Response of Filefish to the Colored Lights (색광에 대한 쥐치의 반응)

  • The author carried out an experiment to find out the responsing patterns of filefish, Stepha nolepis cirrhifer (Temminck et Schlegel) to the color lights. The experimental tank (360LX50WX55H cm) was set up in a dark room. Six Longitudinal sections each being 60 em intervals are marked in the tank to observe the location of the fish. Water depth in the tank was kept 50 em level. Light bulbs of 20W were placed at the both ends of the tank to be projected the light horizontally into the tank. Two different colored filters were selected in combination from four' colors-red, blue, yellow, and white, and were placed in front of the light bulbs to make\ulcorner different light of color. Light intensity were controlled by use of auxiliary filters intercepted between the bulb and the filter. The fish were acclimatized in the dark for 40 minutes prior to employ in the experiment. Upon turning on the light, the number of fish in each section was counted 40 times in every 30 seconds, and the mean of the number of fish in each section was given as the gathering rate of the fish. The results obtained are as follows: 1. Color of light, to which the fish gathered abundantly was found in the named order of blue, white, green, and red. 2. The differences of gathering rate upon arbitary combined two color lights were shown significant, and the differences increased remarkably in accordance with the lapse of illuminating period.

한일공동VLBI상관기(KJJVC)의 운용 및 제어를 위한 Graphical User Interface(GUI) 설계 및 개발

  • 한국천문연구원은 한국우주전파관측망(Korean VLBI Network, KVN)에서 시험 운영 중인 한일공동VLBI상관기(Korea-Japan VLBI Correlator, KJJVC)의 핵심인 VLBI상관 서브시스템(VLBI Correlation Subsystem, VCS)을 제어하기 위해 CCS(Correlation Control Software)를 개발하였다. CCS는 명령어의 문법 검사를 하고, VCS와 TCP/IP 소켓으로 통신하여 명령어와 응답(reply)을 주고받으며, VCS의 실시간 요청에 대응하여 입력 지연 파라미터(delay parameter)를 자동으로 전달하는 소프트웨어이다. CCS는 사용자의 명령어를 local UNX FIFO를 통하여, 명령줄(Command Line)로 전달받고, 이러한 명령어들의 조합을 쉘 스크립트로 묶는 형태로 구성 되어있다. 그러나, 이렇게 명령어의 조합을 직접 편집하여 구성할 때, 입력할 명령어나 지연 파라미터의 구성이 복잡해지면, 스크립트에 필요한 명령이 문법에 맞지 않게 사용되거나, 일부 명령어가 누락될 수 있는 위험이 있다. 이러한 오류를 차단하고 작업 효율을 높이기 위하여, 상관 처리의 전체 운용 및 제어를 통합적으로 할 수 있는 사용자 인터페이스가 필요하다. 이를 위하여 KJJVC의 운용 및 제어를 위한 GUI를 설계하고 제작하였다. 개발된 GUI는 Python 언어를 기반으로 하는 wxPython 패키지를 사용하였으며, 사용자(Operator)가 명령어 문법이나 순서를 확인할 필요 없이, 관측 정보와 관측국 정보, 기록 매체 정보만 가지고 손쉽게 명령어의 조합을 생성해 주며, 시스템 초기화와 각종 정보의 입력이 직관적으로 이루어질 수 있도록 해 준다. 본 연구에서는 개발된 GUI의 설계 및 개발 과정과 시험운영에 대해 소개한다.

  • Cobalt(III) complex containing ethylenediamine-triacetate and trimethylenediamine has been isolated from the reaction of ethylenediamine-tetraacetatocobaltate(III) with trimethylene-diamine in aqueous solution by Dowex 50W-X8, cation exchange resin in $H^+$ form. The ethylenediamine-triacetate($EDTRA^{3-}$) ligand coordinates to the cobalt(III) ion as a quadridentate with a free acetate branch. It has been observed that the complex has trans(O-O) (1) structure via the elemental analysis, UV, IR and NMR data.

Flour Quality Characteristics of Korean Waxy Wheat Lines

  • Flour physicochemical properties of six Korean waxy wheat lines and their parental plants, including Kanto 107 and BaiHuo, which have partially null in GBSS (granule bound starch synthase), were evaluated in this study. The very low amylose content (3.20%) of Korean waxy wheat lines, which had been influenced by the null in all three GBSS isoforms encoded by three Wx loci, could result in the higher starch swelling power (25.15%), lower starch and flour pasting temperature (61.37$^{\circ}C$; 65.85$^{\circ}C$), and higher starch pasting peak viscosity and breakdown (246.60 RVU; 161.50 RVU) than those of their parental plants. In addition to high swelling and pasting properties, Korean waxy wheat lines had the higher protein content (12.80%), alkaline water retention capacity (97.39%), SDS sedimentation volume (80.33 $m\ell$) and damaged starch content (4.35 %) than those of their parental plants.

  • In this study, the optimum conditions for mycelium culture of the mushroom Tricholoma matsutake and the inhibitory effect of the mycelium extracts no tyrosinase activity have been examined. When the extracts of the Tricholoma matsutake mycelia were tested for inhibitory activity on tyrosinase, it was found that the components extracted with ethyl acetate and water showed the highest inhibitory activity. The effect of antioxidants on the growth of mycelium and tyrosinase-inhibiting activity was also investigated. The results showed that tocopherol inhibited the growth in a concentration-dependent manner. In terms of tyrosinase-inhibiting activity, however, tocopherol was found to enhance the inhibitory activity.

  • 자유 라디칼을 이용한 RAFT 중합은 성장하는 고분자 반응을 제어할 수 있는 특성이 있어 주목 받고 있는 고분자 합성방법 중 하나이다. 이 반응의 기작은 agent라 불리는 분자를 주축으로 삼아 단량체들을 단계적으로 성장하는 가역적 방법으로 원하지 않는 종결반응으로부터 고분자 라디칼을 보호하는 역할을 수행한다. 보호의 근본적인 원인은 중간체 상태에서의 안정화 정도와 관련이 있으나 안정해진 만큼 반응속도가 느려지는 지연효과가 발생한다. 지연효과를 유도하는 원인은 많은 논란이 있었으며 그 중 하나로 agent에 존재하는 Z 치환기의 영향을 원인으로 지목하고 있다. 본 연구는 Z 치환기의 변화에 따른 안정화 정도를 파악하기 위하여 RAFT agent로 주로 이용하는 것 중 두 개의 황이 있는 dithioester와 xanthate를 WxMacMolplt 7.3.2를 이용하여 propagation 초기 단계를 구현한 후 GAMESS2 프로그램을 이용하여 양자화학적 계산을 수행하였다. 계산결과 안정화 에너지와 경계 궤도함수에서는 phenyl기가 있을 때 공명효과에 의하여 안정화가 이루어졌으며 또한 propyl benzyl에서도 늘어난 알킬 사슬의 donating effect로 인한 안정화 영향의 범위를 발견하였다. PES 기법을 통해 두 methyl 단량체를 움직이면서 반응하는 동안의 에너지 변화를 알아보았으며 그 결과 dithioester는 Z 치환기의 변화에 더 많이 의존한다는 것을 확인하였다. 본 연구를 종합해본 결과 phenyl을 제외한 aryl기가 있는 dithioester는 낮은 addition 퍼텐셜과 안정화 에너지를 가질 수 있을 것이고 지연효과를 줄일 수 있을 것으로 기대된다.

Allelic Variation of Glutenin, Granule-Bound Starch Synthase l and Puroindoline in Korean Wheat Cultivar

  • To investigate the genetic variation of high-and low-molecular-weight glutenin subunits (BMW-GS and LMW-GS), granule-bound starch synthase I (GBSSI) and puroindoline in 24 Korean wheat cultivars. At the BMW-GS compositions, three Glu-A1 alleles, five Glu-B1 alleles and three Glu-D1 alleles were identified. The high frequency of alleles at each locus was Glu-A1c allele (15 cultivars), Glu-B1b allele (16 cultivars) and Glu-D1f allele (16 cultivars). Four alleles were identified at the Glu-A3 and Glu-B3 loci and three at Glu-D3 locus and Glu-A3d, Glu-B3d and Glu-D3a were mainly found at each Glu-3 locus. Glu-A3d, Glu-B3d, Glu-D3b or c (4 cultivars, respectively) and Glu-A3d, Glu-B3d, Glu-D3a and Glu-A3c, Glu-B3d or h, Glu-D3a (3 cultivar, respectively) were predominantly found in Korean wheats. At the GBSS compositions, 2 waxy wheat cultivars, Shinmichal and Shinmichal1, showed null alleles on the Wx loci and other cultivars were wild type in GBSS compositions. At the puroindoline gene compositions, Korean wheat cultivars carried 3 genotypes, which 10 cultivars (41.7%) were Pina-D1a and Pinb-D1a, 11 cultivars (45.8%) had Pina-D1a and Pinb-D1b and 3 cultivars (12.5%) carried Pina-D1b and Pinb-D1a. These genetic variations could present the information to improve flour and end-use quality in Korean wheat breeding programs.

  • Gamadi, a native rice cultivar from Nepal in which the panicle remains enclosed within its flag leaf sheath upto maturity, was crossed with different genetic marker testers of 12 linkage groups in order to analyze its linkage relationship. The results obtained from the experiment were summarized as follows: Normal segregations of all the genetic marker genes used in this experiment viz Cl, wx and Pla of linkage group I, Pn, Rd and Pub of linkage group III, and lg, g, Ps, gh, Hla, la, nl, bl, be and gl of linkage groups II, IV, V, VI, VII, VIII, IX, X, XI, and XII respectively confirmed the previous results, and also strongly indicated that the genetic constituent of the Gamadi and marker testers is same. 'Gamadiness' (the panicle enclosing character) was controlled by two complementary dominant genes with the segregation ratio of 9 Gamadi to 7 normal panicle-exserting types. These genes have been temporarily proposed as G-a and G-b for gamadiness. G-a gene was found to be linked with the neckleaf gene (nl) of linkage group Ⅸ with the crossover value of 0.3733$\pm$0.027. G-b gene appeared to be associated with the brittle culm gene (bc) of the linkage group XI with the crossover value of 0.2725$\pm$0.061.TEX>0.061.

  • In order to improve the efficiency of F$_1$ hybrid seeds production(KF 114), some experiments were carried out in a greenhouse this year. Mother plant(MSNC567) and pollen plant(NC) were grown in some pots (30 x 30cm, WxH). The gathered pollens were mixed with celite, pollen deluent dusts and stored in refrigerator at l$0^{\circ}C$ before pollination. To establish the critical range of the mixed rate and the storage period of pollens, the change in a percent of capsule set, number of seed and seed weight per capsule, weight of 1000 seeds and germination percent of seed resulted of pollination with pollen deluent dusts were investigated. The results are as follows; Percent of capsule set showed the mixed rate 1:5 resulted in no difference and those of 1:10 and 1:20 decreased about 5 ~15% when compared with trials using pollen alone. There was no difference among duration of storage in a same mixed rate. Numbers of seed per capsule showed a significant difference among the mixed rates and among durations of storage. Numbers of seed per capsule were decreased about 55 ~ 90 % as the mixed rate increase, about 2 ~ 28 % as the duration of storage increase. Weight of seed per capsule decreased about 16 ~ 23 % as the duration increase and decreased about 3 ~ 89 % as the mixed rate increase when compared with trials using pollen alone. Weight of 1000 seeds showed non significant difference among duration of storage but were higher than trials used pollen alone as the mixed rate increase.

Crystal structure of CodW in Bacillus Subtilis - the first N-terminal serine pretense

  • CodWX, encoded by the cod operon in Bacillus subtilis, is a member of the ATP-dependent protease complex family, and is homologous to the eukaryotic 26S proteasome. It consists of two multimeric complexes: two hexameric ATPase caps of CodX and a protease chamber consisting of CodW dodecamer. Prior structural studies have shown that the N-terminal threonine residue is solely functional as a proteolytic nucleophile in ATP-dependent proteases such as HslV and certain β-type subunits of 20S proteasome, which have a primary sequence similarity of -50% and -20% with CodW respectively. Here we present a 3.0 Å resolution crystal structure of CodW, which is the first N-terminal serine protease among the known proteolytic enzymes. In spite of the same fold and the conserved contacts between subunits with HslV in E. coli and H. influenza, this structure shows the five additional residues extending from conserved Thr1 among the other ATP-dependent pretense and extraordinary basic proteolytic chamber.
