Optimization of soaking in aqueous ammonia pretreatment of canola residues for sugar production (당 생산을 위한 카놀라 부산물의 암모니아 침지 전처리 공정의 최적화)

  • Bioenergy production from lignocellulosic biomass and agriculture wastes have been attracted because of its sustainable and non-edible source. Especially, canola is considered as one of the best feedstock for renewable fuel production. Oil extracted canola and its agriculture residues are reuseable for bioethanol production. However, a pretreatment step is required before enzymatic hydrolysis to disrupt recalcitrant lignocellulosic matrix. To increase the sugar conversion, more efficient pretreatment process was necessary for removal of saccharification barriers such as lignin. Alkaline pretreatment makes the lignocellulose swollen through solvation and induces more porous structure for enzyme access. In our previous work, aqueous ammonia (1~20%) was utilized for alkaline reagent to increase the crystallinity of canola residues pretreatment. In this study, significant factors for efficient soaking in aqueous ammonia pretreatment on canola residues was optimized by using the response surface method (RSM). Based on the fundamental experiments, the real values of factors at the center (0) were determined as follows; $70^{\circ}C$ of temperature, 17.5% of ammonia concentration and 18 h of reaction time in the experiment design using central composition design (CCD). A statistical model predicted that the highest removal yield of lignin was 54% at the following optimized reaction conditions: $72.68^{\circ}C$ of temperature, 18.30% of ammonia concentration and 18.30 h of reaction time. Finally, maximum theoretical yields of soaking in aqueous ammonia pretreatment were 42.23% of glucose and 22.68% of xylose.

Screening of Lactic Acid Bacteria for the Development of Probiotics and the Effect of Cryoprotectant Agents (생균제 개발을 위한 유산균의 선별 및 동결건조 보호제의 효과)

  • For the developement of probiotics, three strains haying psychorophilic and salt tolerant characteristics were isolated from Kimchi. Among the isolated strains, MGl9, MG89 and MG208 were identified as Lactobacillus brevis, Enterococcus faecium and Lactobacillus plantarum, respectively. The relationship between cryoprotectant and the viability of freeze dried strains has been studied. The most effective cryoprotectant for MGl9 was found to be 10% skim milk contained 1% soluble starch haying the survival rate of 71.4%. In MG89, 10% skim milk contained 1% sucrose and 10% skim milk contained l% fructose in MG208 were showed effective cryoprotectant having the survival rate 68.9% and 64.7%. respectively These results suggest that these isolated strains can play an important role as probiotics in aquaculture.

Effect of Glucose, Its Analogs and Some Amino Acids on Pre-steady State Kinetics of ATP Hydrolysis by PM-ATPase of Pathogenic Yeast (Candida albicans)

  • Fast kinetics of transient pH changes and difference spectrum formation have been investigated following mixing of ADP/ATP with partially purified plasma membrane PM-ATPase of the pathogenic yeast Candida albicans in the presence of five nutrients: glucose, glutamic acid, proline, lysine, and arginine and two analogs of glucose: 2-deoxy D-glucose and xylose. Average $H^+$- absorption to release ratio, indicative of population of ATPase undergoing complete hydrolytic cycle, was found to be 0.27 for control. This ratio varied between 0.25 (proline) to 0.36 (arginine) for all other compounds tested, except for glucose. In the presence of glucose, $H^+$- absorption to release ratio was exceptionally high (0.92). While no UV difference spectrum was observed with ADP, mixing of ATP with ATPase led to a large conformational change. Exposure to different nutrients restricted the magnitude of the conformational change; the analogs of glucose were found to be ineffective. This suppression was maximal in the case of glucose (80%); with other nutrients, the magnitude of suppression ranged from 40-50%. Rate of $H^+$- absorption, which is indicative of E~P complex dissociation, showed positive correlation with suppression of conformational change only in the case of glucose and no other nutrient/analog. Mode of interaction of glucose with plasma membrane $H^+$-ATPase thus appears to be strikingly distinct compared to that of other nutrients/analogs tested. The results obtained lead us to propose a model for explaining glucose stimulation of plasma membrane $H^+$-ATPase activity.

표고버섯 균사체 배양 및 그 추출물의 생리학적 특성

  • 한국산 표고버섯 균사체를 액체배양하여 천연항암물질로 알려진 단백다당체를 추출한 후 그 물질의 특성에 대하여 검토하였다. 균사체의 최적 재양조건을 TGY배지로 조사한 바 온도 $25^{\circ}C$, 배양초기 pH4.0, 교반속도 300rpm, 균사배지 접종량을 10.0%로 하고 산소 통기량을 1.0volume of ait/volume of medium/mimute으로 하였을때 가장 양호한 조건이였으며, 대량생산 하기 위한 SCM배지에서 최적의 C/N비는 13.1오써 7일간 배양하였을때 18.8g/L의 균사령을 얻었으며, 이때 생산수율은 0.46으로 나타났다. 발효가 끝난 배양액에서 균사, 여액 그리고 배양액의 전체에서 단백다당체를 분리한 결과 각 분획에서 단백다당체가 각각 0.55%, 0.12%, 0.69%가 회수되어 배양액 전체에서 단백다당체를 추출하는 것이 바람직하며, 추출방법으로 열수추출, glass bead추출 및 cellulasa처리를 하여 단백다당체의 수율을 비교한 결과 0.25-0.5mm glass nead로 30분간 균사체를 분쇄한 다음 열수추출을 1시간을 하였을때 990mg/100ml의 단백다당체를 얻을 수 있었다. 고단백다당체를 1차 단백질 가수분해 효소로 분해하고, EDAE cellulose 및 Sepadex G-100 column chromatography로 정제한 후, TLC/FLD, ultracentrifugation한 결과 순수한 물질임을 알 수 있었다. 단백다당체의 항암효과 조사중 in vitro배양에서 $P_{388}$$L_{1210}$에 대한 단백다당체의 활성단위 1 unit는 1mg정도였으며, 인체의 장암세포인 HCT-48, HRT-18, HT-29 밀 간암세포인 Hep G2 대한 생육저해 단위는 각각 4.4, 3.6, 6.6, 2.6mg이었다. HCT-48과 Hep G2 세포의 크기 분포도는 대조군에 비하여 시간이 경과함에 따라, 그리고 단백다당체의 농도가 증가함에 따라 peak가 작은 size 쪽으로 이동하였다. 또, 단백다당체를 첨가 배양한 HCT-48과 Hep G2세포의 현미경 관찰에서 본래의 암세포 형태가 변형되고 크기가 감소하며 세포사이의 경계막이 흐트러지면서 세포수가 감소하고 사멸하였다. In vivo실험에서는 대조군보다 단백다당체를 첨가한 군에서 항체 형성능력이 대조군에 비하여 형질세포가 2배로 증가하였다. 단백다당체의 화학적 성분 분석에서 다당함량은 46.1%이면 구성다당류는 glucose, galactose, mannose, xylose로 구성되었고 단백질의 함량은 7.28%이며, 구성아미노산은 15종의 아미노산으로 되었다. 또 무기물은 Na, K, Zn, Ca등의 순으로 이루어 짐을 알 수 있었다.

Characteristics and Antimicrobial Resistance Patterns of Staphylococcus aureus Isolated from Horse (국내 말에서 분리된 Staphylococcus aureus의 특성 및 약제 내성 양상)

  • This study carried out to investigate the antimicrobial resistance and biochemical characteristics of Staphylococcus aureus (S. aureus) isolated from reproductive and respiratory tract in Thoroughbred horse. The specimens were collected from equine vaginal mucosa and upper respiratory tract from March to December 2006 using a culture swab in Korea. S. aureus suspected colonies on blood agar plates were selected and identified as standard biochemical tests and PCR (Applied Biosystems, USA). Antimicrobial resistance test of S. aureus isolates was performed with 30 antimicrobial agents (BBL, USA) by using the agar disk diffusion method. S. aureus isolates were isolated 58 (39.2%) strains of 148 samples: wound 64.7% (11/17), genital discharge 37.0% (37/100) and nasal discharge 32.2% (10/31). Almost isolates showed high resistance to spectinomycin, sulfonamides, erythromycin, tetracyelin, ciprofloxacin and penicillin. These results may provide the basic information to establish strategies for treatment and prevention of reproductive and respiratory disease in Thoroughbred horses in Korea.

Inhibition of Enzymatic Browning of Crown Daisy by Maillard Reaction Products (마이얄반응생성물(Maillard reaction product)에 의한 쑥갓의 효소적갈면 억제)

  • The study was conducted to investigate the effect of Maillard reaction products (MRPs) on enzymatic browning of crown daisy (Chrysanthmum coronarium var. spatiosum). The MRPs prepared by heating various amino acid and sugar at $90^{\circ}C$ caused a strong inhibitory effect on crown daisy polyphenol oxidase (PPO, ${\sigma}$-diphenol oxygen oxidoreductase, EC 1.10.3.1). As the reaction time of the solution containing glycine and glucose increased at $90^{\circ}C$, the production of MRPs was increased, whereas the amounts of glycine and glucose were decreased. Accordingly, the inhibitory effect of crown daisy PPO activity by MRPs was increased as the amounts of synthesized MRPs were increased. The MRPs synthesized from the various amino acids and sugars significantly reduced the PPO activity, particularly MRPs prepared by glutamine and xylose. The Michealis-Menten constant value ($K_m$) of crown daisy PPO with catechol as a substrate was 22.0 mM, and MRPs were a noncompetitive inhibitor against crown daisy PPO.

Furfural Production and Recovery by Two-stage Acid Treatment of Lignocellulosic Biomass (Two-stage 산 처리에 의한 목질계 바이오매스로부터 푸르푸랄 생산과 회수)

  • In this study, we investigated optimal reaction conditions for furfural production from lignocellulosic biomass by two-stage acid treatment. Furfural produced by this method was recovered using XAD-4 resin. Oxalic and sulfuric acid were used as catalysts for the first stage of treatment. The concentration of xylose in the hydrolysate obtained from the first stage was $18.86g/{\ell}$ with oxalic acid and $19.35g/{\ell}$ with sulfuric acid. The concentration of oligosaccharide was relatively high when sulfuric acid was used. Maximum yield of furfural, that is, 55.10% ($6.71g/{\ell}$), was obtained when oxalic acid was used for the first stage and $0.1m{\ell}$ of sulfuric acid was used for the second stage of treatment for 90 min. Furfural production yield increased with increasing the reaction time. Most of the furfural produced by this two-stage treatment method was recovered using XAD-4 resin.

Enhancing Enzymatic Saccharification of Corn Stover by Aqueous Ammonia Soaking Pretreatment (옥수수 줄기의 암모니아수 침지 전처리에 의한 효소 당화 향상)

  • Enhancing enzymatic saccharification of corn stover by aqueous ammonia soaking pretreatment was investigated on chemical compositional changes and enzymatic hydrolysis characteristics. At three different levels of aqueous ammonia soaking temperature and time ($140^{\circ}C$-1 h, $90^{\circ}C$-16 h and $50^{\circ}C$-6 days), higher temperature and shorter treatment time led to more xylan and lignin removal based on overall composition analysis and carbohydrate compositional analysis. More xylan and lignin removal in higher temperature treatment led to higher enzymatic saccharification of cellulose and xylan to monosaccharide by commercial cellulase mixtures (Celluclast 1.5L and Novozym 342 from Novozyme, Denmark).

Kinetic Behavior of Salmonella on Low NaNO2 Sausages during Aerobic and Vacuum Storage

  • This study evaluated the growth kinetics of Salmonella spp. in processed meat products formulated with low sodium nitrite (NaNO2). A 5-strain mixture of Salmonella spp. was inoculated on 25-g samples of sausages formulated with sodium chloride (NaCl) (1.0%, 1.25%, and 1.5%) and NaNO2 (0 and 10 ppm) followed by aerobic or vacuum storage at 10℃ and 15℃ for up to 816 h or 408 h, respectively. The bacterial cell counts were enumerated on xylose lysine deoxycholate agar, and the modified Gompertz model was fitted to the Salmonella cell counts to calculate the kinetic parameters as a function of NaCl concentration on the growth rate (GR; Log CFU/g/h) and lag phase duration (LPD; h). A linear equation was then fitted to the parameters to evaluate the effect of NaCl concentration on the kinetic parameters. The GR values of Salmonella on sausages were higher (p<0.05) with 10 ppm NaNO2 concentration than with 0 ppm NaNO2. The GR values of Salmonella decreased (p<0.05) as NaCl concentration increased, especially at 10℃. This result indicates that 10 ppm NaNO2 may increase Salmonella growth at low NaCl concentrations, and that NaCl plays an important role in inhibiting Salmonella growth in sausages with low NaNO2.

First Report on Isolation of Salmonella Enteritidis from Eggs at Grocery Stores in Korea

  • Salmonella Enteritidis is responsible for causing foodborne diseases upon consumption of egg products. While cases of S. Enteritidis isolation from eggs have been reported in other countries, no such cases have previously been reported in Korea. In this study, we report the first isolation and identification of S. Enteritidis from domestically distributed eggs in Korea. Eggs were collected from eight countrywide grocery stores during different seasons between 2011 and 2012. Egg contents and washing solution of egg shells were incubated in buffered peptone water, and the enriched broth was further enriched in tetrathionate broth and Rappaport-Vassiliadis. The secondary enriched broth was streaked on xylose lysine desoxycholate agar. The suspected colonies were confirmed to S. Enteritidis by a biochemical test, serotyping, and PCR test. Genetic relatedness among the isolates was analyzed using Diversilab Salmonella kit. Three strains of S. Enteritidis were isolated from egg contents and egg shells collected from grocery stores of the Eumseong-city in the fall of 2011. All three stains showed resistance to chloramphenicol, streptomycin, nalidixic acid, and ampicillin by the disk diffusion method. In addition, the isolates showed more than 99% DNA homology, indicating that they were presumably identical strains. Therefore, there is a requirement to monitor and control against S. Enteritidis from eggs in Korea.
