheat (Triticum aestivum L.) as forage has been extensively used in the world. In this study, the effects of N application rates on the yields, nutritive value and silage quality were investigated. The N application rates were 0, 75, 150, 225, and 300 kg/ha. The research results indicated that the dry matter yield of whole-crop wheat increased significantly with increasing N rate up to 150 kg/ha, and then leveled off. The crude protein content and in vitro dry matter digestibility of whole-crop wheat increased significantly with increasing N up to 225 kg/ha, while they no longer increased at N 300 kg/ha. On the contrary, the content of various fibers tended to decrease with the increase of N application. The content of lactic acid, acetic acid and propionic acid in silages increased with the increase of N rate (p<0.05). The ammonia-N content of silages with higher N application rates (${\geq}225kg/ha$) was significantly higher than that with lower N application rates (${\leq}150kg/ha$). Whole-crop wheat applied with high levels of N accumulated more nitrate-N. In conclusion, taking account of yields, nutritive value, silage quality and safety, the optimum N application to whole-crop wheat should be about 150 kg/ha at the present experiment conditions.

Effects of Management Practices on the Quality of Round Baled Oat Silage (제조 방법이 라운드베일 연맥 사일리지의 품질에 미치는 영향)

  • This experiment was canied out to determine the effect of management practices on the quality of round baled oat silage at experimental field of Grassland and Forage Crops division, National Livestock Research Institute, RDA, Suwon from 1997 to 1998. The experiments are consist of randomized block design with 3 replications. The treatments are 3 wilting dates(0, 2 and 4 days), 3 wrap colors(white, black and green and 3 inoculant(untreated, Inoculant A and Inocuant B). The crude protein(CP) content was increased by prolonged wilting periods, but the effect of wrap color and inoculant were not founded. Acid detergent fiber(ADF) and neutral detergent fiber(NDF) content of all silages were not founded significant difference, but in vitro dry matter digestibility of oat silage with inoculant was significantly higher compare with control. Wilting treatment increased the mean silage acidity compare with control and inoculant treatment significantly reduced silage acidity. Wrap color did not influence the silage acidity. Wilting or inoculant treatments increased lactic acid content but, decreased the content of acetic and butyric acid. The quality grade of all silage were grade 3, except inoculant treated silage. Wilting or inoculant decreased silage DM loss, but wrap color did not effect on siage DM loss. The result of this study indicate that wilting for 2-4 days and inoculant will improve the silage fermentation and quality of round baled oat silage. Characteristics of Flour Ferment Using Lactobacillus acidophilus as Starter (Lactobacillus acidophilus로 발효시킨 밀가루 발효물의 특성)

  • Growth of Lactobacillus acidophilus in flour was investigated for production of noodle and bread. L. acidophilus grew when fermented in flour, and growth continued upon fermentation with salt for 72 hr. pH of L. acidophilus-fermented flour with salt decreased up to 72 hr, reaching 3.06. Fermented flour with salt showed no decomposition as compared to that without salt. In flour fermented by L. acidophilus, amounts of lactic and acetic acids produced increased with incubation time, and reached, after 72 hr incubation, 6,821 and 0.191 mg/g, respectively, resulting in significantly higher production of lactic acid. Viscosity of fermented flour with salt increased, whereas that without salt decreased with incubation time. Results reveal L. acidophilus-fermented flour with salt could be applied as effective agent in noodle and bread productions.
