Biological conversion of biomass to succinic acid

  • Batch cultivations of Anaerobiospirillum succiniciproducens have been systematically studied for the economical production of succinic acid from wood hydrolysate with corn steep liquor(CSL) as a nitrogen source. CSL was found to be an alternative complex nitrogen source for A. succiniciproducens when glucose and wood hydrolysate were used as carbon sources. Compared with polypeptone and/or yeast extract, CSL had similar effects on fermentation performance such as succinic acid yield and a ratio of succinic acid to acetic acid in the fermentation of wood hydrolysate as well as glucose. This means that succinic acid can be produced more economically from wood hydrolysate and CSL than relatively expensive carbon and nitrogen sources. Besides its low cost, the alternative medium served as a green technology for succinic acid production because it gives a net-zero effect on global warming.

Effects of nitrogen sources on cell growth and biochemical composition of marine chlorophyte Tetraselmis sp. for lipid production

  • Nitrogen is one of the most critical nutrients affecting cell growth and biochemical composition of microalgae, ultimately determining the lipid or carbohydrate productivity for biofuels. In order to investigate the effect of nitrogen sources on the cell growth and biochemical composition of the marine microalga Tetraselmis sp., nine different N sources, including NaNO3, KNO3, NH4NO3, NH4HCO3, NH4Cl, CH3COONH4, urea, glycine, and yeast extract were compared at the given concentration of 8.82 mM. Higher biomass concentration was achieved under organic nitrogen sources, such as yeast extract (2.23 g L−1) and glycine (1.62 g L−1), compared to nitrate- (1.45 g L−1) or ammonium-N (0.98 g L−1). All ammonium sources showed an inhibition of cell growth, but accumulated higher lipids, showing a maximum content of 28.3% in ammonium bicarbonate. When Tetraselmis sp. was cultivated using yeast extract, the highest lipid productivity of 36.0 mg L−1 d−1 was achieved, followed by glycine 21.5 mg L−1 d−1 and nitrate 19.9 mg L−1 d−1. Ammonium bicarbonate resulted in the lowest lipid productivity of 14.4 mg L−1 d−1. The major fatty acids in Tetraselmis sp. were palmitic, oleic, linoleic and linolenic acids, regardless of the nutritional compositions, indicating the suitability of this species for biodiesel production.

Controllable Biogenic Synthesis of Intracellular Silver/Silver Chloride Nanoparticles by Meyerozyma guilliermondii KX008616

  • Intracellular synthesis of silver/silver chloride nanoparticles (Ag/AgCl-NPs) using Meyerozyma guilliermondii KX008616 is reported under aerobic and anaerobic conditions for the first time. The biogenic synthesis of Ag-NP types has been proposed as an easy and cost-effective alternative for various biomedical applications. The interaction of nanoparticles with ethanol production was mentioned. The purified biogenic Ag/AgCl-nanoparticles were characterized by different spectroscopic and microscopic approaches. The purified nanoparticles exhibited a surface plasmon resonance band at 419 and 415 nm, confirming the formation of Ag/AgCl-NPs under aerobic and anaerobic conditions, respectively. The planes of the cubic crystalline phase of the Ag/AgCl-NPs were confirmed by X-ray diffraction. Fourier-transform infrared spectra showed the interactions between the yeast cell constituents and silver ions to form the biogenic Ag/AgCl-NPs. The intracellular Ag/AgCl-NPs synthesized under aerobic condition were homogenous and spherical in shape, with an approximate particle size of 2.5-30nm as denoted by the transmission electron microscopy (TEM). The reaction mixture was optimized by varying reaction parameters, including temperature and pH. Analysis of ultrathin sections of yeast cells by TEM indicated that the biogenic nanoparticles were formed as clusters, known as nanoaggregates, in the cytoplasm or in the inner and outer regions of the cell wall. The study recommends using the biomass of yeast that is used in industrial or fermentation purposes to produce Ag/AgCl-NPs as associated by-products to maximize benefit and to reduce the production cost.

옥살산 전처리 옥수숫대를 이용한 동시당화발효 최적 조건 탐색 (Optimal Condition for Simultaneous Saccharification and Fermentation Using Pretreated Corncob by Oxalic Acid)

  • 본 연구는 옥살산으로 전처리를 수행한 후 얻어진 옥수숫대를 이용하여 동시당화발효를 위한 최적조건을 탐색하였다. Pichia stipitis CBS 6054를 이용한 동시당화발효에서 독립변수인 반응온도($25.8{\sim}34.2^{\circ}C$)와 교반속도(80~220 rpm)에 대한 에탄올 생산량은 각각 99% 신뢰구간을 가졌다. 종속변수로 에탄올 생산량을 적용하였을 때 $30^{\circ}C$, 170 rpm에서 최대의 에탄올 생산을 예측할 수 있었다(22.5 g/L). 최적의 온도 및 교반속도에서 최적 질소원을 조사한 결과 yeast extract (1.25 g/L)와 urea (1.25 g/L)를 혼합하여 사용하였을 경우 에탄올 생산량은 증가하였으며 trace metal 성분과 비타민은 첨가하지 않았을 때 에탄올 생산이 촉진되었다. 동시당화 발효를 위한 $KH_2PO_4$, $MgSO_4{\cdot}7H_2O$의 최적 농도는 각각 1 g/L, 0.25 g/L로 나타났다.

감귤과피 압착액을 기질로 한 SCP 생산 (SCP Production from Mandarin Orange Peel Press Liquor)

  • The bioconversion of mandarin orange peel press liquor to single cell protein (SCP) by two yeast strains, F-60, and C-7, which were isolated from mandarin orange peel was carried out and compared with that of using Candida utilis IFO 0598. Experiments were directed toward the high yield of biomass and high protein in cultures of the strains mentioned above. Candida utilis IFO 0598, F-60 and C-7 strains were cultivated at 3$0^{\circ}C$, pH 5.2 for 3 days in shaking flasks. The effects of some nutrients on cell growth were studied. Cell mass and protein content per cell mass were increased by addition of urea 1%, KH$_2$PO$_4$ 0.1% and MgSO$_4$ㆍ7$H_2O$ 0.05%, When the F-60 strain cultured under the optimal conditions, cell mass, growth yield and protein content were 41.2g/l, 53.9%, 59.7%, respectively. Cell mass was also increased up to 15% by modifying the fermentation condition on the bench type 20l jar fermentor. Crude fat content (10.3%) of dried C-7 cell was higher than those of C. utilis and F-60, 4.9% and 5.6% respectively. Total protein content of the F-60 strain was 59.7% per dry weight. And we compared their amino acid compositions with that of FAO provisional pattern. In the case of the F-60 strains, amino acid contents such as lysine, leucine and isoleucine were much higher than those of methionine, cystine and tryptophan.

  • Starch is an abundant resource in plant biomass, and it should be hydrolyzed enzymatically into fermentable sugars for ethanol fermentation. A genetic recombinant yeast, Saccharomyces cerevisiae GA-7458, was constructed by integrating the structural gene of both $\alpha$-amylase from Bacillus stearothermophilus and the gene (STA1) encoding glucoamylase from S. diastaticus into the chromosome of S. cerevisiae SH7458. The recombinant yeast showed active enzymatic activities of $\alpha$-amylase and glucoamylase. The productivity of ethanol fermentation from the pH-controlled batch culture (pH 5.5) was 2.6 times greater than that of the pH-uncontrolled batch culture. Moreover, in a fed-batch culture, more ethanol was produced (13.2 g/L), and the production yield was 0.38 with 2% of corn starch. Importantly, the integrated plasmids were fully maintained during ethanol fermentation.

Effect of Glucose Concentration on the Production of Erythritol by Trichosporon sp.

  • The effect of glucose concentration on the production of erythritol by Trichosporon sp. was mainly studied. The specific growth rate and production rate of erythritol gave the highest values of $0.23 h^{-1}\; and\; 4.2 g/\ell/h,\; respectively,\; on\; 100 g \; glucose/\ell$ of medium. The conversion yield of erythritol during the exponential phase and the stationary phase was constantly maintained at 19% and 51 %, respectively, while the glucose concentration in the medium varied from 100 g/$\ell$ to 400 g/$\ell$. The maximum overall erythritol conversion yield of 47% was obtained when the glucose concentration in the medium was 400 g/$\ell$. It corresponded to a 74% increase compared with the 100 g/$\ell$ glucose medium. The diauxy growth of this microbe was also observed. It grew exponentially consuming glucose, then after the second lag phase, biomass slowly increased using glycerol and erythritol.

Submerged Culture of Phellinus linteus for Mass Production of Polysaccharides

  • In order to increase the mycelial production of Phellinus linteus, which exhibits potent anticancer activity, some ingredients of the medium used to culture P. linteus were investigated. The optimal medium composition for the production of Phellinus linteus was determined to be as follows: fructose, 40 g/l; yeast extract, 20 g/l; $K_2HPO_4$, 0.46 g/l; $K_2HPO_4$, 1.00 g/l; M$MgSO_4\cdot7H_2SO$, 0.50 g/l; $FeCl_2\cdot6_2O$, 0.01 g/l; $MnCl_2\cdot4H_2O$, 0.036 g/l; $ZnCl_2$, 0.03 g/l; and $SuSO_4\cdot7H_2O$, 0.005 g/l. The optimal culture conditions were determined to be as follows: temperature, 28$^{\circ}C$; initial pH, 5.5; aeration, 0.6 vvm; and agitation, 100 rpm, respectively. Under optimal composition and conditions, the maximum mycelial biomass achieved in a 5 l jar fermentor was 29.9 g/l.

Biologics For The Protection Of Forests On The Basis Of Mushroom Phlebiopsis Gigantea With Deep Cultivation On Alcohol Stillage Production

  • In the Republic of Belarus as well as in the world acute problem of protecting forests from diseases and pests. The damage caused by root rot is essential, therefore, the problem of forest protection is an urgent task. The biologics has the greatest prospects in according with traditional methods of struggle. Deep method of cultivation of a mushroom Phlebiopsis gigantea with use of nutrient mediums on the basis of ethanol stillage and its components (fugat) is researched. Feasibility of use stillage as raw materials in production of a biological product for the wood protection against root decay is shown. The effect of different additives (sawdust, fodder yeast) on the accumulation of reactive biological product - oidy has been studed It was determined that the deep cultivation using sawdust of the highest accumulation oidy (1.5 $10^6units/ml$). It was also found that the stillage is the best breeding ground for fungus biomass accumulation (7.9 9.8 g / l) versus fugat (6.0 6.6 g / l). On the basis of research work the technological scheme for production of a biological product were developed. Based on the conducted studies, a technological scheme was proposed for obtaining a biological preparation by deep cultivation of the fungus Phlebiopsis gigantea.

  • L- arabinose residues are widely distributed in plant cell walls, where they are present in polymers such as arabinans, arabinoxylans, arabinogalactans and arabinogalactan proteins. L-arabinose suppress intestinal sucrase and decrease the adsorption of sugar in the small intestine, consequently, weight loss and fatness prevent. Now, xylose be used replacement sugar and arabinose be utilized fatness prevent of our time. Various Agricultural surplus like com fiber, contain $20\;{\sim}\;40%$ of hemicellulose. Corn fiber from Agricultural Renewable Biomass was chosen the best suitable material for arabinose production. In this work, we searched about for L-arabinose gene in compost, metagenome pool and indonesian soil. So, the B1029 TS2-8 of L-arabinase gene in compost was selected by YNB media(5% yeast nitrogen base, 5% arabinogalactan). After enzyme reaction with corn fiver, B1029 TS2-8 produced 2.15 g/L of L-arabonose.
