실리마린의 항산화 및 항염증 효과 (Antioxidant and anti-inflammatory effects of silymarin)

  • 본 연구는 실리마린의 항산화 및 항염증 활성을 알아보고자 하였다. 항산화 활성은 2,2-diphenyl-1-picrylhydrazyl (DPPH)와 2,2'-azino-bis (3-ethylbenzothiazoline5 6-sulfonic acid) (ABTS) 라디칼에 대한 소거능을 측정하여 확인하였다. 실리마린은 1 mg/mL의 농도에서 DPPH 라디칼을 71%, ABTS 라디칼을 78% 소거하여 우수한 항산화 효과를 나타냈다. 실리마린은 DNA의 산화적 손상을 효과적으로 억제하였고 사람 혈청 단백질과 Cu,Zn-SOD의 산화적 변형을 억제하였다. 또한 실리마린은 H2O2와 LPS에 의한 세포사멸, ROS 생성 및 DNA fragmentation을 억제하였다. 본 연구 결과들을 통해 실리마린은 효과적인 천연 항산화 및 항염증 소재로 적용될 수 있음을 제시하였다.

Transduced HSP27 protein protects neuronal cell death by enhancing FALS-associated SOD1 mutant activity

  • Familial Amyotrophic lateral sclerosis (FALS) is a progressive neurodegenetative disorder induced by mutations of the SOD1 gene. Heat shock protein 27 (HSP27) is well-defined as a stress-inducible protein, however the its role in ALS protection has not yet been established. To investigate the role HSP27 may have in SOD1 mutant-mediated apoptosis, human SOD1 or HSP27 genes were fused with a PEP-1 peptide in a bacterial expression vector to produce a genetic in-frame fusion protein, which was then transduced into cells. We found the purified PEP-1-HSP27 fusion proteins can be transduced efficiently into neuronal cells and protect against cell death by enhancing mutant SOD1 activity. Moreover, transduced PEP-1-HSP27 efficiently prevents protein aggregation produced by oxidative stress. These results suggest that transduced HSP27 fusion protein may be explored as a potential therapeutic agent for FALS patients.

문주란의 항산화효소 활성과 isoenzyme 패턴의 계절적 변화

  • 본 연구는 항산화효소의 효율적 생산을 위한 적절한 수확시기의 결정 및 산업적 가치를 검토하고자, 제주도에 자생하는 문주란 (Crinum asiaticum var. japonicum)에서 항산화효소 (SOD, peroxidase, catalase, APX) 활성과 isoenzyme 패턴의 계절적 변화양상을 조사하였다. SOD isoenzyme 패턴을 전기영동으로 살펴보면, 전체적으로 7개의 isoenzyme이 검출되었으며, 여름철과 겨울철에 있어서 뚜렷한 차이가 없을 뿐 아니라 일변화에 있어서도 큰 차이를 보이지 않았다. 그리고 이들 isoenzyme은 $H_2O$$_2$와 KCN에 의한 선택적 저해로부터 1개의 Mn-SOD와 6개의 CuZn-SOD로 구분할 수 있었다. Catalase는 단일밴드로 나타났으며, 여름철이 겨울철에 비해 높은 활성을 보였다. Peroxidase는 전체적으로 4개의 isoenzyme이 검출되었다. 이 중 peroxidase 1은 효소활성에는 차이가 있지만 여름철과 겨울철에 모두 검출되었으며, 3개의 isoenzyme (peroxidase 2-4)은 겨울철에만 특이적으로 검출되는 것으로 나타났다. 그리고 여름철에는 낮시간에 다소 높은 활성을 보였으며, 겨울철에는 낮시간보다는 새벽과 밤에 높은 활성을 보였다. APX는 8개의 isoenzyme이 검출되었으며, 여름철과 겨울철에 있어서 뚜렷한 차이를 보이지 않았다. 이상의 결과를 종합해보았을 때 SOD, catalase, peroxidase, APX 등 항산화효소의 산업적 생산을 위한 채취는 수확량을 감안하였을때 여름철이 적정 채취시기로 보인다. 그리고 문주란의 catalase와 peroxidase는 SOD나 APX는 달리 단일 밴드 또는 주요 밴드가 있고 높은 활성을 보여 정제시 유리하게 작용할 것으로 보인다. prospects will be also discussed.behaviors to ferromagnetic behavior was observed. Tunneling barrier called "decay length for tunneling" for the films having the thickness of Co layer from 1.4 to 1.6 nm was measured to be ranged from 0.004 to 0.021 ${\AA}$$\^$-1/.문에 기업간 관계를 연구하는 측면에서는 탐험적 연구성격이 강하다. 더 나아가 본 산업의 주된 연구가 질적이고 기업내부만을 연구했던 것에 비교하면 시초적이라고 할 수 있다. 또한 관계마케팅, CRM 등의 이론적 배경이 되고 있는 신뢰와 결속의 중요성이 재확인하는 결과도 의의라고 할 수 있다. 그리고 신뢰는 양사 간의 상호관계에서 조성될 수 있는 특성을 가진 반면, 결속은 계약관계 초기단계에서 성문화하고 규정화 할 수 있는 변수의 성격이 강하다고 할 수가 있다. 본 연구는 복잡한 기업간 관계를 지나치게 협력적 측면에서만 규명했기 때문에 많은 측면을 간과할 가능성이 있다. 또한 방법론적으로 일방향의 시각만을 고려했고, 횡단적 조사를 통하고 국내의 한 서비스제공업체와 관련이 있는 컨텐츠 공급파트너만의 시각을 검증했기 때문에 해석에서 유의할 필요가 있다. 또한 타당성확보 노력을 기하였지만 측정도구 면에서 엄격한 개발과정을 준수하지는 못했다. 향후에는 모바일 컨텐츠 파트너의 기업의 특성을 조사하여 관계성 변수와의 상호관련연구를 진행할 필요가 있다. 관계기간, 의존성, 거래처의 단/복수여부, 서비스 범주 등의 제반 변수를 고려하여 이러한 변수가 양사와의 관계성 변수에 어떤 영향이 있는가를 검증할 필요가 있다. 또한 신뢰, 결속 등 다차원의 개념

흰쥐 간조직의 활성산소 및 제거효소에 미치는 목초액의 영향 (Effect of Pyroligneous Liquor on Oxygen Radicals and Their Scavenger Enzymes in Liver of CD Rats)

  • 시판 목초액 A (원액 35%)을 사용하여 음용수에 0%, 1.0%, 25.0%, 50.0%, 75.0%가 되도록 조제한 다음, CD계 수컷 흰쥐 (150 ${\pm}$ 10 g)에 2개월 동안 조제사료와 함께 자유 음용케 하여 간조직 중의 활성산소로서 수퍼옥시드 라디칼(O$_{2}\;^-$), 히디록시 라디칼, 과산화수소 및 제거효소로서 수퍼옥시드 디스무타아제 (SOD), 글루타치온 퍼옥시다아제 (GPx) 및 카탈라아제 (CAT)에 미치는 영향을 평가하였다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미토콘드리아 획분에서는 O$_{2}\;^-$의 생성량은 대조군 대비 12${\sim}$14%의 O$_{2}\;^-$의 생성량 억제효과가 나타났고, 간조직의 마이크로솜획분에서는 대조군 대비 각각 11.0${\sim}$15.0의 O$_{2}\;^-$의 생성량 억제효과가 나타났다. 간조직 미토콘드리아획분에서 ${\cdot}$OH 생성량이 목초액 PL-25 및 PL-50 투여군에서 대조군 대비 12${\sim}$20%의 ${\cdot}$OH 생성 억제효과가 나타났고, 간조직의 마이크로솜획분에서는 대조군 대비 17${\sim}$20%의 ${\cdot}$OH 생성 억제효과가 나타났다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미토콘드리아획분에서는 대조군 대비 12${\sim}$15%의 유의적인 H$_{2}$O$_{2}$의 생성 억제효과가 나타났고, 간조직의 마이크로솜획분에서는 대조군 대비 20${\sim}$22%의 상당히 유의적인 H$_{2}$O$_{2}$의 생성 억제효과가 나타났다. 목초액 PL-25 및 PL-50 투여군의 간조직의 Mn-SOD 활성은 대조군 대비 15${\sim}$25%나 유의적인 활성 증가효과가 인정되었고, 간조직의 Cu/Zn-SOD 활성은 대조군 대비 11${\sim}$16%의 유의적인 SOD활성 증가효과가 인정되었다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미토콘드리아획분의 GPx 활성은 대조군 대비 10${\sim}$17%의 GPx 활성 증가효과가 인정되었고, 간조직의 마이크로솜획분의 GPx 활성 증가효과가 인정되었다. 목초액 PL-25 및 PL-50 투여군의 간조직의 미트콘드리아획분의 CAT 활성은 대조군 대비 12${\sim}$14%의 유의적인 CAT 활성 증가효과가 나타났고, 간조직의 시토졸 1획분에서는 대조군 대비 15${\sim}$27%의 CAT 활성 증가효과가 인정되었다. 이상의 결과에서 목초액의 장기간 투여는 간조직 중의 활성산소의 억제효과뿐만 아니라 방어시스템으로서 활성산소 제거효소의 역할도 충실히 수행하여 노화를 효과적으로 예방하고 억제할 수 있을 것으로 기대된다.

Supplementation of nano-zinc in lower doses as an alternative to pharmacological doses of ZnO in weanling pigs

  • A set of studies was performed to determine the influence of dietary ZnO concentration and source during two phases (days 0 to 14 and days 15 to 28). Experiment 1: 168 weaned piglets were allocated to four treatment groups in six replicates. The treatments included a basal diet without ZnO supplementation (control), 2,500 mg ZnO/kg (In2500), 500 mg nano-ZnO/kg (N500), and 150 mg nano-ZnO/kg (N150). Experiment 2: 168 weaned piglets were divided into three treatment groups with eight replicates. The treatments included control, In2500, N300, and 150 mg nano-ZnO/kg (N150). An in vitro trial showed that the growth of Listeria monocytogenes, Escherichia coli, and Salmonella typhimurium was inhibited when exposed to 300 and 500 ppm of ZnO after 24 h of incubation. In experiment 1, the average daily gain (ADG) by the pigs was improved in the N500 and IN2500 treatment groups. Colonization of coliforms and Clostridium spp. significantly decreased in the pigs fed the N500 and IN2500 diets in phase 1. The total plasma antioxidant capacity was greater in the IN2500 and N500 treatment groups than in the control. Superoxide dismutase (SOD) activity was greater in pigs fed the IN2500 (phase 1) or the IN2500 and N500 (phase 2) diets than in the control and N150 treatment group. In experiment 2, pigs in the N300 treatment group showed a higher ADG and lower fecal score colonization of coliforms and Clostridium spp. compared with those in the N150 treatment group. In conclusion, nano-ZnO at a dose of 300 ppm showed the same growth as the pharmacological dose of Zn. This provides an option to the pharmacological dose.

Effect of Rhodiola Extract Supplementation on Blood Lipid Concentrations and Anti-Oxidant Status in Rats Fed Highly Oxidized Linoleic Acid Diets

  • The influence of Rhodiola extract on tissue antioxidant status, plasma lipid levels, cholesterol contents of liver and fores were investigated in rats find oxidized linoleic acid. Groups of five-week old male Sprague-Dawley rats fed ad libitum with a diet containing 20% oxidized linoleic acid with or without 300 mg/kg body weight freeze-dried Rhodiola water extract. The antioxidant effect of dietary Rhodiola extract supplementation on the peroxidation potential of rats was investigated. The microsomal thiobarbiruric acid reactive substance (TBARS) contents were changed significantly by Rhodiola extract supplementation. Hepatic Catalase activities were increased in Rhodiola supplemented rats, whereas hepatic Manganese Superoxide Dismutase (MnSOD) or Copper Zinc Superoxide Dismutase (CuZnSOD) were not elevated. In addition, plasma cholesterol lowering effect was observed along with the stimulated excretion of cholesterol through the feces were observed with Rhodiola feeding. Supplementation with Rhodiola extract did not alter high density lipoprotein (HDL) cholesterol. These results support that Rhodiola extract may be effective in protection against oxidative stress, and prevention and treatment of blood dyslipidemia. It demonstntes that Rhodiola extract has a potential to exert anti-atherogenic properties antioxidative capacities .

헛개나무(Hovenia dulcis Thunberg) 추출물이 활성 산소종과 관련한 효소에 미치는 영향 (Effects of Hovenia dulcis Thunberg Extract on Enzymes Related Reactive Oxygen Intermediate)

  • In order to investigate the effects of 70% EtOH extract obtained from Hovenia dulcis Thunberg on enzymes relating reactive oxygen intermediate, cancer-stricken animals induced by DEN (N,N-diethylnitrosamine) were recovered by administering the extract of Hovenia dulcis Thunberg. It showed that there was no effect on the generation of superoxide radical by the extract of Hovenia dulcis Thunberg. However, considering the increase of the activity of Cu, Zn-SOD and Mn-SOD in the tested animal class, the extract of Hovenia dulcis Thunberg could participate directly in removing of superoxides. The experimented-animals treated with the extract of Hovenia dulcis Thunberg showed an increase in the activity of the enzymes, catalase and glutathione peroxidase, which can eliminate hydrogen peroxide pertained in liver tissue. The extract of Hovenia dulcis Thunberg seemed to have some factors that accelerate the oxidation. Also, the extract of Hovenia dulcis Thunberg showed effects on the enzymes relating to the active oxygen toxicity which could be an indicator of aging and body toxicity.

Transcriptional Activation of CuIZn Superoxide Dismutase And Catalase Genes by Panaxadiol Ginsenosides Extracted From Panax ginseng

  • Superoxide dismutase (SOD) and catalase constitute the first coordinated unit of defense against reactive oxygen species. Here, we examined the effect of ginseng saponins on the induction of SOD and catalase gene expression. To explore this possibility, the upstream regulatory promoter region of Cu/Zn superoxide dismutase (SODI) and catalase genes were linked to the chloramphenicol acetyl-transferase (CATI structural gene and introduced into human hepatoma HepG2 cells. Total saponin and panaxatriol did not activate the transcription of SODI and catalase genes but panaxadiol increased the transcription of these genes about 2-3 fold. Among the Panaxadiol ginsenosides, the Rb2 subtraction appeared to is a major induce of SODI and catalase genes. Using the deletion analyses and mobility shift assays, we showed that the 5051 gene was greatly activated by ginsenoside Rba through transcription factor AP2 binding sites and its induction. We also examined the effect of the content ratio of panaxadiol extracted from various compartment of ginseng on the transcription of 5031 gene. Saponin extract that contains 2.6-fold more PD than PT from the fine root Increased the SODI induction about 3-fold. These results suggest that the panaxadiol fraction and its ginsenosides could induce the antioxidant enzymes, which are important for maintaining cell viability by lowering level of oxygen radical generated from intracellular metabolism.

과산화지질의 투여가 흰쥐 간의 산화와 항산화계에 미치는 영향 (The Degree of Lipid Oxidation of Rat Liver Fed Peroxidized Lipid and Its Effects on Anti-Oxidative System)

  • Accumulation of peroxidized lipid, fed or injected in the body of rats was investigated and the effect of peroxidized lipid on the antioxidative system was studied also. Three groups each having six of Sprague-dawley rats were raised for 8 weeks. the peroxide value(POV) of diet fed to the control and the peroxidized group was 5.47 and 22.14meq/kg , respectively. Injected group was given the control diet and peroxidized linoleic acid(POV 31.81meq/kg) was injected into the peritoneal area three times a week. The POV, MDA, and protein carbonyl values of the peroxidized and the injected group (experimental groups) were significantly higher (p<0.05) than those of the control group. Cu, Zn-SOD and M-SOD activity of the experimentla groups increased 1.6 times that of control group at 4 th week. and decreased by 60% of their activityafter 8 weeks of feeding (p<0.05) . Catalase activity, glutathione and Vt, E contents of the experimental groups were significantly lower (p<0.05) than those of the control group during 8 weeks. The accumulation of peroxidcized lipid in liver were ovserved both in the fed or the injected group. The increased of enzyme activity of the experimental group during 4 weeks suggests ianadaptation of antioxidative system to get rid of the peroxidized lipid. Decrease of enzyme activity and glutathione observed as the peroxidized lipid lipid accumulation proceeded further, however, seems to indicate the oxdiative damage of enzyme and protein . Determination of the protein carbonyl content may be used as a method for measuring the oxidative damaging effect of peroxidized lipid.

뇌조직의 활성산소 및 그 제거효소에 미치는 뽕잎 추출물의 영향 (Effects of Mulberry (Morus alba L.) Leaf Extract on Oxygen Radicals and Their Scavenger Enzymes in Brain of SD Rats)

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in brain membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) lecels resulted in significant decreases (13.4% and 21.1%, 12.0% and 13.4%, respectively) in brain mitochondria and microsome of MLE-100 and MLE-300 groups compared with control group. Superoxide radical ($O_2$) levels were significantly decreased about 12% in brain cytosol of MLE-300 group compared with control group. Lipid peroxide (LPO) levels were effectively inhibited (18.1% and 12.3%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Oxidized protein (OP) levels were significantly decreased (14.2%, and 10.9%, respectively) in brain mitochondria and microsomes of MLE-300 groups compared with control group. Mn-SOD activities in brain mitochondria were significantly increased (13.5% and 18.6%, respectively) in MLE-100 and MLE-300 groups, and Cu/Zn-SOD activities in brain cytosol were also effectively increased (about 17.7%) in MLE-300 groups compared with control group. GSHPx activities in brain cytosol were remarkably increased (17.2% and 23.9%, respectively) in MLE-100 and MLE-300 groups compared with control group. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes in brain.
