Antioxidant Activities and Xanthine Oxidase Inhibitory Effects of Hot-water Extracts from Fruits of Elaeagnus multiflora Thunb. in Maturity (성숙에 따른 뜰보리수(Elaeagnus multiflora Thunb.) 열수추출물의 항산화 활성과 Xanthine Oxidase 저해효과)

  • This study was conducted to analyze the antioxidant activity and xanthine oxidase inhibitory effect of hot-water extracts from Elaeagnus multiflora Thunb. to provide fundamental data for the development of functional materials. The antioxidative activities of hot-water extracts from E. multiflora Thunb. were analyzed by electron donating ability (EDA) using 1,1-diphenyl-2-picryl hydrazyl (DPPH), superoxide dismutase (SOD) -like activity by pyrogallol and nitrite scavenging ability. EDA of extracts from unripe, ripe, and overripe fruits were 77.7%, 36.0%, and 23.7% at 100 ${\mu}g/mL$, respectively, and EDA of extract in unripe fruits was higher than those of others. At 1,000 $\mu$g/mL, the SOD-like activities were 32.8%, 11.2% and 5.0% for unripe, ripe, and overripe fruits, respectively. The SOD-like activity was increased along with the increase of unripe and ripe extract concentrations. The nitrite scavenging ability of unripe fruit extracts in 100, 300, 500 ${\mu}g/mL$ of extraction solution in pH 1.2 and 3.0 was higher than those of others. The nitrate scavenging ability of all extracts was decreased according to the increase of pH. Xanthine oxidase inhibitory activities of unripe, ripe and overripe fruits were 30.0%, 28.2%, and 18.2% at 1,000 ${\mu}g/mL$, respectively.

  • Objectives : To compare the anti-oxidant effects of Ligusticum chuanxiang and Cnidium officinale extracts and their mixture with Angelica gigas, we investigated the anti-oxidative activities using rat liver tissues. Methods : We investigated the anti-oxidative activities by analysis of lipid peroxidation, xanthine oxidase activity, aldehyde oxidase activity, hydroxyl radical scavenging activity, superoxide dismutase activity and catalase activity in rat liver tissues. Results : Both Ligusticum chuanxing and Cnidium offieinale inhibited the lipid peroxidation compared to the control, there is no significant differences between two groups. Cnidium officinale showed strong inhibitory effects on xanthine oxidase and aldehyde oxidase activities compared with that of Ligusticum chuanxing. Ligusticum chuanxing and Cnidium officinale were scavenged the hydroxy radicals, and increased SOD and catalase activities. These results suggested that Ligusticum chuanxing and Cnidium officinale were showed antioxidant activity, especially Cnidium officinale showed higher activity than Ligusticum chuanxing. Conclusions: These results suggest that Ligusticum chuanxing could be replaced by Cmdium afficinale in clinical trial.

Antioxidative Constituents from the Twigs of Vitex rotundifolia

  • In the course of screening for antioxidant compounds by measuring the radical scavenging effect on DPPH (1,1-diphenyl- 2-picrylhydrazyl), a total extract of the twigs of Vitex rotundifolia (Verbenaceae) was found to show potent antioxidant activity. Subsequent activity-guided fractionation of the methanolic extract led to the isolation of three iridoid compounds, 10-O-vanilloylaucubin (1), 10-O-p-hydroxybenzoylaucubin (2) and aucubin (3), two C-glycoside flavones, vitexin (4) and orientin (5), and a quinic acid derivative, 3,4-di-O-caffeoylquinic acid (6). Their structures were elucidated by spectroscopic studies. Among them, compounds 5 and 6 showed the significant antioxidative effects on DPPH free radical scavenging test. In riboflavin-NBT-light and xanthine-NBT-xanthine oxidase systems, compounds 5 and 6 exhibited the formation of the blue formazan in a dose-dependent manner. Compounds 5 and 6 showed better superoxide quenching activities than vitamin C.

돼지 액상정액에서 $Barodon^{(R)}$의 항산화 효과에 관한 연구

  • 세포에 대한 산화스트레스는 세포의 대사와 기능저하의 원인이 되고 있으며 이를 줄이기 위한 항산화 물질의 첨가가 연구되고 있다. 본 연구는 비특이면역증가제이며 다목적 고기능성 알칼리용액 조성물인 Barodon의 항산화 효과를 돼지정자를 이용하여 조사하여, 돼지 액상정액의 보존성 향상을 위한 Barodon의 이용성을 알기 위하여 시도하였다. 실험구로서 무첨가구와 활성산소 인위발생구(xanthine+xanthine oxidase, X-XO), X-XO구에 superoxide dismutase, cataiase, Barodon(2종류)의 단독처리구 및 X-XO구에 이들 항산산제의 복합처리구로 나누어 항산화제 처리효과에 따른 정자운동특성을 CASA로 분석하였다. 또한 돼지 액상정액에서 Barodon의 항산화 효과를 무첨가구, catalase구 및 Barodon구로 나누어 정액의 보존기간별 정액성상 변화(정자활력, 생존성, 첨체이상)를 조사하였다.

Protective Effects of Opuntia Ficus-Indica and Saururus Chinensis on Free Radical-Induced Neuronal Injury in Mouse Cortical Cell Cultures (생쥐 피질세포배양에서 Free Radical 유발 신경손상에 대한 손바닥선인장 및 삼백초의 보호효과)

  • The author examined whether the methanol extracts of Opuntia ficus-indica fruit and Saururus chinensis have the inhibitory action on xanthine/xanthine oxidase (X/XO)-, $FeCl_2/ascorbic$ acid- and arachidonic acid-induced neurotoxicity in mouse cortical cell cultures. The methanol extracts ($10\;{\mu}g/ml{\sim}1\;mg/ml$) of Opuntia ficus-indica and Saururus chinensis were exhibited 53-89% and $48{\sim}100%$ inhibitory action on X/XO-induced neurotoxicity, respectively. At the range of same concentration, both extracts also attenuated the $FeCl_2/ascorbic$ acid-induced neurotoxicity by $35{\sim}100%$ and $15{\sim}98%$, respectively. In arachidonic acid neurotoxicity, the methanol extract (1 mg/ml) of Opuntia ficus-indica and Saururus chinensis reduced neuronal injury by 22% and 38%, respectively. These results suggest that Opuntia ficus-indica fruit and Saururus chinensis may contribute the neuroprotection in certain free radical-mediated neuronal injury.

Effect of Alkaloidal Fraction from Cynanchi Radix on Lipid Peroxidation (우피소근(牛皮消根)의 알칼로이드 분획이 과산화지질 생성에 미치는 영향)

  • The crude alkaloidal fraction of the root of Cynanchum caudatum Max.(Asclepiadaceae) was tested for the effects on the activities of free radical generating enzymes and the formation of lipid peroxide. Aldehyde oxidase was strongly inhibited to about 90% of the activity by treating 1.0 mg/ml of alkaloidal fraction, corresponding to competitive inhibition. Moreover, the formation of lipid peroxide which causes damage of cell membrane was reduced in proportion to the increasing alkaloid concentration. However, xanthine oxidase of which structure and function are similar to those of aldehyde oxidase was not inhibited by the alkaloidal fraction.

  • The methanolic extracts of some marine natural products were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzymes. The methanolic extracts of Styela clava, Ecklonia stolonifera, Pachymeniopsis elliptica and Hypnea charoides which decreased the formation of lipid peroxide inhibited the activity of xanthine oxidase about 41, 20, 20 and 21% by adding $100\;{\mu}g/ml$ of each methanolic extracts, respectively. However, the four extracts didn't inhibit the activity of aldehyde oxidase.

  • Degradation of deoxyribonucleic acid(DNA) and ribonucleic acid(RNA) by cell-free extract of mixed rumen protozoa of buffalo rumen was investigated. DNA was observed to be degraded rapidly during an initial incubation period of 2 hr with simultaneous appearance of degradation products. RNA on the other hand recorded a rapid degradation during an initial incubation period of 1 hr. RNA degradation products appeared upto an incubation period of 2 hr. DNA was observed to degrade into oligo- and mononucleotides. pyrimidine nucleosides, purine nucleoside adenosine and bases xanthine, hypoxanthine and thymine. Degradation products of RNA comprised of pyrimidine nucleosides, purine nucleoside, adenosine and bases xanthine, hypoxanthine and uracil besides oligo- and mononucleotides.

  • 산소라디칼-질병모델을 보다 간편한 방법으로 만들어 생화학적 발병 기전을 추구하므로서 신약창출에 필요한 기초자료를 제시하고자 본 실험을 실시하였다. 미세혈관의 혈류이상은 과잉의 활성산소를 생산하게되며 이로 인해 여러 종류의 난치성 질환 및 염증을 수반하는 많은 질병이 발병하게 된다. 일반적으로 흔히 이용되어지는 허혈-재관류모델이 아닌 환경독성물질을 활용한 병태모델을 만들어 이 모델조건에서 활성산소생성과 밀접한 관계를 갖는 잔틴 옥시데이즈(Xanthine Oxidase)의 변화와 과산화지질생성정도를 연관지어 검토하였다. 수은, 구리, 몰리브덴등의 금속이온들은 잔틴 디하이드로저네이즈(Xanthine dehydrogenase)로 부터 옥시데이즈(Oxidase)형으로의 전환을 촉진시켰으며 막독성의 지표로 이용되어지는 과산화지질의 생성도 현저히 증가시켰다. 또한 알데하이드류의 첨가실험에서도 잔틴산화효소의 형전환이 촉진되었으며 첨가한 알데하이드의 탄소수와 수소수 그리고 탄소와 탄소의 결합상태와도 밀접한 관계가 있음이 관찰되었다.

RNAi-mediated reduction of xanthine dehydrogenase results in increased biomass of Arabidopsis seedlings

  • Xanthine dehydrogenase (XDH), a classic enzyme involved in purine catabolism, can catalyze the formation of redox-signaling reactive oxygen and nitrogen species such as superoxide and nitric oxide. We generated transgenic plants of Arabidopsis in which XDH was knocked out by introduction of hairpin RNA-expression vector. Expression analysis by reverse transcription-PCR and in-gel staining of XDH activity revealed that transgenic lines efficiently suppressedXDH expression at the transcriptional level, demonstrating that RNA interference was successfully induced. XDH-suppressed transgenic lines exhibitedincreased biomass production during the growth of seedlings.
