In this study, we evaluated the fruit quality indices and the incidence of physiological disorders of Korean new pear cultivar 'Whasan' to determine appropriate harvest time for exportation and to enhance storability and shelf-life of the fruits during simulated exportation and extending market environment under different temperature conditions. In the experiment of simulated exportation headed for North America including Canada and U.S.A., the fruits transported at $1^{\circ}C$ showed less weight loss than those of $5^{\circ}C$. Market temperature appeared as a key factor for keeping freshness of exported pear fruits rather than transportation temperature. Quality factors such as high flesh firmness and low incidence of fruit rot and physiological disorders including core breakdown and pithiness were attained at the fruits maintained at $18^{\circ}C$ when we compared with $25^{\circ}C$. The fruits of harvested early maturity at 135 day after full bloom showed 28.6 N of flesh firmness when the fruits stored at $1^{\circ}C$ of transportation and $18^{\circ}C$ of market temperature, while the fruits progressed $5^{\circ}C$ of transportation and $25^{\circ}C$ of market temperature dropped to 24.2 N during 30 days of shelf life. Also, a high incidences of physiological disorders and of fruit decay rates were obvious in the fruits distributed at $25^{\circ}C$ were observed approximately two times higher than the those of $18^{\circ}C$. Therefore, temperature management during marketing resulted as an important point for maintaining fruits quality in the process of pear fruit exportation.
Purpose: Resonance frequency analysis, Periotest, and removal torque (RT) test were known as the methods to assess implant stability. The results of these methods are affected by the bone condition, implant diameter and shape. The purpose of this study is to access the meaning and the correlationship of the resonance frequency analysis, Periotest and RT test in osseointegration simulated acrylic resin when the engaged bone thickness and peri-implant bone defect are changed. Materials and methods: To simulate osseointegration, the fixture was fixed to an aluminum mold with a screw. Acrylic resin powder and liquid were poured into the mold for polymerization. The engaged resin thickness with implant was controlled. Simulated cortical bone thicknesses were 1, 3, 5 and 10 mm. Additional 1, 3 and 5 mm peri-implant bone defects were simulated. Three types of implants were used; 4 mm diameter implants of straight shape, 4 mm diameter implants of tapered shape and 5 mm diameter implants of tapered shape. Five fixtures per each type were tested in respective bone condition. Resonance frequency analysis and Periotest were evaluated in all bone conditions. Peak removal torque was measured at simulated cortical bone thicknesses of 1 and 3 mm. The statistical analysis was performed with the Kruskal-Wallis test, Mann-Whitney U test, and Spearman test using a 95% level of confidence. Results: With increasing engaged bone depth, the Implant Stability Quotient (ISQ) values increased and the Periotest values (PTVs) decreased (P<.001, P<.001). With increasing peri-implant bone defect, ISQ values decreased and PTVs increased (P<.001). When the diameter of implant increased, ISQ values increased and Periotest values (PTV) decreased (P<.001). There was a strong correlation between ISQ values and PTVs (r = -0.99, P<.001). Furthermore, the peak removal torque values had weak correlations with both ISQ values and PTVs (r = 0.52, P<.001 ; r = -0.52, P<.001). Conclusion: This study confirmed favorable implant stability with increasing engaged bone depth and implant diameter and decreasing peri-implant bone defect. ISQ values and PTVs showed strong correlation with each other and not with the peak removal torque values.
It was aimed to study the effects of ${\alpha}_2-adrenoceptor$ agonists on the sleeping time in $one{\sim}two-day-old$ chickens. Furthermore, it was also evaluated whether ${\alpha}_1-adrenoceptor$ agonist and antagonist might affect the sleeping in the chickens and discussed in relation with opiate receptor. 1) Guanabenz, clonidine, guanfacine and B-HT 933 decreased the latency of the loss of righting reflex in a dose-dependent manner, but B-HT 920 and oxymetazoline slightly prolonged it. 2) ${\alpha}_2-Adrenoceptor$ agonists produced dose·related increase in sleeping time. The potency was guanabenz>clonidine>oxymetazoline${\geq}$B-HT 933${\geq}$B-HT 920>guanfacine in this order. 3) ${\alpha}_2-Adrenoceptor$ antagonists decreased guanabenz-induced sleeping time in a dose ·dependent manner. The rank order of ${\alpha}_2-adrenoceptor$ antagonists was yohimbine>rauwolscine>piperoxan${\geq}$RX 781094. 4) Sleeping time caused by both ethanol and hexobarbital was not affected by yohimbine in chickens. 5) Methoxamine and phenylephrine showed little significant effect on the guanabenz-induced sleeping time. However, prazosin increased it. Paradoxically, corynanthine rather caused to decrease it. These results suggest that the stimulation of central ${\alpha}_2-adrenoceptor$ mediates sleeping, however it is remained uncertain in the role of central ${\alpha}_1-adrenoceptor$ in chickens. In addition, the one~two-day-old chickens may be considered as a useful, inexpensive and simple experimental model to evaluate the in vivo pharmacological action of the ${\alpha}_2-adrenoceptor$ agonist and antagonist related to sedation.
Lentinus lepideus, known as train wrecker fungus, has been used for nutritional and medicinal purposes. Recently, commercial cultivation technique and a new cultivar of the mushroom were developed. To investigate the genetic diversity and phylogenetic relationship for identifying the mushroom strains and cultivar, one commercial and 13 strains of Lentinus lepideus from different geographical regions of Korea were analyzed by ITS regions of rDNA and RAPD of genomic DNA. Three strains of Lentinus edodes were also used for the analysis. The size of the ITS1 and ITS2 regions of rDNA from the different strains varied from 173 to 179 bp and 203 to 205 bp, respectively. The sequence of ITS1 was more variable than that of ITS2, while the 5.8S sequences were identical with 156 base pairs. A phylogenetic tree based on the ITS region sequences indicated that selected strains could be classified into four clusters, while 3 strains of L. edodes was divided into a new cluster. Ten primers out of 20 arbitrary primers used in the RAPD-PCR efficiently amplified the genomic DNA. The numbers of amplified DNA bands varied with the primers and strains, with polymorphic DNA fragments in the range from 0.2 to 2.6 kb. The results showed that phylogenetic relationship among Korean strains of Lentnus lepideus is high, but genetic diversity is low.
To determine the distribution and recovery of fall-applied N in various parts of satsuma mandarins (Citrus unshiu Marc). $19.68g\;N\;tree^{-1}$ as urea containing 5 atom % $^{15}N$ and 58 kg$K_2O$$ha^{-1}$ were broadcast-applied to 11 years old 'Miyagawa Wase' grown at a spacing of $2.7{\times}2.7m$ on 18 November 1998. Nitrogen, $P_2O_5$, and $K_2O$ were applied at 104, 308, and $62kg\;ha^{-1}$ on 22 March and N and $K_2O$ at 42 and $83kg\;ha^{-1}$ on 15 June 1999. Two trees were excavated on 15 June and 8 December 1999, respectively. In mid-June, whole tree contained $168.2g\;N\;tree^{-1}$ of which 11.9, 42.1, 29.7, and 16.3% were in fruits, leaves, stems, and roots, respectively. In early December, total tree N averaged $169.8g\;tree^{-1}$ and fruits accounted for 27.6%, leaves 36.4%, stems 22.8%, and roots 13.2% of total tree N. Regardless of harvest date, N derived from fertilizer was highest in newly developed tissues. In mid June, the tree recovered 18.5% ($3.63g\;tree^{-1}$) of fertilizer N. Fruits accounted for 21.1%, leaves for 50.4%, stems for 21.5%, and roots for 7.9% of fertilizer-derived N in the tree, respectively. In early December, the tree recovered 17.0% of fertilizer N. Fruits contained 39.6%, leaves 40.5%, stems 14.5%, and roots 5.3% of fertilizer-derived N in the tree respectively. Comparing with total tree N, a higher proportion of fertilizer-derived N was allocated to metabolically active tissues while a less proportion to old tissues regardless of harvest date.
Osteosarcoma (OS) is the most common and malignant bone tumors. Although many types of resection surgery and experimental agents were developed, median survival and clinical prognosis are poorly investigated. Recently, several researches have reported that Eucheuma cottonii has potent as protective effects of coal dust-induced lung damage via inhibition of malondialdehyde (MDA) and oxidative stress in bronchoalveolar lavage fluids (BALF). However, anti-cancer effects and specific molecular mechanism of extract from Eucheuma cottonii (EE) has not been clearly studied yet. This study evaluated that anti-cancer potential of EE in human osteosarcoma Saos-2 cells. EE indicated cytotoxicity on Saos-2 cells in a dose-dependent manner. Morphological degradation and nucleic condensation were also observed under the EE treatment. However, it did not significantly affect on non-cancerous kidney HEK-293 cells under the same concentration which is shown cytotoxicity on Saos-2 cells. The phosphorylation of Fas-Associated Death Domain (FADD) and expression of cleaved caspase-8, -7 and -3 were upregulated in a dose-dependent manner. In immunofluorescence staining, expression level of Fas and cleaved PARP were upregulated by EE treatment. Furthermore, treatment of EE induces upregulation of sub G1 phase by flow cytometry analysis. The results demonstrated that EE has a therapeutic potential against osteosarcoma via FADD mediated caspase cascade apoptosis signal pathway.
Purpose: To evaluate the clinical stability and function after arthroscopic anterior cruciate ligament(ACL) reconstruction using fresh-frozen tibialis tendon allograft. Materials and Methods: Of the patients who underwent ACL reconstruction using tibialis tendon allograft from July 2002 to June 2003, thirty-one patients could be evaluated and the mean follow-up period was 19 months. Evaluations included were Lysholm knee score, 2000 International knee Documentation Committee (IKDC) subjective knee score, Lachman test, pivot shift test, KT-1000 arthrometer measurement and 2000 IKDC knee examination. Results: The mean Lysholm score was 88. Twenty-eight patients (90.3%) were good or exellent for the measured parameters. Twenty-seven patients(87.1%) was over 70 in IKDC subjective knee score. Thirty patients (96.8%) had 1+ firm end or negative Lachman test. 27 patients (87.1%) had a negative pivot shift. Thirty patients (96.8%) had less than 5mm difference of maximal manual difference by KT-1000 arthrometer. Twenty -nine patients (93.5%) were nearly normal or normal grade by 2000 IKDC knee examination. Complications were 1 case of failure and 1 case of infection. Conclusion: ACL reconstruction with the double-stranded fresh-frozen tibialis tendon allograft resulted in a reliable and predictable outcome after short-term follow-up.
There are relatively many reports have been issued on he active movement of cuticle of larva. which tend to protect their body, however, only a few reports have been disclosed on the cuticle of pupal body except the small portion of rectangle which shown tortoise-shell shape. In this connection, many portion of the external structure of pupal cuticle has been studied and the following. results were found: 1) No. spot of rectangle which is sculptured in the surface of cuticle that born by branching. out of the development of cell in imaginal bud of antenna and head were found. However, in the compound eye of net shaped sculpture was found in the (equation omitted) shaped parts which holding. the diameter of about 8u and the surrounding area has the small bump and the one is dark brown coloured comparing with shape. 2) The sculpture shape of thorax is a little different than in the head. However, (equation omitted) portion is varies from the segment to segment. In general, it is not very clear than the compound eye in the head, the dark brown bump shape is slowly fade a from the prothorax, mesothorax to metathorax. 3) The surface of intersegment membrane is colourless or slightly yellow, and the entire surface has stripped marking with thine lines. 4) In the abdominal segment, there are many and small sculptures in net shape around the (equation omitted) shape portions. 5) The size of sculpture in (equation omitted) portion of abdomen is smaller than one in thorax, and in the same segment, the dorsal is smaller than abdomen and the rear portion of the segment is larger than the front of segment. 6) After the 7th abdominal segment, no intersegmental membrane is found and the cuticle of the external structure is the same as external structure of the segment. 7) The seta is not found in head, compound eye, antenna and wing which portions were subdivided by development of imagined bud of the cell, no seta is found in cuticle of the segment in the general cell of the larva stage and also in the dorsal and intersegmental membrane.

Off-Site Construction (OSC) is a new construction method based on factory production. OSC (Off-Site Construction) is a new construction method based on factory production. Researches such as OSC-related design and production standardization, transport methods are actively conducted in the U.S., UK and other parts of the world as this new method has an edge over existing methods in terms of productivity, economy and quality. As the emergence of this new area requires reasonable management, an analysis of the scope of construction project management is required accordingly. Therefore, this research analyzed the study trends and relationships at the CM/PM range's "Operation level" to identify areas of study, relationship between studies and deficiencies in current research. This study carried out a comprehensive literature review of the OSC (CM/PM) research by analyzing 94 papers in Operation level as of September 3, 2018, and the analysis results are as follows. (1) Working stage level researches have been increasing rapidly since 2006. (2) Non-volumetric type is contributing most significantly at work stage level. In the building sector, it has been identified that problems such as residential: living, quality issues, non-residential: economic difficulties, factory: productivity problems have been addressed. (4) The Non-volumetric pre-assembly type dealt with the economic feasibility of residential and non-residential buildings, whereas the modular type was studied in regards to assembly quality. (5) From 2006, project management areas (e.g., quality, human resources, risks) have been expanded. It is expected that this research will help find new areas of research for OSC. If the analysis is carried out to the level of the industrial, corporate and project phases in the future, it is deemed that the overall research flow and area of the OSC industry can be identified.

The first small interfering RNA (siRNA) therapeutics have recently been approved by the Food and Drug Administration in the U.S., and the demand for a new RNA therapeutics bioanalysis method-which is essential for pharmacokinetics, including the absorption, distribution, metabolism, and excretion of siRNA therapeutics-is rapidly increasing. The stem-loop real-time qPCR (RT-qPCR) assay is a useful molecular technique for the identification and quantification of small RNA (e.g., micro RNA and siRNA) and can be applied for the bioanalysis of siRNA therapeutics. When the anti-HPV E6/E7 siRNA therapeutic was used in preclinical trials, the established stem-loop RT-qPCR assay was validated. The limit of detection was sensitive up to 10 fM and the lower limit of quantification up to 100 fM. In fact, the reliability of the established test method was further validated in three intra assays. Here, the correlation coefficient of $R^2$>0.99, the slope of -3.10 ~ -3.40, and the recovery rate within ${\pm}20%$ of the siRNA standard curve confirm its excellent robustness. Finally, the circulation profiles of siRNAs were demonstrated in rat serum, and the pharmacokinetic properties of the anti-HPV E6/E7 siRNA therapeutic were characterized using a stem-loop RT-qPCR assay. Therefore, the stemloop RT-qPCR assay enables accurate, precise, and sensitive siRNA duplex quantification and is suitable for the quantification of small RNA therapeutics using small volumes of biological samples.
