Differential Effect of Copper (II) on the Cytochrome P450 Enzymes and NADPH-Cytochrome P450 Reductase: Inhibition of Cytochrome P450-Catalyzed Reactions by Copper (II) Ion

  • Inhibitory effects of Cu$\^$2+/ on the cytochrome P450 (P450)-catalyzed reactions of liver microsomes and reconstituted systems containing purified P450 and NADPH-P450 reductase (NPR) were seen. However, Zn$\^$2+/, Mg$\^$2+/, Mn$\^$2+/, Ca$\^$2+/, and Co$\^$2+/ had no apparent effects on the activities of microsomal P450s. Cu$\^$2+/ inhibited the reactions catalyzed by purified P450s lA2 and 3A4 with IC$\sub$50/ values of 5.7 and 8.4 ${\mu}$M, respectively.(omitted)

Nutritional value and the kaempferol and quercetin contents of quinoa (Chenopodium quinoa Willd.) from different regions

  • This study compared the nutritional value of quinoa cultivated in different regions, i.e., Peru (PQ), United States (UQ), and Korea (KQ), focusing on their proximate and nutrient compositions and functional components. Moisture, protein, lipid, and ash contents were highest in KQ, and the carbohydrate content was the highest in UQ. KQ had the highest amount of total amino acids, especially lysine. KQ had the lowest levels of Na but the highest levels of K, P, Fe, Mg, Zn, and Mn. The antioxidant compounds, quercetin and kaempferol were not detected in KQ, which consequently had the lowest total phenolic and total flavonoid contents (TPC and TFC, respectively). These values were comparatively higher in UQ. Meanwhile, PQ had the highest TPC and TFC values as well as kaempferol content, but lacking quercetin. These results demonstrate that the nutritional value of quinoa varies according to the region in which it is cultivated.

Novel Alkali-Stable, Cellulase-Free Xylanase from Deep-Sea Kocuria sp. Mn22

  • A novel xylanase gene, Kxyn, was cloned from Kocuria sp. Mn22, a bacteria isolated from the deep sea of the east Pacific. Kxyn consists of 1,170 bp and encodes a protein of 390 amino acids that shows the highest identity (63%) with a xylanase from Thermohifida fusca YX. The mature protein with a molecular mass of approximately 40 kDa was expressed in Escherichia coli BL21 (DE3). The recombinant Kxyn displayed its maximum activity at $55^{\circ}C$ and at pH 8.5. The $K_m,\;V_{max}$, and $k_{cat}$ values of Kxyn for birchwood xylan were 5.4 mg/ml, $272{\mu}mol/min{\cdot}mg$, and 185.1/s, respectively. Kxyn hydrolyzed birchwood xylan to produce xylobiose and xylotriose as the predominant products. The activity of Kxyn was not affected by $Ca^{2+},\;Mg^{2+},\;Na^+,\;K^+$, ${\beta}$-mercaptoethanol, DTT, or SDS, but was strongly inhibited by $Hg^{2+},\;Cu^{2+},Zn^{2+}$, and $Pb^{2+}$. It was stable over a wide pH range, retaining more than 80% activity after overnight incubation at pH 7.5-12. Kxyn is a cellulase-free xylanase. Therefore, these properties make it a candidate for various industrial applications.

Utilization of Dietary Nutrients, Retention and Plasma Level of Certain Minerals in Crossbred Dairy Cows as Influenced by Source of Mineral Supplementation

  • Nutrient utilization and plasma level of minerals were studied in lactating crossbred cows fed diet supplemented from different source of minerals. Twelve crossbred cows of first to third lactation during their mid stage of lactation were distributed equally under two groups and were fed concentrate mixture, green fodder (para grass) and finger millet straw as per requirement. Cows in group I were fed concentrate mixture supplemented with 1% mineral mixture whereas cows in group II were not supplemented with mineral mixture in the concentrate mixture but were offered additional quantity of green fodder (1 kg DM/cow/day) to compensate for the requirement of minerals. Balance study conducted towards the end of 120 days of feeding experiment indicated that the total dry matter intake in both the groups did not differ significantly. The cows in group II offered additional quantity of green fodder consumed significantly (p<0.05) more of green fodder (5.11 vs. 4.51 kg /animal/day) and the cows in group I consumed significantly (p<0.01) more of finger millet straw (1.71 vs. 0.92 kg/animal/day). The digestibility of major nutrients did not differ between the groups except for ether extract which was significantly (p<0.05) lower in cows fed additional green fodder. The total daily intake of P, Cu, Fe and Co did not differ significantly in both the groups whereas significantly higher intakes of Ca, Mg, Zn and Mn were observed in cows supplemented with inorganic source of minerals. However, supplementation through both the sources could meet the mineral requirement in group I and group II. The gut absorption (%) of all the minerals was comparable between the groups except for Mg which was significantly (p<0.05) higher in cows supplemented mineral mixture. The net retention of all the minerals was significantly more in the group supplemented with inorganic source of minerals. Except for P, Mg and Co the retention as percentage of total intake was comparable for all minerals in both the groups. Irrespective of source of mineral supplementation the average monthly blood plasma mineral levels (Ca, P, Mg, Cu, Zn, Fe) were within the normal range and comparable between the groups. Lower level of Cu observed at the start of the experimental feeding in both the groups increased with the advancement of supplemental feeding. The plasma Zn and Fe content in cows of both the groups did not vary. The blood plasma level of some minerals (Ca, P, Mg and Cu) was significantly higher towards the end of experimental feeding as compared to the initial values due to the reduction in milk yield with advancement of lactation or due to supplemental effect of minerals. It could be concluded that supplementation of minerals through inorganic source is better utilized in terms of retention as compared to green fodder (para grass), which is a moderate source of most nutrients.

Degradation of Phytate Pentamagnesium Salt by Bacillus sp. T4 Phytase as a Potential Eco-friendly Feed Additive

  • A bacterial isolate derived from soil samples near a cattle farm was found to display extracellular phytase activity. Based on 16S rRNA sequence analysis, the strain was named Bacillus sp. T4. The optimum temperature for the phytase activity toward magnesium phytate (Mg-$InsP_6$) was $40^{\circ}C$ without 5 mM $Ca^{2+}$ and $50^{\circ}C$ with 5 mM $Ca^{2+}$. T4 phytase had a characteristic bi-hump two pH optima of 6.0 to 6.5 and 7.4 for Mg-$InsP_6$. The enzyme showed higher specificity for Mg-$InsP_6$ than sodium phytate (Na-$InsP_6$). Its activity was fairly inhibited by EDTA, $Cu^{2+}$, $Mn^{2+}$, $Co^{2+}$, $Ba^{2+}$ and $Zn^{2+}$. T4 phytase may have great potential for use as an eco-friendly feed additive to enhance the nutritive quality of phytate and reduce phosphorus pollution.

Growth and Foliar Constituents of Mulberry ($M_5$) Cultivated under Organic Based Nutrient Management

  • A field experiment to evaluate the effect of application of different organic manures and inorganic fertilizers on growth, yield and quality of leaf was studied during 2004-05 has showed that, the application of 10 kg each of Azospirillum brasilense and Aspergillus awamori+20% each of recommended N through compost+green manure (Glyricidia maculata)+castor cake+vermicompost+Urea and remaining P and K through fertilizers ($T_{11}$) has recorded significantly higher leaf yield (250 g/ plant and 34.70 tonnes/ha/yr, respectively) with improvement in growth characters as compared to control. Leaf quality status was also improved in terms of N (3.19%), P (1.97%), K (1.28%), total soluble protein (8.39 mg/ml), total soluble sugars (14.40 mg/ml), secondary nutrients viz., Ca (3.00%), Mg (0.60%), S (0.35%) and micronutrients viz., Cu (0.410 ppm), Mn (0.454 ppm) and Zn (0.112 ppm) contents. The mulberry grown with 20 tonnes of compost+300:120:120 Kg of NPK/ha/year through fertilizer ranked second for growth and foliar constituents.

Evaluation of Physical Property and Material Characteristics for Stained Glass in the Yakhyeon Catholic Church, Korea (약현성당 스테인드글라스의 재료학적 특성과 물성평가)

  • The Yakhyeon catholic church (Historic Site No. 252 in Korea) that was constructed in 1892 has been the first western brick structure to ever have existed and one of the most important historical materials in the Korean modern architecture. After a fire had broken out at the catholic church in 1998, the stained glass on the back wall, the slab glass (dalle de verre) introduced by Lee Nam Guy in 1974, was repaired in 1999 because of singe scorch and water leakage. An analysis of the coloration elements showed that yellow, red and green included Zn, K and Cd, respectively. The glass of red contained Se, dark green contained Mn and Cr, and blue contained Pb and S. According to material analysis, the masonry joint was identified dolomite ($CaMg(CO_3)_2$) and calcite ($CaCO_3$), which was observed plate, columnar, rhombic and square of crystalline particles. Meanwhile, ultrasonic velocity in the stained glass recorded low speed in the middle and lower right of the window (an average of 4,130 m/s). And the joint was measured the lowest physical properties of the top left and middle of the window (an average of 2,053 m/s). This study have showed that extensive physical damage was founded to the left and middle rather than the right side. In this respect, more research in needed to conserve the correlation between color and physical properties.

Protease Properties of Protease-Producing Bacteria Isolated from the Digestive Tract of Octopus vulgaris (Octopus vulgaris의 장관으로부터 분리한 단백질 분해효소 생성 균주와 생성된 효소의 특성)

  • A high protease-producing strain was isolated and identified from the digestive tract of octopus vulgaris by detecting a hydrolysis circle of protease and its activity. The strain was identified by morphology observation, biochemical experiments, and 16S rRNA sequence analysis. The protease obtained from the strain was purified by a three-step process involving ammonium sulfate precipitation, carboxy methyl-cellulose (CM-52) cation-exchange chromatography, and DEAE-Sephadex A50 anion-exchange chromatography. The properties of protease were characterized as well. The strain Bacillus sp. QDV-3, which produced the highest activity of protease, was isolated. On the basis of the phenotypic and biochemical characterization and 16S rRNA gene-sequencing studies, the isolate was identified as follows: domain: Bacteria; phylum: Firmicutes; class: Bacilli; order: Bacillales; family: Bacillaceae; and genus: Bacillus. The isolate was shown to have a 99.2% similarity with Bacillus flexus. A high active protease designated as QDV-E, with a molecular weight of 61.6 kDa, was obtained. The enzyme was found to be active in the pH range of 9.0-9.5 and its optimum temperature was $40^{\circ}C$. The protease activity retained more than 96% at the temperature of $50^{\circ}C$ for 60 min. Phenylmethylsulfonyl fluoride (PMSF) inhibited the enzyme activity, thus confirming that this protease isolated from Bacillus sp. QDV-3 is an alkaline serine protease. Metal ions, $Mn^{2+}$ and $Mg^{2+}$, were determined to enhance the protease activity, whereas $Ba^{2+}$, $Zn^{2+}$, and $Cu^{2+}$ were found to inactivate the enzyme.

Studies on the Change in Chemical Composition of Strawberry during Maturing (딸기 성숙 과정중의 성분변화에 관한 연구)

  • Three types of Korean strawberries, Gardian cultivated in vinyl house, Gardian cultivated on the field and Bogyo, at early variety, on the field were harvested after $10{\sim}34$ days from falling of flowers to investigate the chemical changes of fruits during maturing. The initial pH range after 10 days from falling of flowers was $4.13{\sim}4.28$, which was decreased to $3.40{\sim}3.49$ with maturing. The total acid was steadily increased for 26 days and then decreased thereafter. Vitamin C, not detected until 14 days, was gradually increased to $15.83{\sim}24.90mg%$ in 34 days. Reducing sugar content was increased for 24 days and then decreased, while total sugar, glucose and fructose were generally increased during maturing. Xylose was detected a little. For minerals, potassium was measured the highest value, followed by magnessium, and sodium. A trace amount of manganese, zinc and copper were detected. The strawberries had the content range of $6.00{\sim}24.60mg%$ of methyl alcohol and $6.00{\sim}24.40mg%$ of ethyl alcohol.

Trace Metal Contents in Cultured and Wild fishes from the Coastal Area of Tongyeoung, Korea and their Safely Evaluations (통영연근해역 양식산 및 자연산 어류 중의 미량금속 함량 및 안전성 평가)

  • The content of the trace metals in the cultured and wild fishes were determined. The tested fishes were genuin porgy (Pagrus major) and black porgy (Acanthopogrus schlegeli). The samples of the cultured and wild fishes were collected from slices of raw fish in shops, during 2003 to 2004. The samples were digested with acids, then analyzed by ICP (inductively Coupled plasma Spectrometer) and AAS (Automic Absorption Spectrometer) for the content of lead (Pb), cadmium (Cd), arsenic (As), copper (Cu), manganese (Mn) and zinc (Zn). The content of mercury (Hg) was determined using mercury analyzer. The mean contents of trace metals in cultured and wild fish was 0.031,0.047mg/kg far total-mercury,0.321,0.407 for Pb, 0.048,0.063 for Cd, 1.006, 1.132 for As, 0.467,0.806 for Cu, 0.233, 0.293 for Cr, 9.69, 12.20 for Zn,0.798, 0.624 mg/kg far Mn, respectively. The content of all the trace metals except manganese in wild fish was more than that in cultured fish. The highest level of total-mercury, lead, cadmium, zinc, chromium and arsenic in the samples analyzed were all below the quarantine limit of Korean regulation and guideline established by the U.S. Food and Drug Administration f3r human consumption. The level of the trace metals in the samples was negligible, which could be endogenous. Our data obtained in this study showed that the average weekly intakes of lead, cadmium and mercury from cultured and wild fishes takes about $6\∼13\%$ of Un(Provisional Tolerance Weekley Intakes) that FAO/WHO Joint Food Additive and Contaminants Committee has set to evaluate their safeties.
