  • Single spore isolates of Plasmodiophora brassicae e4 and e9 obtained from diseased Chinese cabbage were identified as race 4 and race 9, respectively, by the Williams' differential variety set. To confirm the possibility of variation in same generation and progeny of a single spore isolate of P. brassicae, random amplified polymorphic DNA (RAPD) analysis was conducted using the URP 3, 6 and OPA 7 primers. There was no difference in band type at each part of the gall of Chinese cabbage obtained by inoculation of e4 and e9 and amplification using the URP 3 and 6 primers when the same generation was analyzed. In addition, the progeny analysis, which was expanded to the third generation and conducted using the URP 3 and OPA 7 primers, revealed no differences in the band type of the e4 isolate. Based on these results, the single spore isolate of P. brassicae was genetically stable.

Development of a highly effective T-DNA inserted mutant screening method in a Chinese cabbage (Brassica rapa L. spp. pekinensis) reverse genetics system

  • We present a highly effective T-DNA inserted gene screening method as part of a reverse genetics model system using the Chinese cabbage (Brassica rapa L. spp. pekinensis). Three-step two-dimensional (2D) matrix strategies are potentially accurate and useful for the identification of specific T-DNA inserted mutants from a large population. To construct our Chinese cabbage model, we utilized a forward genetics screening approach for the abnormal phenotypes that were obtained from transgenic plants of Brassica rapa generated with Agrobacteria tumefaciens containing the pRCV2 vector. From one transgenic plant with an abnormal phenotype, we observed that the st1 gene (which is related to senescence-associated process proteins) contained a T-DNA fragment, and that its expression level was decreased. This T-DNA insert was then used as a control to construct an effective screening pool. As a result, the optimum template concentration was found to be 0.1-1 ng in our PCR strategy. For other conditions, positive changes to the Gibbs free energy prevented the formation of oligo dimers and hairpin loop structures, and autosegment extension gave better results for long fragment amplification. Using this effective reverse genetics screening method, only 23 PCR reactions were necessary to select a target gene from a pool of 100 individual DNAs. Finally, we also confirmed that the sequence we obtained from the above method was identical to the flanking sequence isolated by rescue cloning.

Effects of Rice and Vegetable Restricted Diet on Albino Rats (쌀 야채 식이와 식이제한이 흰쥐 성장에 미치는 영향)

  • The effect of dietary restriction was obserbed in Albino rats. Dietary restriction was at 2 levels: 10%, 20%, 30 males and same number of females of Albino rats, aged $30{\sim}40$ days were devided into following 6 groups, 5 rats each. 1 group; rice standard group 2 group; 10% restriction of rice standard 3 group; 20% restriction of rice standard 4 group; 65% rice+35% chinese cabbage group 5 group; 10% restriction of 65% rice+35% chinese cabbage 6 group; 20% restriction of 65% rice+35% chinese cabbage The rats were kept in individual cage and given 6 different diet for 7 weeks. The results of this study were elucidated as follow. 1. Final body weight was not obserbed any significant differences between non-restriction group and 10% restriction group, but 20% restriction group revealed the decrease of body weight. 2. According to dietary restriction, final organ weight was almost decreased in the restriction group of rice+chinese cabbage, and in rice standard group, there were no significants differences between none-restriction group and 10% restriction group, but organ weight was sharply decreased such as Liver, Heart, Kidney, Adrenal, Spleen in 20% restriction group. Sex organ weight of female was increased but weight of male was decreased in 20% restriction group of rice standard group. 3. The result of biochemical analysis such as nitrogen retention, serum cholesterol, glucose of urine, feces, and serum, and lipid of feces and liver, was not revealed any significant diffences, but in 20% restriction group, urinary glucose and serum glucose levels were somewhat decreased, and liver nitrogen level was increased. 4. Therefore there is no effect on albino rats by 10% restricted diet. It can be concluded that it is possible to restrict the diet within 10%.

Physicochemical Properties of a Mixture of Dried Food Waste Powder with Organic Fertilizer and Effects on the Growth of Major Leafy Vegetable (음식물류 폐기물 건조분말과 유기질비료 혼합물의 이화학적 특성 및 주요 엽채류 생육에 미치는 영향)

  • This study was conducted to investigate the application effects of dried food waste powder (FWP) and mixtures of FWP and organic fertilizer (MFOs) on the growth of lettuce and Chinese cabbage. The physicochemical properties of FWP and MFOs were analyzed. As FWPs containing 3.11% NaCl was applied at 250 kg, 500 kg and 1,000 kg per 10a, growth factors of lettuce and Chinese cabbage, such as a number of leaf, leaf length, leaf width, and dry weight were decreased than those of control appling organic fertilizer. The higher the FWP ratio in the MFOs, the lower the concentrations of N and P2O5 in the MFOs, while the higher the concentration of NaCl. As compared to dry weight of control after treating MFOs, lettuce were not significant in MFOs treatments containing 10~30% FWP, and Chinese cabbage in MFOs treatment containing 10% FWP.

Characteristics of Crude Lipoxygenase in Chinese Cabbages (배추 Lipoxygenase 의 특성)

  • Inactivation of lipoxygenase activity in Chinese cabbage was shown after salting and heat treatments. Crude lipoxygenase was obtained from treatment of $(NH_{4})_{2}SO_{4}$. Lipoxygenase activity in Chinese cabbage was about 50% after 20 hrs of salting in 13% (w/v) concentration. After heating at $90^{\circ}C$ for 15 min, residual activity of lipoxygenase was about 50%. Inactivation of lipoxygenase was highly accelerated by increasing temperature and heating time. Decimal reduction time (D-value) were 42, 20 and 14 min at 70, 80 and $90^{\circ}C$, respectively. When cabbage was soaked in 0.05 M $CaCl_{2}$ and heated at $55^{\circ}C$ for 1.5 hr, higher activity of crude lipoxygenase was found compared with the heat treatment without $CaCl_{2}$.

Differential expression of pistil genes induced by $CO_2$ treatment in chinese cabbage ($CO_2$ 처리에 의한 배추 화주 유전자의 특이적 발현 연구)

  • Chinese cabbage (Brassica campestris), one of the major vegetable crops in Korea, undergoes self-incompatible pathway for reproduction. To maintain inbred lines of chinese cabbage, a method in that $CO_2$ gas is treated to the pistils to break the self-incompatibility and thereby self-pollens can successfully make germination and fertilization has been selectively used in speed company. In this study, the pistil genes induced by the $CO_2$ treatment was investigated by mRNA differential display (DD-PCR) method. The result shows PCR products amplified in a differential pattern from both $CO_2$ gas treated- and untreated-pistil mRNAs, suggesting that the pistil genes are probably regulated positively and also negatively by the $CO_2$ gas.

Changes in the quality of frozen vegetables during storage (저장기간에 따른 동결채소의 품질 변화)

  • This study was conducted to assess the possibility of preserving frozen vegetables (Aster scaber, soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion) for a long period and of using them after such storage by measuring changes in quality due to their preservation. Various freshly harvested vegetables were blanched under optimal conditions (that were determined in a preliminary experiment), quick-frozen at $-40^{\circ}C$ for 24 h, and stored at $-20^{\circ}C$. The change in the chromaticity of the frozen A. scaber, soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion did not vary. The hardness of the frozen A. scaber, green pumpkin, Chinese cabbage and Welsh onion did not change during the preservation period, whereas the hardness of the cotyledon and hypocotyl of the soybean sprouts significantly increased on the sixth month of their storage. The total bacterial counts of the A. scaber during the preservation period remained constant at $10^3$ CFU/g, whereas those of soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion decreased slightly to $10^2$ CFU/g. Coliform was not detected in any of the samples. The sensory evaluation showed that the preservation period that was used in this study did not significantly affect the marketability of the frozen vegetables. Therefore, it was considered that A. scaber, soybean sprouts, Chinese cabbage, green pumpkin, and Welsh onion can be safely preserved by freeze-storing them for up to 12 months.

Evaluation of the Quality Characteristics Chinese Cabbage from Two Geographic Origins during Fermentation of Kimchi (국산 및 수입산 배추김치의 품질 특성 연구)

  • This study was investigated for quality characteristics of cabbage from two geographic origins during fermentation of kimchi at $4^{\circ}C$ for 50 days. The pH of kimchi made from cabbage of domestic and imported was decreased with increased fermentation period, while total acidity was increased with increased fermentation period In two samples, the best pH was when it became the 10th of the fermentation period. All fermentation period, vitamin C content is gradually increasing, decreasing after the maximum value. And 40 day fermentation period expect that kimchi made using domestic cabbage, kimchi made using imported cabbage had a higher content of vitamin C contents. Hardness and chewiness of two samples decreased during fermentation. Change of hardness was lowed kimchi made using domestic cabbage. The results of sensory evaluation, texture and overall acceptability were higher in both the kimchi made using domestic cabbage.

Effects of Postharvest Predrying on Storability of 'Norang' Chinese Cabbage (수확 후 예건이 배추 '노랑' 품종의 저온저장에 미치는 영향)

  • This study investigated the effects of conventional predrying and modified atmosphere (MA) on the head quality and storability of Chinese cabbage 'Norang' cultivar. Immediately after harvest, heads were predried for 2 days and MA stored in $50{\mu}m$ PE film packages at $4^{\circ}C$. MA packaging restrained Hunter L and b values of Chinese cabbage more effectively than non-packaging during storage at $4^{\circ}C$. But there was little change between the two treatments. Fresh weight decreased less in heads treated with predrying and MA than non-treatment during storage. Predried Chinese cabbage heads kept a high level of soluble solids in 4 weeks of storage, while non-packaging maintained high contents of soluble solids for 6 weeks of storage period. Chinese cabbage heads contained 7.0 mg/gFW glucose, 6.3 mg/gFW fructose, and 0.6 mg/gFW sucrose as major soluble sugars at harvest. The content of sugars decreased immediately after predrying and increased steadily after 2 weeks storage. It was found inappropriate to assess head quality of Chinese cabbage by investigating was investigated by Hunter a, firmness, dry matters content, pH, and soluble sugars after predrying and MA package. Marketability of Chinese cabbage was lost when heads were stored at room temperature in 2 weeks. It showed poor appearance of heads stored at $4^{\circ}C$ in 7 weeks. Decay occurred in Chinese cabbage stored in MA under excessive relative humidity. Predried head showed good appearance during storage at $4^{\circ}C$ for 7 weeks. As a result, Chinese cabbage heads can be successfully stored for at least 7 weeks with predrying and MA storage.

Screening of effective control agents against bacterial soft rot on Chinese cabbage in alpine area (고랭지 배추 무름병 방제를 위한 우수약제 선발)

  • Bacterial soft rot by Erwinia carotovora subsp. carotovora is one of the diseases causing the biggest problem in Chinese cabbage. Chemical screening was conducted to select effective agents for controlling bacterial soft rot. Control effect of antibiotics, plant activator, and Biokeeper (avirulent Erwinia) to soft rot were tested by in vitro assay, nursery test, and field experiment. The in vitro assay was done by paper disc method and potato slice method. The nursery test was performed by using mineral oil inoculation method with consistent disease induction. The in vitro assay showed that streptomycin, oxolinic acid, bronopol, and copper hydroxide significantly suppressed the growth of pathogenic bacterium and the decomposition of potato slice. However, plant activators including acibenzolar-S-methyl did not show the suppressive effect on the growth of pathogenic bacterium and the decomposition of potato slice. When applied by the nursery test condition using mineral oil inoculation method with Chinese cabbage 'Kangruckyeurum', Biokeeper, oxolinic acid, antibiotics streptomycin, validamycin, and copper compound provided 83.5%, 95.2%, 91.2%, 57.5% and 79.9% in control efficacy, respectively. However, the control effect of acibenzolar-S-methyl showed to be low to cause phytotoxicity. Also acibenzolar-S-methyl showed a significant control effect in the field experiment with Chinese cabbage 'Sanchon' in 2000, but the field experiment with Chinese cabbage 'Kangruckyeurum' in 2001 revealed it had phytotoxicity to Chinese cabbage. Such a difference was considered to be caused by differences in phytotoxic reaction of Chinese cabbage cultivars to the chemical. Streptomycin+copper, copper hydroxide and Biokeeper showed 79.7%, 71.9% and 60.9% in control efficacy, respectively, in the field experiment with Chinese cabbage 'Sanchon' in 2002.
