Purification of Isoflavone from Soybean Hypocotyls using Various Resins

  • Isoflavone was extracted with various concentration of aqueous methanol using whole hypocotyls as the starting material. Whole hypocotyls were preferred as the raw material because the residue could be easily removed from the solvent after the extraction process. Extraction yield was almost constant at the methanol concentration of $20-80\%$. Most of the isoflavone was extracted within 1 hr, and the extraction yield remained almost constant thereafter. When the concentration of methanol was $80\%$, the content of total solid was reduced due to the reduced extraction of contaminating protein as the result of protein insolubilization. Among resins tested, Diaion HP-20, Amberlite XAD-16, and Amberlite IRC-50 showed the highest capacity to absorb the compound. Open column chromatography with Diaion HP-20 showed that $80\%$ aqueous ethanol was most efficient as the eluting solvent with final recovery of the phytochemical being more than $95\%$. Maximum adsorption of the phytochemical occurred at the acidic pH 2-4. When the spatial velocity was increased to 15 and more, the degree of adsorption was decreased, whereas below spatial velocity of 15, the adsorption capacity of isoflavone to the resin was almost constant. The purity of the isoflavone purified by column chromatography was $78\%$.

The Flavonoids from Caragana Chamlagu Leaves (골담초엽의 플라보노이드)

  • The phytochemical studies of the leaves of Caragana chamlagu were carried out as a sieries of the investigation of medicinal resources. The roots of Caragana chamlagu have been used as neuralgia, arthritis and migraine in the folk medicines of Korea. The methanolic extract of the leaves of Caragana chamlagu was suspended with water and then separated with chloroform. Compound I was isolated from precipitates of these water fraction by recrystalization. The aqueous fraction of MeOH extract was performed to column chromatography on Amberlite XAD-4 and Sephadex LH-20, and three compounds, compound II, compound III, and compound IV were isolated. The structures of the four compounds were elucidated by spectroscopic data of $^1H-NMR$, ^{13}C-NMR$, IR, and FAB-MS. Compound I-IV were tilianine ($acacetin-7-O-{\beta}-D-glycopyranoside$), rutin($quercetin-3-O-{\alpha}-L-rhamnopyranosy(1{\rightarrow}6)-{\beta}-D-glu-copyranoside$), $kaempferol-3-O-{\alpha}-L-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-D-galactopyranoside$, and apigetrin, ($apigenin-7-O-{\beta}-D-glycopyranoside$), respectively.

Escherichia coli에서 발현된 재조합 인간 상피세포 증식인자의 정제 및 특성

  • Recombinant human epidermal growth factor (rhEGF) was produced by E. coli BL21 harboring a plasmid pYHB101. The maximum production was 68.7 mg/l when the E. coli strain was cultured at 25$\circ$C for 48 hours in the modified MBL medium containing 10 g/l glucose with 1 mM IPTG induction at 2 hours after inoculation. The rhEGF was purified upto 267 folds by Amberlite XAD- 7 chromatography, ultrafiltration, and DEAE Sepharose fast flow ion exchange chromatography with an overall yield of 66.6%. The purified rhEGF was further separated into two fractions by HPLC. The N-terminal amino acid sequence of the second fraction was Asn-Ser-Asp-Ser-Glu-Cys-Pro-Leu-Ser-His. The effect of rhEGF on the DNA synthesis was examined using in vitro biological assay based on the incorporation of 5'-bromo-2'- deoxy-uridine (BrdU). The purified rhEGF shows no difference with natural human epidermal growth factor (nhEGF) in N-terminal amino acids residues and biological activity. From the results, we concluded that rhEGF produced from E. coli harboring the plasmid pYHB101 was apparently the same as nhEGF.

Speciation of Arsenic from Soil Organic Matter in Abandoned Gold and Silver Mines, Korea

  • Organic forms of arsenic (As) were determined through fractionation procedure of soil organic matter (SOM) in soil, sediments and mine tailing samples from the Myungbong, Dongil, and Okdong mining areas of southern Korea. An alkaline extraction method was applied to soil samples followed by the fractionation procedures of SOM by the DAX-8 and XAD-4 resin adsorption method. Major fraction of organic As species (42% to 98%) was found in acid-soluble fraction, whereas minor fraction (0.1 % to 67.8%) was present in the humic-associated As. In acid-soluble fractions, the transphillic- and hydrophilic-associated As were dominant in addition to As binding with humic and fulvic SOM. Arsenic binding was the strongest between pH 6 to 8 and reduced to about 70% at both low and high pH regions. The amount of both transphillic and hydrophillic associated As was less changed than humic and fulvic-associated As, in both low and high pH regions. This apparently indicates that As has stronger affinity towards hydrophillic rather than hydrophobic organics. From the experimental observation of As-binding SOM in natural soil, the ligand exchange model may be a feasible explanation of transphillic and hydrophillic affinity of As.

Endocrine Disruptive Potentials in Surface Water Samples from Taihu Lake, Yangtze Delta

  • Taihu Lake is a major water source for part of Yangtze Delta, which is one of the most urbanized and economically prosperous areas in China. In last couple of decades, some parts of the lake were highly polluted due to eutrophication. This study analyzed dioxin-like potential and mutagenic potential in surface water samples from Taihu Lake. The samples were prepared by XAD-2 resin procedure. A batch of biological assays, including dioxin-like potential microassay with the rat hepatocyte cell line H411E, and Ames test was employed in the research. Results showed that jour water samples have high content of dioxin-like biological potential, the highest activity TEQ to 2,3,7,8-TCDD was 48 pg/ι in sample 1. The mutagenic effect with reading-frame shifting mechanism was confirmed in 3 of 4 samples. The effective sewage treatment facilities and reliable monitoring surveillance system are urgently needed for this area.

  • As a series of the studies on the specific and indigenous plants of Mt. Halla, the constituents from stem of Rhamnus taquetii (Rhamnaceae) were investigated. From the water fraction of the MeOH extract, a new flavonol glycoside, kaempferide-3-O-${\alpha}$-L-rhamnopyranosyl(1->3)-${\alpha}$-L-rhamnopyranosyl(1-6)-${\beta}$-D-galactopyranoside, was isolated with three known compounds, quercetin, quercetin-3-methylether, kaempferol, by column chromatographic separation using Amberlite XAD-2, ODS-gel and Sephadex LH-20 and elucidated physico-chemical evidences($^1H-NMR,\;^{13}C-NMR$,IR, EI-Mass, FAB-Mass, and G.C.),respectively.

Biological Monitoring on the Absorption of Antineoplastic Drugs in Nursing Personnel (항암제 취급간호사의 항암제 체내흡수에 관한 조사)

  • Antineoplastic agents may exhibit effects not only in patients therapeutically exposed, but also in health workers who prepare and administer these drugs. This study was done to clarify whether nurses who handle anticancer drugs show signs of drug absorption. The experimental group was 14 nurses handling anticancer drugs at three medical wards of a hospital in J city ; the control group was 12 psychiatric nurses at the same hospital. The test materials were the nurses' 24hr urine specimens, which were concentrated by XAD-2 column chromatograpy. Tester strains were TA98(±S9mix), TA100(±S9 mix), TA1535(±S9 mix) and TA1537(±S9 mix) : the salmonella mammalian microsomal test (Ames test) was used for the urinary mutagenicity assay. The results are summarized as follow : 1. In qualitative analysis of the results, both experimental group and control group showed 15.4% urine toxicity. 2. The experimental group revealed significantly higher urinary mutagenicity both in the activation method test and non-activation method test of the tester strains TA98, TA100 and TA1535. In the case of TA1537, the two groups showed no difference in the non-activation method test, but the activation method revealed a difference. 3. In urinary mutagenicity of the experimental group by ward career, there was a significant difference between the group with more than 20 months experience and the group with less than 20 months on the tester strains TA98, TA100, and TA1537. No Significant difference was found between two groups by the tester strain TA1535.

Inhibition of Porcine Pancreatic Elastase (PPE) by Korean Mistletoe (Viscum album var.coloratum) Fractions

  • The serine proteases such as human leukocyte elastase (HLE) and porcine pancreatic elastase (PPE) are classified in the chymotrypsin family, and possibly the most destructive enzymes having the ability to degrade virtually all of the connective components in the body. In the present study, the extracts of water and methanol of Korean mistletoe (Viscum album var. coloratum) inhibited significantly the PPE activity. The fractions eluated on Amberlite XAD-2 from methanol extract were further purified on the repeated $SiO_2$ column chromatography and the fractions A, B and C were eluated. The fractions A, B and C at 3 mg/ml inhibited significantly the PPE activity up to 66%, 95% and 85%, respectively. In conclusion, the fraction A assumed as lignans or phenylpropanes, and fraction B and C assumed as triterpenoids showed the PPE inhibitory effects on the PPE and that these compounds in mistletoe may be used for treatment of pathological processes such as age-dependent tissue loss or inflammation.

Application and Optimization of the IsoButoxycarbonyl Derivatization method to the Analysis of Trace Level Phenols in Environmental Samples (환경시료로부터 미량 페놀류의 분석을 위한 isoButoxycarbonyl 유도체화 분석방법 적용 및 최적화)

  • Eleven phenols including two chlorophenols, eight alkylphenols and bisphenol A were derivatized with isobutylchloroformate to form their isobutoxycarbonyl derivatives. Standard phenol mixture was concentrated for the isobutoxycarbonyl (isoBOC) derivatization and analysed by gas chromatography/mass spectrometry (GC/MS). The recoveries of the derivatization method of alkylphenols, chlorophenols, and bisphenol A were calculated by gas chromatography/mass spectrometry-selected ion monitoring mode using two work-up methods for comparison; shaking and heating method. The linear detector responses were obtained in the concentration range of 5∼400 ng, with correlation coefficients varying from 0.9755∼0.9981. Recoveries of the alkylphenols, chlorophenols, and bisphenol A were determined by gas chromatography/mass spectrometry-selected ion monitoring mode using two work-up methods for comparison ; the US-EPA method and the isoBOC derivatization method, Eleven phenols in water samples were extracted with dichloromethane and then concentrated. Also, solid-phase extraction (SPE) with XAD-4 and subsequent conversion to isobutoxycarbonyl derivatives for sensitive analysis with the selected ion-monitoring (SIM) mode. The recoveries were 85.1∼109.9% and 90.3∼126.6% for the US-EPA method and the isoBOC.

Anti-Herpes Simplex Virus type I (HSV-1) Effect of Isorhamnetin 3-0-beta-D-Glucopyranoside Isolated from Brassica rapa (무청으로부터 분리된 이소람네틴 3-O-beta-D글루코피라노사이드의 항헤르페스 바이러스 1형(HSV-1) 효과)

  • In the course of our search for anti-Herpes simplex virus type I (HSV 1) substances from natural sources, we screened crude drugs for their antiviral activity using SRB assay. T he methanol extract from herb of Brassica rapa (Cruciferae) was found to inhibit HSV-1. Though bioassay-directed fractionation of the extract, anti-HSV-1 agent was isolated by chromatographic separation using Amberlite XAD-4 and Sephadex LH-20. The structure of compound I was elucidated by spectral means including $^1H-^1H$ COSY, HMQC and HMBC to be isorhamnetin 3-O-${\beta}$-D-glucopyranoside (compound I). Compound I was active against HSV-1 with the 50% effective concentration of O.42mg/ml and the 50% cytotoxicity of 5.0mg/ml.
