Effect of Lactic Acid Bacteria Inoculation on Fermentation Characteristics of Whole Crop Barley Silage

  • An experiment was carried out to determine the homofermentative activity of Lactobacillus plantarum KCC-10 and KCC-19 on the ensiling of whole crop barley (WCB). The crude protein in the silages was slightly higher in the KCC-10 and KCC-19 treatments compared to the control, but there was no significant difference between the two inoculant-treated silages. Nutrient parameters such as acid detergent fiber, neutral detergent fiber and in vitro dry matter digestibility in L. plantarum KCC-10 and KCC-19 treated silages did not differ from those in the control silage. The lactic acid content increased in KCC-10 and KCC-19 treated silage when compared with the control silage but the contents of acetic acid and butyric acid produced in KCC-10 and KCC-19 treated silages were similar with the control silage. Further, the number of lactic acid bacteria (LAB) in KCC-10 treated silage demonstrated a significant increase when compared to the control. Especially, KCC-19 treated silage showed greater lactic acid bacterial growth potential. Other microbes such as yeast and fungi were not detected in KCC-10 and KCC-19 treated WCB silages. Hence, this study suggests that the addition of L. Plantarum KCC-10 and KCC-19 to the WCB silage can improve fermentation quality for the production of high-quality silage.

Whole Genome Resequencing of Heugu (Korean Black Cattle) for the Genome-Wide SNP Discovery

  • Heugu (Korea Black Cattle) is one of the indigenous cattle breeds in Korea; however there has been severe lack of genomic studies on the breed. In this study, we report the first whole genome resequencing of Heugu at higher sequence coverage using Illumina HiSeq 2000 platform. More than 153.6 Giga base pairs sequence was obtained, of which 97% of the reads were mapped to the bovine reference sequence assembly (UMD 3.1). The number of non-redundantly mapped sequence reads corresponds to approximately 28.9-fold coverage across the genome. From these data, we identified a total of over six million single nucleotide polymorphisms (SNPs), of which 29.4% were found to be novel using the single nucleotide polymorphism database build 137. Extensive annotation was performed on all the detected SNPs, showing that most of SNPs were located in intergenic regions (70.7%), which is well corresponded with previous studies. Of the total SNPs, we identified substantial numbers of non-synonymous SNPs (13,979) in 5,999 genes, which could potentially affect meat quality traits in cattle. These results provide genome-wide SNPs that can serve as useful genetic tools and as candidates in searches for phenotype-altering DNA difference implicated with meat quality traits in cattle. The importance of this study can be further pronounced with the first whole genome sequencing of the valuable local genetic resource to be used in further genomic comparison studies with diverse cattle breeds.

Division Algorithm in SuanXue QiMeng

  • The Division Algorithm is known to be the fundamental foundation for Number Theory and it leads to the Euclidean Algorithm and hence the whole theory of divisibility properties. In JiuZhang SuanShu(九章算術), greatest common divisiors are obtained by the exactly same method as the Euclidean Algorithm in Elements but the other theory on divisibility was not pursued any more in Chinese mathematics. Unlike the other authors of the traditional Chinese mathematics, Zhu ShiJie(朱世傑) noticed in his SuanXue QiMeng(算學啓蒙, 1299) that the Division Algorithm is a really important concept. In [4], we claimed that Zhu wrote the book with a far more deeper insight on mathematical structures. Investigating the Division Algorithm in SuanXue QiMeng in more detail, we show that his theory of Division Algorithm substantiates his structural apporaches to mathematics.

Photoreversibility of Fruiting and Growth in Oriental Melon (Cucumis melo L.)

  • BACKGROUND: Photoreversibility, a reversion of the inductive effect of a brief red light pulse by a subsequent far red light pulse, is a property of photo responses regulated by the plant photoreceptor phytochrome B. Plants use photoreceptors to sense photo signal and to adapt and modify their morphological and physiological properties. Phytochrome recognizes red light and far red light and plays an important role in regulating plant growth and development. METHODS AND RESULTS: The reversal responses of growth and fruiting characteristics were investigated to increase the yield of oriental melon (Cucumis Melo L. var. Kumsargakieuncheon) by means of controlling light quality in a plastic house. Red (R:660nm) and far red (FR:730nm) lights were subsequently irradiated on the whole stems and leaves of the oriental melon plant during growing periods, using red and far red LEDs as light sources, from 9:00 PM daily for 15 minutes. The intensities of R and FR light were 0.322-0.430 μmol m-2s-1 and 0.250-0.366 μmol m-2s-1, respectively. Compared to R light irradiation, combination of R and FR light irradiation increased the length of internode, number of axillary stems, number of female flowers, and fruit number of oriental melons. The results of treatment with R were similar to R-FR-R light irradiation in terms of length of internode, number of axillary stems, number of female flowers, and number of fruits. When FR treatment was considered, R-FR and R-FR-R-FR light irradiation had similarities in responses. These reversal responses revealed that oriental melon showed a photoreversibility of growth characteristics, flowering, and fruiting. CONCLUSION: These results suggested the possibility of phytochrome regulation of female flower formation and fruiting in oriental melon. The fruit weight of the oriental melon was the heaviest with the R light irradiation, while the number of fruits was the highest with the FR light. With the FR light irradiation, the fruit weight was not significantly higher compared to that of the control. Meanwhile, the yield of oriental melon fruits increased by 28-36% according to the intensities of the FR light due to the increases of the number of fruits.

Visibility Evaluation of Various Retroreflective Fabric Types and LED Position on Safety Life Jacket (안전기능성 구명복에 적용된 재귀반사 종류별 및 LED 위치별 가시성능 평가)

  • This study prepared 5 retroreflective fabrics of glass beads and colors of silver, gold and purple in order to characterize morphologies, luminance and illuminance properties. Retroreflective fabrics were prepared according to division and area by Samp.1 (Division 2, area $80cm^2$), Samp.2 (Division 2, area $60cm^2$), Samp.3(Division 3, area $60cm^2$), Samp.4 (Division 5, area $50cm^2$), and Samp.5 (Division 1, area $100cm^2$). The results of the morphologies indicate that the back side of retroreflective fabrics was the 3M-Sv, RF-Sv. The RF-Gd of the fabrics were woven fabrics and the SRF-Sv, SRF-Pp was knitting. The largest glass bead size was 3M-Sv and the smallest was SRF-Pp. RF-Sv was the thinnest and SRF-Sv, SRF-Pp were the thickest. The results of luminance properties indicated a tendency to decrease slightly as the number of divisions and area increased, except for Samp.2 to Samp.3. The result of illuminance properties tend to increase as the number of LED becomes greater than the number of divisions in the same area; in addition, the higher the luminance as a whole increases. The results show that it is preferable to use a silver-colored RF-Sv which has the highest luminance and physical properties when a retroreflective fabric is applied to clothes.

  • Antibiotics have been used in livestock production for not only treatment but also for increasing the effectiveness of animal feed, aiding animal growth, and preventing infectious diseases at the time when immunity is lowered due to stress. South Korea and the EU are among the countries that have prohibited the use of antibiotics for growth promotion in order to prevent indiscriminate use of antibiotics, as previous studies have shown that it may lead to increase in cases of antibiotic-resistant bacteria. Therefore, this study evaluated the number of antibiotic resistance genes in piglets staging from pre-weaning to weaning. Fecal samples were collected from 8 piglets just prior to weaning (21 d of age) and again one week after weaning (28 d of age). Total DNA was extracted from the 200 mg of feces collected from the 8 piglets. Whole metagenome shotgun sequencing was carried out using the Illumina Hi-Seq 2000 platform and raw sequence data were imported to Metagenomics Rapid Annotation using Subsystem Technology (MG-RAST) pipeline for microbial functional analysis. The results of this study did not show an increase in antibiotic-resistant bacteria although confirmed an increase in antibiotic-resistant genes as the consequence of changes in diet and environment during the experiment.

Whole Exome Sequencing in Patients with Phenotypically Associated Familial Intracranial Aneurysm

  • Objective: Familial intracranial aneurysms (FIAs) are found in approximately 6%-20% of patients with intracranial aneurysms (IAs), suggesting that genetic predisposition likely plays a role in its pathogenesis. The aim of this study was to identify possible IA-associated variants using whole exome sequencing (WES) in selected Korean families with FIA. Materials and Methods: Among the 26 families in our institutional database with two or more IA-affected first-degree relatives, three families that were genetically enriched (multiple, early onset, or common site involvement within the families) for IA were selected for WES. Filtering strategies, including a family-based approach and knowledge-based prioritization, were applied to derive possible IA-associated variants from the families. A chromosomal microarray was performed to detect relatively large chromosomal abnormalities. Results: Thirteen individuals from the three families were sequenced, of whom seven had IAs. We noted three rare, potentially deleterious variants (PLOD3 c.1315G>A, NTM c.968C>T, and CHST14 c.58C>T), which are the most promising candidates among the 11 potential IA-associated variants considering gene-phenotype relationships, gene function, co-segregation, and variant pathogenicity. Microarray analysis did not reveal any significant copy number variants in the families. Conclusion: Using WES, we found that rare, potentially deleterious variants in PLOD3, NTM, and CHST14 genes are likely responsible for the subsets of FIAs in a cohort of Korean families.

First Annulus Formation and Age Determination for Otoliths of Chub Mackerel Scomber japonicus (고등어(Scomber japonicus) 이석의 초륜 형성 및 연령 사정)

  • First annulus formation and age determination of otoliths were examined for chub mackerel Scomber japonicus collected in Korean waters over the one year from January to December in 2009. Translucent zone was regarded as an annual mark. Age interpretation criteria was based on the data of the number of translucent zone, capture date, and edge type of the otolith, assuming the nominal birthday to be 1 January. Monthly changes in mean marginal index indicated that translucent zone was formed once a year, mainly in June. The otolith of 0-ring group was detected comparing the progression by month of the smaller fish length, appearing to be a single first opaque zone. The average distance from the core to the first translucent zone was ~1.77 mm, provided as supplementary information to increase ageing accuracy. The ageing criteria for chub mackerel was made to determine correct year-class with the purpose of effective stock assessment. This method using nominal birthdate and edge type analysis could estimate age of fish closer to the true age than purely counting the number of translucent zone on a whole otolith.

Friction and Wear Characteristics of Friction Material with the Content of Hexamethylenetetramine (Hexamethylenetetramine의 함량에 따른 마찰재의 마찰.마모 특성)

  • The friction characteristics of phenolic resin and model friction materials were investigated with the content of hexamethylenetetamine (HEXA). At 10 wt.% of HEXA, the phenolic resin and model friction materials showed the most stable friction coeffcient in constant temperature test at various test conditions because of its good thermal stability and proper curing reaction. It was found from constant interval test in mild condition that the friction coeffcients of friction materials cured with 10 wt.% of HEXA was the highest and stable values in the whole range of braking operations. However, at the severe condition in constant interval test, the friction coefficient of friction materials cured with 10 wt.% to of HEXA was lowered and as the number of braking operation increased, the values became stable. In order to obtain the thormal stable friction materials, the content of HEXA from 5 to 10 wt.% could be recommended.

Symmetry Exploitation of Diffraction Gratings to Enhance the Spectral Resolution

  • A diffraction grating is a highly symmetric optical element with a physical structure that is invariant under translational spatial movements. The translational symmetry is reflected in the fields that are diffracted from the grating. Here, we introduce a plane-parallel mirror pair onto the grating, which translates the fields through double reflections, and we describe a method of exploiting the symmetry to enhance the spectral resolution of a diffraction grating beyond the limit that is set by the number of grooves. The mirror pair creates another virtual grating beside the original one, effectively doubling the number of grooves. Addition of more mirror pairs can further increase the effective number of grooves despite the increased complexity and difficulty of experimental implementation. We experimentally demonstrate the spectral linewidth reduction by a factor of four in a neon fluorescence spectrum. Even though the geometrical restriction on the mirror deployment limits our method to a certain range of the whole spectrum, as a practical application example, a bulky spectrometer that is nearly empty inside can be made compact without sacrificing the resolution.
