생물학적 노출평가를 통한 타겟 제조업 근로자의 공정별 인듐 노출위험성 조사 (Investigating the potential exposure risk to indium compounds of target manufacturing workers through an analysis of biological specimens)

  • Objectives: Along with the several cases of pulmonary disorders caused by exposure to indium that have been reported in Japan, China, and the United States, cases of Korean workers involved in processes that require handling of indium compounds with potential risk of exposure to indium compounds have also been reported. We performed biological monitoring for workers in various target manufacturing processes of indium, indium oxide, and indium tin oxide(ITO)/indium zinc oxide(IZO) in domestic factories. Materials: As biological exposure indices, we measured serum concentrations of indium using inductively coupled plasma mass spectrometry, and Krebs von den Lungen 6(KL-6) and surfactant protein D(SP-D) using enzyme-linked immunosorbent assays. We classified the ITO/IZO target manufacturing process into powdering, mixing, molding, sintering, polishing, bonding, and finishing. Results: The powdering process workers showed the highest serum indium level. The mixing and polishing process workers also showed high serum indium levels. In the powdering process, the mean indium serum concentration in the workers exceeded $3{\mu}g/L$, the reference value in Japan. Of the powdering, mixing, and polishing process workers, 83.3%, 50.0%, and 24.5%, respectively, had values exceeding the reference value in Japan. We suppose that the reason of the higher prevalence of high indium concentrations in powder processing workers was that most of the particles in the powdering process were respirable dust smaller than $10{\mu}m$. The mean KL-6 and SP-D concentrations were high in the powdering, mixing, and polishing process workers. Therefore, the workers in these processes who were at greater risk of exposure to indium powder were those who had higher serum levels of indium, as well as KL-6 and SP-D. We observed significant differences in serum indium, KL-6, and SP-D levels between the process groups. Conclusions: Five among the seven reported cases of "indium lung" in Japan involved polishing process workers. Polishing process workers in Korea also had high serum levels of indium, KL-6, and SP-D. The outcomes of this study can be used as essential bases for establishing biological monitoring measures for workers handling indium compounds, and for developing health-care guidelines and special medical surveillance in Korea.

A Study on Characteristics of Atmospheric Heavy Metals in Subway Station

  • In this study, we investigated the atmospheric heavy metal concentrations in the particulate matter inside the subway stations of Seoul. In particular, we examined the correlation between the heavy metals and studied the effect of the heavy metals on cell proliferation. In six selected subway stations in Seoul, particulate matter was captured at the platforms and 11 types of heavy metals were analyzed. The results showed that the mean concentration of iron was the highest out of the heavy metals in particulate matter, followed by copper, potassium, calcium, zinc, nickel, sodium, manganese, magnesium, chromium and cadmium in that order. The correlation analysis showed that the correlations between the heavy metals was highest in the following order: (Cu vs Zn), (Ca vs Na), (Ca vs Mn), (Ni vs Cr), (Na vs Mn), (Cr vs Cd), (Zn vs Cd), (Cu vs Cd), (Ni vs Cd), (Cu vs Ni), (K vs Zn), (Cu vs K), (Cu vs Cr), (K vs Cd), (Zn vs Cr), (K vs Ni), (Zn vs Ni), (K vs Cr), and (Fe vs Cu). The correlation coefficient between zinc and copper was 0.937, indicating the highest correlation. Copper, zinc, nickel, chromium and cadmium, which are generated from artificial sources in general, showed correlations with many of the other metals and the correlation coefficients were also relatively high. The effect of the heavy metals on cell proliferation was also investigated in this study. Cultured cell was exposed to 10 mg/l or 100 mg/l of iron, copper, calcium, zinc, nickel, manganese, magnesium, chromium and cadmium for 24 hours. The cell proliferation in all the heavy metal-treated groups was not inhibited at 10 mg/l of the heavy metal concentration. The only exception to this was with the cadmium-treated group which showed a strong cell proliferation inhibition. This study provides the fundamental data for the understanding of simultaneous heavy metal exposure tendency at the time of particulate matter exposure in subway stations and the identification of heavy metal sources. Moreover, this study can be used as the fundamental data for the cell toxicity study of the subway-oriented heavy metal-containing particulate matter.

  • Effects of exposure to a neurotoxicant, fenitrothion on antioxidant enzyme activities in Chironomus riparius Mg. (Diptera, Chironomidae) larvae were evaluated under laboratory conditions. Exposure to this chemical led to an increase of cupper, zinc type superoxide dismutase and manganese type superoxide dismutase activities and to a decrease of glutathion peroxidase activity. An activation of catalase was observed in the larvae exposed to high fenitrothion concentration. The response of superoxide dismutase was rapid and sensitive to low chemical concentrations, but changes in catalase, total peroxidase and glutathion peroxidase were less sensitive. In this study, antioxidant enzyme activities in Chironomus riparius larvae were identified as pertinent biomarkers for environmental monitoring.

  • Mancozeb is a protective fungicide on plants and a polymeric complex of ethylene bisdithiocarbamate manganese with zinc salt. It is reported to induce teratogenic and carcinogenic effect in laboratory animals. But the immunomodulating effects of Mancozeb exposure have not been systemically evaluated. The purpose of this study was to investigate the effects of Mancozeb on cell-mediated immunity in mice. For ex vivo assessment, mice were orally exposed to Mancozeb dissolved in distilled water as concentrations of 2,500, 5,000, 10,000 mg/kg for single occasion (acute exposure) or 250, 1,000, 1,500 mg/kg/day 5 days a week for 30days(subacute).   • The thermal spray coating process is being used to protect the metals and alloys from wear, abrasion, fatigue, tribology, and corrosion failure. Therefore, in the present study, Al and Zn was deposited by plasma arc thermal spray process onto the steel substrate and their performance was assessed. The bond adhesion result shows that Al coating has higher value attributed to compact, dense, and less porous compared to Zn coating which contain defects/pores and uneven morphology assessed by scanning electron microscopy (SEM). Electrochemical results show that the Al coating exhibited higher impedance value compared to Zn in artificial ocean water solution at prolonged exposure periods. However, both coatings show the increment in polarization resistance with exposure periods which reveal that porosity of coatings is filled by the corrosion products.

카드뮴 및 아연 전처리가 흰쥐의 급성카드뮴 중독 및 Metallothionein생성에 미치는 효과 (Effects of Cadmium and Zinc Pretreatment on the Acute Cadmium Toxicity and Metallothionein Induction in Rats)

  • Tolerance to toxic effects of cadmium(Cd), including lethality has been shown following pretreatment with cadmium and zinc. This study was designed to determine if tolerance also develops to Cd-induced hepatotoxicity and renal toxicity. Three groups of rats(A, B, C), each consisting of 52 rats, were studied and each group was divided into three subgroups(1,2,3), 28 rats for each subgroup. Rats were subcutaneously pretreated with saline(A), $CdCl_2$(0.5 mg/kg, B), and $ZnCl_2$(13.0 mg/kg, C) during time periods of 5 days. At the end of the period, rats were challenged with $CdCl_2$(3.0 and 6.0 mg/kg) by intraperitoneal injection. As for the cadmium levels in rat tissues after 1,3,5,6 days of pretreatments, it was highest in the liver. Then kidney, heart, blood and muscle followed it in that order. After 24, 48 and 96 hours of intraperitoneal injection by challenge doses the concentration of cadmium in liver and kidney increased proportionally to the increase of challenge dosage. However metallothioneins in liver and kidney were increased by the pretreatment of cadmium and zinc. These data indicate the liver is a major target-organ of acute Cd poisoning, and suggest that cadmium induced hepatic injury, via release of Cd-MT, may play an important role in the nephrotoxicity observed in response to short-term exposure to cadmium. This result suggest that increasing cadmium concentrations, gradually accumulating in liver and kidney as the result of the pretreatment, served to induce the synthesis of metallothionein, thus making them resistant to the challenge from cadmium.

고농도 아연 조건에서 수수-수단그라스 교잡종의 생장, 광합성 및 아연 제거능 (Growth, Photosynthesis and Zinc Elimination Capacity of a Sorghum-Sudangrass Hybrid under Zinc Stress)

  • Plant biomass, photosystem II (PSII) photochemical activity, photosynthetic function, and zinc (Zn) accumulation were investigated in a sorghum-sudangrass hybrid (Sorghum bicolor ${\times}$ S. sudanense) exposed to various Zn concentrations to determine the elimination capacity of Zn from soils. Plant growth and biomass of the sorghum-sudangrass hybrid decreased with increasing Zn concentration. Symptoms of Zn toxicity, i.e., withering and discoloration of old leaves, were found at Zn concentrations over 800 ppm. PSII photochemical activity, as indicated by the values of $F_v/F_m$ and $F_v/F_o$, decreased significantly three days after exposure to Zn concentrations of 800 ppm or more. Photosynthetic $CO_2$ fixation rate (A) was high between Zn concentrations of 100-200 ppm ($22.5{\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$), but it declined as Zn concentration increased. At Zn concentrations of 800 and 1600 ppm, A was 14.1 and $1.8{\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. The patterns of stomatal conductance ($g_s$), transpiration rate (E), and water use efficiency (WUE) were all similar to that of photosynthetic $CO_2$ fixation rate, except for dark respiration ($R_d$), which showed an opposite pattern. Zn was accumulated in both above- and below-ground parts of plants, but was more in the below-ground parts. Magnesium (Mg) and iron (Fe) concentrations were significantly low in the leaves of plants, and symptoms of Mg or Fe deficiency, such as a decrease in the SPAD value, were found when plants were treated with Zn concentrations above 800 ppm. These results suggest that the sorghum-sudangrass hybrid is able to accumulate Zn to high level in plant body and eliminate it with its rapid growth and high biomass yield.

아연결핍이 흰쥐에서 metallothionein의 발현과 카드뮴의 흡수 및 분포에 미치는 영향 (The Effects of Dietary Zinc Deficiency on the Expression of Metallothionein, Absorption and Distribution of Cadmium in Rats)

  • Zinc (Zn) is an essential element in biological process, however inadequate Zn status in general population have been recognized. To update the knowledge for Zn-cadmium (Cd) interaction, we studied the intestinal uptake and transport, and the expression of metal transporter proteins (divalent metal transporter 1, DMT1 ; metal transporter protein 1, MTP1 ; zinc transporter 1, ZnTl ; metallothionein 1 , MT1) in duodenum after Cd exposure using Zn deficient animal model. Rats were led Zn deficient (ZnD, 0.5-1.0 mgZn/kg) or Zn supplemented (ZnS, 50mg Zn/kg) diet for 4 weeks, and followed single administration of $^{109}$ CdCl$_2$orally. The body Zn flatus and tissue Cd concentration were determined at 24 hrs after Cd administration. Total body burden of Cd and Cd absorption index (AI, %) were estimated based on the tissue Cd analyzed. DMT1, MTP1, ZnTl and MT1 mRNA were analyzed by using RT-PCR method. Feeding of Zn deficient diet for 4 weeks produced a reduced body weight gain and a depletion of body Zn. Tissue Cd concentration, body burden of Cd and Cd absorption index were higher in the ZnD diet fed rats than the ZnS diet red rats. Especially, Cd concentration in the small intestine (duodenum, jejunum and ileum) and the colon of FeD diet fed rats were higher markedly than in the FeS diet group. The expression levels of DMT1, MTP1 and ZnT1 mRNA in FeD diet fed rats were similar to the FeS diet. The level of MT1 mRNA expression was significantly lower in the FeD than the FeS diet fed rats. Taken together, theses results indicate that Zn deficiency in diet induce an increased intestinal absorption and tissue retention of Cd, and down -regulate the MT1 expression in the intestine which might be play a part of role in Cd absorption and transport in mammalian. These findings suggest that deficiency of essential metal could be enhanced the toxicity of toxic, non-esstial metals through the metal-metal interaction.

Dietary Zinc Effects on Growth Performance and Immune Response of Endotoxemic Growing Pigs

  • A $2{\times}3$ factorial arrangement of treatments was used in a completely randomized design to determine the effects of dietary Zn on performance and immune response of acutely endotoxemic growing pigs (n=96, mean BW=24.9 kg). Factors included 1) intramuscular injection of $10{\mu}g/kg$ BW of Escherichia coli lipopolysaccharide (LPS) or control and 2) supplemental Zn at 10, 50, or 150 ppm. Diets were fed beginning after weaning (initial body weight=7.6 kg) in the nursery and continued for 16 d into the grower phase. The basal corn-soybean meal grower diet contained 1% lysine and 34.3 ppm Zn. Pigs were acclimated for 12 d in the growerfinishing facility before LPS treatment on d 13. Gain, feed intake, and feed efficiency were unaffected by dietary Zn. Feed intake decreased (p<0.10) and gain/feed was greater (p<0.10) from d 13 to d 16 for pigs injected with LPS. Serum Zn and alkaline phosphatase activity increased (p<0.05) with increasing Zn levels. The febrile response to LPS peaked at 6 h post exposure and pigs were afebrile within 12 h. Rectal temperature was greater (p<0.05) in pigs receiving 50 and 150 ppm Zn than in pigs supplemented with 10 ppm Zn. In vivo cellular immune response, measured on d 13 by skin thickness response to phytohemagglutinin (PHA), was greater after 6 h (p<0.05) in pigs fed 10 ppm Zn and exposed to LPS compared to all other treatments, but was not affected at 12, 24 or 48 h. Zinc did not affect mitogen induced lymphocyte proliferation. Zinc supplemented at 50 or 150 ppm resulted in an enhanced febrile response in pigs subjected to iatrogenic endotoxemia, but did not affect pig performance or immune response measurements.
