Screening of Wood-Rot Fungi Based on RBBR Decolorization and Its Laccase Activity (RBBR 탈색능을 이용한 목재부후균의 선발 및 이들 균의 Laccase 효소활성)

  • This study was to screen white-rot fungi possesing strong lignin degrading enzymes, glucose-1 oxidase (GOD), laccase (LAC) and Mn-peroxidase (MnP), based on their decolorization activity of Remazol Brilliant Blue R (RBBR). In the midst of 20 tested fungi, 9 isolates were shown 4 kinds of activities such as RBBR decolorization, GOD, LAC and MnP. Relatively high active strains were identified as Phlebia radiata, Trametes versicolor, Abortiporus biennis, Gleophyllum odoratum and Cerrena unicolor. In particular, T. versicolor, G. odoratum, and C. unicolor, which have high activities of LAC, were used to confirm the optimal temperature and pH and to evaluate the effect of inducer, 2,5-xylidine on their LAC activity. The optimum temperatures for mycelial growth were $28^{\circ}C$ for T. versicolor and G. odoratum, and $25^{\circ}C$ for C. unicolor. The optimum pH for mycelial growth was 5.5. Three strains showed the increase of LAC enzyme activity by the addition of 2,5-xylidine. T. versicolor had the highest LAC activity of $22,700nkat/{\ell}$, corresponding to 11.3 times, G. odoratum $15,400nkat/{\ell}$, 9 times and C. unicolor $17,330nkat/{\ell}$, 5.5 times higher than those of the control.

Expression of a Manganese Peroxidase Gene (mnp5) from White rot fungus Phanerochaete chrysosporium in the Pichia pastoris (백색부후균 Phanerochaete chrysosporium에서 유래한 Manganese Peroxidase Gene(mnp5)의 Pichia pastoris에서의 이종발현)

  • The manganese peroxidase (mnp5) from white-rot fungus Phanerochaete chrysosporium has been heterologously expressed in the methylotrophic yeast Pichia pastoris. The majority of the rMnP5 (recombinant MnP5) produced by P. pastoris exhibited an approximate molecular mass 45 kDa considerably larger than that of the predicting mnp5 due to two glycosylation sites of mnp5. After site direct mutation treatment, the effect of N-linked hyperglycosylation was examined by enzyme activity. Analysis by sodium dodesyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Coomassie Brilliant Blue (CBB) staining revealed a major protein band with a molecular mass of 37 kDa. Enzyme activity of M-rMnP5 (mutant recombinant MnP5) was similar to that of rMnP5, indicating that hyperglycosylation did not affect the active site. In this work, active mnp5 was successfully expressed in P. pastoris, suggesting that P. pastoris has potential capability of producing active heme-containing proteins.

Changes in Activities of Lignin Degrading Enzymes and Lignin Content During Degradation of Wood Chips by Polyporus brumalis (겨울우산버섯에 의한 목재칩의 리그닌 분해 효소 활성 및 리그닌 함량 변화)

  • In this study, laccase activity, rate of weight loss and degree of lignin degradation of pine wood chips were determined during the liquid and solid state incubation with Polyporus brumalis. The results showed that laccase enzyme activity at untreated wood chip was gradually decreased after 20 days, but enzyme activity with wood chip treatment showed 10 times higher than untreated ones at 60 incubation days. Rate of weight losses of pine chip and rate of lignin loss were 23.4% and 6.3% by P. brumalis during 80 incubation days. Gene expression of pblac1 from P. brumalis was 3 times increased under pine chip treatment at 40 incubation days. Consequently, laccase activity of white rot fungi, P. brumalis, was increased at incubation with wood chip and pblac1 gene was important factor of lignin degradation. Therefore, to regulate lignin degrading enzyme gene expression by using the tools of biotechnology will be able to develop superior strains and it will be useful for pretreatment of lignocellulosic biomass at bioethanol production.

Biodegradation of aromatic dyes and bisphenol A by Trametes hirsuta (Wulfen) Pilat (흰구름버섯에 의한 방향족 염료와 비스페놀 A의 분해)

  • Trametes hirsuta, a white rot fungus, exhibits the ability to degrade synthetic aromatic dyes such as congo red (CR), methylene blue (MB), crystal violet (CV), and remazol brilliant blue R (RBBR). The mycelia of T. hirsuta degraded RBBR and CR more efficiently than CV and MB in the PDB liquid medium (supplemented with 0.01% 4 aromatic dyes). In these mycelia the activities of three ligninolytic enzymes-laccase, manganese peroxidase (MnP), and lignin peroxidase (LiP)-were observed. Among these, laccase was identified to be the major enzyme responsible for the degradation of the four aromatic dyes. The degradation of bisphenol A was also investigated by culturing the mycelia of T. hirsuta in YMG medium supplemented with 100 ppm bisphenol A. The mycelia of T. hirsuta were found to degrade bisphenol A by 71.3, 95.3, and 100 % within incubation periods of 12, 24, and 36 hr, respectively. These mycelia also showed ligninolytic enzyme-like activities including those similar to laccase, MnP, and LiP. Therefore, these results indicate that T. hirsuta could emerge as a potential tool for the remediation of environmental contamination by aromatic dyes and bisphenol A.

Diseases of the cultivated Porphyra at seocheon area (서천 해역 김 양식장의 갯병에 관한 연구)

  • Disease occurrence and dead cell ratio of Porphyra cultivated at Seocheon area were studied. Seaweed samples were collected monthly at 12 cultivating sites of Porphyra from Nov. 2008 to Mar. 2009. Although disease occurrence was low at the beginning of cultivation at the most of collecting sites, the occurrence increased with time. The most common symptom of Porphyra disease in this area was diatom attachment, followed by white rot disease. Licmophora sp. was the most common species of diatom and its attachment was the highest on November, the beginning of cultivation. Although the occurrence ratios of green spot disease and white rot disease were high on November, chytrid disease was increased at the end of cultivation period. Dead cell ratio of Porphyra thalli was different at each study site and cultivating time, but normally the ratio was high at the end of cultivating season. The changes of dead cell ratio could be related to the nutrition supplement and currents, and the more precise reason of dead cell ratio would be revealed by further research related to cultivating environment conditions.

Synthetic aromatic dye degradation by white rot fungus, Pleurotus eryngii (큰느타리(Pleurotus eryngii)의 방향족 합성염료 분해 특성)

  • Pleurotus eryngii, a white rot fungus, produces two extracellular lignin-degrading enzymes, laccase and manganese peroxidase (MnP). Owing to these enzymes, P. eryngii efficiently degrades synthetic chemicals such as azo, phthalocyanine, and triphenyl methane dyes. In this study, we investigated the degradation processes of four aromatic dyes, congo red (CR), methylene blue (MB), crystal violet (CV), and malachite green (MG), by P. eryngii under solid and liquid culture conditions. CR and MG were the most quickly degraded under solid and liquid culture conditions, respectively. However, compared to CR, CV, and MG, MB was not degraded well under both culture conditions. The activities of ligninolytic enzymes (laccase and MnP) were also investigated. Laccase was identified to be the major enzyme for dye degradation. A positive relationship between decolorization and enzyme activity was observed for CR, MB, and CV degradation. In contrast, decolorization of MG ensued after high enzyme activity. These results indicate that the degradation process differs between MG and the other aromatic dyes. Therefore, P. eryngii could be a potential tool for the bioremediation of synthetic aromatic dye effluent.

Effect of Incubation Temperature and pH on Chlamydospores Germination of Cylindrocarpon destructans Causing Root Rot of Panax ginseng (인삼 뿌리썩음병균 Cylindrocarpon destructans의 후막포자 발아에 미치는 배양온도 및 pH의 효과)

  • Effects of incubation temperature and pH on chlamydospore germination of Cylindrocarpon destrcutans (isolate CY-9802) causing root rot of Panax ginseng were studied. Germination rate of the chlamydospores on Czapek solution agar(CSA) was higher than on potato dextrose agar(PDA) at the incubation temperatures tested. The chlamydospores were able to be germinated at range of 5$\^{C}$ to 30$\^{C}$ after 48 hours incubation on CSA. Germination rate was 53.2∼6.27% at range of 15$\^{C}$ to 25$\^{C}$, and the optimum temperature was 20$\^{C}$, whereas they were very low at 30$\^{C}$ on PDA. Germination rate was 43.6% to 47.9% at range of 10$\^{C}$ to 20$\^{C}$, and the optimum temperature was 20$\^{C}$ as well. They were able to be germinated at pH of 5.2 to 8.1 on CSA and 5.2 to 7.2 on PDA. Optimum pHs for the germination on CSA and PDA were from 6.4 to 8.2 and from 5.2 to 6.0, respectively. Mycelial color of the fungus on CSA was pale brown at pH from 5.2 to 6.0 and white from pH 6.4 to 8.1, while it was typical dark brown ar range of pH 5.2 to 7.1 and brown at pH 7.2 on PDA after 21 days incubation.

Density, Bonding Strength, Bending strength and Decay Resistance of Radiata Pine Laminated Veneer Lumber (라디에타소나무 단판적층재의 밀도·접착·강도성능 및 내부후성)

  • In this study, LVLs of radiata pine were fabricated with non-preservative treated veneers, CuAz treated veneers, and ACQ treated veneers, using aqueous vinyl urethane adhesive and phenol modified resorcinol resin adhesive. Then density gradient, bonding strength, bending properties and decay resistance of LVLs were evaluated. As results, the cone-shaped and higher density gradient pattern was found in layer close to glueline. After cyclic water boiled test, the LVL bonded with aqueous vinyl urethane resin adhesive was delaminated in all layers or partly delaminated including check, chasm in glueline layer. In the case of LVL bonded with phenol modified resorcinol resin adhesive, despite slight cupping due to great glueline stress and vertical check between glueline layers, it was observed that the bonding strength to delamination was higher, owing to most absence of delamination through overall glueline. On the other hand, in the decay test, mass loss by brown rot fungi was greater than white rot fungi in LVL bonded with aqueous vinyl urethane resin adhesive. However, in LVL bonded with phenol modified resorcinol resin adhesive, the mass loss by brown rot fungi was slight and non-preservative treated LVL was low. The mass loss of preservative-treated LVL was 0 (zero), showing the high decay resistance effect.

Flower Rot of Cotton Rose (Hibiscus mutabilis) Caused by Choanephora cucurbitarum (Choanephora cucurbitarum 에 의한 부용 꽃썩음병)

  • In July 2001, rotting and shivering flowers of cotton rose (Hibiscus mutabitis) were fecund in the flower beds along the roadsides in Jinju area. The disease first started as water-soaking, dark-green lesions on the petals, and then whole flower was rotted rapidly, Whitish mycelia and monosporous sporangiophore with monosporous sporangiola were formed abundantly on the lesions. Colony appeared as white to pale yellowish brown mycelia on potato dextrose agar medium (PDA). Monosporous sporangiophore was long slender and branched at the apex, each branch bearing a head of sporangiospores. Sporangium was subglobose in shape and was 42.6-114.2$\mu$m in size. Monosporous sporangiola were elliptic, fusiform or ovoid, and brown in color and 12.3~21.6 $\times$8.3~11.6$\mu$m Um in size. Sporangiospores were elliptic, fusiform or ovoid in shape, dark brown or brown in color and 16.3~23.8$\times$8.2~13.6$\mu$m in size, and they had three or more appendages at bipolar end. Zygospores were mostly globose, dark black colored and sized was 46.2-78.4$\mu$min diameter, The fungus grew on PDA between at 15 to 4$0^{\circ}C$, and the optimum temperature was 3$0^{\circ}C$. This is the first report on the flower rot of cotton rose caused by C. cucurbitarum in Korea.

Review of Disease Incidence of Major Crops in 2001 (2001년 농작물 병해 발생개황)

  • Climate in the year of 2001 could be summarized as severe drought from March to May, unusually high temperature in whole plant growth periods and clear weather condition especially in harvesting time of September and October without any typoons. In rice, major diseases such as leaf and panicle blast, bacterial blight, sheath blight and bacterial grain rot developed little due to unfavorable weather conditions, however, brown spot occurred severely due partly to the reduced use of N-fertilizer, Rice stripe virus infection was unusually high mainly in the west coast areas with four times more diseaseD area campared to the previous year, In pepper phytophthora blight was extremely severe in Cheonnam and Cheonbuk provinces, where had frequent rainfalls during growing period. Incidence of major diseases of tomato and cucumber in 2001 was relatively mild. In watermelon, penicillium fruit rot, one of the peculiar disease, spread over major production areas, whereas CGMMV, usually severe in every years was much reduced. Watermelon plants growing in open-fields were more severely diseased than those in plastic houses. Major diseases of chinese melon and strawberry occurred slightly and in particular, sudden wilt syndrome of chinese melon which was severe in 2000 showed mild infections in 2001. Incidence of white rot of garlic and onion was much lower unlike other years due mainly to spring drought and high temperature in the growing period. Infected area of potato bacterial wilt tended to increase in 2001 while fusarium wilt of sweetpotato was decreased. Apple diseases were generally mild, but powdery mildew of pear increased sharply. Barley scab was not severe as seen in other years.
