Inhibition Activity Against Pathogenic Organism of Probiotic Bacteria and Characterization of Inhibition Activity of Isolated Bacteria from Calf Dejecta (Probiotic균주의 Pathogenic Organism에 대한 억제 활성과 송아지분변 분리균주의 억제활성 특성)

  • This study was conducted to investigate the inhibitory activity of Lactobacillus spp., Bacillus ssp., and calf fecal isolates against pathogenic Salmonella typhimurium, E. coli, Listeria monocytogenes, and Staphylococcus aureus. Among thirteen strains of Lactobacillus ssp. tested, Lactobacillus helveticus CU631 showed the highest inhibition against three pathogens, whereas Bacillus spp. showed a weak inhibitory activity. Four calf fecal isolates were identified as Lactobacillus pentosus CU13, CU05, Pediococcus pentosaceus CUR02, and Lactobacillus lactis ssp. lactis CUM14. The whole cell and cell wall components of L. rhamnosus CU02 and L. pentosus CU13 were active in the inhibition of L. monocytogenes. The medium components and levels, which affect on the inhibitory activity, were revealed as Tween 80 1.0%, peptone 3.0%, yeast extract 3.0%, glucose 3.0%, beef extract 3.0%, and NaCl 1.0~3.0%, respectively. Inhibitory activity of the supernatant culture medium was not affected by catalase and proteinase K treatment but affected by heat treatment at 80℃ and netralization, which implies that the inhibitory activity is due to the production of organic acids during the growth. L. pentosus CU13 and L. rhamnosus CU02 exhibited broad inhibition spectrum against 16 out of 21 strains including some pathogens. Oral administration of L. rhamnosus CU02 to the mice infected with E. coli O157:H7 was proven to be effective to recover their body weight during the experimental period.

Empirical Orthogonal Function Analysis of Surface Pressure, Sea Surface Temperature and Winds over the East Sea of the Korea (Japan Sea) (한국 동해에서의 해면기압, 해수면온도와 해상풍의 경험적 직교함수 분석)

  • The seasonal variability of the sea surface winds over the last Sea of Korea (Japan Sea) is investigated by means of empirical orthogonal function (EOF) analysis. The combined representation of fields of three climatic variables by empirical orthogonal functions is discussed. The eigenvectors are derived from daily sea level pressure, wind speed and 10-day mean sea surface temperature (SST) during 15 years $(1978\~1992)$. The spatial patterns of the mean pressure are characterized by the high pressure in the western part and the low pressure in the eastern part. The spatial distribution of the standard deviation (SD) of pressure are characterized by max SD of 6.6 mb near the Vladivostok, and minima along the coast of the Japan. In Vladivostok, the maxima of SD of SST and south-north wind (WV) were also occurred. The representation of fields of individual meteorological variables by EOF shows that the first mode of the west-east wind (WU) explain over $47.3\%$ of the variance and the second mode of WU represents $30\%$. Especially, the first mode of the WV explain $70.9\%$ of the variance and their time series coefficients show 1-cpy, 0.5-cpy frequency spectrum. The spatial distribution of the first mode eigenvectors of SST are characterized by maximum near Vladivostok. The combined representation of fields of several variables (pressure, wind, SST) reveals that the first mode magnitudes of the variance of the combined eigenvectors (WU-PR) are increased. By means of this result, the 1-year peak and the 6-months peak are remarkable. In the three combined patterns (wind, pressure, SST), the second mode of the eigenvector (wind) is affected by the SST. Their time coefficients of the first mode show noticeable 1-year peak. The spectral analysis of the second mode shows broad seasonal signal with the period of 4-months and a significant peak of variability at 3-month period.

Photoluminance Properties of ${Al_3}{GdB_4}{O_{12}}$ Phosphors Activated by $Tb^{3+} and Eu^{3+}$ ($Tb^{3+}$ 와 Eu^{3+}$로 활성화시킨${Al_3}{GdB_4}{O_{12}}$ 형광체의 발광 특성)

  • The new green $Al_3GdB_4O_{12}:Tb^{3+} and red Al_3GdB_4O_{12}:Eu_{3+}$ phosphors were synthesized and then characterized their optical properties for PDP application. And also the photoluminescence properties of these phosphors were compared with the commercial green $Zn_2SiO_4:Mn^{2+} and (Y,Gd)BO_3: Eu^{3+}$ red PDP phosphors. The phosphors were synthesized by solid state reaction at 115$0^{\circ}C$ for 4hr. It was found that the emitting brightness of $Al_3GdB_4O_{12}:Tb^{3+}$(15mol%) green phosphor under 147nm excitation was higher than that of commercial $Zn_2SiO_4: Mn^{2+}$ green PDP phosphor. However, the color coordinate of this new green phosphor was inferior to the commercial one. On the other hand, the emitting intensity of $Al_3GdB_4O_{12}:Eu^{3+}$(15mol%) red phosphor was smaller than the $commercial(Y,Gd)BO_3: Eu^{3+}$ red one, but the CIE coordinate was slightly improved. The excitation spectrum showed that $Al_3GdB_4O_{12}$ phosphors had a strong excitation band at $\lambda=160nm$ associated with the host absorption. And the photoluminance excitation (PLE) intensity in VUV range for $Al_3GdB_4O_{12}:Tb^{3+}$ green phosphor was higher than that of $Zn_2SiO_4: Mn^{2+}$, but the PLE intensity of $Al_3GdB_4O_{12}:Eu^{3+}$ red phosphor was smaller than $(Y,Gd)BO_3: Eu^{3+}$.

Cervical Tuberculous Lymphadenitis : Clinicopathological Features and AFB Positivity (결핵성 경부 림프절염의 병리소견과 항산균 검출 및 임상소견에 관한연구)

  • Background : Histological analysis of tuberculosis shows a spectrum of findings, from well formed granulomatous inflammation with few bacilli in patient with normal immune response to M. tuberculosis to poorly formed granulomatous inflammation with many bacilli in patient with defective immune response. To evaluate the degree of immune response to M. tuberculosis, we studied the histologic features, including the presence of acid fast bacilli(AFB) in lymph node of patients with cervical tuberculous lymphadenitis, and compared them with clinical characteristics. Methods : We reviewed the histologic features of 33 cases of cervical tuberculous lymphadenitis and processed the excised nodes for auramine-rhodamine staining to detect AFB. The AFB positivity in tissue was compared with the histologic features(degree of granuloma formation, presence of caseation necrosis, presence of neutrophilic infiltration) and clinical characteristics(lymph node size, duration of symptom, presence of local symptom or radiologic evidence of pulmonary tuberculosis). Results : 1) The mean age at diagnosis was 42.4 years, and male to female ratio was 1:4.5. 2) Histologically, all cases showed well formed granuloma and variable degrees of caseation necrosis, and 39% of the cases showed neutrophilic infiltration in the granulomatous inflammation. 3) AFB were confirmed in 52% of the cases, and they were found extracellularly and at the periphery of caseation necrosis. 4) There was no association between AFB positivity and histological features or clinical characteristics. Conclusion : Cervical tuberculous lymphadenitis showed well formed granulomatous inflammation with caseation necrosis, and there was no association between AFB positivity in the tissue and histological or clinical characteristics.

Antimicrobial Activity of Extracts from Different Parts and Essential Oil from Pinus densiflora on Skin Pathogens (소나무 부위별 추출물 및 essential oil의 피부상재균에 대한 항균 활성)

  • This study was carried out to examine the antimicrobial activity of 70% ethanol and hot water extracts from different parts and essential oil in Pinus densiflora on skin pathogens such as Staphylococcus epiermidis, S. aureus, Propionibacterium acnes, Candida albicans, C. tropicalis, and Trichophyton rubrum. The antimicrobial activities of extracts and essential oil were tested by paper disc assay and minimum inhibitory concentration test. The ethanol extract of pine pollen showed antimicrobial activity against C. tropicalis and T. rubrum. Ethanol extract of pine needle and pine gnarl showed antimicrobial activity against S. aureus, C. tropicalis, and P. acnes. Essential oil from pine needle exhibited antimicrobial activity against C. albicans, C. tropicalis, and P. acnes. The minimum inhibitory concentration of ethanol extracts of pine needle and pine gnarl against P. acnes and C. tropicalis ranged from 0.002% to 0.0063%. The lowest minimum inhibitory concentration (0.025%) against C. albicans and C. tropicalis was obtained from essential oil. These results indicate that ethanol extracts of pine pollen, pine needle, pine gnarl, and essential oil of pine needle could be applicable to control the skin infection pathogens. Especially, ethanol extract of pine gnarl had a broad spectrum of antimicrobial activity and pine extracts and essential oil exhibited higher antimicrobial activity with Candida sp. and P acnes.

Textural and Geochemical Characteristics and their Relation of Spinel Peridotite Xenoliths from Jeju Island (제주도 첨정석 페리도타이트 포획암의 조직 및 지화학적 특성과 그 관련성)

  • Abundant spinel lherzolite xenoliths showing distinctively different textural types such as protogranular, porphyroclastic, and mylonitic texture are trapped in the basaltic rocks from southeastern part of Jeju Island. These xenoliths show the textural spectrum from coarse-grained protogranular through porphyroclastic with bimodal grain size to fine-grained and foliated mylonitic texture. They tend to decrease in grain sizes and show more linear grain boundaries and more frequent triple junctions from protogranular through porphyroclastic to mylonitic. Spinel has different occurrence mode according the textural type. Spinel is always associated with orthopyroxene in protogranular texture, whereas it is scattered and independent of orthopyroxene in mylonitic texture. Additionally, porphyroblast from porphyroclastic and mylonitic textures has internal deformation features such as kink band, undulatory extinction and curved lamella, whereas neoblast is strain-free. These textural features indicate increasing degree of static/dynamic recrystallization from protogranular through porphyroclastic to mylonitic texture. The mg#[$=100{\times}Mg/(Mg+Fe_t)$] of olivine, orthopyroxene and clinopyroxene is relatively constant (ol: 88-91; opx: 89-92; cpx: 89-92) regardless of textural differences. The mg# of constituent minerals, NiO content (0.3~0.4 wt%) and MnO content (0.1~0.2 wt%) of olivine are similar to those of mantle xenoliths worldwide, also indicating that studied spinel lherzolite xenoliths were mantle residues having experienced 20~25% partial melting. The geochemical and textural characteristics have close relations showing that LREE and incompatible trace elements content of orthopyroxene and clinopyroxene increases from protogranular through porphyroclastic to mylonitic. These observations suggest that the studied mantle xenoliths experienced metasomatism by LREE enriched melt or fluid after partial melting, indicating a close relation between deformation and metasomatism. The metasomatism was possibly confined to narrow shear zones from where porphyroclastic and mylonitic textured xenoliths originated. These shear zones might favorably drive the percolation of LREE-enriched melts/fluids responsible for the metasomatism in the lithospheric mantle below the Jeju Island.

Modification and Validation of Analytical Method for Oxypaeoniflorin and Paeoniflorin in Moutan Cortex Radicis Extract (목단피 추출물의 Oxypaeoniflorin 및 Paeoniflorin의 분석법 개선 및 검증)

  • The aim of this study was the validation of a modified analytical method for determination of oxypaeoniflorin and paeoniflorin in Moutan Cortex Radicis extract. For validation of the analytical method, we modified established analytical methods and validated improvement. For validation, the specificity, linearity, precision, accuracy, limit of detection (LOD), and limit of quantification of oxypaeoniflorin and paeoniflorin were measured by high performance liquid chromatography. The results show that the correlation coefficients of the calibration curve for oxypaeoniflorin and paeoniflorin were 1.0000 and 0.9998, respectively. The LOD for oxypaeoniflorin and paeoniflorin were $0.23{\mu}g/mL$ and $0.25{\mu}g/mL$, respectively. The inter-day and intra-day precision values of oxypaeoniflorin and paeoniflorin were 0.70~3.19% and 1.74~2.43%, and 0.32~0.92% and 0.62~2.28%, respectively. The inter-day and intra-day accuracies of oxypaeoniflorin and paeoniflorin were 98.33~102.11% and 97.72~118.12%, and 98.44~101.56% and 97.10~112.00%, respectively. Therefore, the analytical method was validated for the detection of oxypaeoniflorin and paeoniflorin in Moutan Cortex Radicis.

The usefulness of diagnostic tests in children with language delay (언어 발달지연 환아에서 진단적 검사의 유용성)

  • Purpose : To assess the usefulness of magnetic resonance imaging (MRI), karyotyping, brainstem auditory evoked potential (BAEP), electroencephalogram (EEG), tandem mass screening test, and newborn metabolic screening test in children with language delay for diagnosing underlying diseases. Methods : From January 2000 to June 2007, a retrospective chart review was performed for 122 children with language delay who visited the Child Neurology Clinic at Yeungnam University Hospital and who underwent neuropsychologic tests and other diagnostic evaluations for underlying diseases. They were grouped into phenomenological diagnostic categories, and test results were analyzed according to the underlying diseases. Results : Of 122 patients, 47 (38.5%) had mental retardation, 40 (32.8%) had developmental language disorders, 23 (18.9 %) had borderline IQ, and 12 (9.8%) had autism spectrum disorder. In 26 (21.3%) cases, the causes or relevant clinical findings to explain language delay were found. Eight (10.4%) of 77 MRIs, 6 (8.0%) of 75 EEGs, and 4 (5%) of 80 BAEPs showed abnormal results. Results directly attributed to diagnosing underlying diseases were 2 hearing defects in BAEPs and 1 bilateral perisylvian cortical dysplasia in MRIs. No abnormal results were found in karyotyping, tandem mass screening tests, and new-born screening tests. Conclusion : Commonly used tests to diagnose the cause of language delay are not very effective and should only be used selectively, according to patient characteristics. However, despite the low diagnostic yields from these tests, because many patients show abnormal results, these tests are useful when conducted in complete evaluation.

Compression Sensing Technique for Efficient Structural Health Monitoring - Focusing on Optimization of CAFB and Shaking Table Test Using Kobe Seismic Waveforms (효율적인 SHM을 위한 압축센싱 기술 - Kobe 지진파형을 이용한 CAFB의 최적화 및 지진응답실험 중심으로)

  • The compression sensing technology, CAFB, was developed to obtain the raw signal of the target structure by compressing it into a signal of the intended frequency range. At this point, for compression sensing, the CAFB can be optimized for various reference signals depending on the desired frequency range of the target structure. In addition, optimized CAFB should be able to efficiently compress the effective structural answers of the target structure even in sudden/dangerous conditions such as earthquakes. In this paper, the targeted frequency range for efficient structural integrity monitoring of relatively flexible structures was set below 10Hz, and the optimization method of CAFB for this purpose and the seismic response performance of CAFB in seismic conditions were evaluated experimentally. To this end, in this paper, CAFB was first optimized using Kobe seismic waveform, and embedded it in its own wireless IDAQ system. In addition, seismic response tests were conducted on two span bridges using Kobe seismic waveform. Finally, using an IDAQ system with built-in CAFB, the seismic response of the two-span bridge was wirelessly obtained, and the compression signal obtained was cross-referenced with the raw signal. From the results of the experiment, the compression signal showed excellent response performance and data compression effects in relation to the raw signal, and CAFB was able to effectively compress and sensitize the effective structural response of the structure even in seismic situations. Finally, in this paper, the optimization method of CAFB was presented to suit the intended frequency range (less than 10Hz), and CAFB proved to be an economical and efficient data compression sensing technology for instrumentation-monitoring of seismic conditions.

Posttransplantation Lymphoproliferative Disorder after Liver Transplantation in Pediatric Patients: Report from a Single-center Over 21 Years (간 이식 소아에서 발생한 이식 후 림프 증식 질환: 단일 기관에서의 21년 경험)

  • Purpose: To analyze the clinical spectrum of posttransplantation lymphoproliferative disorder (PTLD) after liver transplantation in children. Methods: From January 1988 to June 2009, we retrospectively reviewed the medical records of 8 PTLD cases among 148 pediatric patients underwent liver transplantation. The age at transplantation, time of presentation after transplantation, clinical manifestations, histologic diagnosis, results of EBV (Epstein-Barr virus) assessments, managements and outcomes of PTLD were investigated. Results: The prevalence of PTLD in liver transplant pediatric recipients was 5.4% (8 of 148). The mean age of patients was 25.4${\pm}$21.3 months (range 10 to 67 months). Seven of 8 patients (87.5%) underwent liver transplantation before 1 year of age. The common clinical presentations were persistent fever (8 of 8, 100%) and bloody diarrhea (6 of 8, 75%). PTLD was diagnosed with gastrointestinal endoscopic biopsies in five patients and surgical biopsies in three. Histologic findings showed early lesion in three patients, polymorphic in two, and monomorphic in three. Burkitt lymphoma and lymphoblastic lymphoma were found in two of 3 monomorphic patients. Seven of 8 patients were found with EBV-positive. Eight patients were treated with dose reduction of immunosuppressants and infusion of ganciclovir. Rituximab was added to four patients. PTLD were successfully managed in all patients except one who died of sepsis during chemotherapy. Conclusion: Major risk factor of PTLD was to undergo liver transplantation before 1 year of age. Continuous monitoring for EBV viral load and gastrointestinal endoscopic biopsy may be useful to early detection of PTLD.
