Development of Molecular Markers for the authentication of Zanthoxyli Pericarpium by the analysis of rDNA-ITS DNA barcode regions (rDNA-ITS DNA 바코드 부위 분석을 통한 산초(山椒) 기원종 감별용 유전자 마커 개발)

  • Objectives : Due to the morphological similarity of the pericarp and description of multi-species in National Pharmacopoeia of Korea and China, the Zanthoxylum Pericarpium is difficult to authenticate adulterant in species levels. Therefore, we introduced the sequence analysis of DNA barcode and identification of single nucleotide polymorphism(SNP) to establish a reliable tool for the distinction of Zanthoxylum Pericarpium from its adulterants. Methods : To analyze DNA barcode region, genomic DNA was extracted from twenty-four specimens of authentic Zanthoxylum species and inauthentic adulterant and the individual internal transcribed spacer regions (rDNA-ITS and ITS2) of nuclear ribosomal RNA gene were amplified using ITS1, ITS2-S2F, and ITS4 primer. For identification of species-specific sequences, a comparative analysis was performed using entire DNA barcode sequences. Results : In comparison of four Zanthoxylum ITS2 sequences, we identified 16, 4, 6, and 4 distinct species-specific nucleotides enough to distinguish Z. schinifolium, Z. bungeanum, Z. piperitum, and Z. simulans, respectively. The sequence differences were available genetic marker to discriminate four species. Futhermore, phylogenetic relationship revealed a clear classification between different Zanthoxylum species showing 4 different clusters. These results indicated that comparative analysis of ITS2 DNA barcode was an useful genetic marker to authenticate Zanthoxylum Pericarpium in species levels. Conclusions : The marker nucleotides, enough to distinguish Z. schinifolium, Z. piperitum, Z. bungeanum, and Z. simulans, were obtained at 30 SNP marker nucleotides from ITS2 sequences. These differences could be used to authenticate official Zanthoxylum Pericarpium from its adulterants as well as discriminating each four species.

Length-weight Relationship and Condition Factor of Zacco platypus in the Lake Hoengseong (횡성호에 분포하는 피라미 (Pale chub: Zacco platypus) 개체군의 Length-weight Relationship 및 Condition Factor)

  • The dynamics of Zacco platypus population of upstream and downstream in the lake Hoengseong, Korea were investigated from April to October 2005. Length-weight relationship, condition factor (K) and relative condition factor $(K_n)$ of Z. platypus were compared by the study stations. The equations based on length-weight relationship in the lake Hoengseong were $Log(T_w)=-2.2s+3.18{\cdot}Log(T_L)\;(r^2=0.99)$. The result in comparison of variations of Z. platypus populations, in lake was more remain to better than in upstream and downstream them. Also the b value, assessed by Length-weight relationship in lake was 3.36, in upstream and downstream were 3.09, 3.15, respectively indicating the fish in lake better than stream. The slopes of population condition controlled by K factor also showed positive relationship. It was higher in lake environment than in stream sample, reflecting that population of Z. platypus distributed in lake Hoengseong was favorable and stable condition. The lake environment seems to be providing more favorable condition for Z. platypus population.

Resilience as a Moderator and Mediator of the Relationship between and Emotional Labor and Job Satisfaction among Nurses working in ICUs (중환자실 간호사의 감정노동과 직무만족도 관계에서 회복탄력성의 조절 및 매개효과)

  • Purpose : The purpose of this study was to identify the moderating and mediating effects of resilience in the relationship between emotional labor and job satisfaction among nurses working in intensive care units (ICUs). Method : The participants were 144 ICU nurses from three university hospitals. Data were collected using structured questionnaires and analyzed by t-test, ANOVA, $Scheff{\acute{e}}$ test, Pearson correlation coefficients, and multiple regression analysis, using SPSS 25.0. The mediating effect of resilience in the relationship between emotional labor and job satisfaction was analyzed by multiple regression analysis according to Baron and Kenny's procedure. Results : Statistically significant negative correlations were found between emotional labor and resilience (r=-.21, p<.014) and between emotional labor and job satisfaction (r=-.34, p<.001). A significant positive correlation was found between resilience and job satisfaction (r=.31 p<.001). A partial mediating effect by resilience was found between emotional labor and job satisfaction (Z=-2.11, p=.034), but no moderating effect was found. Conclusion : To improve the job satisfaction of ICU nurses, evaluation of their emotional labor, resilience, and interventions are necessary to alleviate emotional labor and improve resilience.

Quantifying how urban landscape heterogeneity affects land surface temperature at multiple scales

  • Background: Landscape metrics have been widely applied to quantifying the relationship between land surface temperature and urban spatial patterns and have received acceptable verification from landscape ecologists but some studies have shown their inaccurate results. The objective of the study is to compare landscape metrics and texture-based measures as alternative indices in measuring urban heterogeneity effects on LST at multiple scales. Results: The statistical results showed that the correlation between urban landscape heterogeneity and LST increased as the spatial extent (scale) of under-study landscapes increased. Overall, landscape metrics showed that the less fragmented, the more complex, larger, and the higher number of patches, the lower LST. The most significant relationship was seen between edge density (ED) and LST (r = - 0.47) at the sub-region scale. Texture measures showed a stronger relationship (R2 = 34.84% on average) with LST than landscape metrics (R2 = 15.33% on average) at all spatial scales, meaning that these measures had a greater ability to describe landscape heterogeneity than the landscape metrics. Conclusion: This study suggests alternative measures for overcoming landscape metrics shortcomings in estimating the effects of landscape heterogeneity on LST variations and gives land managers and urban planners new insights into urban design.

The Effect of Emotional Labor on Job Satisfaction in Dental Hygienists: Focusing on the Mediation Effects of Positive Psychological Capital

  • Background: This study investigated the effects of positive psychological capital on the relationship between emotional labor and job satisfaction among dental hygienists. Methods: Data were collected from 296 dental hygienists working at clinical sites in Korea between July 25 and August 15, 2022. Participants were surveyed using structured questionnaires. Statistical analysis was performed using an independent t-test, ANOVA, descriptive statistics, and stepwise multiple regression analysis using SPSS/WIN 21.0. The mediating effect of positive psychological capital on the relationship between emotional labor and job satisfaction was tested using Baron and Kenny's 3-step hierarchical regression analysis. Results: Emotional labor was negatively correlated with positive psychological capital (r=-0.148, p=0.011) and job satisfaction (r=-0.214, p<0.001). Positive psychological capital had a positive correlation with job satisfaction (r=0.441, p<0.001). In addition, the positive psychological capital, had a partial mediating effect (z=-2.472, p=0.013) on the relationship between emotional labor and job satisfaction. Conclusion: Attitudes that reduce emotional labor, which adversely affects job satisfaction, and bolster positive psychological capital are needed.

Transmittance Characteristics by candlepower of Incandescent Lamp (백열등의 광력에 따른 수중 투과특성)

  • The anchovy scoop net fishery is one of the important in the South sea and coastal of Jeju of Korea. They are using incandescent lamp as a fishing lamp at night to gather anchovy shoals in the water surface. Fishing lamp (AC 100 V 500 W$\times$2~3 or AC 100 V 1 ㎾$\times$1) was installed at 1 m ahead of the prow and 1.5 m height from the water surface. The fishing lamp let anchovy shoals rise to the water surface and are attracted to bag net. On this study, the distribution of submarine illumination of 1㎾ and 2 ㎾ incandescent lamp were analyzed and discussed to investigate the ability of fishing lamp which can gather anchovy shoals effectively. The submarine illumination of incandescent lamp showed peak in wave length 690 nm. The relationship between submarine illumination (L) and water depth (Z) of 1 ㎾ and 2 ㎾ incandescent lamp in vertical light is 1 ㎾ : L = 3851. 9 $e^{-1.4587Z}$ $R^2$=0.9952 2 ㎾ : L= 8211.9 $e^{-1.2852Z}$ $R^2$=0.9977 The submarine illumination of 2 ㎾ incandescent lamp of 0~4 m layers appeared to be 3 to 4 times higher than 1 ㎾ incandescent lamp, and in more deep layers than 6 m appeared to be equal value of each lamp. The light of incandescent lamp (1 ㎾) pass through much better into vertical direction than horizontal, and submarine illumination of 20 m layers was 1.0 l$\chi$. Therefore, fishing lamp power is thought that 1 ㎾ incandescent lamp is more efficient than 2 ㎾ to gather anchovy shoals in depth of 15~20 m to water surface.

EFFECT OF THE SHAPE OF IMPINGEMENT PLATE ON THE VAPORIZATION AND FORMATION OF FUEL MIXTURE IN IMPINGING SPRAY

  • The effect of the shape of the side wall on vaporization and fuel mixture were investigated for the impinging spray of a direct injection(DI) gasoline engine under a variety of conditions using the LIEF technique. The characteristics of the impinging spray were investigated under various configurations of piston cavities. To simulate the effect of piston cavity configurations and injection timing in an actual DI gasoline engine, the parameters were horizontal distance from the spray axis to side wall and vertical distance from nozzle tip to impingement plate. Prior to investigating the side wall effect, experiments on free and impinging sprays for flat plates were conducted and these results were compared with those of the side wall impinging spray. For each condition, the impingement plate was located at three different vertical distances(Z=46.7, 58.4, and 70 mm) below the injector tip and the rectangular side wall was installed at three different radial distances(R=15, 20, and 25 mm) from the spray axis. Radial propagation velocity from spray axis along impinging plate became higher with increasing ambient temperature. When the ambient pressure was increased, propagation speed reduced. High ambient pressures tended to prevent the impinging spray from the propagating radially and kept the fuel concentration higher near the spray axis. Regardless of ambient pressure and temperature fully developed vortices were generated near the side wall with nearly identical distributions, however there were discrepancies in the early development process. A relationship between the impingement distance(Z) and the distance from the side wall to the spray axis(R) was demonstrated in this study when R=20 and 25 mm and Z=46.7 and 58.4 mm. Fuel recirculation was achieved by adequate side wall distance. Fuel mixture stratification, an adequate piston cavity with a shorter impingement distance from the injector tip to the piston head should be required in the central direct injection system.

It is regarded that there will be a clinical availability which can analogize the result of a part by using the result of the other part. It will be able to determine an auxiliary role in the clinical diagnosis. Correction factor to the equation Y = 1.341X + 0.146 is recommended.

Effects of Health Perceptions and Social Support on Health Promotion Behaviors among College Students: the Mediating Effect of Self-efficacy (대학생의 건강지각, 사회적 지지가 건강증진행위에 미치는 영향: 자기효능감의 매개효과)

  • We attempted to identify the mediating effect of self-efficacy on the relationship between health perception, social support, and health promotion behaviors among university students and to provide basic data to improve health promotion behaviors. Data were collected using a structured questionnaire from a sample of 173 university students in City C. Health promotion behaviors according to general characteristics were found to have statistically significant differences in gender(t=3.892, p<.001) and major satisfaction(F=5.867, p=.003) .There was no statistically significant difference in age, grade, major field, hospitalization or surgery experience in the last three years, and economic activity. Health promotion behavior was positively correlated with health perception (r=.439, p<.001), social support (r=.532, p<.001), and self-efficacy (r=.533, p<.001) and was statistically significant. It was found that self-efficacy had a partial mediating effect (Z=3.964, p<.001) on the effect of health perception on health promotion behavior, and self-efficacy had a partial mediating effect (Z=4.24, p<.001) on the effect of social support on health promotion behavior. Therefore, it is necessary to identify the factors affecting health support, social support, and self-efficacy among college students and prepare intervention measures to improve health promotion behavior.

Characteristics of Fish Communities and Length-weight Relationships of Dominant Species (Zacco platypus) in Seomjin River (섬진강 수계 어류군집 특성과 우점종 피라미(Zacco platypus)의 전장 - 체중 관계 분석)

  • As fish communities are sensitive to natural environmental changes as well as anthropogenic pressures, their composition and characteristics help us to assess the aquatic ecosystem health. The fish fauna and the length-weight relationship of Zacco platypus in the Seomjin River system were investigated from July 2018 to May 2019. The collected species during the survey period were 49 species belong to 14 families including 19 Korean endemic and two endangered species. According to Bray-Curtis similarities, 14 sites were divided into four groups based on the fish community composition; two groups(group A, B) and two uncategorized sites(group C, D). There were significant differences between groups A and B(analysis of similarities, R=0.722, p=0.002). A similarity percentage analysis revealed that Z. koreanus(8.55%), Micropterus salmoides(6.90%), and Lepomis macrochirus (4.90%) contributed to these differences of fish assemblages between group A and B. Group A showed higher relative abundances of exotic species such as M. salmoides and L. macrochirus, while Z. platypus and Z. koreanus which are the common species in Korea, were the most dominant species in group B. Based on the length-weight relationship of Z. platypus, b values were varied from 2.82 to 3.80, indicating that growth patterns of the species were spatially different. This study could be served as baseline data for understanding fish fauna, assessing habitat characteristics based on the fauna, and identifying health conditions of Z. platypus in the Seomjin River system.
