Silencing of NbNAP1 Encoding a Plastidic SufB-like Protein Affects Chloroplast Development in Nicotiana benthamiana

  • It was previously shown that AtNAP1 is a plastidic SufB protein involved in Fe-S cluster assembly in Arabidopsis. In this study, we investigated the effects of depleting SufB protein from plant cells using virus-induced gene silencing (VIGS). VIGS of NbNAP1 encoding a Nicotiana benthamiana homolog of AtNAP1 resulted in a leaf yellowing phenotype. NbNAP1 was expressed ubiquitously in plant tissues with the highest level in roots. A GFP fusion protein of the N-terminal region (M1-V103) of NbNAP1 was targeted to chloroplasts. Depletion of NbNAP1 resulted in reduced numbers of chloroplasts of reduced size. Mitochondria also seemed to be affected. Despite the reduced number and size of the chloroplasts in the NbNAP1 VIGS lines, the expression of many nuclear genes encoding chloroplast-targeted proteins and chlorophyll biosynthesis genes remained unchanged.

Two Groups of Phytoplasma from Chrysanthemum (Dendranthema grandiflorum) Distinguished by Symptoms and 16S rRNA Gene Sequence in Korea

  • Two groups of phytoplasma were identified in chrysanthemum(Dendranthema grandiflorum) cv. Chunkwang showing distinct symptoms. Isolate Ph-ch1 showed symptoms of dwarf, witches'-broom, rosette and root death. The other isolate, Ph-ch2, revealed symptoms of dwarf, yellowing, leaf cupping, vein clearing and root death. The presence of phytoplasma structures in chrysanthemum leaf tissue was confirmed by transmission electron microscopy. The 16S rRNA gene was amplified from isolates Ph-ch1 and Ph-ch2 by PCR and cloned, and the nucleotide sequences were determined. In RFLP analysis, isolate Ph-ch2 showed profiles identical to Ph-ch1, except with restriction enzymes HhaI and MseI. The sequence data showed that isolate Ph-ch1 was most closely related to the aster yellows (AY) phytoplasma, and isolate Ph-ch2 was more closely related to stolbur phytoplasma than to AY phytoplasma. This is the first reported observation of stolbur phytoplasma in chrysanthemum species.

  • Phenol-free resin was synthesized and its printing ink properties were investigated. The phenol-free resin was produced by esterification of poly phthalate and Diels-Alder adduct of rosin anhydride. Compared to rosin modified phenolic resin, eco-friendly phenol-free resin showed better vehicle properties in terms of gloss, yellowing, runability, and storage stability. The results suggest the utility of phenol-free resin instead of conventional rosin modified phenolic resin.

  • Thermally expandable microspheres were used as post-treatment initiators of potassium persulfate, sodium bisulfite, and sodium sulfide in order to improve the foaming ability and whiteness when foaming a mixture of thermally expandable microsphers and poly(vinyl chloride). Potassium persulfate showed no significant influence on the foaming behavior, foam expansion, whiteness, and yellowing, whereas in the case of using sodium bisulfite. In particular, sodium bisulfite demonstrated the best efficiency with 2 wt% treatment. The thermally expandable microspheres prepared herein can provide excellent foamability and whiteness, and are expected to be applicable in various fields such as general coating and wallpaper.

  • Cultivar Yeollow Special A, the most leading Korean economic leaf tobacco in Korea, were field-cultivated in four different localities of Korea for the period of two years (1970 and 1971) and sprayed at varied levels of 2-chloroethyl phosphonic acid (CEPA) for foliar application few days after topping. While no striking difference in leaf yield by weight was obtained among the treatments when compared with control, leaf quality as expressed for shipment price in won tended when compared with control, leaf quality as expressed for shipment price in won tended to improve. The treated leaves with 300 to 900 ppm of CEPA (approximately 140 1/acre of 500 ppm) not only showed yellowing and accelerated maturity to pick 4 to 5 days with practicable optimal level earlier than control, but also speeded up to take nearly with practicable optimal level earlier than control, but also speeded up to take nearly last half of the total time required for the five stages of flue-curing. It is therefore considered that CEPA is as effective maturity-accelerating agent and useful as known for other solanaceous plants showing climacteric stage respiration, and discussions were made about physiological actions of ethylene gas released from CEPA at plant tissues sprayed.

Leaf Blight of Kudzu (Pueraria lobata) caused by Fusarium solani

  • Leaf blight of kudzu ( Pueraria lobata ) was found in Jeonbuk province in 2002. The main symptoms appeared as leaf blight and showed yellowing and wilting. The causal pathogen of the leaf blight was isolated from symptomed kudzu leaf and produced white to cream, usually floccose mycelium. It readily formed reddish orange mycelium on PDA. It produced typical microconidia and macroconidia. The microconidia were the reniform. The macroconidia were wide, slightly curved, usually 3 to 4 septate and size was 45 ∼ 85 ${\times}$ 5 ∼ l0$\mu\textrm{m}$. The pathogen produced chlamydospore singly on short hyphal branches within 2 to 3 weeks, which was hyaline, globose, and smooth walled. The pathogen was, therefore, identified as Fusarium solani based on cultural and morphological characters. This is the first report on the leaf blight of kudzu caused by Fusarium solani in Korea.

Candidatus Phytoplasma trifolii Associated with Witches' broom of Lespedeza cyrtobotrya M.

  • The Symptoms of witches' broom disease caused by phytoplasma including general stunting and yellowing, were observed in leafy lespedeza (Lespedeza cyrtobotrya M.) on Doam-myeon, Pyeongchang-gun, in 2006. Based on the sequence analysis of PCR-amplified 16S ribosomal DNA and 16S-23S spacer region DNA products using universal phytoplasma primers, the phytoplasma associated with leafy lespedeza witches' broom (LLWB) disease was identified as a member of Candidatus Pytoplasma trifolii. It was most closely related to alsike clover proliferation phytoplasma (99.8% similarity, accession no. AY390261), Candidatus Pytoplasma trifolii strain. RFLP patterns generated with AluI, HpaII clearly differentiated LLWB phytoplasma from the referenced phytoplasma strains, water dropwort witches' broom, mulberry dwarf, glehni aster yellow dwarf and jujube witches' broom. This paper is the first report on Candidatus Phytoplasma trifolii in leafy lespedeza identified at a molecular level.

  • In order to investigate the crystallization in silk fibroin induced by acid treatment and its photo-yellowing reduction effect. silk fibroin were immersed in PH 3.5~4.0 sulfuric acid and 0.01N hydro-chloric acid at 2$0^{\circ}C$, and the changes of crystallinity were measured by infrared spectroscopy, X-ray diffraction. Yellowness index caused by UV irradiation were also observed. The results are as follows : 1. ${\beta}$-form transition occurs when the silk fibroin are immersed at 2$0^{\circ}C$ in pH 3.5~4.0 sulfuric acid and 0.01N hydro-chloric acid. Formation of new hydrogenbond in molecules on the amorphous region induced ${\beta}$-form structure, and increased crystallinity. 2. Crystallization owing to $\beta$-form transition reduced the initial yellowness index by ultraviolet irradiation.
