The application of potassium above the optimum level may cause the inhibition of plant growth, fertilizer loss, and environmental pollution. Therefore, application rate of K fertilizer should be recommended on the basis of soil test. In order to determine critical K content in soils causing growth inhibition of vegetables, $1m^2-pot$ experiments with Chinese cabbage and radish were accomplished with various K-application rates. The threshold concentrations of exchangeable potassium causing the inhibition of plant growth were $0.96cmol_c\;kg^{-1}$ for Chinese cabbage in spring, and $1.28cmol_c\;kg^{-1}$ for radish in autumn. Above those concentration levels, the yields of them were decreased with the increase of potassium levels in soils. Germination rate of Chinese cabbage in spring decreased with increase of the electrical conductivity (EC) of soils due to application of potassium fertilizer. In the harvesting stage, the potassium contents of plant were increased with the increase of K application rate while plant uptake of nutrients was decreased at the K adjustment level of over $2.0cmol_c\;kg^{-1}$.
The pupose of the present paper is to estimate the effects of constraint condition of pile head, ground condition(dry unit weight. $\gamma_4$) and embedded pile lengths on the behavior of single pile which is embedded in normally consolidated clay. BBperiment functions can be quantified to these effects obtained from the results of model teats. The ground of model tests is normally consolidated( NC ) clay under three kinds of effective vertical stress. The results of the model tests using the steel pile of two different embedded pile length and of free-head and fired-head show that the lateral load-deflection relationship is to be elasto plastic behavior below $\gamma_d/\gamma_{dmax}$: 0.84 and that the reduction of lateral load of beyond maximum lateral load($Q_{max}$) at each model test is significantly time-dependent. In this study, it is shown that the displacement relationship can be fitted to exponential function of time by model best results. The effect of ground conditions on the ultimate and yield lateral load is fitted to exponential function including the ratio of dry unit weight to maximum dry unit weight. When tests by results are compared with those from Broms and Budhu et at., the predicted results are over-estimated about 27-87 ayo. In effectivity of constraint condition of pile head on the lateral load-deflection response, the $Q_{fixed}/Q_{gree}-y/D$ relationship is highly non-linear and fitted to parabolic function.
This study was conducted to evaluate the effect of dietary antioxidant and energy density on performance and antioxidative status in transition cows. Forty cows were randomly allocated to 4 dietary treatments in a $2{\times}2$ factorial design. High or low energy density diets (1.43 or 1.28 Mcal $NE_L$/kg DM, respectively) were formulated with or without antioxidant (AOX, a dry granular blend of ethoxyquin and tertiary-butylhydroquinone; 0 or 5 g/cow per d). These diets were fed to cows for 21 days pre-partum. During the post-partum period, all cows were fed the same lactation diets, and AOX treatment followed as for the pre-partum period. Feeding a high energy diet depressed the DMI, milk yield, and 4% fat-corrected milk (FCM) of cows. However, AOX inclusion in the diet improved the milk and 4% FCM yields. There was an interaction of energy density by AOX on milk protein, milk fat and total solids contents. Feeding a high energy diet pre-partum increased plasma glucose and ${\beta}$-hydroxybutyrate, whereas dietary AOX decreased plasma ${\beta}$-hydroxybutyrate value during the transition period. There were also interactions between time and treatment for plasma glutathione peroxidase activity and malondialdehyde content during the study. Cows fed high energy diets pre-partum had higher plasma glutathione peroxidase activity 3 days prior to parturition, compared with those on low energy diets. Inclusion of AOX in diets decreased plasma glutathione peroxidase activity in cows 3 and 10 days pre-partum. Addition of AOX significantly decreased malondialdehyde values at calving. Energy density induced marginal changes in fatty acid composition in the erythrocyte membrane 3 days post-partum, while AOX only significantly increased cis-9, trans-11 conjugated linoleic acid composition. The increase in fluidity of the erythrocyte membrane was only observed in the high energy treatment. It is suggested that a diet containing high energy density pre-partum may negatively affect the anti-oxidative status, DMI and subsequent performance. Addition of AOX may improve the anti-oxidative status and reduce plasma ${\beta}$-hydroxybutyrate, eventually resulting in improved lactation performance; the response to AOX addition was more pronounced on the high energy diet.
This study was conducted to evaluate performance, breast (thigh) muscle yield, nutrient digestibility and serum biochemical parameters of broilers fed low-protein diets supplemented with various threonine to lysine ratios (Thr/Lys). Three hundred and twenty four day-old male Arbor Acres broilers were randomly allotted to six dietary treatments with six replicates per treatment and nine broilers per replicate. Six treatments included one control diet [formulated according to NRC (1994)], and five experimental diets (treatments 1-5). In treatments 1-5, the crude protein level was 2% lower than that of the NRC control diet. The Thr/Lys in treatments 1-5 was 0.65, 0.70, 0.75, 0.80 and 0.85 during the starter and grower phases and 0.70, 0.75, 0.80, 0.85 and 0.90 during the finisher phase with other nutrient levels kept consistent. The results showed that: (1) With increasing Thr/Lys, ADG of treatments 1, 3 and 4 were lower (p<0.05) than those of the NRC control diet during the starter phase; ADG of treatments 1-5 were lower (p<0.05) than those of the NRC control during the grower phase, and ADG of treatment 1 was lower (p<0.05) than that of the NRC control during the finisher phase. Average daily feed intake in treatments 1 and 4 were lower (p<0.05) than that of broilers on the NRC control diet during the grower phase. Feed/gain (F/G) of the NRC control diet was lowest (p<0.05) during the starter phase. F/G in treatments 2 and 5 were higher (p<0.05) than that of NRC during the grower phase. (2) Breast muscle proportion on d 21 increased linearly (p<0.05) in response to graded levels of Thr/Lys. (3) No differences were detected for dry matter, energy and crude protein digestibility among all seven treatments. (4) On d 21, serum triglyceride of broilers on dietary treatments 3 and 5 was higher (p<0.05) than that of broilers in control. The results indicate that the low-protein diets supplemented with an appropriate Thr/Lys could not support the same performance that was achieved by the broilers fed the NRC control diet.

Background: Dendritic cell (DC)-based cancer immunotherapy is studied for several years. However, it is mainly derived from autologous PBMC or leukapheresis from patient, which has limitations about yield and ability of DC production according to individual status. In order to solve these problems, inquiries about allogeneic DCs are performed but there are no preclinical trial answers for effect or toxicity of allogeneic DC to use for clinical trial. In this study, we compared the anti-tumor effect of allogeneic and autologous DCs from mouse bone marrow stem cells in mouse metastatic melanoma model. Methods: B16F10 melanoma cells ($5{\times}10^4$/mouse) were injected intravenously into the C57BL/6 mouse. Therapeutic DCs were differentiated from autologous (C57BL/6: CDC) or allogeneic (B6C3F1: BDC) bone marrow stem cells with GM-CSF, SCF and IL-4 for 13days and pulsed with B16F10 tumor cell lysate (Blys) for 18hrs. DC intra-peritoneal injections began on the 8th day after the tumor cell injection by twice with one week interval. Results: Anti-tumor response was observed by DC treatment without any toxicity especially in allogeneic DC treated mice (tumor burden score: $2.667{\pm}0.184,\;2.500{\pm}0.463,\;2.000{\pm}0.286,\;1.500{\pm}0.286,\;1.667 {\pm}0.297$ for saline, CDC/unpulsed-DC: U-DC, CDC/Blys-DC, BDC/U-DC and BDC/Blys-DC, respectively). IFN-${\gamma}$ secretion was significantly increased in allogeneic DC group stimulated with B16F10 cell lysate ($2,643.3{\pm}5,89.7,\;8,561.5{\pm}2,204.9.\;6,901.2{\pm}141.1pg/1{\times}10^6$ cells for saline, BDC/U-DC and BDC/Blys-DC, respectively) with increased NK cell activity. Conclusion: Conclusively, promising data was obtained that allogeneic DC can be used for DC-based cancer immunotherapy.

This study was undertaken to optimize combining the processes of enzymatic hydrolysis and extraction for lycopene production from autumn olive berry. The autumn olive berry was pulverized and suspended in water, followed by treatment with various hydrolytic enzymes including Ceremix, Celluclast, AMG, Viscozyme, Pectinex, Promozyme, Ultraflo and Tunicase. Reaction solutions were subjected to extraction by applying different organic solvents including acetone, ethyl acetate, hexane and chloroform. Highest yields of lycopene extraction were obtained with the Ceremix (hydrolysis enzyme) and chloroform (extraction solvent) combination. Subsequently, using this ideal combination, enzymatic hydrolysis conditions, including enzyme concentration, pH and temperature, were statistically optimized to 0.58%, 5.5 and 54.4℃, respectively, by applying the response surface method. The lycopene extraction yield increased 2.3-fold (22.6 mg/100g) by using the selected combined process. We propose that these results could be used for the future development of bioactive materials required for bio-health care products.
A green house experiment was conducted to investigate the growth of Chinese cabbage, radish and soybean and change in the chemical properties of the soil after the 10mm application of the simulated acid rain(SAR) of pH 2.7 in the form of sprinkle and mist. It was applied 30 times for Chinese cabbage and radish and 62 times for soybean at the two-day intervals. The results obtained are summarized as follows: 1. Visual damages caused by SAR were dark-brown or red brown leaf spots in Chinese cabbage, and dark-brown and wrinkled leaf margins in radish and soybean. 2. The degree of visual damages became severer as the number of SAR applications increased and it was severer with mist than with sprinkle of SAR. 3. Chlorophyll content was reduced by SAR, but it was not affected by the form of SAR application(sprinkle or mist) in all crops, although it was slightly lower with mist than with sprinkle of SAR. 4. Fresh weight of Chinese cabbage heads and radish roots and grain yield of soybean were reduced by SAR, and were not affected by the form of SAR application. 5. Contents of K, Ca, and Mg in leaves were reduced, while S content increased by SAR in all crops. The forms of SAR application did not affect contents of mineral nutrients in all crops. 6. SAR decreased soil pH and the contents Ca, Mg, and K of soil, while increased $SO_4$ content. However, the contents of soil organic matter, N, and P were not affected by SAR. Forms of SAR application did not affect soil chemical properties either.

Despite the ability of biochar to enhance soil fertility and to mitigate greenhouse gas, its carbon sequestration and profit analysis with arable land application have been a few evaluated. This study was conducted to estimate carbon sequestration and to evaluate profit of greenhouse gas mitigation during corn cultivation periods. For the experiment, the biochar application rates were consisted of pig compost(non application), 2,600(0.2%), 13,000(1%), and 26,000(2%) kg/ha based on pig compost application. For predicting soil carbon sequestration of biochar application, it was appeared to be linear model of Y = 0.5523X - 742.57 ($r^2=0.939^{**}$). Based on this equation, soil carbon sequestration by 0.2, 1 and 2% biochar application was estimated to be 1,235, 3,978, and 14,794 kg/ha, and their mitigations of $CO_2$-eq. emissions were estimated to be 4.5, 14.6, and 54.2 ton/ha, respectively. Their profits were estimated at $14.6 for lowest and $452 for highest. In Korea Climate Exchange, it was estimated that the market price of $CO_2$ in corn cultivation periods with 0.2, 1 and 2% biochar application was $35.6, $115.3 and $428.2 per hectare, respectively. For the plant growth response, it was observed that plant height and fresh ear yield were not significantly different among the treatments. Therefore, these experimental results might be fundamental data for assuming a carbon trading mechanism exists for biochar soil application in agricultural practices.
Mastitis is an inflammation of the mammary gland that develops as a response to pathogenic microorganisms. Somatic cell count (SCC) and bacteriological analysis are the most accepted tools to determine udder health. This study evaluated the effects of intra-mammary gland infusion of silver ion against clinical mastitis in Holstein cows. Silver ion (20 ${\mu}g$/ml) was infused through the intra-mammary gland in quarters having clinical mastitis and milk was collected to determine SCC, and levels of lactoferrin and bovine serum albumin (BSA). Silver ion infusion decreased udder inflammation, firmness and swelling, and reduced clots, BSA, lactoferrin and SCC in milk. However, milk yield and circulating blood cells remained unchanged. The silver ion-mediated reductions of BSA and SCC indicate reduced inflammation and bacterial activity in silver ion-treated mammary glands in Holsteins with mastitis, which may be exploited in a curative strategy.
This study used response surface methodology (RSM) in an effort to optimize the hot water extraction conditions of dandelion leaves in order to increase antioxidant activity in the extract. A central composite design was applied to investigate the effects of independent variables, which included the ratio of solvent to sample ($X_1$), extraction temperature ($X_2$), and extraction time ($X_3$), on dependent variables of the extracts, including soluble solid ($Y_1$), total polyphenols ($Y_2$), total flavonoid ($Y_3$), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability ($Y_4$), 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical scavenging ability ($Y_5$), and superoxide radical scavenging ability ($Y_6$). The estimated optimal conditions were as follows: $83.77{\pm}1.07^{\circ}C$ of the extraction temperature, $20.85{\pm}0.24 mL/g$ of solvent per sample, and $1.59{\pm}0.12$ hr of extraction time. At the optimal conditions, the predicted characteristic values were: a yield of 38.98%, a total polyphenol level of $74.28{\mu}g/mg$, a total flavonoid level of $74.00{\mu}g/mg$, a DPPH radical scavenging ability ($IC_{50}$) of 0.14 mg/mL, a ABTS radical scavenging ability ($IC_{50}$) of 3.24 mg/mL, and a superoxide radical scavenging ability ($IC_{50}$) of 2.49 mg/mL.
