Positivity and Intensity of Gnathostoma spinigerum Infective Larvae in Farmed and Wild-Caught Swamp Eels in Thailand

  • From July 2008 to June 2009, livers of the swamp eels (Monopterus alba) were investigated for advanced third-stage larvae (AL3) of Gnathostoma spinigerum. Results revealed that 10.2% (106/1,037) and 20.4% (78/383) of farmed eels from Aranyaprathet District, Sa Kaeo Province and those of wild-caught eels obtained from a market in Min Buri District of Bangkok, Thailand were infected, respectively. The prevalence was high during the rainy and winter seasons. The infection rate abruptly decreased in the beginning of summer. The highest infection rate (13.7%) was observed in September and absence of infection (0%) in March-April in the farmed eels. Whereas, in the wild-caught eels, the highest rate (30.7%) was observed in November, and the rate decreased to the lowest at 6.3% in March. The average no. (mean${\pm}$SE) of AL3 per investigated liver in farmed eels ($1.1{\pm}0.2$) was significantly lower (P=0.040) than those in the caught eels ($0.2{\pm}0.03$). In addition, the intensity of AL3 recovered from each infected liver varied from 1 to 18 ($2.3{\pm}0.3$) in the farmed eels and from 1 to 47 ($6.3{\pm}1.2$) in the caught eels, respectively. The AL3 intensity showed significant difference (P=0.011) between these 2 different sources of eels. This is the first observation that farmed eels showed positive findings of G. spinigerum infective larvae. This may affect the standard farming of the culture farm and also present a risk of consuming undercooked eels from the wild-caught and farmed eels.

Morphological Observation of Pre- and Postovulated Eggs from Artificially Maturated Wild and Feminized Eels (성성숙 유도된 자연산과 자성화 뱀장어의 채란 전·후 난의 형태학적 관찰)

  • This study about morphological observation of pre- or post ovulated eggs obtained from artificially maturated female eels. Female eels were divided with two groups as wild eels from nature and feminized eels from farm. Artificial maturation had been conducted with the established methods in this laboratory, and then maturate eggs sampled at fixed 3 times and these were observed with stereomicroscope. Rate of increased body weight (RIW) were measured with 2 times. Egg diameters and development of oil droplets are determined for standardization of egg maturation degree, and the transparency of egg cytoplasm and the homogeneous degrees of egg size were referred to determine of egg quality. Rate of increased body weight (RIW) were good in range about 10 % at final salmon pituitary extracts (SPE) injection time and in range about 20 % at $17{\alpha}$, $20{\beta}$-dihydroxy-4-pregnen-3-one (DHP) injection time. Good matured egg for fertilization had $900-1000{\mu}m$ diameter, and they had about 50 oil droplets in size about $100{\mu}m$ diameter. There were not predominant differences at egg size and oil droplet development between wild female and feminized eels.

Nutritional Properties of Body Composition Based on Captured Location and Size in Wild Eels, Anguilla japonica (자연산 뱀장어의 채집지역 및 크기에 따른 체성분의 영양학적 특성)

  • The present study provides nutritional characteristics of body composition based on the different collected locations and body sizes in wild eels, Anguilla japonica. Eels were collected in Kyungho river, near the Sachun bay (Gyeongnam) and Inchun river, near Gochang, Jeonnam. Eels were divided by size into the following groups: less than 100 g (Gochang: 75.8$\pm$17.6 g, Sachun: 89.3$\pm$6.42 g) and more than 200 g group (Gochang: 215$\pm$8.95 g, Sachun: 236$\pm$11.7 g). The moisture content of individuals in Gochang area (Gw) was higher than that in Sacheon area (Sw). In the results of proximate analysis, the ranges of moisture, crude protein, crude fat, and crude ash were 63∼72, 16∼20, 6.1∼15.4 and 2.2∼2.8%, respectively. Moisture and crude fat were significantly different (P<0.05) based on the captured locations. In fatty acid composition, the ranges of saturates, monoenes, polyenes, and n-3 PUFA content were 38.0∼48.0, 51.5∼58.5, 1.95∼4.12 and 0.68∼1.53%, respectively. It was significantly different (P<0.05) based on the captured locations. In the amino acid composition, the ranges of essential and non-essential amino acids were 7.11∼12.5 and 8.34∼ 12.9%, respectively. These values were not significantly different (P>0.05) based on the captured locations, however these were significant different (P<0.05) based on their body weight. Therefore, these results indicated that the different body composition depends on the different captured locations and body sizes. Furthermore, it is necessary to examine more varied locations and body sizes for their body composition in eels.

Plasma Sex Steroid Hormone Profiles in Artificially Maturing Wild Eel, Anguilla japonica (자연산 뱀장어의 인위적인 성숙 유도에 따른 혈중 성호르몬 변동)

  • To understand the changes in plasma levels of sex steroids in the wild Japanese eel Anguilla japonica during artificially maturing process, eels received weekly intraperitoneal injections of a water-in-oil (W/O) type emulsion with Freund`s incomplete adjuvant containing salmon pituitary extract (SPE; 20 mg pituitary powder/fish) were examined. In the weekly Eel's Ringer-treated control wild eels, the body weight (BW) changes of fish decreased slowly during the experiment period. Plasma testosterone (T), $estradiol-17{\beta}\;(E_2)$ and $17a,20{\beta}-dihydroxyprogesterone$ (DHP) levels did not change significantly at the end of the experiment. In the weekly SPE-treated silver eels, however, rapid increase in BW changes occurred after 6 to 10 weeks, and the oocytes of all fish were observed to be in the migratory nucleus stage. Furthermore, significant increase in sex steroid hormones (T and $E_2$) levels occurred from 6 weeks. In the weekly SPE-treated yellow eels, the BW changes of fish increased slowly at 6 weeks and then increased. In these fish, the oocytes were at the tertiary yolk globule stage even at the end of the experiment. Plasma sex steroid hormones profiles revealed individual variability in SPE-treated yellow eels. Plasma T and $E_2$ levels significantly increased at 8 weeks and after 6 weeks, respectively, in SPE-treated yellow eels. In the weekly SPE-treated wild eels (silver and yellow eels), however, plasma DHP levels did not change significantly during the experiment period. In silver eel, final maturation could be induced by weekly administration of SPE using W/O type emulsion.

Feeding Habits of the Glass Eel Anguilla japonica Determined by C and N Stable Isotopes in the Nakdong River Estuary of the Korean Peninsula (안정동위원소를 이용한 낙동강 하구 실뱀장어의 먹이 습성 분석)

  • Wild glass eels found in the Korean peninsula are thought to migrate along the North Equatorial Current and undergo the processes of spawning and incubation in the Mariana Trench. Juveniles of the wild glass eels are collected from the southern and western coasts of the Korean peninsula and used as seeds for aquaculture. To investigate the feeding behavior of wild glass eels, we collected glass eels from the Nakdong River estuary during March and April 2014 and analyzed the total length, wet weight, dry weight, carbon and nitrogen contents, and stable isotope ratio of eels as well as water temperature and salinity. Water temperature in the Nakdong River estuary was $13.2{\pm}1.0$ (mean ${\pm}$ 1SE; range, $10.1{\sim}15.7)^{\circ}C$ and salinity was $24.8{\pm}2.4$ (13.2~34.0), showed a lower range from 13.2 to 30.0 (a mean of 21.2) when the floodgates were opened. The total length of glass eels was $56.5{\pm}0.2$ (51.0~63.6) mm, wet weight was $70.9{\pm}1.4$ (33.6~133.2) mg, and dry weight was $16.5{\pm}0.3$ (10.1~29.1) mg. Carbon and nitrogen contents of glass eels were $51.0{\pm}0.8%$ and $13.9{\pm}0.1%$, respectively. Mean ${\delta}^{13}C$ and ${\delta}^{15}N$ values of glass eels collected from the Nakdong River estuary were -20.9±0.2‰ and 6.1±0.1‰, displaying similar values to those of leptocephalus, glass eel larvae collected from the North Equatorial Current. Therefore, this result suggest that the glass eels collected from the Nakdong River estuary do not feed on prey after metamorphosis from eel larvae to glass eels their migration.

Studies on Lipids in Fresh-Water Fishes 7. Comparison of Lipid Components among Wild and Cultured Eel (Anguilla japonica), and Conger Eel (Astroconger myriaster) (담수어의 지질에 관한 연구 7. 천연 및 양식 뱀장어와 붕장어의 지질성분 비교)

  • This study was designed to compare the lipid components among wild and cultured eel, Anguilla japonica, and conger eel, Astroconger myriaster. The lipid components of cultured eel were analyzed and compared with those of wild and conger eel. In the content of total lipid, the lipid content in cultured eel was slightly higher than that in wild one, but 2 times higher than that in conger eel. The lipid contents in edible portion of wild and cultured eel were 5 times higher than those in viscera, but the lipid content in edible portion of conger eel showed a similar trend to that in viscera. In the fatty acid composition of neutral lipid in edible portion, percentages of $C_{14:0},\;C_{16:0}\;and\;C_{18:1}$ in cultured eel were higher than those in wild one, while percentages of $C_{16:1},\;C_{18:2},\;C_{18:3},\;C_{20:4},\;C_{20:5},\;C_{22:5}\;and\;C_{22:6}$ lower, and percentages of $C_{18:0},\;C_{20:4}\;and\;C_{22:6}$ in conger eel were noticeably higher than those in wild and cultured eels. In the case of phospholipid in edible portion, percentages of $C_{18:0}\;and\;C_{18:2}$ in cultured eel were higher than those in wild one, while percentages of $C_{16:0},\;C_{16:1},\;C_{18:1},\;C_{18:3},\;C_{20:4},\;C_{20:5},\;C_{22:5}\;and\;C_{22:6}$ lower. The unsaturation (TUFA/TSFA) of neutral lipid was no significant difference among wild and cultured eel, and conger eel, but that of phospholipid in wild eel was higher than that in cultured eel and conger eel. The essential fatty acid content(TEFA) of neutral lipid in edible portion of wild eel was 3 times higher than that of cultured one. but the TEFA of phospholipid in edible portion was no significant difference among wild and cultured eels, and conger eel. The w3 highly unsaturated fatty acid content (w3 HUFA) of neutral lipid in edible portion of wild eel was 2.0 to 2.5 times higher than that of cultured eel and conger eel, but the w3 HUFA of phospholipid in edible portion of wild eel was noticeably higher than that of cultured eel and conger eel. In the ratio (A/B) of fatty acid content (A) in cultured eel to that (B) in diet, the A/B ratios of $C_{18:2}\;w6,\;C_{18:3}\;w3,\;C_{20:5}\;w3\;and\;C_{22:6}\;w3$ were 0.23 to 0.48 much lower than the other fatty acid. Consequently, it is considered that the ratios of w3 HUFA is related to the biosynthesis of polyenoic acid and growth rate of cultured eel.

Correlationship between Artificial Maturation Season and Reproduction Coefficient in the Cultured Eel Anguilla japonica (양식산 뱀장어 Anguilla japonica에 있어서 인공성성숙시기와 번식률과의 상관관계)

  • This study investigated the correlationship between artificial maturation season and reproduction coefficient of cultured eel Anguilla japonica from May (spring) to next January (winter). The brood stock, female eels ($400{\sim}600\;g$) were artificially matured by weekly intramuscular injections of salmon pituitary extracts (SPE, 20 mg/fish) to induce a completion of vitellogenesis. After completion of vitellogenesis, final oocyte maturation and ovulation was induced by injection of $17{\alpha}$, $20{\beta}-dihydroxyprogesterone$ (DHP) at about $2\;{\mu}g/g$ body weight. Most fish ovulated $15{\sim}18\;h$ following the DHP injection. The ovulated fish were induced to natural spawning or artificial fertilization by the dry method. Males ($200{\sim}350\;g$) were received weekly intramuscular injections of human chorionic gonadotropin (HCG) at a dosage of 1 IU/g body weight to induce testicular maturation and spermiation. Seasonal reproduction coefficient which includes the rate of ovulation, buoyancy, fertilization and hatching of eggs in the artificially matured eel during spring to summer ($May{\sim}July$) were significantly higher than the other season, while there were no significant difference among spring and summer (P<0.05). Furthermore, the number of eggs spawned and larvae hatched in the artificially maturated eel during spring to summer ($May{\sim}July$) were significantly higher than the other season, while there were no significant difference in spring and summer (P<0.05). These results indicate that artificial maturation by hormone treatment of A. japonica was successful only during spring to summer, which is the maturation period in the wild stock in nature. Consequently, it is possible to determine the period of artificially induced sexual maturity by the reproduction coefficient which includes the rate of ovulation, buoyancy, fertilization and hatching of eggs in the cultured eel A. japonica.
