Risk Assessment of Drometrizole, a Cosmetic Ingredient used as an Ultraviolet Light Absorber

  • As the use of cosmetics has greatly increased in a daily life, safety issues with cosmetic ingredients have drawn an attention. Drometrizole [2-(2'-hydroxy-5'-methylphenyl)benzotriazole] is categorized as a sunscreen ingredient and is used in cosmetics and non-cosmetics as a UV light absorber. No significant toxicity has been observed in acute oral, inhalation, or dermal toxicity studies. In a 13-week oral toxicity study in beagle dogs, No observed adverse effect level (NOAEL) was determined as 31.75 mg/kg bw/day in males and 34.6 mg/kg bw/day in females, based on increased serum alanine aminotransferase activity. Although drometrizole was negative for skin sensitization in two Magnusson-Kligman maximization tests in guinea pigs, there were two case reports of consumers presenting with allergic contact dermatitis. Drometrizole showed no teratogenicity in reproductive and developmental toxicity studies in which rats and mice were treated for 6 to 15 days of the gestation period. Ames tests showed that drometrizole was not mutagenic. A long-term carcinogenicity study using mice and rats showed no significant carcinogenic effect. A nail product containing 0.03% drometrizole was nonirritating, non-sensitizing and non-photosensitizing in a test with 147 human subjects. For risk assessment, the NOAEL chosen was 31.75 mg/kg bw/day in a 13-week oral toxicity study. Systemic exposure dosages were 0.27228 mg/kg bw/day and 1.90598 mg/kg bw/day for 1% and 7% drometrizole in cosmetics, respectively. Risk characterization studies demonstrated that when cosmetic products contain 1.0% of drometrizole, the margin of safety was greater than 100. Based on the risk assessment data, the MFDS revised the regulatory concentration of drometrizole from 7% to 1% in 2015. Under current regulation, drometrizole is considered to be safe for use in cosmetics. If new toxicological data are obtained in the future, the risk assessment should be carried out to update the appropriate guidelines.

The Effects of New Nonspecific Immunostimulators in Pig (면역기능 증강성 신물질에 대한 돼지에서의 면역 증강성 실험)

  • New nonspecific immunostimulators (Koko enterprise Co., Ltd., Korea) were evaluated for its effectiveness as a nonspecific immunostimulator in pigs. The effects of new nonspecific immunostimulators were determined by analysis of cytokines using ELISA and blood-chemistry. IFN-r which is one of the cell mediated immune cytokines significantly increased in DIR-vitamineral 0.2% group posttreatment 4 weeks and was significantly higher IMF 0.2%, DIR-vita 0.1% and DIR-vitamineral 0.2% groups than control group in 3 months. DIR-vitamineral 0.2% had a most strong effectiveness as a nonspecific immunostimulator in our treatment materials in pigs. IMF 0.2% and DIR-vita 0.1% were seen that there had effectiveness of a nonspecific immunostimulator in posttreatment 3 months. IgG IgM and Total Ig which were humoral immune globulins, were not significantly changed in posttreatment 4 weeks and 3 months. In conclusion, this study has demonstrated that new nonspecific immunostimulators had an immunostimulatory effect on pigs through cell mediated immune response.

Influence of Houttuynia cordata Thnub Powder Mixtures on Plasma Lipid Composition in Pigs (어성초 분말 혼합 식이가 돼지의 혈장 지질성분에 미치는 영향)

  • This study was designed to investigate the influence of Eosungcho (Houttuynia cordata Thunb) mixture on the plasma lipids of pigs. Experimental groups were divided into 3 groups: control group(0%), 5% and 10% Eosungcho feeding groups. Plasma concentration of total cholesterol, atherosclerotic index, free cholesterol and cholesteryl ester were significantly lower in the Eosungcho powder 5% and 10% feeding groups than control group. Plasma contents of HDL-cholesterol, cholesteryl ester ratio, triglyceride and phospholipid were significantly different between 10% Eosungcho feeding group and control group. Plasma concentration of LDL-cholesterol was significantly love in 10% Eosungcho feeding group($76.5{\pm}5.4mg/dL$) than in control group($86.5{\pm}5.6mg/dL$). Plasma concentration of VLDL and chylomicron were showed similar tendency to LDL-cholesterol. Also, blood sugar, GOT and GPT concentration were significantly lower in 10% Eosungcho feeding group than in control group. In control group, the ratio of saturated fatty acid to unsaturated fatty acid was 58.1 : 51.9, but in 5% and 10% Eosungcho feeding groups, those were 42.1 : 57.9 and 41.0 : 59.0, respectively. The component of main fatty acid was different between control group and Eosungcho feeding groups. In Eosungcho feeding groups, main unsaturated fatty acids were oleic acid(18:1) and linoleic acid(18:2). From the results, addition of Eosungcho powder might be effective in improving the plasma lipid components in pigs.

The Study on the Amount and Major Compositions of Excreta from Swine (돼지의 분뇨 배설량 및 분뇨 성분 조사)

  • This work was carried out to investigate the quantity of excreta and its composition in crossbred pigs (Yorkshire ${\times}$ Landrace ${\times}$ Duroc) at different stages of growth. Twelve young piglets (average BW weight of $19.0{\pm}0.33kg$) were used in this study. Pigs were divided into four phases during growing time and two phases during finishing time. The average excreta production for growing pig was 3.46 kg/head/day (feces: 1.07 kg, urine: 2.39 kg). The average moisture contents of feces and urine were 70.54% and 97.39%, respectively. Contents of Calcium, Magnesium, Copper, Plumbum, and Arsenic were 1.00%, 0.26%, 10.47 mg/kg, 2.43 mg/kg, and 1.02 mg/kg, respectively. The concentration of the water pollutants like Biochemical Oxygen Demand ($BOD_5$), Chemical Oxygen Demand (COD), Suspended Solid (SS), Total Nitrogen (TN) and Total Phosphorus (TP), excreted from pig were 96335, 61073, 207466, 8104 and 4209 mg/L in feces and 7364, 7149, 2715, 10110 and 613 mg/L in urine at the end of test, respectively. The daily loading amount of water pollutants ($BOD_5$, COD, SS, TN, and TP, respectively) in pig excreta were 102.1, 61.8, 221.6, 8.7, and 3.9 g/head/day in feces, and 19.3, 16.7, 8.0, 22.2, and 1.3 g/head/day in urine, respectively. The Nitrogen, $P_2O_5$, and $K_2O$ contents in the excreta of pigs were 0.96, 0.83 and 0.42% in feces, and 0.80, 0.09 and 0.53% in urine, respectively. Finally, this work was suggested to give basic information to swine farms.

  • Salmonella (S.) Enterica infection ranks among the most common food borne bacterial infections worldwide. Although there are six subspecies of S. Enterica, the vast majority of human and animal infections are caused by strains belonging to subspecies 1 serovar Typhimurium and Enteritidis. Recent reports on antibiotic resistance of Salmonella spp. are rising steadily. The increasing problem of antibiotic resistance has rekindled interest in bacteriophage to therapy. Therefore, we investigated the efficacy of bacteriophage in S. enterica serovar Enteritidis infected mice and pigs by measuring of body condition, body weight, bacterial colonization and weight of organs based on the in vitro analysis. In vitro experiment, phage cultured with S. Enteritidis showed clear lysis pattern, the plaque forming unit (PFU) of our phage culture was $1.5{\times}10^{11}PFU/mL$, and phage showed its maximum activity at 4 h post inoculation. In mouse experiment, there was no significant difference among experimental groups in the general body conditions and body weight of mice. However, there was difference in weight of liver and spleen depending on the experimental group (p < 0.05). The weight of liver and spleen were reduced by the phage treatment. Also bacterial colonization in spleen and liver were significantly reduced by the phage treatment. In pig experiment, the general body conditions and body temperature exhibited not much difference among the pigs except few pigs in group 3 which showed poor body conditions. From the feces in each group, we could isolate the S. Enteritidis only from group 3. Bacterial enrichment culture was necessary for isolating the bacteria from 5 dpi and 10 dpi, however direct isolation was possible from 15 dpi feces. In phage treated group, postmortem lesion was better than non-phage treated group. Recently, antibiotic resistance concerns on the food-borne bacterial pathogens have been increasing because of the wide spread of the antibiotics resistance genes. This concern is widely transmitted to the human related public health. As one of the alternative treatments on the bacterial pathogens, attempt using phages have been made to control the bacterial diseases. The positive possibility of the trail using phage was observed to control the S. enterica serovar Enteritidis in this study even though the further analysis has been remained.

Association with Kinetic Characteristics of sperm in Duroc Boar and the Zygote Arrest 1 gene Polymorphism (g.2540T>C) (Zygote arrest 1 유전자 변이(g.2540T>C)와 두록 정액의 운동학적 특성과의 연관성 분석)

  • The sperm quality is determined by the kinetic characteristics and acrosome integrity of the sperm. In the previous studies, analysis of semen quality had large errors because those experiments by using microscope had been conducted by people. In recent years, the molecular biological methods have been newly developed to complement the previous techniques. The ZAR1 gene is known to be a gene that affects early embryonic development in vertebrates, but there is no study of the association with semen. In this study, we analyzed the association between the kinetic characteristics and ZAR1 single nucleotide polymorphism (SNP) genotype. To detect the SNPs, we performed sequencing using genomic DNA from the whole bloods of Duroc pigs. We identified an SNP in the ZAR1 gene g.2540T>C. ZAR1 SNP genotypeing in 105 pigs revealed that the major and minor alleles were T and C, respectively. After we analyzed the association between the kinetic characteristics of sperm and the ZAR1 SNP genotype, we found a significant association in MOT (p<0.01), VSL (p<0.05) of the kinetic characteristics in the Duroc boars. It was confirmed that the boars with T allele were lower in MOT and VSL than C allele. Therefore, pigs with C allele are judged to be better at the MOT and VSL of semen. Based on these results, ZAR1 SNP genotyping may be a useful molecular biomarker to improve semen quality by applying molecular breeding technology.

Effect of Feeding Probiotics on Physico-chemical Properties and Sensory Evaluation of Pork (생균제의 급여가 돈육의 이화학적 성상 및 관능에 미치는 영향)

  • A total of 120 pigs were used to investigate the effect of feeding probiotics on physico-chemical properties and sensory evaluation of pork loin. About 50kg pigs were randomly alloted into one of six experimental diet groups (C1:commercial diet feed the gilt; C2:commercial diet feed the barrow; T1:$0.5\%$ YC2000 feed the gilt T2:$0.5\%$ YC2000 feed the barrow; T3:$0.1\%$ YC2000 + $0.3\%$ KBC1121 feed the gilt; T4:$0.1\%$ YC2000 + $0.3\%$ KBC1121 feed the barrow). Pigs were slaughtered at approximately 110kg live weight. Crude fat and crude ash were not difference among the treatments. However, water content was higher in T1 and T2 compared to other treatment and the protein level of T3 was higher than those of other treatments. All of dietary probiotic groups showed higher pH compared to control. Especially, pH of T1 and T2 were higher among the dietary probiotic groups. Cholesterol level of dietary probiotic groups were lower compared to control. In meat color, $a^{*}$ was higher in T1 and $b^{*}$ was lower in T2 compared to other treatments. In sensory evaluation of cooked meat, aroma, flavor, tenderness, juiciness and overall palatability were higher in control, whereas T3 and T4 showed higher score in tenderness, juiciness and overall palatability. T3 had higher myristic acid. palmitoleic acid and oleic acid, whereas arachidonic acid was lower in T3. In conclusion, dietary probiotic groups were much better than other treatments in cholesterol, color, tenderness and juiciness. But drip loss of dietary probiotic groups showed higher due to lower pH compared to control.

About 6 kg pigs were randomly alloted into one or three experimental diet groups (C: commercial diet feed; T1: 0.1% KBC1121 feed; T2: 0.1% YC2000+0.1% KBC1121 feed). Pigs were slaughtered at approximately 110 kg live weight and chemical composition and physico-chemical characteristics were measured in pork loin. Moisture, crude protein and crude ash were not differences among the treatments. However, crude fat content of T2 was significantly higher than that of other treatments. All of dietary probiotic groups showed significantly higher pH than control. WHC was significantly higher in T1 than other treatments. Cooking loss, shear force value and cholesterol content were not differences among the treatments. In meat color, $L^*$ value was not difference among the treatments, but $a^*\;and\;b^*$ value were lower in T1 than other treatments. In textrure properties of cooked meat, brittleness, hardness, gumminess and chewiness value were significantly higher in T1 than other treatments. Sensory evaluation was not difference among the treatments. The myristic, stearic and oleic acid content of T2 were significantly higher than those of other treatments. Whereas linoleic acid was significantly lower than other treatments. Unsaturated fatty acid (UFA) was significantly higher in T1 than T2. Essential fatty acid (EFA) and EFA/UFA were higher in the order of T1 > C > T2. In amino acid composition, total and essential amino acid, aspartic acid, threonine, serine, glutamic acid, valine, isoleucine, leucine and lysine level were lower in T2 than other treatments.

Comparison of Meat Quality Characteristics between Crossbreeds (돼지 품종의 교배조합간 육질특성 비교)

  • This study was conducted to investigate the meat quality characteristics of pork loin in 3 crossbred groups (30 pigs/group). The crossbreeds were LY female${\times}$D male, Y female${\times}$B male and L female${\times}$B male (L: Landrace, Y: Yorkshire, B: Berkshire, D: Duroc). Pork loins were collected from animals ($110{\times}120kg$ body weight) slaughtered in a commercial slaughter house and stored at $-3^{\circ}C$. Samples were analyzed for general composition, physico-chemical properties, meat and fat color, texture characteristics, sensory evaluation and fatty acid composition at 24 hrs postmortem, and TBARS (Thiobarbituric Acid Reactive Substances) values were determined at various storage times. The LYD pigs showed a higher back fat thickness than the YB and LB pigs. There were no significant differences in intramuscular fat, intermuscular fat, subcutaneous fat and springiness among the 3 crossbreeds. In addition, there were no significant differences in general composition, pH, drip loss, cooking loss and cooked sensory evaluation among the 3 crossbreeds (p>0.05). The shear force value of LB pork was significantly lower than other crossbreeds (p<0.05). Total heme pigment was significantly higher in YB pork compared to LYD and LB pork (p<0.05). The TBARS value of YB pork was significantly lower than that of LYD pork, except after the I day of storage (p>0.05). TBARS values increased significantly during storage for all three crossbreeds. The lightness (CIE L) of YB pork was significantly lower, while the redness (CIE a) was significantly higher in YB pork relative to the other crossbreeds. With regard to textural properties, the hardness and adhesiveness values were significantly lower in YB pork compared to LB pork. In fresh meat, the color of YB pork was significantly higher than the other breeds (p<0.05). The content of myristic acid (C14:0) was significantly lower, while the content of arachidonic acid (C20:4) was significantly higher in LB pork (p<0.05). These results show that the YB breed has the highest quality meat of the three crossbreeds.

Analysis of Reproductive and Growing Performances by Sow Farm Sizes (국내 양돈장의 모돈규모별 번식 및 육성성적 분석)

  • This study was carried out to investigate the reproductive and growing performances in sow farms located in Chungnam. Data collected from a total of 15 sow farms divided into 3 farm groups for 1 year were analyzed. The results obtained were as follows; The average of sow at small and large farms were 114.25 and 487.88 heads, respectively. And then, the difference among 3 groups in sow farm was not found significantly in farrowing rate. The significant difference among 3 farm groups was significantly found(p<0.05) in total litter size. The litter size born alive at sucking in small sow farm and middle farms were 9.93, and 10.48 pigs. The difference between small and large farms in number of pigs at weaning were significantly shown 8.89, and 9.35 pigs(p<0.05), respectively. The difference among 3 sow farms for ages at weaning showed significantly (p<0.05). The cycles of farrowing rate per year in small, large and middle farms were 2.17, 2.23 and 2.32, respectively. The significant difference among 3 sow groups was found in farrowing rate(p<0.05). The growing rate up to weaning for middle farms was significantly the higher level(94.70%) than that for other farms(p<0.05). The differences among 3 farm groups for marketing weight were not significantly shown.
