Development of Wheat Breeding Material Mediated wide Hybridization Response to Climate Change

  • This study is to develop new wheat breeding material through wide hybridization with wild species harboring useful characteristics such as salt, heat, and drought tolerance. Leymus mollis, wild rye was used to improve wheat genetic quality. L. mollis, is a perennial plant harboring tolerance against salt, heat, and drought because L. mollis distributes on the seaside. The F1 hybrids were produced by crossing between common wheat (Triticum aestivum L., Chinese Spring) and L. mollis. Genomic in situ hybridization revealed that the F1 hybrids have L. mollis genome. For the evaluation of salt and drought tolerance, seeds from the F2 were used. Under 2% NaCl solution, the F3 wheat-Leymus addition plants with salt tolerance showed more tillering and longer roots than other F3 plants without salt tolerance. Also, the F3 plants with salt tolerance showed better shallow-rooted than other F3 plants without salt tolerance. Finally, the F3 plants with salt tolerance made seed-setting under 2% NaCl condition, but other F3 plants without salt tolerance were not. Under drought conditions, the F3 plants with drought tolerance showed longer culm and spike length than other F3 plants without drought tolerance and even those of Chinese Spring under well-water conditions. We evaluated and selected the F3 plants with salt or drought tolerance for generation advancement.

조류 유래 재조합 H7N1 인플루엔자 바이러스의 분자적 특성 규명 (Molecular Characterization of an Avian-origin Reassortant H7N1 Influenza Virus)

  • 최근에 재조합 H7Nx 인플루엔자 바이러스가 산발적으로 인체 감염 사례가 보고되고 있으며 이러한 바이러스는 조류 종으로부터 지속적으로 분리되고있다. 본 연구에서는 조류에서 유래된 H7N1 인플루엔자 바이러스를 분리하여 A/wild bird/South Korea/sw-anu/2023로 명명하였고, 전장유전체 분석과 분자적 특성을 분석하였다. 계통발생학적 분석 결과 A/wild bird/South Korea/sw-anu/2023는 유라시아 혈통에 속하는 H7N1 인플루엔자 바이러스로 확인되었다. A/wild bird/South Korea/sw-anu/2023 바이러스의 polymerase basic 1(PB)2, PB1, polymerase acidic (PA), nucleoprotein (NP) 유전자는 야생 조류에서 분리되었던 조류 인플루엔자 바이러스유전자와 밀접한 관련이 있는 것으로 밝혀졌으며, hemagglutinin (HA), neuraminidase (NA), matrix (M), nonstructural (NS) 유전자는 집오리에서 분리되었던 조류 인플루엔자 바이러스와 유사하였다. 이러한 결과는 동아시아-호주 이동 경로를 따라 이동하는 야생 조류들 사이에서 새롭게 유전자가 재배열된 재조합 H7N1 조류 인플루엔자 바이러스가 순환되고 있음을 시사하고 있다. 따라서, H7Nx 인플루엔자 바이러스는 전 세계적으로 순환하며, 돌연변이된 H7N1 조류 인플루엔자 바이러스는 인간을 감염시킬 수 있으므로 야생 조류 및 가금류에서 H7N1 조류 인플루엔자 바이러스의 지속적인 감시가 필요할 것이다.

Saxifraga cortusifolia (Saxifragaceae): A new record for the Korean flora from Jejudo Island

  • Saxifraga cortusifolia Siebold & Zucc. (Saxifragaceae, sect. Irregulares, ser. Rufescentes), a species considered endemic along mountain streams throughout Honshu, Shikoku, and Kyushu in Japan, was discovered as a wild population in the Cheona Valley in Haean-dong, Jeju-si on Jeju-do Island of Korea. The population consisted of three subpopulations occupying narrow areas 10 × 10 m2, 5 × 5 m2, and 10 × 5 m2 in size. The number of individuals in the three subpopulations was confirmed to exceed 100. Most of the individuals were on or below rocks at the edge of the valley. A morphological description, detailed photos, and a key to related taxa in Korea are presented.

Peroxiredoxin I participates in the protection of reactive oxygen species-mediated cellular senescence

  • Peroxiredoxin I (Prx I) plays an important role as a reactive oxygen species (ROS) scavenger in protecting and maintaining cellular homeostasis; however, the underlying mechanisms are not well understood. Here, we identified a critical role of Prx I in protecting cells against ROS-mediated cellular senescence by suppression of $p16^{INK4a}$ expression. Compared to wild-type mouse embryonic fibroblasts (WT-MEFs), Prx $I^{-/-}$ MEFs exhibited senescence-associated phenotypes. Moreover, the aged Prx $I^{-/-}$ mice showed an increased number of cells with senescence associated-${\beta}$-galactosidase (SA-${\beta}$-gal) activity in a variety of tissues. Increased ROS levels and SA-${\beta}$-gal activity, and reduction of chemical antioxidant in Prx $I^{-/-}$ MEF further supported an essential role of Prx I peroxidase activity in cellular senescence that is mediated by oxidative stress. The up-regulation of $p16^{INK4a}$ expression in Prx $I^{-/-}$ and suppression by overexpression of Prx I indicate that Prx I possibly modulate cellular senescence through $ROS/p16^{INK4a}$ pathway.

Variation in the number of nucleoli and incomplete homogenization of 18S ribosomal DNA sequences in leaf cells of the cultivated Oriental ginseng (Panax ginseng Meyer)

  • Background: Wild ginseng, Panax ginseng Meyer, is an endangered species of medicinal plants. In the present study, we analyzed variations within the ribosomal DNA (rDNA) cluster to gain insight into the genetic diversity of the Oriental ginseng, P. ginseng, at artificial plant cultivation. Methods: The roots of wild P. ginseng plants were sampled from a nonprotected natural population of the Russian Far East. The slides were prepared from leaf tissues using the squash technique for cytogenetic analysis. The 18S rDNA sequences were cloned and sequenced. The distribution of nucleotide diversity, recombination events, and interspecific phylogenies for the total 18S rDNA sequence data set was also examined. Results: In mesophyll cells, mononucleolar nuclei were estimated to be dominant (75.7%), while the remaining nuclei contained two to four nucleoli. Among the analyzed 18S rDNA clones, 20% were identical to the 18S rDNA sequence of P. ginseng from Japan, and other clones differed in one to six substitutions. The nucleotide polymorphism was more expressed at the positions 440-640 bp, and distributed in variable regions, expansion segments, and conservative elements of core structure. The phylogenetic analysis confirmed conspecificity of ginseng plants cultivated in different regions, with two fixed mutations between P. ginseng and other species. Conclusion: This study identified the evidences of the intragenomic nucleotide polymorphism in the 18S rDNA sequences of P. ginseng. These data suggest that, in cultivated plants, the observed genome instability may influence the synthesis of biologically active compounds, which are widely used in traditional medicine.

A phylogenetic analysis of the Korean endemic species Paraphlomis koreana (Lamiaceae) inferred from nuclear and plastid DNA sequences

  • Paraphlomis koreana (Lamiaceae) was newly named and added to Korean flora in 2014. Paraphlomis belongs to the tribe Paraphlomideae, along with Ajugoides and Matsumurella. However, a recent study has suggested that P. koreana is morphologically similar to Matsumurella chinensis, making them difficult to distinguish from each other. Therefore, we aimed to examine the phylogenetic placement of P. koreana within the tribe and compare its genetic relationship with M. chinensis. We sequenced an additional complete plastid genome for an individual of P. koreana and generated sequences of nuclear ribosomal (nr) DNA regions of internal and external transcribed spacers (ITS and ETS) for two individuals of P. koreana. Maximum likelihood analyses based on two nrDNA regions (ITS and ETS) and four plastid DNA markers (rpl16 intron, rpl32-trnL, rps16 intron, and trnL-F) covering 13 Paraphlomis species and M. chinensis were conducted. Phylogenetic analyses concordantly supported that P. koreana forms a monophyletic group with M. chinensis. Moreover, our study revealed that P. koreana includes nrDNA sequences of M. chinensis as minor intra-individual variants, suggesting that the genetic divergence between the two taxa is incomplete and may represent intraspecific variation rather than distinct species. In conclusion, our findings suggest that the independent species status of P. koreana within Paraphlomis should be reconsidered.

Analysis of in planta Expressed Orphan Genes in the Rice Blast Fungus Magnaporthe oryzae

  • Genomes contain a large number of unique genes which have not been found in other species. Although the origin of such "orphan" genes remains unclear, they are thought to be involved in species-specific adaptive processes. Here, we analyzed seven orphan genes (MoSPC1 to MoSPC7) prioritized based on in planta expressed sequence tag data in the rice blast fungus, Magnaporthe oryzae. Expression analysis using qRT-PCR confirmed the expression of four genes (MoSPC1, MoSPC2, MoSPC3 and MoSPC7) during plant infection. However, individual deletion mutants of these four genes did not differ from the wild-type strain for all phenotypes examined, including pathogenicity. The length, GC contents, codon adaptation index and expression during mycelial growth of the four genes suggest that these genes formed during the evolutionary history of M. oryzae. Synteny analyses using closely related fungal species corroborated the notion that these genes evolved de novo in the M. oryzae genome. In this report, we discuss our inability to detect phenotypic changes in the four deletion mutants. Based on these results, the four orphan genes may be products of de novo gene birth processes, and their adaptive potential is in the course of being tested for retention or extinction through natural selection.

Effect of Lactobacillus dominance modified by Korean Red Ginseng on the improvement of Alzheimer's disease in mice

  • Background: Gut microbiota influence the central nervous system through gut-brain-axis. They also affect the neurological disorders. Gut microbiota differs in patients with Alzheimer's disease (AD), as a potential factor that leads to progression of AD. Oral intake of Korean Red Ginseng (KRG) improves the cognitive functions. Therefore, it can be proposed that KRG affect the microbiota on the gut-brain-axis to the brain. Methods: Tg2576 were used for the experimental model of AD. They were divided into four groups: wild type (n = 6), AD mice (n = 6), AD mice with 30 mg/kg/day (n = 6) or 100 mg/kg/day (n = 6) of KRG. Following two weeks, changes in gut microbiota were analyzed by Illumina HiSeq4000 platform 16S gene sequencing. Microglial activation were evaluated by quantitative Western blot analyses of Iba-1 protein. Claudin-5, occludin, laminin and CD13 assay were conducted for Blood-brain barrier (BBB) integrity. Amyloid beta (Aβ) accumulation demonstrated through Aβ 42/40 ratio was accessed by ELISA, and cognition were monitored by Novel object location test. Results: KRG improved the cognitive behavior of mice (30 mg/kg/day p < 0.05; 100 mg/kg/day p < 0.01), and decreased Aβ 42/40 ratio (p < 0.01) indicating reduced Aβ accumulation. Increased Iba-1 (p < 0.001) for reduced microglial activation, and upregulation of Claudin-5 (p < 0.05) for decreased BBB permeability were shown. In particular, diversity of gut microbiota was altered (30 mg/kg/day q-value<0.05), showing increased population of Lactobacillus species. (30 mg/kg/day 411%; 100 mg/kg/day 1040%). Conclusions: KRG administration showed the Lactobacillus dominance in the gut microbiota. Improvement of AD pathology by KRG can be medicated through gut-brain axis in mice model of AD.

Rice 7-Hydroxymethyl Chlorophyll a Reductase Is Involved in the Promotion of Chlorophyll Degradation and Modulates Cell Death Signaling

  • The loss of green coloration via chlorophyll (Chl) degradation typically occurs during leaf senescence. To date, many Chl catabolic enzymes have been identified and shown to interact with light harvesting complex II to form a Chl degradation complex in senescing chloroplasts; this complex might metabolically channel phototoxic Chl catabolic intermediates to prevent oxidative damage to cells. The Chl catabolic enzyme 7-hydroxymethyl Chl a reductase (HCAR) converts 7-hydroxymethyl Chl a (7-HMC a) to Chl a. The rice (Oryza sativa) genome contains a single HCAR homolog (OsHCAR), but its exact role remains unknown. Here, we show that an oshcar knockout mutant exhibits persistent green leaves during both dark-induced and natural senescence, and accumulates 7-HMC a and pheophorbide a (Pheo a) in green leaf blades. Interestingly, both rice and Arabidopsis hcar mutants exhibit severe cell death at the vegetative stage; this cell death largely occurs in a light intensity-dependent manner. In addition, 7-HMC a treatment led to the generation of singlet oxygen ($^1O_2$) in Arabidopsis and rice protoplasts in the light. Under herbicide-induced oxidative stress conditions, leaf necrosis was more severe in hcar plants than in wild type, and HCAR-overexpressing plants were more tolerant to reactive oxygen species than wild type. Therefore, in addition to functioning in the conversion of 7-HMC a to Chl a in senescent leaves, HCAR may play a critical role in protecting plants from high light-induced damage by preventing the accumulation of 7-HMC a and Pheo a in developing and mature leaves at the vegetative stage.

Perenniality-Potential and challenges for future sustainable crop production

  • The most drought resistant among the five most important cereal crops, and a key dual-use (grain and biomass) crop in regions containing some of the world's most degraded soils, sorghum has inherent climate resilience that is likely to become more important under environmental conditions that are projected by many climate change models. The importance of sorghum might be further elevated by the development of productive genotypes that increase the extent and duration of soil cover beyond those of conventional annual crops, mitigating or even reversing losses of ecological capital through multiple crops from single plantings. Rich genetic and genomic resources have been developed to link Sorghum phenotypic diversity to its molecular basis, and in particular the genus has become a model for dissecting the molecular control of perenniality. Nature has made Sorghum perennial at least twice, and crosses between wild perennials and cultivated sorghums show the feasibility of developing genotypes with varying degrees of investment in perenniality while still providing harvestable food, feed, sugar and/or cellulose. Genetic analysis of progeny from these crosses is revealing the hereditary basis of traits related to ratooning and perenniality and providing diagnostic DNA markers. One perennial Sorghum species has adapted to continents and latitudes far beyond the reach of its progenitors, surviving stresses year after year that are only periodically experienced by conventional (annual) sorghum, and may also harbor novel alleles that may mitigate production challenges in conventional annual sorghums.
