Research on Functional Paper by using Traditional Dyestuffs (전통 염색 재료를 활용한 기능성 종이 연구)

  • Dyestuffs extracted from fifteen kinds of natural dyeing materials were researched on their antimicrobial performance against 3 kinds of bacteria and 2 kinds of fungi in order to develop conservation/storage materials of traditional and natural dyed papers. The three types of mordants were evaluated by using the same method and then mixed with the dyestuffs to research their antimicrobial performance. With those results, dyed paper was made by mixing mordants with 9 kinds of dyestuffs with excellent antimicrobial performance. Among the dyed papers, Cotinus coggygria, Coptis chinensis, Phellodendri amurense, Rhus javanica and Acer ginnala multiple mordant papers were found to have an antibacterial quality against fungi, while, in the insect repellent experiment using termites, Coptis chinensis and Phellodendri amurense dyed papers were found to have insect repellent qualities. Also, with regard to the research result about the affect of dyeing paper on other materials like metal, Coptis chinensis have relatively stable qualities. Also, the three types of dyeed paper by Cotinus coggygria, Coptis chinensis and Phellodendri amurense, were found to have a tendency to remove harmful gases. Not only can these dyed papers be used for making storage box for cultural properties to prevent various organic artifacts from bio-damaging, but they can also be utilized as functional finishing materials in various spaces.

Myeloid-specific SIRT1 Deletion Aggravates Hepatic Inflammation and Steatosis in High-fat Diet-fed Mice

  • Sirtuin 1 (SIRT1) is a mammalian $NAD^+$-dependent protein deacetylase that regulates cellular metabolism and inflammatory response. The organ-specific deletion of SIRT1 induces local inflammation and insulin resistance in dietary and genetic obesity. Macrophage-mediated inflammation contributes to insulin resistance and metabolic syndrome, however, the macrophage-specific SIRT1 function in the context of obesity is largely unknown. C57/BL6 wild type (WT) or myeloid-specific SIRT1 knockout (KO) mice were fed a high-fat diet (HFD) or normal diet (ND) for 12 weeks. Metabolic parameters and markers of hepatic steatosis and inflammation in liver were compared in WT and KO mice. SIRT1 deletion enhanced HFD-induced changes on body and liver weight gain, and increased glucose and insulin resistance. In liver, SIRT1 deletion increased the acetylation, and enhanced HFD-induced nuclear translocation of nuclear factor kappa B (NF-${\kappa}B$), hepatic inflammation and macrophage infiltration. HFD-fed KO mice showed severe hepatic steatosis by activating lipogenic pathway through sterol regulatory element-binding protein 1 (SREBP-1), and hepatic fibrogenesis, as indicated by induction of connective tissue growth factor (CTGF), alpha-smooth muscle actin (${\alpha}$-SMA), and collagen secretion. Myeloid-specific deletion of SIRT1 stimulates obesity-induced inflammation and increases the risk of hepatic fibrosis. Targeted induction of macrophage SIRT1 may be a good therapy for alleviating inflammation-associated metabolic syndrome.

Assessment of Apprehensive Area of Non-Point Source Pollution Using Watershed Model Application in Juam Dam Watershed (주암댐 유역 비점오염부하량 우심지역 평가를 통한 오염물질 저감시설 최적 설치지점 선정 연구)

  • This study analyzes potentially hazardous sub-watersheds from non-point source pollution areas using an HSPF model. The watershed of the Juam dam reservoir was divided into 29 sub-watersheds, and the flow, BOD, TN and TP concentration for the Juam dam watershed were evaluated from 2009 to 2012 using a watershed model, with a warming period from 2009 to 2010. The results of the watershed model agreed well with the flow and water quality field measurements. The calculated average non-point source loadings were BOD of 8.8 and $9.1kg/day/km^2$ in 2011 and 2012, respectively; TN of 9.7 and $10.1kg/day/km^2$ in 2011 and 2012, respectively; and TP of 0.30 and $0.33kg/day/km^2$ in 2011 and 2012, respectively. The non-point source loading of the Bonghwa stream watershed was calculated, and predominantly assessed upstream of the Boseong river. Additionally, the Miryeok, Jangpyeong, Yuleo, Guam, Seokgyo, Mundeok, Incheon, and Bongnae stream watersheds, with extensive agricultural areas, were assessed to be potentially hazardous areas in terms of non-point source management. In this study, HSPF model was applied in order to aid in the selection of non-point source reduction facilities for the Juam dam watershed, where they were evaluated as to whether they would be applicable for non-point source management.

The Present State of Occupational Injuries and Prevention on East Side of Korea Fishing (동해안 연근해 어업의 산재현황과 예방대책)

  • Fishermen have higher risk of occupational injuries and disease due to frequent machine usage and direct contact to live biological materials. Moreover, growing elderly workers makes the susceptibility to occupational injuries and disease higher. This study was performed to investigate the occupational safety and health status among fishermen. The interview was carried out at Jumunjin and Geojin ports which were representative port at North East side of Korea. The structured questionnaire were used to interview the fishermen from AM 6:00 to PM 11:00 and total respondent were 97 workers. The results were followed; 7 fishermen of all respondents experienced occupational injuries during their work, 5 fishermen were needed to admission longer than 4 days. The injury types were contusion(4 persons), fracture(1 persons) and amputation(2 persons). The cause of injury might be the lack of caution and the unstability of working condition. These results suggested the ergonomical evaluation of working condition and proper management. But there was limited concern and studies on the policy on occupational safety and health on fishermen. So, further study was required to establish the sound policy of fishermen's occupational safety and health.

Comparison of Opened Rates and Quality Characteristics of Frozen Baby-clam In-shell Tapes philippinarum Prepared by Different Processing Method (제조방법을 달리하여 제조한 껍질붙은 냉동바지락(Tapes philippinarum)의 껍질 개패율 및 품질특성 비교)

  • We compared two different processing methods for preparing high quality frozen in-shell baby clam products. In the first method, sand and mud were removed from the clams, then they were vacuum packed in polyethylene film, boiled at $97^{\circ}C$ for 6 min, and snap frozen in a cold air blast freezer (sample 1). The second processing method was similar, except the boiling process was excluded (sample 2). Both frozen products were boiled for 4 min, and then shucked and minced. Various quality metrics, such as the opening rates of shells, chemical composition, pH, volatile basic nitrogen (VBN), salinity, thiobarbituric acid (TBA), amino-N, total amino acids and free amino acids were measured, and sensory evaluation was conducted. The opening rates of shells of sample 1 and sample 2 were 98.3% and 4.67%, respectively. The proximate composition of sample 1 and sample 2 was 75.2% and 78.7% moisture, 19.7% and 16.2% crude protein, 2.45 and 2.2% crude lipid, 2.8% and 2.1% ash, and 2.1% and 1.9% salinity, respectively. The L, a, b and ${\Delta}E$ values were similar: 48.6 and 49.2, 3.9 and 3.9, 15.7 and 15.5, and 50.7 and 50.1 for sample 1 and sample 2, respectively. The sensory evaluation score of sample 1 was higher than that of sample 2. Sample 1 was deemed to be superior to sample 2; therefore, we determined that the boiling process is needed for manufacturing high-quality frozen clam products.

A XML Based Framework for Automatically Generating Control and Monitoring Software (제어 및 모니터링 소프트웨어 자동 생성을 위한 XML 기반 프레임웍)

  • In this paper, we present a framework which is used to develop, modify, maintain and extend a control and monitoring software easily for any kind of automatic instruments. The proposed framework is composed of three XML documents (IID, MAP, CMIML) and two tools (Virtual Instrument Wizard, Generator). Interface information of behaviors and states of instrument is written on IID. Mapping information between the interface information in IID and API of a real instrument driver is written on MAP Final information of the control and monitoring software is written on CMIML, IID, MAP and CMIML are written by XML format to provide a common usage and platform independence of the proposed framework. Vl Wizard generates CMIML intermediate platform independent document using IID and existing CMIML, and Generator generates the source code of a control and monitoring software platform dependent code automatically using CMIML and MAP. The suggested framework provides an easy development and maintenance because it automatically generates a control and monitoring software in GUI environment and it also provides common usage and platform independence in virtue of using description document of XML format. Also, reusability can be increased by reusing platform independent software description document and not reusing platform dependent software code.

A Study on the Traditional Industrial Landscape Valued as Scenic Site - With the Focus on Daranginon, Dok-sal, Saltern - (전통산업경관의 명승적 가치 - 다랑이 논, 독살, 염전을 중심으로 -)

  • This study aimed to discover new types of landscape resources that have not designated as cultural heritages through a survey of the nationwide traditional industrial landscape resources whose original forms have been lost or damaged in the aspect of management and conservation. It also discovered and analyzed the values of the traditional industrial landscape considered to be humanistic and cultural values. Among the traditional industrial landscape resources distributed nationwide, this study mainly investigated Daranginon, Dok-sal(Korean traditional stone fishing weir) and saltern. Make a list of traditional industrial landscape based on the scenic resources and analyzed the value as scenic spot by conducted basic research and field work. In case some resources were highly valued, this study aimed to rediscover the value as a cultural heritage of the traditional industrial landscape that was a basis of our ancestors' lives and has represented our traditional agricultural and fishing activities done by them over a long period of time by exploring the possibilities of being designated as cultural heritages. Continuous discoveries and studies of traditional industrial landscapes, conservation and usage of traditional industrial landscapes by being designated as a cultural heritage required to be done will be required to be done. Also, this study was supposed to be used as a basic data.

Somatic Embryo Germination and the Related Biochemical Changes of Liriodendron tulipifera by Bioreactor Immersion Time (생물반응기 내 침지시간에 따른 백합나무 체세포배 발아 및 생화학적 변화)

  • To determine physical and physiological factors for Liriodendron tulipifera L. somatic embryo germination, temporary immersion bioreactor (TIB) system was investigated. It was designed to immerse liquid media with plantlets so that it was able to adjust the immersion time. Immersion of 120 minutes every 4 hours and 60 minutes every 4 hours was found to be effective in germination (91.64%, 85.67%, respectively). However, hyperhydricity of the plantlets was higher in short immersion time (15 minutes every 6 hours) and long immersion time (120 minutes every 4 hours) (51.61%, 34.28%, respectively). Immersion of 60 minutes every 4 hours showed the lowest hyperhydric plantlets, and also it showed the lowest activities of abscisic acid (ABA), superoxide dismutase (SOD), and catalase. The overall results implied that immersion time of media affected germination and growth of somatic embryo, and it was able to make use of germination and growth of L. tulipifera somatic embryos.

Effect of polysaccharides from a Korean ginseng berry on the immunosenescence of aged mice

  • Background: Korean ginseng has been widely evaluated to treat human diseases; however, most studies on Korean ginseng have focused on its root. In this study, polysaccharides [acidic-polysaccharide-linked glycopeptide (APGP) extracted with 90% ethanol and hot water] were prepared from Korean ginseng berries, and their effect on immunosenescence was explored. Methods: The effect of APGP on thymic involution was evaluated by measuring the size of thymi dissected from aged mice. The effect of APGP on populations of immune cells, including natural killer (NK) cells, dendritic cells, age-correlated CD11c-positive B cells, and several subtypes of T cells [CD4-positive, CD8-positive, and regulatory (Treg) T cells] in the thymi and spleens of aged mice was analyzed by fluorescence-activated cell sorting analysis. Serum levels of interleukin (IL)-2 and IL-6 were evaluated by enzyme-linked immunosorbent assay analysis. Profiles of APGP components were evaluated by high-performance liquid chromatography (HPLC) analysis. Results: APGP suppressed thymic involution by increasing the weight and areas of thymi in aged mice. APGP increased the population of NK cells, but showed no effect on the population of dendritic cells in the thymi and spleens of aged mice. APGP decreased the population of age-correlated CD11c-positive B cells in the spleens of aged mice. APGP showed no effect on the populations of CD4- and CD8-positive T cells in the thymi of aged mice, whereas it increased the population of Treg cells in the spleens of aged mice. APGP further decreased the reduced serum levels of IL-2 in aged mice, but serum levels of IL-6 were not statistically changed by APGP in aged mice. Finally, HPLC analysis showed that APGP had one major peak at 15 min (a main type of polysaccharide) and a long tail up to 35 min (a mixture of a variety of types of polysaccharides). Conclusion: These results suggested that APGP exerted an anti-immunosenescent effect by suppressing thymic involution and modulating several types of immune cells.

In Vitro Metabolism of a New Cardioprotective Agent, KR-33028 in the Human Liver Microsomes and Cryopreserved Human Hepatocytes

  • KR-33028 (N-[4-cyano-benzo[b]thiophene-2-carbonyl]guanidine) is a new cardioprotective agent for preventing ischemia-reperfusion injury. This study was performed to identify the metabolic pathway of KR-33028 in human liver microsomes and to compare its metabolism with that of cryopreserved human hepatocytes. Human liver microsomal incubation of KR-33028 in the presence of NADPH and UDPGA resulted in the formation of four metabolites, M1, M2, M3, and M4. M1 and M2 were identified as 5-hydroxy-KR-33028 and 7-hydroxy-KR-33028, respectively, on the basis of LC/MS/MS analysis with the synthesized authentic standard. M3 and M4 were suggested to be dihydroxy-KR-33028 and hydroxy-KR-33028-glucuronide, respectively. Metabolism of KR-33028 in cryopreserved human hepatocytes resulted in the formation of M1, M2, and M4. These data show a good correlation between major metabolites formed in human liver microsomes and cryopreserved human hepatocytes. In addition, KR­33028 was found to inhibit moderately the metabolism of CYP1A2 substrates. Based on the results obtained metabolic pathway of KR-33028 is proposed.
