Phenolic acids of white ginseng were extracted and fractionated into free, esterified, and insoluble-bound forms. The contents of individual phenolic acids in different forms were quantified by gas liquid chromatography. Nine different phenolic acids as free, esterified, and insoluble-bound forms were identified in white ginseng. Total phenolic compounds in different forms of extracts was 0.309% (free form), 0.230% (esterified form) and 0.138% (insoluble-bound form), respectively. Total phenolic acid contents in free, esterified and insoluble-bound form were 889.3, 356.8, 1,176.9 mg/100g fraction, respectively. Ferulic acid was the predominant phenolic acid, representing 63.7% and 50.9% of total phenolic acids in esterified fom and insoluble-bound form, respectively. While caffeic acid was only detected in esterified form. At 10 mg/ml insoluble-bound form quenched 95.9% ABTS free radicals generated from 2,2-azobis(2-amidinopropane) dihydrochloride (AAPH). Also, electron donating ability and lipid peroxidation inhibitory activity of insoluble-bound fom were higher than other fraction. All phenolic acid fractions scavenged over 80% of hydroxyl radical at 10 mg/ml.
Succinate dehydrogenase was activated by ethyl acetate extract from the methanol extract of white ginseng previously treated with petroleum ether and ethyl ether to remove all highly nonpolar components, and the residual aqueous solution from the ethyl acetate extraction. Also, all of the extracted fractions of red ginseng except the ether extract gave positive results. On the contrary to some suggestions by other workers that alkaloids of ginseng may enhance the succinate degydrogenase activity, our results show that the alkaloids may have exhibited some inhibitory eject on this enzyme.
The purpose of the present study was to find the preferences of college students and to understand trends in their consumption patterns. This survey was conducted using a questionnaire containing 50 questions answered by 614 sudients of 4 universities in Daejeon and Seoul and the following result were obtained . 1. The majority of students favored coffee, yulmy tea, ginseng tea, and lllack tea in decreasing order. Only 49.0% of the students believe that ginseng was beneficial to the human life as a health, fond . 2. 54% of the students had experience in taking fresh ginseng and 68% had used in white ginseng. 3. Their expectations for the efficacy of ginseng were as remedies for hang-over syndrom, high blood pressure, heart trouble, liver trouble, and gastronic trouble, in decreasing order. 4. Most of the students took soup made by adding ginseng to chicken broth (SamGyetang) when taken ginseng. 5. The percent of students who had taken coffee was 97.7%, whereas only 48.3% had used processed ginseng products. The female students especially disliked ginseng tea. 6. The preferences of the students indicate that the likelihood that they will be major consumers of ginseng in the future is very low.
Red ginseng (RG) and fermented red ginseng (FRG) are produced from ginseng (GS) via certain biological processes. The main difference between three ginsengs is the composition of ginsenosides known as major metabolites having several biological activities. The concentration of the metabolites has been known to be dependent on the methods which make RG and FRG In this study, we investigated the effects of WG, RG and FRG on the productions of inflammatory proteins (NF-${\kappa}B$, iNOS, COX-2) and cytokines (TNF-$\alpha$, INF-$\gamma$) in LPS-stimulated RAW 264.7 cells. The levels of NO production and iNOS expression were suppressed by the treatment of white ginseng (WG), RG and FRG in LPS-stimulated cells. Also, the production of TNF-$\alpha$ and INF-$\gamma$ was decreased in the cells by all of them. It was indicated that the inhibition of NF-${\kappa}B$ activation in LPS-stimulated cells treated with three kinds of ginsengs was resulted from the suppression of the level of COX-2 expression and the phosphorylation of IkB by LPS. The present study indicated that RC showed the best biological activity among them and FRG was better than WG. The better activity of RG on the inhibition of NO production is considered to be caused by the difference of ginsenoside composition produced during their preparations. In order to elucidate the mechanism, animal test should be performed with three ginsengs.
Ginseng plants in the 2nd year were grown under the shade (90-95%) of colored cellophane films at $15^{\circ}C$ in a natural light phytotron. Relative root grown (final/initial) was in decreasing order of blue, green, white, yellow and red. The growth of aerial part, especially stem length was poorest in blue light with relative root growth negatively correlated with stem length. In this relation each color showed specific domain. Stem length showed positive linear correlation with leaf area per plant in each and among light though it was not significant in red light. The content and ratio of chlorophyll a and b in leaf were in the decreasing order of White, Blue, Green, Red and Yellow. There is positive correlation between chlorophyll a and chlorophyll b, and between total chlorophyll and chl:a/b ratio. Blue color shade is expected to be beneficial for ginseng production.
The present study was carried out to investigate the effects of Panax ginseng saponin extracts on the dexamethasone-induced apoptosis of mouse thymus in vivo and mouse thymocytes in vitro. The saponin fractions of red ginseng (R-SAP) and white ginseng (Wl-SAP) were provided by the Korea Ginseng & Tobacco Research Institute, and the other saponin fraction of white ginseng (W2-SAP) was extracted in our laboratory. 1. The male ICR mice (3~4 wk old; weighing 15$\pm$2 g) were given by each saponin fraction of 5 mg/kg/ day for 4 days, and at one hour after the last treatment, they were injected by deuamethasone (5 mg/kg : DX). The mouse thymus was extracted at 6 hours after DX injection, and they were stained with hematoxylin-eosin reagents and an Apop-Tag kit, respectively, and the thymocytes prepared from it were labelled with anti-mouse FITC-anti-CD4 and anti-mouse PE-anti-CD8 and then analyzed by fluorescence activated cell sorter (FACS). DX-induced reduction of thymus weight was significantly attenuated by W2- SAP but was not affected by other saponin fractions. And DX-induced apoptotic death of thymocytes, appeared in the histologic findings of the thymus, was inhibited by the saponin fractions and the order of these inhibitory potencies was R-SAP》W2-SAP>Wl-SAP. However, in respect of T cell receptors, the differentiation of thymocytes seems not to be changed by treatments with DX or/and the saponin fractions. 2. In the primary thymocyte culture, the DX-induced reduction of thymocyte MTT values was rather greater in RPMI 1640 medium of IWc fetal bovine serum (FBS) or horse serum (HS). In addition, the DX-Induced MTT reduction was significantly inhibited by R-SAP or W2-SAP, in the culture using that medium of 5% FBS or HS. But these saponin fraction did not effected the DX-induced reduction of thymocyte MTT value in primary culture of 10% FBS or 10% HS. These results suggest that R-SAP and some W-SAP fractions may protect thymocyte from stress or glucocorticoisteroid-induced death of them.
This study was conducted to investigate the characteristics of aboveground plants and red ginseng of polystem ginseng in 6 years of age having two or more stems in a plant. Total leaf weight and area of polystem ginseng were larger, while its stem diameter, and the leaf weight and area of the biggest stem in each plant were decreased with increase the stem numbers in a plant. The ratio of shoot weight to root weight in the polystem ginseng with three or more stems was higher than that in the monostem ginseng and the polystem ginseng with two stems, In ginseng plants with no more than 2 stems, there were positive correlations between root weight and total leaf weight, and leaf area, but not between leaf weight and area of the biggest stem. Inner cavity and inner white, limiting factors for redginseng quality grade, occurred more in tri-stem ginseng than mono- and di-stem one. Percentages of Heaven (1st grade) and Earth (2nd grade) red ginseng in tri-stem ginseng were decreased compared with mono stem and di-stem ginseng.
Ginseng shows protective and trophic effects in neurodegenerative diseases in experimental models, and showed cognitive improvement in normal population. To investigate the efficacy of ginseng in patients with Alzheimer's disease, patients, who met NINDS-ADRDA criteria for AD were studied Subjects were randomly assigned to ginseng group and control group, and ginseng group was treated with Korean white ginseng powder (4.5 g/day) for 12 weeks. Efficacy variables included changes in mini-mental status exam (MMSE) and cognitive subscales of Alzheimer's disease assessment scale (ADAS-cog) at 4 weeks and 12 weeks. Baseline MMSE and ADAS scores showed no difference between the two groups. Results showed that ginseng improved ADAS-cog compared to the control group at 12 weeks (p<0.05). MMSE was also increased by ginseng treatment compared to the control at 12 weeks (p<0.01). This study suggests the symptomatic efficacy of ginseng in patients with Alzheimer's disease.
Among 131 isolates of Strptomyces obtained from ginseng cultivating soil, the two isolates ST59 and ST129 showing high antagonistic activity to Fusarium solani(Mart.) Appel & We. causing ginseng root rot were identified. The two isolates were identified Streptomyces alboniger Porter, et al. and Strptomyces reseolilacinus Pridham, et al., respectively, based on mrophology, cultural, and physiological characteristics on various culture media. Spore chains of ST59 and ST129 were flexuous(RF) and coiled(S). Spore surfaces of two isolates were all smooth. Aerial mass color of ST59 was white series and ST129 red series.
