Molecular Characterization of Hypernodulation in Soybean

  • SS2-2, a hypernodulating soybean mutant was isolated by EMS mutagenesis from Sinpaldalkong 2. This auto-regulation mutant showed greater number of nodules and smaller plant size than its wild type Sinpaldalkong 2. SSR markers were used to identify DNA variation at SSR loci from different soybean LG. The only SSR marker that detected a length polymorphism between SS2-2 and its wild type ancestor was Satt294 on LG C1 instead of LG H, locating a hypernodulating gene. Sequencing data of flanking Satt294 indicated that the size variation was due to extra stretch of TTA repeats of the SSR motif in SS2-2, along with $A\longrightarrow$G transversion. In spite of phenotypic differences between the wild type and its hypernodulating mutants, genomic DNA poly-morphisms at microsatellite loci could not control regulation of nodule formation. The cDNA-AFLP method was applied to compare differential display of cDNA between Sinpaldalkong 2 and SS2-2. After isolation and sequence comparison with many AELP fragments, several interesting genes were identified. Northern blot analysis, immunolocalization and/or the yeast two-hybrid system with these genes might provide information on regulation of nodule development in SS2-2.

Assessment of environmental impact of vitamin A-enhanced soybeans and hybrid soybeans

  • An understanding of safety problems pursuant to environmental release of GM (Genetically Modified) crops is considered important. Among the recognized safety problems, the possibilities of weediness and ecosystem invasion are constantly being validated. We herein compared the growth characteristics and germination rate of soybeans formed by hybridization with vitamin A-enhanced soybeans carrying an introduced gene that increases β-carotene content. We also examined overwintering, survival, and weed competitiveness to evaluate hybrid ecological impact on long-term unmanaged cultivatable land. These studies revealed that the hybrid soybeans exhibited intermediate growth characteristics and germination rate compared with the vitamin A-enhanced soybeans and wild soybeans, or exhibited traits similar to those of the maternal strain. Overwintering experiments were conducted by planting seeds at depths of 0, 5, 10, and 20 cm and recovering them after three or five months. After five months, all seeds at depths more than 5 cm lost viability. Among seeds recovered after three months, only wild soybeans retained viability at depths of more than 5 cm. Survival and weed competitiveness were assessed by sowing each type of seed and performing no irrigation, or pest or weed control. Quantitative assessment of numbers of individual soybean plants that appeared in the experimental plot revealed that all plants germinated after sowing, but only wild type plants survived overwintering. These studies suggest that both GM soybeans and hybrid soybeans cannot survive in uncultivated land even if they are released into the environment, which indicates less possibility of ecosystem invasion and weediness.

숙성 기간 및 절임원에 따른 고사리장아찌의 이화학적 특성 변화 (Physiochemical Properties of Fernbraken Jangachi during Korean Traditional Pickling Process)

  • Fernbraken is a popular and well-known wild grass, but the physiochemical properties of Korean Traditional pickling (Jangachi) during aging have been little reported. Therefore, this study was carried out to investigate the physiochemical properties of Fernbraken Jangachi treated with a soybean sauce mixture for 6 weeks (1st pretreatment) as well as fermented with soybean paste, red hot pepper paste and soybean sauce for 7 weeks. Total polyphenol contents, acidity, salinity, sweetness (Brix), and ${\alpha}$-tocopherol of Fernbraken Jangachi all increased with aging period. On the other hand, pH and total acidity of Fernbraken Jangachi did not change much. Among the three kinds of Jangachies, Jangachi fermented with soybean sauce showed 2 fold higher salinity and total acidity levels compared to the others. Therefore, the physiochemical properties of Fernbraken Jangachi were dependent on the pickling properties, such as soybean paste, hot pepper paste or soybean sauce, and these properties maintained a certain level after 5~6 weeks of aging. Further, proper aging period for Fernbraken Jangachi was suggested as 10 weeks.

  • Wildbean(Glycine soja Siebold et Zucc.) is known as the orgin of soybean (Glycine max Merr.). Based on the hypothesis that the secondary transformation of chemical compound in wildbean might have occurred during its breed improvement to soybean. this study was carried out to compare the antioxidant activities and chemical composition in wildbean and soybean. The present study demonstrates that 1) Antioxidant activity was much higher in EtOAc, extract of wildbean than in soybean. 2) strong antioxidant activity observed in EtOAc extract of wildbean was due to the presence of (-)-epicatechin, which was not present in the extract of soybean but isolated, for the first time, from the extract of wild bean, and 3) antioxidant activity of the isolated (-)-epicatechin was greater than that of tocopherol, the previously known antioxidant.

Genetic Mapping of Hypernodulation in Soybean Mutant SS2-2

  • Hypernodulation soybean mutant, SS2-2, is characterized with greater nodulation and nitrogen fixing ability in the root nodule than its wild type, Shinpaldalkong 2. The present study was performed to identify a genetic locus conferring hypernodulation in soybean mutant SS2-2 and to determine whether the gene controlling the hypernodulation of SS2-2 is allelic to that controlling the supernodulation of nts382 mutant. Hybridization studies between SS2-2 and Taekwangkong revealed that the recessive gene was responsible for the hypernodulation character in soybean mutant SS2-2. Allelism was also tested by crossing supernodulating mutant nts382 and hypernodulating mutant SS2-2 that both hypernodulation and supernodulation genes were likely controlled by an identical locus. Molecular marker mapping of hypernodulation gene in SS2-2 using SSR markers confirmed that the gene conferring hypernodulation was located at the same loci with the gene conferring supernodulation. It is interesting to note that the same gene controlled the super- and hyper-nodulation characters, although SS2-2 and nts 382 exhibited differences in the amount of nodulation in the root system. Further genetic studies should be needed to clarify the genetic regulation of super- and hyper-nodulation in soybean.

  • 대두 경남 장려품종인 장백, 광교, 강림 및 남천등의 근류로부터 Rhizobium japonicum을 분리하여 그 중 질소고정력이 R. japonicum 61A 76이나 3 I1110보다 강한 JB 101을 선별하였다. 선별된 JB 101을 N-methy1-N'-nitro-N-nitrosoguanidine 처리 및 UV조사에 많은 돌연변이체를 얻고 이들로 부터 경남 장려품종인 장백에 접종시근류 생성력 및 질소고정력등 JB 101에 비하여 훨씬 높을 뿐만아니라 SM 35에 비해서도 다소 높은 JB 65를 선별하여 이 균주의 근류생성력, 질소고정력, 고정된 암모니아가 질소고정에 미치는 영향, hydrogenase 활성등 공생관련 제 특성과 아울러 이 균주 단백질의 아미노산 조성, 2D-polyacrylamide gel을 이용한 전기영동등의 생화학적 특성도 아울러 조사하였다.

항생물질(抗生物質) 표식(標識) 근류균(根瘤菌)의 조제(造製)와 그 접종효과 (Preparation of Antibiotic-Resistant Bradyrhizobium japonicum and Its Inoculation Effects on Soybean [Glycin max(L.) Merr])

  • 현재 대두근류군 접종제로서 이용되고 있는 Bradyrhizobium japonicum의 토양중에서 그 추이를 밝히고자 항생제 내성균주를 조제하여 그들의 특성을 모균주와 비교하고 아울러 토양에서 접종효과를 추적한 결과는 다음과 같다. 1. USDA110K-$STR^r$, USDA110N-$STR^r$, R138-$STR^r$은 pH9에 내성이 약했으며, R214-$STR^r$, $NAL^r$, USDA110N-$STR^r$은 sorbitol에 R138-$STR^rNAL^r$은 xylose에 대한 자화성이 불분명했다. 2. Serogroup에 의해 각 균주들은 서로 다른 균주로 동정되었으나 원균주와 표식균주(標識菌株)간에는 거의 차이가 없었다. 3. Plasmid size는 생육 촉진형 군이 지연형보다 약간 컸으나, 야생균주와 그들의 표식균간(標識菌間)에는 각각 차이가 없었다. 4. 5년이상 대두 재배 토양에 표식균주(標識菌株) 접종시(接種時) 0~12.5%, 대두 무재배 토양에서는 5~22.5%의 접종률을 보였다. 5. 원균주와 표식균주(標識菌株)를 각각 접종시 5년이상 대두재배 토양에서는 접종효과가 없었으나 대두 무재배 토양에서는 상당한 접종효과가 있었으며 원균주가 표식균주(標識菌株)보다 더 큰 효과를 나타냈다.

옥수수와 사료용 콩 혼작에 의한 조사료 수량 및 품질 (Yield and Quality of Forage Produced by Mixed Planting of Soybean and Corn)

  • The soybean [Glycine max (L.) Merr.], an edible legume, has a high protein content in both its hay and grain, so it is often used as a supplement for other forages that have a deficient protein concentration. Therefore, this study investigated the forage quality and yield in the case of mixed planting of soybean and corn. The forage yield and quality were assessed for three cropping patterns: soybean mono planting, corn mono planting, and mixed planting of soybean and corn. For planting, this study used a forage corn cultivar, Kwangpyeongok, and three recombinant inbreed lines, W2, W4, and W11, selected from Glycine soja (PI483463)${\times}$G. max (Hutcheson). The mixed planting of soybean and corn produced a higher forage yield than the corn mono cropping. The crude protein and crude fat content were also increased with the mixed planting of soybean and corn when compared with the corn mono cropping. Some decrease of ADF and NDF, and increase for RFV in mixed planting of soybean and corn than corn mono cropping. Therefore, the results show that mixed planting of soybean and corn is an effective cropping system to improve the forage quality.

한국, 일본 및 중국 지린성 야생콩(Glycine soja Sieb. and Zucc.)의 SSR마커에 의한 유전적 다양성과 유연관계 (Genetic diversity and relationships of Korean, Japanese, and Chinese Jilin provincial wild soybeans (Glycine soja Sieb. and Zucc.) based on SSR markers)

  • 한국의 농업유전자원센터로부터 분양받은 한국 야생콩, 일본의 Biological Resource Center in Lotus and Glycine, Frontier Science Research Center, University of Miyazaki로부터 분양 받은 일본 야생콩, 그리고 중국 지린성에서 수집되어진 야생콩 의 유전적 다양성과 유연관계를 SSR마커로 분석하여 콩 육종의 유전적 변이 확대를 위한 기초자료로 이용하고자 수행한 연구결과를 요약하면 다음과 같다. 1. 한국 야생콩 67종, 일본 야생콩 71종 및 중국 지린성의 야생콩 46종을 포함한 총 184종을 23개의 SSR마커로 유전적 다양성과 유연관계를 분석한 결과, 총 964개의 대립인자가 확인되었고, 평균 41.9개의 대립인자가 확인되었다. SSR마커별로 대립인자 수는 최소 23개(Satt635)에서 최대 56개(Satt157)까지 확인되었으며, PIC 값의 범위는 0.880~0.968로 평균 0.945이었다. 2. 한국 야생콩은 대립인자의 수는 총 513개이었고 평균 22.3개 이었으며, 일본 야생콩은 대립인자의 수는 총 511개이었고 평균 22.2개이었으며, 중국 지린성 야생콩의 대립인자의 수는 총 312개 이었으며 평균 13.6개이었다. 평균 유전적 다양성(PIC 값)은 한국 야생콩이 0.905, 일본 야생콩이 0.897 및 중국 지린성의 야생콩이 0.850로서 큰 차이가 없었다. 3. 한국, 일본 및 중국 지린성의 야생콩들은 SSR마커를 이용한 유전적 거리에 따른 군집분석에서 3그룹으로 구분되었다. I그룹은 중국 지린성의 야생콩만이 분포하였고, II그룹은 대부분이 일본의 야생콩이 분포하였는데 한국의 야생콩 5종이 포함되었으며, III그룹은 대부분이 한국의 야생콩이 분포하였으며 일본의 야생콩 6종과 중국 지린성의 야생콩 1종이 포함되었다. I그룹은 동일그룹 내에서 다른 군이 형성되지 않았으나, II그룹과 III그룹은 동일그룹 내에서 각각 몇 개의 군으로 분류되었다. 4. SSR마커에 의한 유연관계는 한국과 일본의 야생콩 간의 유전적 거리가 한국과 중국 지린성 및 일본과 중국 지린성의 야생콩 간의 유전적 거리보다 더 가까웠다.
