Objective: The objective of this experiment was to investigate the effects of fat-soluble vitamin injection on plasma and tissue vitamin status in nursery pigs. Methods: A total of 16 pigs (initial body weight: 7.15±1.1 kg) were allotted to 2 treatments at d 7 post-weaning. Pigs were fed a corn-soybean meal-based basal diet with no supplemental vitamin A and i.m. injected with 300,000 IU of retinyl palmitate, 900 IU of d-α-tocopherol and 30,000 IU of vitamin D3 with control pigs having no vitamin injection. Blood (d 0, 3, 7, and 14 post-injection) and tissue samples (liver, brain, heart, lung, and muscle; d 7 and 14 post-injection) were collected from pigs. Retinyl palmitate, retinol, and α-tocopherol concentrations were analyzed in plasma and tissues, while plasma was assayed for 25-hydroxycholecalciferol (25-OHD3). Results: Plasma retinol and 25-OHD3 concentrations increased by the vitamin injection from d 3 to 14 post-injection (p<0.05) whereas plasma retinyl palmitate was detected only in the vitamin treatment at d 3 and 7 post-injection (115.51 and 4.97 ㎍/mL, respectively). Liver retinol, retinyl palmitate, and retinol+retinyl palmitate concentrations increased by retinyl palmitate injection at d 7 and 14 post-injection (p<0.05) whereas those were not detected in the other tissues. The d-α-tocopherol injection increased α-tocopherol concentrations in plasma at d 3 and 7 post-injection (p<0.05) and in liver, heart (p<0.10), and muscle (p<0.05) at d 7 post-injection. Conclusion: Fat-soluble vitamin injection increased plasma status of α-tocopherol, retinol, retinyl palmitate and 25-OHD3. As plasma levels decreased post-injection, vitamin A level in liver and vitamin E level in muscle, heart and liver increased. The α-tocopherol found in plasma after injection was distributed to various tissues but retinyl palmitate only to the liver.
Studies were carried out to determine the effect of feeding diet containing 5% canola oil on growth, feed efficiency, and fatty acid profile of bacon in finishing pigs and of longissimus muscle in horses fattening for meat production. In experiment 1, twenty cross-bred barrows and twenty cross-bred gilts (average weight, 80 kg) were blocked by sex and weight, and five barrows or five gilts were allotted to one of eight pens $(6.25m^2/pen)$, respectively. Four pens (two with barrows and two with gilts) randomly selected were assigned to a control diet containing 5% tallow and the remaining four pens to a diet containing 5% canola oil. The average daily weight gain, daily feed intake and feed efficiency over a 6-wk feeding period were not different (p>0.05) between the two diets, nor was backfat thickness. Fatty acid profile in bacon fat showed that the 0-3 fatty acid ($\alpha-linolenic$ acid) content in pigs fed diet containing 5% canola oil was approximately three times (P<0.01) as much as in pigs fed tallow. In experiment 2, thirty-two Jeju horses (average $weight{\pm}SE,\;244{\pm}5kg$) were blocked by sex and weight, and two horses of the same sex and similar body weight were allotted to one $(15m^2/pen)$ of eight pens. Eight pens (four with males and four with females) selected randomly were assigned to a control diet containing 5% tallow and the remaining eight pens to a diet containing 5% canola oil. The average daily weight gain, daily feed intake and feed efficiency for concentrates without roughages over a 5-month feeding period were not different (P>0.05) between the two diet groups. Fatty acid profile in the muscle fat showed that the 0-3 fatty acid (a-linolenic acid) content in horses fed diet containing 5% canola oil was approximately two times (P<0.01) that in horses fed tallow. The increased (P<0.01) 0-3 fatty acid content in pigs and horses fed canola oil decreased the ratio of n-6 to n-3 fatty acids compared to the control, indicating a significant improvement in pork and horsemeat fatty acid profile for health benefit. Our study demonstrated that feeding diet containing 5% canola oil may help produce pork and horsemeat with more health benefit, increasing their $\alpha-linolenic$ acid content without deleterious effects on growth of pigs and horses.
Somatic cell nuclear transfer (SCNT) for miniature pig has been developed for xenotransplantation and many other biomedical experiments. However, the efficiency of SCNT is still very low due to many factors. To optimize the surrogate mother condition for improvement of cloned miniature pigs efficiency, we investigated the effect of the status of surrogate mother on pregnancy, farrowed rate in SCNT pigs. After SCNT with mesenchymal stem cells as donor cells, the SCNT embryos were surgically transferred into the oviduct of surrogated pigs. To compare the effects of status of surrogate pigs on pregnancy, surrogate pigs were prepared by artificial abortion at day 20~29 (Group 1), 30~39 (Group 2), and 40~45 (Group 3) of gestation. After SCNT embryos transfer in three different status of surrogate pigs, Group 2 (56.3%) and 3 (55.6%) had significantly ($p$ <0.05) higher the pregnancy rate than group 1 (0%) at day 30 of gestation. The status of ovulation in surrogate pig also was investigated. Post-ovulation status (54.8%) had higher proportion than pre-ovulation status (38.7%) and ovulation status (6.5%). We obtained 19 cloned miniature piglets from seven surrogate gilts and five piglets are living healthy but fourteen piglets died soon after birth or stillbirth. The weights of piglets greatly differ from 254 to 1,296 g. Microsatellite analysis showed that cloned piglets were genetically different from the surrogate mother and cloned piglets were genetically equal to the donor cell. In conclusion, the present result indicates that artificially abortion method can improve the efficiency of pregnancy after SCNT in pigs. This study will provide available method for the further study and application in the field of xenotransplantation.

In a trial involving 120 pigs, the growth performance of finishing pigs and proximate composition, physicochemical properites and fatty acid composition of loin muscle were investigated by feeding the high-energy density diet and low-energy density diet. The treatments included feeding 1) the low-energy density diet (3,290 cal/kg DE, 14.50% CP, 0.70% lysine and 0.78% Ca) for 38 days and 2) the hight-energy density diet (3,350 cal/kg ME, 15.0% CP, 0.70% lysine and 0.78% Ca) for 27 days. The average daily gains were significantly higher for pigs fed the high-energy density diet than those fed the low-energy density diet, but average daily feed intake and feed conversion rate were significantly lower than those fed the low-energy density diet (p<0.05). The cooking loss and yellowness of pork were significantly (p<0.05) lower for pigs fed the low-energy density diet than those fed the high-energy density diet. There were no significant differences in carcass characteristics, proximate composition and fatty acid composition of pork. In conclusion, the results of the experiment suggested that the high-energy density diet for pigs tended to improve the growth performance, but the cooking loss and yellowness of pork were higher for pigs fed the high-energy density diet than those fed the low-energy density diet.

To examine the in vivo immunostimulating effect of conjugated linoleic acid (CLA) in pigs, the change of peripheral blood cells and the phagocytic response of phagocytes were evaluated. Spayed male pigs, 80 kg of average body weight, fed a diet containing either 0.5% 10t-12c CLA or 0.5% CLA mixture (mostly 9c-11t CLA and 10t-12c CLA) for 4 weeks. The change of blood cell values (PCV, WBC, differential count of WBC) and the phagocytic activities of phagocytes were evaluated on week 0, 2, 4, and 5, respectively. There were no change in the PCV values regardless of CLA supplement. The number of WBC, especially neutrophils, in pigs fed a diet with CLA was significantly increased (p<0.05 to 0.01) when compared with control pigs fed a diet without CLA. The phagocytosis of peripheral blood mononuclear cell (MNC) and peripheral blood polymorphonuclear cells (PMN) were analyzed by a flow cytometry system. There was no change in the phagocytic activity of MNC and monocyte-rich cells regardless of CLA supplement. However, the phagocytic activity of PMN composed by approximately 95% neutrophils was remarkably increased (p < 0.05 to 0.01) on week 2, 4, and 5 as compared wth control pigs. These results suggested that supplement of CLA into pigs induces the increase of neutrophil number and the enhancement of neutrophil phagocytosis.
We have previously found that mycelia culture broth of eight kinds of traditional herbal extracts fermented with Phellinus linteus (previously named as 8-HsPLCB) not only inhibited melanin and tyrosinase activity, but also reduced the contents of melanogenesis-related proteins, including tyrosinase and microphthalmia-associated transcription factor, in 3-isobutyl-1-methylxanthine-stimulated B16F0 melanoma cells. For a further study, the effect of 8-HsPLCB against skin pigmentation in brown guinea pigs with ultraviolet B (UVB)-induced hyperpigmentation was investigated. 8-HsPLCB (3%) and arbutin (2%) as positive controls were applied topically twice daily for 4 weeks to the hyperpigmented areas. 8-HsPLCB showed skin-lightening effect as effective as arbutin, one of the most widely used in whitening cosmetics. Melanin index values as the degree of pigmentation showed a significant reduction week by week post 8-HsPLCB treatment and then substantially reduced by 4 weeks. The degree of depigmentation after 4 weeks of topical application with 8-HsPLCB was 32.2% as compared with before treatment (0 week). Moreover, using Fontana-Masson staining and hematoxylin-eosin staining, 8-HsPLCB reduced melanin pigmentation in the basal layer of the epidermis and epidermal thickness changes exposed to the UV-B irradiation as compared with non-treatment and vehicle treatment. The intensity of the skin-lightening effect of 8-HsPLCB was similar to arbutin. These results suggest that the skin-lightening effect of 8-HsPLCB might be resulted from inhibition of melanin synthesis by tyrosinase in melanocytes. To conclude, 8-HsPLCB treatment showed reduction of the melanin pigment and histological changes induced by UV irradiation in brown guinea pigs.
A total of 96 crossbred weanling barrows aged 21 days were randomly allocated to 32 pens of a new nursery to investigate the effects of antibiotics, phytogenics, and probiotics on intestinal growth and development. The animals were fed a set of three-phase basal diets containing 0.3% zinc oxide (CON) or the basal diets supplemented with 353 ppm of a combination of tiamulin, neomycin, chlortetracycline, and oxytetracycline (ANTI), 75 ppm triterpenoid saponin plus 150 ppm mixed saccharides (HERB; Sacchapin$^{(R)}$), or $1{\times}10^7$ brewer's yeasts plus $8{\times}10^7$ spores of each of Bacillus licheniformis and Bacillus subtilis per kilogram feed (PROBIO; Yeasture Plus 2B$^{(R)}$) for five weeks. Thirty-two pigs representing as many pens were slaughtered at the end of the feeding trial, after which morphological measures and digestive enzyme activities of intestinal mucosa were determined. Weight gain and gain:feed of the pigs were not affected by the dietary treatments (TRT) during the overall feeding trial. Total intestinal length was greater in PROBIO than in ANTI (P<0.05). Wet mucosa weight of the duodenum was not affected by TRT. However, jejunal mucosa weight was greater in PROBIO than in any other group sum of mucosa weights of the duodenum and jejunum was greater (P<0.05) in PROBIO than in ANTI and HERB. The height and width of duodenal villus were not affected by TRT, but crypt depth decreased (P<0.05) in response to HERB and PROBIO vs CON. Specific activities of alkaline phosphatase, sucrase, maltase, lactase, and leucine aminopeptidase in the duodenum and jejunum were not changed by TRT. In conclusion, results suggest that the present dietary treatments have no effects on growth performance of weanling pigs and that of PROBIO enhances intestinal growth and development under a clean experimental setting.
Xenotransplantation may help to overcome the critical shortage of human tissues and organs for human transplantation, Swine represents an ideal source of such organs owing to their anatomical and physiological similarities to human besides their plentiful supply, However, the use of organs across the species barrier may be associated with the risk of transmission of pathogens, specially porcine endogenous retroviruses (PERVs).• Although most of these potential pathogens could be eliminated by pathogen-free breeding, PERVs are not eliminated by this treatment. PERVs are integrated into the genome of all pigs and produced by normal pig cells and infect human cells. They belong to gamma retroviruses and are of three classes viruses: A, B and C. In the present study, PCR based cloning was performed with chromosomal DNA extracted from pigs from domestic pigs in Korea. Amplified PCR fragments of about 1.5 Kb, covering the partial env gene, were cloned into pCR2.l-TOPO vectors and sequenced. A total of 91 env clones were obtained from domestic pigs, Berkshire, Duroc, Landrace and Yorkshire in Korea. Phylogenetic analysis of these genes revealed the presence of only PERV class A and B in the proportion of 58 % and 42 %, respectively. Among these, 28 clones had the correct open reading frame: 18 clones in class A and 10 clones in class B. Since both these PERV classes are polytropic and have the capacity to infect human cells, our data suggest that proviral PERVs have the potential to generate infectious viruses during or after xenotransplantation in human.
The supplementation level of barley was limited because of high contents of fiber in monogastric animals. Barley contained high soluble fiber, thus it could prevent to diarrhea of weaning pigs. Moreover, as the barley break down by enzymes, free sugars come out from the barley, which could be used as an energy source in weaning pigs and replace milk by-products in weaning pig's diet. Therefore, present study was conducted to investigate the influence of barley to replace milk by-product in weaning pig's diet on growth performance, blood profile, nutrient digestibility, diarrhea incidence, and economic analysis in weaning pigs. A total of 112 crossbred ($[Yorkshire{\times}Landrace]{\times}Duroc$, weaned at 28 days of age) piglets were allotted to 4 treatments in a randomized complete block (RCB) design. Each treatment has 7 replications with 4 pigs per pen. Pigs were fed each treatment diet which containing different levels of barley (0%, 10%, 20%, and 30%) at the expense of whey powder and lactose. Three phase feeding programs were used for 6 weeks of growth trial (phase 1: 0-2 weeks; phase 2: 3-4 weeks; phase 3: 5-6 weeks). During 0-2 week, body weight (BW), average daily gain (ADG) and G:F ratio were decreased as barley level increased in the diet (linear response, p < 0.01). In blood profile, blood urea nitrogen was decreased as the barley level increased in the diet (linear, p < 0.01). However, no significant differences were observed in blood glucose level. In nutrient digestibility, crude fat digestibility was linearly increased as barley increased (linear, p < 0.01). The incidence of diarrhea was improved as increasing barley contents in all phases (linear, p < 0.01). These results demonstrated that supplementation of barley to replace milk by-product influenced negatively on growth performance during 0-2 week. However, the incidence of diarrhea and later growth performance from 3 week postweaning were improved as dietary barley level increased.
