A protocol was developed for somatic embryogenesis with 100% induction rate from immature zygotic embryo axes of peanut (Arachis hypogaea L. cv. DRG-12) cultured on MS medium containing $18.09\;{\mu}M$ 2,4-D. The frequency of somatic embryogenesis (31.7%) as well as the number of somatic embryos induced per explant (6.6) decreased when the concentration of 2,4-D was increased to $72.4\;{\mu}M$. Morphologically abnormal somatic embryos were observed at a frequency of 43.3% on MS medium containing $72.4\;{\mu}M$ 2,4-D. Somatic embryos isolated from 30-day-old cultures of immature zygotic embryo axes exhibited precocious germination with varied responses when placed on MS basal medium with 3% sucrose. Maximum shoot induction (80.0%) was observed from somatic embryos isolated from 60-day-old cultures of immature zygotic embryo axes when placed as a clump rather than individually on MS medium supplemented with $26.63\;{\mu}M$ BA and $0.54\;{\mu}M$ NAA. Shoots developed from somatic embryos rooted with higher frequency (93.3%) on Blaydes' medium containing $5.4\;{\mu}M$ NAA.
This experiment was conducted to introduce phosphinothricin acetyl -transferase(PAT) gene, resistant to basta and non-selective herbidide, into tobacco(Nicotiana tabacum cv.BY4). For shoot formation,tobacco leaf disks were placed on the MS medium supplemented with 2.0mg/L BA and 0.1mg/L NAA. In this medium condition, tobacco leaf disces were cocultivated with A. tumefaciens MP90 containing NPT IIand PAT resistant to kanamycin and Basta, respectively. Shoots were obtained in the medium containing antibiotics, and those were transferred to rooting medium supplemented with 0.1mg/L NAA and antibiotics. The plants obtaining roots were transplanted into soil. Phenotype of transgenic tobacco plant was mostly as normal plant. However, about 5% was abnormal plant, which did not set seeds. PCR analysis and southern blot were performed to determine transformation. As the results, it was confirmed that PAT gene was stably integrated into tobacco genome.When herbicide, basta, was sprayed to the plants confirmed by PCR, the transgenic plants showed normal growth, whereas normal plants died. Therefore, the result of this experiment show that tobacco transformation for the resistance to basta, non-selective herbicide, was successful because PAT gene was stably integrated into tobacco.
Sulfur, an important component of plants, is regulated by a variety of stresses in sulfate assimilation and metabolism. Increase has been observed in the expression of O-acetylserine(thiol)lyase (OASTL) through two-dimensional electrophoresis with the shoot tips of Arabidopsis infected by beet severe curly top geminivirus (BSCTV). With the three- to six-fold increases in the transcript expression of OASTL, serine acetyltransferase (SAT) and $\gamma$-glutmylcysteine synthetase (GSH) were induced over the mock-inoculated organization in each organization through real-time RT-PCR analysis. The expression of those genes might affect the accumulation of anthocyanin in symptomatic tissues and the induction of abnormal callus-like structures formed by additional cell divisions as typical disease symptoms of BSCTV-infected Arabidopsis. This is the first report to describe the collaborative induction of OASTL, SAT, and GSH in virus-infected plants. The changed expressions of OASTL, SAT, and GSH in Arabidopsis infected with BSCTV raises new aspects regarding the biological function of symptomatic tissues related to sulfate metabolism.

We previously showed that NtCDPK1, a tobacco calcium-dependent protein kinase, interacts with and phosphorylates the Rpn3 regulatory subunit of the 26S proteasome, and that both NtCDPK1 and Rpn3 are mainly expressed in rapidly proliferating tissues, including shoot and root meristem. In this study, we examined NtCDPK1 expression in roots using GUS expression in transgenic Arabidopsis plants, and investigated its function in root development by generating transgenic tobacco plants carrying a sense NtCDPK1 transgene. GUS activity was first detected in roots two days after sowing. In later stages, strong GUS expression was detected in the root meristem and elongation zone, as well as the initiation sites and branch points of lateral roots. Transgenic tobacco plants in which NtCDPK1 expression was suppressed were smaller, and their root development was abnormal, with reduced lateral root formation and less elongation. These results suggest that NtCDPK1 plays a role in a signaling pathway regulating root development in tobacco.
An experiment was conducted to find out effect of soil water condition on shoot and root growth and P, K uptake by soybean plants. Soybean plants were grown under different soil water table levels, 20cm, 40cm and 60cm below the soil surface using minirhizotron with 20cm in diameter, and well irrigated and water stressed conditions using 1/2000a Waganer pots. Three soybean plants, Paldal culfivar, were grown and sampled at the early growing period, 37 days after planting, and at the harvesing period, 115 days after planting. Shoot and root growth were restricted by water stresed condition and by excessive soil water condition with the 20cm water table. Little difference in shoot and root growth were found between well irrigated condition and 40cm or 60cm water table conditions. The P and K contents in shoot under water stressed condition were higher than well irrigated condition at the early growing period but reversed at later harvesting period. The dry weight and length of roots were more severely restricted by water stress than those of shoots. Root morphological difference was found by anatomical observation. Normal cortex was developed under the well irrigated condition, while abnormal cortex with aerenchyma formed by lysis under excessive water conditions of 20 or 40cm water talbes. Aerenchyma was formed at outer skirt of pericycle. Role of formation of aerenchyma of soybean roots might be an adaptation to excessive soil water condition and possibly related to survival of soybean roots growing under near flooding condition on dikes of paddy lands.
Induction of embryogenic callus from Allium wakegi Araki explants was promoted on medium containing 2,4-D, and production of abnormal embryos from the embryogenic callus increased as 2,4-D concentration was raised. Shoot tip was found to be the best explant source for embryogenic callus formation followed in the order by bulb scale and leaf section. Medium containing 0.09 M sucrose was effective for embryogenic callus production. The regenerants from embryogenic callus on medium containing 2,4-D and BA at different concentrations was consisted of diploids, tetraploids and a few mixaploids of 2n+4n, and their chromosomal aberration rate ranged from 8.0 to 33.3%. Frequency of chromosomal aberrants was the highest (18.7%) in the regenerants obtained from bulb scale-derived embryogenic callus among others. Plant regeneration rate was high (33.5%) from the shoot tip-derived embryogenic callus and the frequency of chromosomal aberrants was very low (7.0%). The plantlets regenerated on medium containing 0.26 M sucrose resulted in high chromosomal aberrants. The regenerants on medium containing sucrose at 0.09∼0.20 M produced chromosomal aberrants at around 15.2∼16.6%.

Climate change affects plant responses on physiological characteristics and growth, and Pinus densiflora, one of the major tree species in Korea, are expected to be particularly vulnerable to rising temperature and increased precipitation. This study was conducted to investigate the effects of an open-field warming and precipitation manipulation on physiological characteristics and growth of P. densiflora seedlings. Seedlings of 2-year-old P. densiflora were planted in April, 2013, in open-field nursery located at Korea University. The air temperature of warmed plots had been set to be $3^{\circ}C$ higher than the control plots using infrared lamps. Precipitation was manipulated to be 30% lower or higher than the control, using transparent panels and drip irrigation. Net photosynthetic rate, total chlorophyll content, seedling height, root collar diameter and biomass were measured from April, 2014 to April, 2015. The increase in new shoot biomass from warming was statistically significant, with the biomass in warmed plots about 2-fold higher than in the control plots in 2014 and 2015. This result might be related to advanced bud burst and increased occurrence of abnormal new shoots in warmed plots. Meanwhile, the results of net photosynthetic rate, total chlorophyll content, seedling height, root collar diameter and total biomass from warming and precipitation manipulation were not statistically significant, but tendencies of lower net photosynthetic rate and higher seedling height and biomass in warmed plots compared to the control were shown. Such might be speculated as results of the extended growth period. When root to shoot (R/S) ratio was calculated from the biomass data obtained in April 2014 and April 2015, increased R/S ratio was observed regardless of the treatments applied. Drought tolerance of P. densiflora and particularly low annual precipitation observed in 2014 were suggested as the possible reasons.
Since the peculiar virus disease of chinese date tree (Zizyphus jujuba Mill. var. inermis Rehd.) has been noted in South Korea around 1950, 70% to 80% of the economically important trees have been either completely destroyed or infected with the virus, severe damage has been noted, particularly, across the area ranged from middle east to the middle part of Korea, including Seoul area. Yoon-Koock-Byung in 1958 first reported the disease and descirbed it might be caused by a kinds of yellows. But he did not conform in his paper that the disease is pecisely caused by yellows virus. The authors, hereby intend to identify the true cause of the desease of the chinese data tree by studying the external symptoms of the disease and the internal morphological characteristics of the diseaset plant which shows various abnormalities in contrast to the healthy checks. In view of fact that leaves of the infected plants become yellowish in color similar to the peach yellows, aster yellows, it is likely to be identifiable as the common yellows. Furthermore, the abnormal characteristics observed by the authors are as follow: The floral organs such as petals, sepals, stamens, and pistil turn into vegetative leaves, the leaves on heavily infected plant appear as small sized one and also showing as a common witch's broom like symptom. There are also an occuring of numerous advantitious shoots developed from both of stems and roots. The amount of photosynthetic starch grains increases in parenchymatous cells, necrosis takes place in mesophyll, Particularly, Palisade Parenchyma in the leaves of infected plants are distinguished in contrast to the healthy checks. From the symptoms and the present experimetns described above, the authors are believed that the disease of chinese data tree is not caused by the yellows. It appears the disease is rather similar to the symptoms of sandal spike virus which was noted in India early in this centry. But the host plant of standal disease, Santalum albun L. and the insect vector, Jassus indicus Wal., have never been reported in Korean flora and the founa. The termperature and the otehr environmental factors is quite different Korea and India. Thus the authors believe that the peculiar disease must be an endemic new virus origin in Korea and must be called as "shoot cluster disease of chinese date tree."
Cotyledonary somatic embryos after being cultured in a liquid MS medium for 1 week were subcultured on a solid MS medium and then the embryos germinated at a rate of 92%, but the rate was lowered by extending the culture period of the embryos on a liquid medium: 26% germination on a liquid medium culture for 4 weeks. Somatic embryos subcultured on the liquid medium showed the normal elongation of hypocotyl and radicle but in part showed secondary embryogenesis on hypocotyl and callus formation on and around the root-hypocotyl juncture. Through observation of scanning electron microscope, apical meristem in plumule showed the loose arrangement of cells, and abnormal leaf primordium formation and growth arrest of the primordium or no leaf primordium formation. Therefore, it is suggested that the germination arrest of carrot somatic embryos on liquid medium culture is due to the structural abnormality of the apical meristem in plumule.
The relative aluminium tolerance of 5 Populus species (Populus davidiana, P. maximowiczii, P. nigra, P. alba ${\times}$ P. glandulosa, and P. nigra ${\times}$ P. maximowicizii) was evaluated on Murashige and Skoog medium with 5 levels of aluminium-EDTA concentration (0, 0.1, 0.2, 0.5, 1.0mM). The number of root initiation of 5 Populus species was significantly affected by the medium containing various levels of aluminium concentration. The native poplar, P. davidiana showed the highest aluminium tolerance among 5 species and even stimulated shoot and root growth on the medium containing at the low levels aluminium, 0.1 to 0.2 mM concentrations. The medium containing 1.0 mM aluminium inhibited root initiation from plantlets of all species except P. davidiana. Plantlets cultured on the medium with more than 0.5 mM aluminium exhibited retarded growth and abnormal leaf shape. P. nigra ${\times}$ P. maximowiczii exhibited significantly higher aluminium tolerance than its parental species. Silvicultural aspects associated with aluminium tolerance of Populus are also discussed.
