식용식물의 항산화 효과 검색과 산초의 항산화 성분 (Further Screening for Antioxidant Activity of Vegetable Plants and Its Active Principles from Zanthoxylum schinifolum)

  • The antioxidant activity of methanol extracts of thirty plants was tested using the methol of 1, 1-diphenyl-2-pi-cryl hydrazyl (DPPH) reactivity. Four methanol extracts from Zingiber officinale, Piper nigrum , Zanthoxylum schinifolium and Capsocum annuum were found to be the most effective on DPPH radical scavenging activity. The next effective ones were Perilla frutescens , Sedium sarmentosum , Raphnus sativas, aArctium lappa, Beta vulgaris. Brassica oleracea var. Acephala, bBrassica juncea inorder, and the others did not show a considerable activity. The methanol extract obtained from the seed coats of Zanthoxylum schinifolium was fractinated with several sovlents. The interphase materials exhibited the strongest antioxidant activity and was further purified by silica gel and Sephadex LH-20 column chormatography. Two active principles were isolated and identified as quercetin -3-O-$\alpha$-L-rhamonopyranoiside(quercitrin) and quercetin 3-O-$\alpha$-D-galactopyranoside (hyperoside) by ultraviolet(UV), proton nuclear magetic resonance (1H-NMR) and carbon nuclear magnetic resonance (13C-NMR). Its antioxidative activity was a little higher that that of L-ascorbic acid.

산초 정유성분의 식중독균에 대한 항균 활성 (Antibacterial Activities of Essential Oil from Zanthoxylum schinifolium Against Food-Borne Pathogens)

  • In this study, the antibacterial activities of essential oil from Zanthoxylum schinifolium against four Gram-positive bacteria and six Gram-negative bacteria were investigated. The antibacterial activity of the oils was determined using the agar-well diffusion assay, MIC (minimum inhibitory concentration) and MBC (minimum bactericidal concentration). In particular, essential oil from Z. schinifolium showed higher antibacterial activity against Gram-positive bacteria than against Gram-negative bacteria. Essential oil from Z. schinifolium displayed large inhibition zones especially against Bacillus cereus (31 mm). At concentrations between 0 and $20\;{\mu}g/mL$ the oils showed an antibacterial effect against both Gram-negative and Gram-positive bacteria. The minimum inhibitory concentration (MIC) values against nine bacteria ranged from 1.25 to $5\;{\mu}g/mL$. The minimum bactericidal concentration (MBC) values against eight bacterial ranged from 2.5 to $20\;{\mu}g/mL$, except Shigella sonnei. Furthermore, our finding on the antibacterial activities of essential oils from Zanthoxylum schinifolium validated the use of this plant for medical purposes.

산초와 초피 잎의 화학성분 (Chemical Components of Zanthoxylum schinifolium and Zanthoxylum piperitum Leaves)

  • To study the potential of the Zanthoxylum schinifolium and Z. piperitum leaves, as raw materials for functional food and medicine, apart from male and female, chemical components were carried out. Among general components of sancho and chopi leaves, moisture and crude protein were higher sancho leaf than chopi leaf, but total sugar and crude fat were higher chopi leaf than sancho leaf and the components of major minerals were K, Ca, Mg and Na. Among free sugars, glucose(0.24% and 0.21%) and sucrose(0.19% and 0.27%) were the highest contents in sancho and chopi leaves(male and female), respectively. The organic acid were isolation and identification as malic acid and citric acid, citric acid is higher than malic acid. The total amino acid of sancho and chopi leaves contained proline and glutamic acid in male and female sancho and female chopi leaves, glutamic acid and aspartic acid in male chopi leaf highly in order. The fatty acid contents of four samples were high 15.16%, 9.76%, 8.78% and 9.29% of linolenic acid, respectively. Among many volatile compounds, limonene(13.25% and 19.16%) and citronellal(34.37% and 29.66%) were predominant flavor compounds in sancho and chopi leaves(male and female), respectively.

A study of analgesic effect of Zanthoxylum bungeanum Maxim pharmacopuncture

  • Objectives : This study was carried out to evaluate analgesic effects of Zanthoxylum bungeanum Maxim (ZM) pharmacopuncture on formalin-induced pains in Sprague-Dawley (SD) rats and ICR-mice. Methods : The subjects were divided 8 weeks aged rats with constant pain sensitivity into five groups; normal (treated with normal saline at Taegye (KI3) and before injected with normal saline at hindpaw), Con-1 (treated with normal saline at KI3 before injected with formalin at hindpaw), Lido-1 (treated with lidocaine at KI3), ZMWG-1 (treated with Hot water extraction pharmacopuncture of Zanthoxylum bungeanum Maxim at KI3), ZMEG-1 (treated with ethanol extraction pharmacopuncture of Zanthoxylum bungeanum Maxim at KI3). After 35 minutes, we measured ultrasonic vocalization (USV) and enzyme activities of both Aspartate aminotransferase (AST), Alanine aminotransferase (ALT) in rat serum. In addition, Tail flick test is performed by injecting ICR mice at 5 weeks of age. And it classified into 4 groups (Con-2, Lido-2, ZMWG-2, ZMEG-2) according to the kind of drug (normal saline, lidocaine, ZMW, ZME). After each drug injection, we examined the reaction by placing the tail in water at $50^{\circ}C$. Results : ZME had analgesic effects in the early and late phase of USV during the formalin test. There were no significant differences between ZMEG-1 and Lido-1 in early and late phase of USV. Also, No significant differences observed in serum AST and ALT activity in ZMWG-1 and ZMEG-1 compared with Con-1. For tail-flick test, analgesic effect on warmth significantly increased in Lido-2 and ZMEG-2 compare to that of Con-2. Conclusion : ZME pharmacopuncture had analgesic effects on formalin-induced pain without liver toxicity. Also, tail-flick test suggest that ZME pharmacopuncture could be useful technique on analgesic effect on warmth and treatment of pains.

초피(Zanthoxylum piperitum DC)의 과피와 잎의 방향성분 (Flavor Components of the Fruit Peel and Leaf Oil from Zanthoxylum piperitum DC)

  • 9월 중에 채취한 성숙한 초피 (Zanthoxylum piperitum DC)의 과피와 잎성분의 변성을 고려한 방법인 gas co-distillation으로 추출하고, GC/MS와 함께 workstation BASIC program들을 사용하여 극성이 다른 두 column(FFAP, HP-1)에 대한 authentic standard들의 retention index(RI) 값들을 구하여 HP-1 및 FFAP, RI reference library를 만든 후 초피의 정유성분들의 두 column library 탐색을 병행하여 분석함으로써, 과피에서 myrcene외 14 hydrocarbon들, linalool외 8 alcohol들, citronellal외 3 aldethyde들, carvone외 2 ketone들, methyl salicylate외 7 ester들 그리고 1,8-cineol 외 3 oxide를 확인하였고, 잎에서 limonene외 7 hydrocarbon들, citronellol외 7 alcohol들, cumin aldehyde외 1 aldehyde, carvone, 그리고 estragole외 1 oxide를 확인하였다. 과피에서는 1,8-cineol(25.47%), limonene(11.9%), geranylacetate(9.01%), myrcene(6.15%), geraniol(2.88%), citronellal(2.25%)등이 주성분이었고, 잎에서는 citronellal(23.11%), 1,8-cineol(18.38%), citronellol(6.04%), methyl cinnamate(4.08%)등이 주성분이었다.

인플루엔자 바이러스에 대한 Zanthoxylum속의 항바이러스 효과 검정 (Antiviral Activity of Zanthoxylum Species against Influenza Virus)

  • We aimed to investigate the antiviral activity of Zanthoxylum species against influenza virus A/WS/33, A/PR/8 and B/Lee/40 used by sulforhodamine B (SRB) assay and the action of leaves extracts of Zanthoxylum piperitum on life cycle of influenza virus A/WS/33. Among the twelve extracts, only the leaf extract of Z. piperitum exhibited strong antiviral activity at low concentration of less than 10${\mu}g/m{\ell}$ with no citotoxicity (50${\mu}g/m{\ell}$) against all of three viruses. In addition, only oseltamivir showed antiviral activity with $IC_{50}$ of 65.3${\mu}g/m{\ell}$ against influenza A/WS/33 among the viruses. Furthermore, the leaf extract of Z. piperitum suppressed infection of influenza virus A/WS/33, when added just prior (-1 hr) or after virus inoculation (0 hr). Leaf extract of Z. piperitum directly affect the infectivity of influenza virus A/WS/33 particles. Therefore, Leaf extract of Z. piperitum exhibited higher antiviral activity against three influenza viruses than that of the oseltamivir, which directly interacts with influenza A/WS/33 particles, affecting the initial stages of infection such as receptor binding and virus entry.

산초(Zanthoxylum schinifolium) 추출물과 초피(Zanthoxylum piperitum) 정유의 꽁치과메기 산패 억제 효과 (Suppressive Effects of the Extract of Zanthoxylum schinifolium and Essential Oil from Zanthoxylum piperitum on Pacific Saury, Coloabis saira Kwamegi)

  • 본 연구는 산초 잎의 에탄올 추출물과 초피 과피에서 얻은 정유를 과메기 제조 시 처리하였을 때 산패 억제와 품질 향상 효과를 조사하였다. 산초의 에탄올 추출물(ZS)은 1% 에탄올에, 초피 정유(ZP)는 20% 에탄올에 용해시켜 길이로 2등분하여 탈수시킨 꽁치 1 kg 당 산초 추출물은 0, 0.125, 0.25, 1 ppm 되게, 초피 정유는 0, 0.25, 0.5, 1, 2 ppm 되게 분무하여 과메기를 자연 건조하여 제조하고 다층 필름포장제로 진공 포장하였다. 포장된 과메기를 실험 시까지 -20${^{\circ}C}$에 보관하였다가 개봉한 후 4${^{\circ}C}$ 냉장고에서 1, 3, 7일 보관하며 산패 정도와 아민류의 변화, 관능검사를 실시하였다. 과메기의 산가, 과산화물가 및 TBA값은 개봉 후 보관기간이 길어질수록 증가하였으며 산초 추출물 0.125 ppm 이상, 초피 정유 0.25 ppm에서 효과가 나타났으며 이 효과는 농도 의존적이며 보관기간이 길어질수록 더 뚜렷하였다. 산초 추출물과 초피 정유 처리에 의하여 과메기 내의 dimethylamine과 trimethylamine(TMA)의 증가도 억제하였으며 trimethylamine oxide의 함량은 높게 유지되었고 3일 보관한 과메기에서 TMA<4.5 mg/100 g을 유지시킬 수 있는 산초 추출물의 농도는 0.25 ppm, 초피 정유는 0.5 ppm이었다. 3일 보관된 과메기에 대한 관능검사 결과 초피정유를 0.5-1ppm 처리한 시료에서 가장 좋은 점수를 나타내었다. 결론적으로 소량의 산초추출물과 초피 정유는 과메기의 산패를 억제하여 품질을 개선시키는 효과가 있다고 사료된다.

국내산 초피와 산초의 산업적 활용 가능성: 고당으로 유도된 뇌신경세포 독성에 대한 추출물의 보호 효과 (Industrial potential of domestic Zanthoxylum piperitum and Zanthoxylum schinifolium: Protective effect of both extracts on high glucose-induced neurotoxicity)

  • 본 연구에서는 국내 주요 토종 향신료로 활용되는 초피(Zanthoxylum piperitum)와 산초(Zanthoxylum schinifolium)가 갖는 in vitro 항당뇨 활성과 뇌신경세포 보호 효과를 확인하고 이에 영향을 주는 주요 생리활성물질을 분석하고자 하였다. 추출 용매에 따른 차이를 비교하기 위하여 총 페놀함량을 측정한 결과, 공통적으로 40% 에탄올의 초피 추출물(EZP)과 산초 추출물(EZS)에서 뛰어난 함량을 나타냈으며, ABTS/DPPH 라디칼 소거 활성과 MDA 생성 억제 효과에 대해서도 상대적으로 우수한 항산화 활성을 보였다. 초피 추출물(EZP)와 산초 추출물(EZS)의 당뇨 관련 효소에 대한 저해 효과를 비교한 결과, 초피 추출물(EZP)는 α-amylase 및 α-glucosidase와 같은 효소를 직접적으로 억제하는 능력이 뛰어났으며, 산초 추출물(EZS)는 비효소적 반응으로 단백질과 당의 결합을 막아 최종당화산물의 생성을 억제하는 능력이 뛰어난 것으로 나타났다. 또한, MC-IXC 뇌신경세포에 고당을 처리하여 산화적 스트레스를 유발시켰을 때 생성되는 ROS의 함량과 뇌신경세포 사멸에 대해 우수한 보호 효과를 보여주었다. 이러한 초피 추출물(EZP)와 산초 추출물(EZS)의 생리활성물질을 확인하기 위해 UPLC IMS-QTOF/MSE 분석을 실시한 결과, 초피 추출물(EZP)의 경우 3-CQA, 4-CQA, quercetin-3-O-glucoside 및 quercetin-3-O-rhamnoside가 주요 물질임을 확인하였으며, 산초 추출물(EZS)의 경우 protocatechuic acid glucoside, 5-CQA 및 rutin이 주요 물질임을 확인하였다. 이상의 결과로부터 초피와 산초는 식후 혈당의 급격한 상승을 지연 또는 개선하고 이로 인해 야기되는 산화적 스트레스로부터 뇌신경세포를 보호하여 고혈당으로 인한 퇴행성 뇌질환과 같은 당뇨 및 그 합병증을 예방할 수 있는 천연 유래 건강기능식품 소재로의 산업적 활용 가능성을 확인하였다.

Volatile Compounds of Zanthoxylum piperitum A.P. DC.

  • Volatile compounds, isolated from Chopi (Zanthoxylum piperitum A.P. DC.) using steam distillation, were analyzed by gas chromatography/mass spectrometry-olfactometry (GC-MS-O). Forty-six volatile compounds, consisting of 12 hydrocarbons, 8 aldehydes, 5 esters, 12 alcohols, 4 ketones, 4 oxides and 1 acid, were tentatively identified from the essential oil of Chopi. Unidentified compounds constituted 7.2% of the total peak area. Limonene was the most abundant compound, followed by geranyl acetate, citronellal, cryptone and ${\beta}$-myrcene. In addition, aroma-active compounds, in particular citronellal and limonene, which are related to the citrus and Chopi flavors of Chopi essential oil, were detected. The aroma of Chopi essential oil had a score of 4.8 on the preference test (neither like nor dislike) and a score of 5.97 on the intensity test (slightly strong) using the 9-point hedonic scale.

한국산 산초열매와 나무의 휘발성 향기성분 (Volatile Flavor Components of Korean Sancho Fruit and Tree(Zanthoxylum schinfolium))

  • An attempt was in this to analyzer volatile flavor components of sancho(Zanthoxylum schinfolium). Essential oils in sancho tree and fruit isolated by a simulataneous steam distillation (SDE) methods using n-pentane/diethyl ether as solvent. A total of 57 and 44 components were identified by gas chromatography(GC) and combined gas chromatography spectrometry (GC-MS), respectively. Granyl acetate(29.23%) geraniol (6.80%, p-isopropyl-2-chclohexenone(5.53%), phellandral (4.10%) in sancho fruit and 4, 6, 6-trimethyl-bicyclo(7.47%), T-carvelo(4.60%, ${\alpha}$-cypernone(3.58%) in sancho tree were found to be major volatile flavor. 22 compnents including myrcene, limonene, 1, 8-cinol in sancho fruit and 10 components including 4-(1-methylethyl)-cyclohexanol, methyl undecyl ketone in sancho tree were identified. The contents of unsaturated fatty acids of palmitic acid(24.34%), myristic acid(3,68%) in sancho tree was higher than that of the sancho fruit.
