Cryopreservation of Mulberry Germplasm Core Collection and Assessment of Genetic Stability through ISSR Markers

  • A simple and reliable cryo technique using desiccation and slow freezing of winter dormant buds was employed for 238 core collection of mulberry germplasm collected from diverse geographical regions and maintained under tropical conditions in the ex situ field gene bank to develop long-term biodiversity conservation for ensuring sustainable utilization of these valuable resources. Desiccation and freezing tolerance of bud grafts and excised shoot apices in the axillary buds of different Morus species under in vivo and in vitro condition indicated species-specific variation and most of the wild Morus species were found sensitive. In vitro regeneration and cryopreservation($-196^{\circ}C$) protocols using differentiated bud meristem like axillary winter dormant buds were worked out for a wide range of Morus species, land races, wild and cultivated varieties. Successful cryopreservation of mulberry winter dormant buds of different accessions belonging to M. indica, M. alba, M. latifolia, M. cathayana, M. laevigata, M. nigra, M. australis, M. bombycis, M. sinensis, M multicaulis and M. rotundiloba was achieved. Among wild species Morus tiliaefolia, and M. serrata showed moderate recovery after cryopreservation. Survival rates did not alter after three years of cryopreservation of different Morus species. ISSR markers were used to ascertain the genetic stability of cryopreserved mulberry, which showed no difference detected among the plantlets regenerated from frozen apices in comparison to the non-frozen material.

  • The chromosome number of Morus bombycis Koidz. and Morus monogolica C.K.Schn. growing wild in the Korea Peninsula is diploid (2n=28) and that of Morus tiliaefolia Makino is hecxaploid (2n=84). The somatic cell division of each species is nomal.

뽕나무 절지의 저장 습도가 새싹의 생장에 미치는 영향 (Effects of Storage Humidity on the Sprout Growth of Mulberry Cut Twigs)

  • 목본류의 절지를 이용한 싹기름 채소생산의 기술을 개발하기 위하여 뽕나무 가지를 2006년 4월 13일에 절지하여 $4{\sim}6^{\circ}C$의 저온저장고에서 습도조건($40{\sim}50%$$80{\sim}90%$)에 따른 생존율과 $1{\sim}3$개씩의 싹을 붙인 가지를 자른 후 싹기름을 했을 때 저온저장 일수에 따른 새싹의 생장 정도를 조사하였다. 뽕나무 절지의 생존율은 저장일수가 길수록 낮아져 저온저장을 하지 않은 채취 당일에 싹기름을 한 것은 97.4%였고, 122일 저온저장한 후 싹기름을 한 절지는 $40{\sim}50%$의 습도조건에서 34.2%, $80{\sim}90%$의 습도조건에서 저장한 것은 85.7%였다. 또한 절지의 저장일수가 길수록 새싹의 잎수가 증가하는 경향을 보였고, 싹의 길이와 폭도 큰 경향을 나타내었다. 따라서 뽕나무의 절지를 $4{\sim}6^{\circ}C$의 저장온도와 $80{\sim}90%$의 습도조건에서 저장한 후 $1{\sim}3$개씩의 싹을 붙인 가지를 잘라 싹기름을 하면 새싹 채소의 생산이 가능한 것으로 나타났다.

대자뽕과 감백뽕의임성 및 오디의 과실적 특성 (Fertility and Characteristics of Mulberry fruits on Daejappong and Garmbacppong (Morus Spp.))

  • 오디용 뽕품종으로 국상20호의 동질4배체인 대자뽕(4원뽕 31호)을 육성하였으며, Turkey 재래종 중에서 3배성인 감백뽕을 선발 육성하였는바 그 주요 특성은 다음과 같다. 1. 염색체수는 대자뽕은 2n=56인 4배체이고, 감백뽕은 2n=42인 3배체 이다. 2. 대자뽕의 종자형성률은 48.2% , 침종자율은 32.7%로 낮았으나 발아율은 90%에 달하였다. 감백뽕의 종자형성율은 41.0% , 침종자율은 1.9%, 발아율은 7.0%에 불과하였다. 3. 대자뽕 오디의 단과중은 4.39 g로 대과충만형이고, 오디색은 연자색이며, 감백뽕 오디는 3.269의 중대형이고 유백색 이다. 4.대자뽕 오디의 당도는 14.2 Brix 총산도는 0.45%이며, 감잭뽕 오디의 당도는 19.3%로서 고당성이고 총산도는 0.15%이다. 5.두 품종 모두 오디의 숙기는 만숙성이고, 오디의 착과정도는 대자뽕은 많으며, 감백뽕은 보통수준이다. 춘기 발아개엽기는 공시품종 모두 중생뽕이다

  • 멧누에는 실내에서의 대량사육이 곤란한 것으로 알려져 왔으나 부화에서부터 100%가까운 다습상태와 $27^{\circ}C$전후의 고온사육조건에서 간단히 사육할 수 있었다. 실내사육결과 전령경과일수는 15~25일의 범위이며 대부분의 유충은 17일 전후이었다. 용화비율은 약40%이며 용기간은 암수모두 13~25일 사이였다. 또한 암수모두 2개의 우화최고점을 가지며 수컷이 암컷에 비해 2~3일정도 빨리 우화하였다.

  • 한반도에 자생하는 돌뽕나무를 수집하여 그 형태학적 특성을 조사한 결과는 다음과 같다 1.체세포의 염색체수는 2n=84인 6배체이다. 2.웅화수는 3.6~6.0cm의 긴 원통형이고 무화주인 자화수의 길이는 18 mm로 왜소하고 자 색 또는 자홍색의 오디는 1.39~l .65 g이다. 3. 동동아는 가지에 비하여 매우 크고 부아는 존재하지 않는다. 4. 잎은 대형의 열엽 또는 전연엽이며, 피목 밀도는 9.42$\pm$1.29개/$\textrm{cm}^2$이다. 5. 잎의 모용 길이는 236~438$\mu\textrm{m}$이고, 이면에 밀도가높은 바 16~37개/25 $\textrm{mm}^2$분포한다. 6. 잎의 표면의 모용형태는 원추형, 이면의 것은 매우 세장한 원추형이다

한국 원예식품의 항산화 비타민 함량 분석 (Determination of Antioxidant Vitamins in Horticultural foods)

  • This study examines the effects of the Korean horticultural foods which are, by traditional wisdom, supposed to contain anti-aging substance. All twenty three kinds of plants are chosen as a sample for the experiment in which the content of three main anti-aging nutrients, -carotene, Vitamin C, and Vitamin E are respectively measured by Nilis, Colorimetric, and AOAC method. The result shows: among the samples, top six plants that contain the most -carotene content are high quality tea(Camellia sinensis), thistle(Cirsium japonicum Var.), Angelica keiskei, mulberry leaves(Morus alba l.), field horsetail(Equisetum arvensa), and carrot(Daucus carota), in order; top four for a Vitamin C contents are low quality tea, mulberry leaves, pine leaves(Pinus densiflora), and high quality tea, in order; top four for a Vitamin E contents are persimmon leaves(Diospyros mongolicum) tea, high quality tea, low quality tea, and pine leaves, in order. Drying or boiling process increases the content of -carotene and Vitamin E in high quality tea, dropwort(Oenanthe javanica), field horsetail, and wild aster(Ligularia stenocephala Marsumum et Koldzumi), while, in case of mulberry leaves, drying process lowers them. Vitamin C content strikingly decreases in heating and drying process.

Regional and Varietal Variation of 1-Deoxynojirimycin (DNJ) Content in the Mulberry Leaves

  • This study was carried out to find out whether there exists any content variation in 1-deoxynojirimycin (DNJ) among regional and varietal mulberry leaves. HPLC analysis was performed for 22 samples collected from several localities in Korea and Tsushima Islands Japan. The highest content was observed in the Cheongilppong sample collected at Youngchun City, Kyungsangbuk Province (1,882.7 $\mu\textrm{g}$/g DW), whereas lowest content was observed in the Cheongilppong sample collected at Jiniu City, Kyungsangnam Province (395.65 $\mu\textrm{g}$/g DW). In the comparison of regional samples of Kaeryangppong variety, DNJ content was highest in the order of Woniu City (1,460.1) > Boungun (999.85) > Hweongsung-gun (939.12) > Cheongju City (688.78). In case of Cheongilppong, DNJ content was highest in the order of Youngchun City (1.882.8) > Sanchung-gun (1,125.3) > Sunchon City (649.17) > Byunsan-myon (441.54) > Jinju City (395.65). Average content of regional samples was higher in Kaeryangppong (1,021.7 $\mu\textrm{g}$/g DW) than that of Cheongilppong (898.87 $\mu\textrm{g}$/g DW). Average DNJ content of the wild mulberry loaves collected from Cheju Island and Tsushima Island (1,012.9$\mu\textrm{g}$/g DW) was high enough to reach to that of the regional Kaeryangppong samples. In the comparison among varietal samples collected in Suwon, the highest DNJ content was observed in YK209 (768.16 $\mu\textrm{g}$/g DW), amongst overall low DNJ content of the varietal samples. Considering a few pieces of information from our data, DNJ content appears not to be influenced solely by annual temperature of the planted areas.

  • The most important factors in environment that influence the physiology of insects are temperature and humidity. Insects display a remarkable range of adaptations to changing environments and maintain their internal temperature (thermoregulation) and water content within tolerable limits, despite wide fluctuations in their surroundings. Adaptation is a complex and dynamic state that widely differs in species. Surviving under changing environment in insects depends on dispersal, habitat selection, habitat modification, relationship with ice and water, resistance to cold, diapause and developmental rate, sensitivity to environmental signals and syntheses of variety of cryoprotectant molecules. The mulberry silkworm (Bombyx mori) is very delicate and sensitive to environmental fluctuations and unable to survive naturally because of their domestication since ancient times. Thus, the adaptability to environmental conditions in the silkworm is quite different from those of wild insects. Temperature, humidity, air circulation, gases and photoperiod etc. shows a significant interaction in their effect on the physiology of silkworm depending upon the combination of factors and developmental stage affecting growth, development, productivity and quality of silk. An attempt has been made in this article to briefly discuss adaptation in insects with special emphasis on the role of environmental factors and their fluctuations and its significance in the physiology of mulberry silkworm, B. mori.

상엽(桑葉) 함유 한약복합제 추출물의 항비만(抗肥滿)효과 연구 (The Study on Anti-obesity Effects of Mulberry Leaves Contained Herbal Mixture)

  • Objectives This experimental study was designed to investigate the effects of Mulberry leaves contained herbal mixture (MLHM) on body weight, serum lipid level and adipocyte differentiation in high fat diet-fed obese mice. Methods Four-week old mice (wild-type C57/BL6) were used for all experiments. Cells were incubated with MLHM at the indicated concentration (0.04-4mg/ml) for 24h, and growth rate was assessed by MTT ((3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. 3T3-L1 preadipocytes were incubated in DMEM for 2 days with the indicated concentrations of MLHM, and on Day 6, the cells were fixed and the cellular lipid contents were assessed by Oil-Red-O staining. The expression of peroxisome proliferator-activated receptor ${\gamma}$ (PPAR ${\gamma}$) and cytidine-cytidine-adenosine-adenosine-thymine (CCAAT)/enhancer-binding proteins ${\alpha}$ (C/EBP ${\alpha}$) as adipocyte-specific proteins were determined by real time RT-PCR and western blotting. In addition, body weight gain and serum lipid levels were measured in the mice with obesity induced by the high fat-diet for four weeks. Results Though MLHM did not show toxicity even at the concentration of 4mg/ml, MLHM significantly inhibited the differentiation of 3T3-L1 preadipocites in a dose-dependent manner. Also, MLHM significantly reduced the expressions of PPAR ${\gamma}$ and C/EBP ${\alpha}$ in a dose-dependent manner. Furthermore, MLHM significantly reduced body weight gain and LDL-cholesterol contents in high fat diet-fed obese mice. Conclusions These results demonstrate that MLHM exerts anti-obesity effect in 3T3-L1 cells and mice with obesity by high-fat diet.
